Congener Specific Profiles and Exposure Pathways of Polybrominated Diphenyl Ethers (PBDEs) in Breast Milk of Chuncheon Area (춘천지역 모유 중 폴리브롬화디페닐메테르(PBDEs)의 이성체별 분포특성과 노출경로)

  • This study was carried out to investigate the exposure pathway of PBDEs in human breast milk collected from new mothers residing in Chuncheon, a comparatively clean area in Korea. The congeners of PBDE in 22 human breast milk samples were analyzed using a high resolution gas chromatography with a high resolution mass detector. The residue level of ${\sum}PBDEs$ was higher in primipara subjects than in multipara subjects (p<0.05). The levels in this study were similar to those of people in some Asian and European countries, but were lower than those of people in north America. In the congener profiles, BDE-47 (mean contribution=36.1%) was predominant, followed by BDE-153 (27%), BDE-99 (11.7%), BDE-100 (11.1%), BDE-28 (7.9%) and BDE-183 (3.5%). The sum of BDE-47 and BDE-153 accounted for more than 50% of ${\sum}PBDEs$ in most samples. BDE-47 was highly correlated with ${\sum}PBDEs$ (r=0.94, p<0.001). No strong trend was observed between PBDE levels and a number of key biological factors (women's age, weight, height and body mass index) examined in this study, however, weak correlations were observed in PBDE levels measured against dietary habits, particularly in fish consumption frequency. It seems that Korean people might be exposed to multiple sources including products of PBDEs, and particularly food resources.

Dietary Behaviors and Total Sugar Intake from Snacks of Female College Students according to Sweet Taste Perception (여대생의 단맛 인지도에 따른 식행동 및 간식류를 통한 당류 섭취량 평가)

  • Increasing sugar intake of population has become a nutritional issue in Korea. Sweet taste perception may be related to behaviors such as eating sweet food including high sugars and total sugar intake. This study aimed to evaluate objective and subjective sweet taste perception and the association among objective sweet taste perception, dietary behaviors related to eating sweet snack food including high sugar, and total sugar intake from the snacks. Participants were 261 healthy female college students (mean age: $21.0{\pm}1.6years$), who were divided into three subgroups based on oral sweet taste evaluation using a sweet taste assessment tool provided by Ministry of Food and Drug Safety: sweet-seeker group (n=139), medium sweet-seeker group (n=54), and unsweet-seeker group (n=68). There was no significant difference in weight and body mass index (BMI) among the three groups; however, the sweet-seeker group had significantly higher sweet taste preference than that of the other groups. Though more people in the sweet-seeker group thought they tended to eat sweet foods than the medium sweet-seeker and unsweet-seeker groups, over half of the sweet-seekers did not think they tended to eat sweet foods. The sweet-seeker group was more likely to eat sweet snacks such as breads, chocolate products, sugar-sweetened milk, and so on than the unsweet-seeker group. Total sugar intake from the selected sweet snacks was 44.4 g for the sweet-seeker group, 34.4 g for the medium sweet-seeker group, and 28.0 g for the unsweet-seeker group with a significant difference. These results indicated the absence of relationship between objective sweet taste perception and the obesity index; however, significant associations were detected among objective sweet taste perception, eating sweet snacks and total sugar intake from the snacks. We also found high disagreement between objective and subjective sweet taste perception of the subjects. The present study provided the novel insight that measuring objective sweet taste perception may be useful for assessing the risk of high sugar consumption and undesirable dietary behaviors.

