Mutagenic effects of industrial wastewaters by using umu-test (umu-test에 의한 일부 배출시설별 폐수의 변이원성 조사연구)

  • Genotoxicity/mutagenicity of organic chemicals in industrial wastewater was investigated using umu-test with a Salmonella typhimurium TA1535 strain. The tester strain was derived by introducing plasmid pSK 1002, which carried a umu C - lac Z fusion gene into S typhimurium TA1535, and tester strain in the presence microsomal activation proved to be the more sensitive maker of genotoxicity. Genotoxic responses were observed in concentrated with a blue-rayon column, from 14 plants tested. The results were as follow; 1. Genotoxic responses were observed in concentrated from nine plants(64.3%) tested. 2. The results show that genotoxic activity was particulary high in the untreated wastewaters and decreased in the treated wastewaters(35.7%) 3. No significant correlation was found between genotoxicity and water ollution indicators, such as COD and BOD.

  • Physicochemical properties of arrowroot starch were investigated. The starch had blue value of 0.397, alkali number of 10.1 and amylose content of 22.0%. The starch was practically insoluble in water up to $55^{\circ}C$. X-ray diffraction was C pattern, but was close to A pattern. The alkali number, X-ray pattern and viscogram of the starch and retrogadation rate of the starch gels indicated that the properties of the arrow root starch were close to those of cereal starches.

  • Self-cleaning and photocatalytic $TiO_2$ thin films were prepared by a facile sol-gel method followed by spin coating using peroxo titanic acid as a precursor. The as-prepared thin films were heated at low temperature($110^{\circ}C$) and high temperature ($400^{\circ}C$). Thin films were characterized by X-ray diffraction(XRD), Field-emission scanning electron microscopy(FESEM), UV-Visible spectroscopy and water contact angle measurement. XRD analysis confirms the low crystallinity of thin films prepared at low temperature, while crystalline anatase phase was found the for high temperature thin film. The photocatalytic activity of thin films was studied by the photocatalytic degradation of methylene blue dye solution. Self-cleaning and photocatalytic performance of both low and high temperature thin films were compared.

  • Curcuma aeruginosa Roxb. (FamilyZingeberaceae), commonly known as 'pink and blue giner' is widely used in Indian system of medicine since time immemorial. The plant is found well in wild habitat and cultivated in southern India. The plant is distinguished by red corolla lobes and ferruginous or greenish-blue rhizome. Rhizoma is used medicinally as anti-diarrheal, anti-fungal; external use as astringent for wounds. A detailed pharmacognostic evaluation of its rhizome showed total ash 6.1%, acid insoluble ash 1.20%, alcohol soluble extractives 3.70% , water soluble extractives 14.50%, sugar 20.93%, starch 41.85%, and tannins 0.68%. Oh hydro distillation, the rhizomes and sessile tubers yielded 0.17% oil.

  • Some properties of starch isolated from gamma-irradiated chestnut at doses of $0{\sim}0.45\;kGy$ were investigated. The treatment increased the water-binding capacity and decreased intrinsic viscosity and amlyograph viscosities. Other properties including swelling power and solubility at $80^{\circ}C$, blue value and percent transmittance of starch dispersion were essentially not affected by irradiation at sprout-inhibition dose.

  • The stoichiometry between the consumption of CO and $O_2$ and the production of $CO_2(2CO+O_2{\rightarrow}2CO_2)$) showed that Pseudomonas carboxydohydrogena grows as a typical aerobic CO oxidizer with CO. The optimal concentration of CO for growth was found to be 30% in gas mixture with air. The initial buffer concentration of the culture medium did not affect the growth of this bacterium. P. carboxydohydrogena is an obligate aerobe and dose not use nitrate as a terminal electron acceptor. The CO dehydrogenase is an inducible and soluble enzyme. The reaction rate and stability were maximal at pH7.5, and the Arrhenius plot revealed an activation energy of 37.7kJ/mol (9.0 Kcal/mol). The crude enzyme used methylene blue, thionin, and toluylene blue as electron acceptors for the oxidation of CO to $Co_2$ under anaerobic conditions. It was found that water must be the source of the second oxygen atom for CO oxidation.

  • This study was carried out to examine the effect of paint dyeing by Gloiopeltis paste in the various painting sizes and concentrations of paste. In the tonality diagram of Lab, surface colors were changed to +a(red), +b(yellow) and -b(blue) by treating of Gloiopelis paste and L value was the tendency of increasing. The penetration decreased with increasing concentration of paste, except for 3% of red and yellow. While it was decreased steadily in the case of Nue. The best penetration of dye was at 3% of concentration of Gloiopeltis paste and it's viscosity was measured 1500cps. As concentration of paste increased, ${\Delta}E_{ab}$increased after 40 hr. irradiation and water fastness and washing fastness test in red and blue. But in the case of ${\Delta}E_{ab}$yellow decreased slightly or not changed.

  • A marine Streptomyces sp., which produce water-soluble blue pigment was isolated from shallow-sea muds. The effect of various nutritional conditions on growth of isolated strain were investigated to facilite the potential use of this organism in industry. The effect of carbon source was characterized as a 10g/1 of soluble starch. Growth was optimum at pH 7.0 and slightly affected with more alkaline range but was shown to decrease dramatically in acidic range. Under the optimum growth conditions, isolated strain produced substantial amounts of blue pigment and biomass.

  • Total polysaccharide contents in Panax ginseng roots were evaluated by a spectrophotometry, utilizing the complex formation of ginseng polysaccharide with alcian blue dye in 50 mM ammonium biphosphate, pH 4.2. The total polysaccharide content in red ginseng was about three times higher than that in fresh ginseng when both were extracted with water, and was increased about two times when red ginseng was extracted with an alkaline solution. The determination of total polysaccharide in various parts of ginseng revealed that main roots contained the component more than fine roots. Fresh ginseng sections stained by the dye showed polysaccharide mainly found in cortex and cambium but not in epidermis. Pattern-analysis on total and acidic polysaccharides from fresh and red ginsengs exhibited that the chemical compositions of the polysaccharides extracted from both ginsengs quite differed from each other.

  • A simple and convenient method of detecting N, N-dimethy laniline (D.M.A.) residues in ampicillin trihydrate (A.T.) was established. D.M.A. was extracted by chloroform from the chloroform presaturated N/10-ammonia water solution of A.T. and chromatographed on silica gel G thin layer. Blue spot appeared in 15minutes after spray of 2, 6-dichloroquinonechlorimide solution was compared with the blue spot of reference concomitantly processed. The developing solvent was prepared by mixing equal volume of cyclohexane and chloroform. To quantitate the amount of D.M.A. in A.T., T.L.C. was performed with the Eastmann Chromatogram sheet, then color density was measured by Cosmo Superclick densitometer. The developing solvent at this time was cyclohexane-chlorofonn (3+7) mixture. The peak areas obtained with the amount of D.M.A ranging from 0.05 to 2.0 .mu.g were linear to color density. Better sensitive results would be available with the densitometer equipped with monochromator.
