Background and Objectives : Cartilage reconstruction is one of medical issue in otolaryngology. Tissue engineering is presently being utilized in part of cartilage repair. Sources of cells for tissue engineering are chondrocyte from mature cartilage and bone marrow mesenchymal stem cells that are able to differentiate into chondrocyte. Recent studies have shown that adipose tissue have mesenchymal stem cells which can differentiate into adipogenic, chondrogenic myogenic osteogenic cells and neural cell in vitro. In this study, we have examined chondrogenic potential of the canine adipose tissue-derived mesenchymal stem cell(ATSC). Materials and Methods : We harvested canine adipose tissue from inguinal area. ATSCs were enzymatically released from canine adipose tissue. Under appropriate culture conditions, ATSCs were induced to differentiate into the chondrocyte lineages using micromass culture technique. We used immunostain to type II collagen and toluidine blue stain to confirm chondrogenic differentiation of ATSCs. Results : We could isolate ATSCs from canine adipose tissue. ATSCs expressed CD29 and CD44 which are specific surface markers of mesenchymal stem cell. ATSCs differentiated into micromass that has positive response to immunostain of type II collagen and toluidine blue stain. Conclusion : In vitro, ATSCs differentiated into cells that have characteristic cartilage matrix molecules in the presence of lineage-specific induction factors. Adipose tissue may represent an alternative source to bone marrow-derived MSCs.
Some crude drugs in ancient literatures have been used as traditional therapeutic agent of leucorrhea mainly caused by Trichomonas vaginalis and Candida albicans. Sixty six kinds of crude drugs in ancient literatures and ten constituents were selected as sample drugs. Trichomycin standard was tested to compare with the above drugs. To determine the anti-fungal effect of these drugs on Candida albicans Yu 1200, a test organism, screening test was conducted. Antifungal activities of crude drug water extracts were observed by means of two test methods : firstly through the agar slant method and secondly the counting chamber method which was used for acknowledged drug agents upon the result of the agar slant method. And in order to improve the fungicidal effect, the organisms were stained with 0.02% methylene blue solution. The results of the above test indicated that Fritillariae Rhizoma has antifungal action in the concentration of 310mcg/ml, Coptidis Rhizoma in 620mcg/ml, Meliae Cortex, Scutellariae Radix both in 5,000mcg/ml. Baicalin, catechol among the pure isolated constituents inhibited in the range of 50mcg/ml. This score was based on 50% inhibition in comparison with amounts of control organisms. Rhei Rhizoma, Mori Radicis Cortex, Linderae Radix, and Amomi globosi Fructus showed the antifungal effect moderately in 5,000mcg/ml, and baicalein and pectolinarin in 50mcg/ml in the limit of between 35% and 50% antifungal activity. Staining with 0.02% methylene blue showed that any of the crude drug extracts was unable to stain the cells, but trichomycin in 0.86unit/ml able to stain 12% of the cells. This result means that crude drugs probably do not have fungicidal but fungistatic action.
본 연구를 통해 BALB/c 마우스에서 quercetin의 경구투여가 PCL로 유도된 접촉성 피부 알레르기의 예방에 미치는 효과에 대해 알아보았다. Quercetin을 투여 농도에 따라 대조군(0 mg/kg), 저용량군(50 mg/kg), 고용량군(100 mg/kg)으로 나누고 8일에 걸쳐 총 8번 경구투여를 실시한 후 알레르기 유발물질인 PCL을 마우스의 양쪽 귀에 감작시켜 접촉성 피부 알레르기를 유발 시켰다. Quercetin의 투여 농도별 ear swelling 변화를 확인한 결과 quercetin을 투여하지 않은 대조군에 비해 quercetin을 투여한 군의 ear swelling 증가폭이 낮게 나타났고, 100 mg/kg (고용량군)에서 농도 유의적으로 ear swelling의 증가폭이 현저히 낮게 나타났다. Quercetin의 투여 농도에 따른 혈청 내 염증성 매개 물질의 농도 변화를 알아보기 위한 IgE 및 histamine level 측정 결과에서 quercetin을 투여하지 않은 대조군에 비해 quercetin을 투여한 50 mg/kg (저용량군), 100 mg/kg (고용량군)에서 낮은 수준의 IgE, histamine 수치가 나왔다. H&E염색과 Toluidine blue stain을 통한 조직병리학적 검사결과에서 quercetin을 투여한 고용량군의 귀 두께가 대조군에 비해 얇게 관찰되었고, 비만세포의 유무를 알아보기 위한 Toluidine blue stain의 결과 대조군에 비해 고용량군에서 적은 수의 비만세포들이 관찰되었다. 따라서 ear swelling과 IgE, histamine level, 조직병리학적 결과를 종합해 봤을 때 식물성 flavonoid 성분인 quercetin은 접촉성 피부 알레르기의 예방에 상당한 효과가 있다고 판단되며, 부작용이 발생하는 기존의 치료제를 대체할 수 있는 후보 물질로써 중요한 가치가 있다고 사료된다.