Effect of Substitution of Fermented King Oyster Mushroom By-Products Diet on Pork Quality during Storage

  • This study was carried out to investigate the effects of substitution of fermented king oyster mushroom (P. eryngii) by-products diet on pork meat quality characteristics, during the storage. A mixture of 40% king oyster mushroom by-products, 28% soybean meal and 20% corn was fermented for 10 d, and the basal diet was then substituted by the fermented diet mixture of up to 20, 50 and 80%, respectively. A total of 96 pigs were fed experimental diet (8 pigs per pen ${\times}$ 4 diets ${\times}$ 3 replication), and eight longissiumus (LD) per treatment were collected, when each swine reached to 110 kg of body weight. The Warner-Bratzler shear forces and cooking loss were significantly lowered in the treatments, while crude protein content and water holding capacity significantly (p<0.05) increased in the treatments than in the control group. The volatile basic nitrogen (VBN), at 1 d of storage, was lower in the treatments, while texture profiles and sensory evaluation did not differ between the control and the treatments (p>0.05). The pH, thiobarbituric acid reactive substances (TBARS), VBN and meat color in all treatments were increased as storage increased. Fermented king oyster mushroom by-products diet effects on lightness (CIE $L^*$), yellowness (CIE $b^*$) and chroma were determined, when LD muscles in T2 and T3 treatments were higher (p<0.05), up to 7 d (p<0.05). Therefore, the results indicate that the substitution of the fermented king oyster mushroom by-products diet to swine diet influenced the quality of the meat and it may be an economically valuable ingredient.

  • Novel food sources have enormous potential as nutritional supplements. For instance, edible insects are considered as an alternative food source due to their higher protein content; moreover, they are economically efficient reproducers and have high in nutritional value. In this study, we investigated the toxicity of the freeze-dried powder of Locusta migratoria (fdLM), known to contain rich proteins as well as fatty acids. The objective of the present study was to evaluate the subacute toxicity of fdLM in male and female Sprague-Dawley (SD) rats. The SD rats were divided into four groups based on the dosage of fdLM administered: dosage of 0 (vehicle control), 750, 1,500, and 3,000 mg/kg/day were administered for 28 days. Toxicological assessments including observations on food consumption, body and organ weights, clinical signs, mortality, ophthalmologic tests, urinalyses, hematologic tests, clinical chemistry tests, gross findings, and histopathology tests were performed. Clinical signs, urinalyses, hematology, serum biochemistry tests, and organ weight examinations revealed no fdLM-related toxicity. The no-observed-adverse-effect level for fdLM was higher than 3,000 mg/kg/day in rats of both sexes; therefore, fdLM, in conclusion, can be considered safe as an edible alternative human and animal food source material.

  • Pharmacological studies and clinical practices have indicated that Radix Astragali, a dried root of Astragalus membranaceus possesses a lot of biological activities, including antioxidant, hepatoprotective, anti-diabetic, tonic, diuretic, antimicrobial, antiviral, and immunological activities. These biological activities approved by the modern pharmacological studies are mainly due to the constituents of Astragalus membranaceus including polysaccharides, saponins, flavonoids, amino acids, and trace elements. In resent, the main constituents in the root part showing a lot of biological activities has been isolated also from the aboveground parts such as leaves and sprouts in our laboratory. However, the safety evaluation for the aboveground parts of Astragalus membranaceus should be checked before expanding their application as one of food. In the study, a 90-day rat oral gavage study has been conducted with the extracts from Astragalus membranaceus-above-ground parts at doses of 1000, 3000, and 5000mg/kg/day. The following endpoints were evaluated: clinical observations, body weight, gross and microscopic pathology, clinical chemistry, and hematology. Based on the analysis of these endpoints, it was estimated that NOEL (no observed effect level) for male rats and NOAEL (no observed adverse effect level) for female rats are 5000mg/kg/day of the water-extracts from Astragalus membranaceus-aboveground parts.

Effects of Skipping Breakfast based on the Frequency of Skipping Breakfast of Middle School Students in Sangju, Gyeongbuk Province (경북 상주 일부 중학생의 아침결식 빈도에 따른 결식 관련요인)

  • This paper examined the frequency of skipping breakfast by middle school students for one week. It carried out the study in order to understand the ways skipping breakfast effected other habits. The subjects were 467 middle school students (219 boys, 248 girls). The results of the survey were as follows; average male students were $171.16{\pm}5.9cm$, $61.45{\pm}10.0kg$, and $20.93{\pm}2.9$ in height, weight and body mass index, while the average female students were $159.27{\pm}6.0cm$, $51.62{\pm}7.9kg$ and $20.33{\pm}2.8$. As for the reasons for skipping breakfast, 58.0% of the subjects responded that they had no time. The frequency of skipping breakfast was divided into the eating group (0~1 time) and the skipping breakfast group (2~3times/week, 4~5 times/week, or 6~7 times/week). As the frequency of skipping breakfast increased, so the rate of the subjects eating alone was higher and eating with family lower(p<.05). The dietary behavior change stage was divided into contemplation stage, preparation stage, action stage, and maintenance stage. The eating group(59.2%) was higher than the skipping breakfast group(29.1%~46.3%) in the rate of subjects maintenance stage(p<.01). Accordingly, skipping breakfast was closely connected with eating habits in their families. A dietitian needs to teach the importance of meals and life habits, so as to enhance public awareness of health and nutrition to the students. Subjects of nutrition education must even enlarge their parents as well as the students.

The Effects of Puerariae Radix Catechins Administration on Liver Function in Carbon Tetrachloride-Treated Rats (사염화탄소를 투여한 흰쥐에 있어서의 간 기능에 미치는 칡 카테킨의 효과)

  • These studies were carried out to investigate the effects of Puerariae radix catechins(PRC) administration on the biochemical parameters of liver function in liver of carbon tetrachloride(CCl4)-treated rats. Thirty six healthy Sprague-Dawley rats weighing about 120g were used for this experiment and divided intot he following 3 groups : normal control group(NCON), $CCl_4$ control group(CCON), PRC treated group(PRC). Fifty percent $CCl_4$ in oil was administered(I.P.) by 2ml per kg body weight two times a week for 3 weeks. PRC treated groups were administered orally at the leaves of 1% per day in distilled water for 8 weeks. Lipid hydroperoxides were analyzed by using chemiluminescence-high performance liquid chromatography(CL-HPLC) method as a phosphatidylcholine hydroperoxide value(PCOOH) in liver tissues. $CCl_4$ treatment significantly(p<0.05) resulted in an increase in GPT & GOT activities and liver hydroperoxide values comparing with those of the untreated control, while administration of PRC to the $CCl_4-treated$ rats significantly(p<0.001) decreased GPT & GOT activities and liver hydroperoxide value. Their ultrastructual changes of hepatocellular organelles were shown to clarify the morphologic nature of protective effects of PRC on hepatocytic injuries. $CCl_4$ treatment observed to change the ultrastructual nature of outer membrane of hepatocytes. However, the hepatic changes on PRC treatment to $CCl_4$ group was not found. PRC administration may inhibit the formatiion of liver lipid hydroperoxides in vivo and were very effective in recovering the liver function in $CCl_4-treated$ rats.

A Study on Intake/Balance of Protein and Zinc in Korean High School Boys (한국인 남자 고등학생의 단백질과 아연 평형에 관한 연구)

  • In this study, the food intake, feces and urine of the seven high school boys were collected and the intake and excretion of protein and zinc were measured. The boys were 16~18 years old and the measurement continued for four weeks during which they maintained their normal living pattern and body weight. Each boy's daily intake and excretion of protein and zinc were measured and apparent digestibility and balance were also studied. The results are as follows. Daily mean intake, fecal loss and apparent digestibility of protein of each boy were $73.89{\pm}1.89g,\;10.86{\pm}0.37g$ and $84.9{\pm}0.5%,$ respectively. The urinary loss of nitrogen was $8.8{\pm}0.2g$ and showed the positive balance of $1.3{\pm}0.3g.$ Daily mean intake, fecal loss and apparent digestibility of zinc of each boy were $14.51{\pm}0.44mg,\;7.82{\pm}0.30mg$ and $43.6{\pm}2.9%,$ respectively. The urinary loss of zinc was $4.96{\pm}0.19mg$ and showed the positive balance of $1.72{\pm}0.58mg.$

Effect of Rheum undulatum Linne extract and Glycyrriza uralensis Fischer extract against arachidonic acid and iron-induced oxidative stress in HepG2 cell and CCl4-induced liver injury in mice (대황과 감초 병용의 항산화 및 간보호효과)

  • Objectives : Rheum undulatum Linne and Glycyrriza uralensis Fischer are widely used herbal medicine. In this study, anti-oxidant and liver protective effects of R. undunlatum extract (RUE) and G. uralensis extract (GUE) were investigated in HepG2 cells, respectively. Oxidative stress and liver fibrosis were induced by arachidonic acid (AA) and iron, and CCl4.Methods : MTT assay was assessed for cell viability, and immunoblotting analysis was performed to detect expression of apoptosis related proteins. In addition, reactive oxygen species (ROS) and mitochondrial dysfunction were measured. In vivo, BALB/c mouse were orally administrated with the aqueous extract of 10 mg/kg RUE and 100 mg/kg GUE for 3 days and then, injected with CCl4 0.5 ml/kg body weight to induce acute liver damage. Serum ALT level was measured, and histological change was observed in Harris's hematoxylin and eosin stainResults : RUE and GUE pre-treatment increased relative cell viability in concentration dependent manner and altered the expression levels of apoptosis-related proteins such as procaspase 3, PARP and Bcl-xL. RUE and GUE also inhibited the mitochondrial dysfunction and excessive reactive oxygen species (ROS) production induced by AA and iron. In addition, RUE and GUE activated liver kinase B1 (LKB1), by increasing phosphorylation. Moreover, RUE and GUE treatment decreased liver injuries induced by CCl4, as evidenced by decreases in histological liver damage as well as serum alanine amino transferase (ALT) level.Conclusions : These data suggest that RUE and GUE has anti-oxidant and liver protective effects against AA and iron-induced oxidative stress and CCl4-induced liver injury.

Experimental Study on the Influence of the Function of Spleen on Learning and Memory (비기능이 학습과 기억에 미치는 영향에 대한 실험적 연구)

  • This study was conducted to prove that there exists a relation between the spleen and learning and memory as Oriental medicine believesTo promote the function of the Spleen, Guibitang was administered to rats in this study. Rats were 250~300g Sprague-Dawley, and were divided into three groups. One was the normal group without any pretreatment. Another was the control group which was administered normal saline and the abdominal injection of L-NAME before learning and memory test. And the 3rd was the sample group, to which was administered Guibitang extract and (no 'the') abdominal injection of L-NAME before the learning and memory test. Each group was made up of 12 rats. Morris water maze and radial arm maze tasks were performed in the learning test and Morris water maze task in the memory test. For 2 days to evaluate the ability of learning in the Morris water maze, 16 trials were carried out and first latency(lapse time to find the escape platform for the first time) was measured. The next day, to evaluate the ability of memory, the escape platform was eliminated from the maze, and total path, target entry number, first latency and memory score were measured. 48hrs before the radial arm maze task was performed, bait was deprived from each group. After learning test, bait was permitted to each group. So 85% of the body weight was maintained for 6 days of the test. Each of the eight arms was baited; correct choice numer and error were counted; each trial was finished when the rat had entered each of the eight arms, or more than 10 minutes had elapsed. The results were as follows: In the learning test, the first latency of the sample group in the Morris water maze showed evident improvement of learning compared to control group at the 11th, 12th, 13th trial of 16 trials, and correct choice number in radial arm maze showed noticeable improvement compared to the control group at 3rd, 4th and 5th; In the memory test, the memory score of the sample group showed evident improvement compared to the control group. From the above results, the administration of Guibitang, which tonifies the function of the Spleen, could enhance the ability of learning and memory. So it was suggested that the Spleen has a relation with learning and memory.