The brilliant cresyl blue (BCB) has been used to select the developmental competent oocytes in pigs, goats and cows. Growing oocytes have a higher level of active glucose-6-phosphate dehydrogenase(G6PDH) compare to mature oocytes and are rarely stained compared to mature oocytes, because G6PDH converts BCB to colorless. First polar body extrusion regard as a guideline of meoisis completion. Selection of polar body extrude oocyte is more developmental competent to blastocyst than unselected. This study was conducted to compare the BCB test to the polar body extrusion on selection of developmental competent porcine oocytes for the production of blastocyst. Cumulus-Oocytes complex were exposed to 26uM BCB stain diluted in NCSU-23 for 90 min. There was no significant difference embryo development to blastocysts between BCB treated and not treated($19.58{\pm}1.99$ vs $18.75{\pm}2.27%$), which means there was no detrimental effect of BCB exposure to oocytes. Normal fertilization is not differed among treatment groups from 70.0 to 78.4% development to blastocyst, beside polyspermy did not. To compare two different selection methods, BCB test and polar body extrusion, evaluate the developmental competent of IVP embryos. BCB+PB+(blue stained and polar body extruded, $20.71{\pm}0.45%$) and BCB-PB+(colorless and polar body extruded, $20.04{\pm}l.29%$) groups are significantly (p<0.05) higher developed than those of BCB+PB-(blue stained and no polar body, $13.24{\pm}0.73%$) and BCB-PB-(colorless and no poladbody, $7.25{\pm}0.77%$). These results showed that selection of polar body extruded oocytes method is more efficient than that of BCB test.
The presence of sialoglycoproteins (SGPs) in the membranes from goat (Capra aegagrus hircus), buffalo (Bubalus bubalis bubalis) and pig (Sus scrofa domestica) erythrocytes was investigated by partial purification with a chloroform-methanol extraction method followed by Sodium dodecyl sulphate - Polyacrylamide gel electrophoresis in comparison to human (Homo sapiens) erythrocytes. The results show that mammalian erythrocytes possess clear differences in the SGPs numbers and molecular weights although all animals studied in this experiment are from the same class i.e. mammalia. The SGPs number in human, goat, buffalo and pig are four (PAS-1 to PAS-4), ten (PAS-GI to PAS-GX), seven (PAS-BI to PAS-BVII) and four (PAS-PI to PAS-IV) respectively as indicated by staining the polyacrylamide gel with sialoglycoprotein-specific Periodic acid-Schiff's (PAS) stain. The new SGPs could be observed only after the partial purification of membrane fractions named as PAS-HI with molecular weight (Mr) 190 kDa and PAS-HII 150 kDa in human, PAS-BIA in buffalo and PAS-PIA and PAS-PIVA in pig. The gels were also stained with Coomassie brilliant blue (CBB) and Silver stain to check the contamination of other membrane proteins in the purified fractions. The quantitative distribution of SGPs was also determined by densitometry. Present study indicates that there are some basic differences in mammalian erythrocyte membrane SGPs, especially with respect to their number and molecular weights indicating major structural variations.
The analysis of digital microscopy images plays a vital role in computer-aided diagnosis (CAD) and prognosis. The main purpose of this paper is to develop a machine learning technique to predict the histological grades in prostate biopsy. To perform a multiclass classification, an AI-based deep learning algorithm, a multichannel convolutional neural network (MCCNN) was developed by connecting layers with artificial neurons inspired by the human brain system. The histological grades that were used for the analysis are benign, grade 3, grade 4, and grade 5. The proposed approach aims to classify multiple patterns of images extracted from the whole slide image (WSI) of a prostate biopsy based on the Gleason grading system. The Multichannel Convolution Neural Network (MCCNN) model takes three input channels (Red, Green, and Blue) to extract the computational features from each channel and concatenate them for multiclass classification. Stain normalization was carried out for each histological grade to standardize the intensity and contrast level in the image. The proposed model has been trained, validated, and tested with the histopathological images and has achieved an average accuracy of 96.4%, 94.6%, and 95.1%, respectively.
The present investigation was focussed on histochemical and electron microscopical observations of utero-vaginal(U-V) glands in U-V junctions of domestic hens. In histochemical observations, fine granules by PAS technique for mucopolysaccharides and nile blue stain for acidic lipids were slightly stained on the cytoplasm of U-V glandular epithelium. Larger granules by Sudan black B stain for neutral fat and phospholipids and Sudan III stain for neutral fat were heavily stained on the perinuclear region of the U-V gland epithelial cells. These positive materials were heavily stained on the U-V glandular epithelium of lowfecundity hens and non-laying hens. In scanning electron microscopic findings of the U-V junction surface, the orifices of U-V glands are seen as the crater-like invagination. The neck of the U-V gland and the epithelium of U-V iunction were covered by ciliated epithelial cells. Aggregates of spermatozoa are observed often to be on the necks of the U-V gland. These spermatozoa heads are embedded in the glandular tubules and many spermatozoa tails are free on the epithelium of uterine surface. In transmission electron microscopic findings, the epithelial cells of the U-V glandular orifices were ciliated, columnar cells. The apical regions of these cells contained numerous electrondense, round secretory granules of uniformly size. The epithelial cells of the U-V glandular tubule were columnar or pyramid shape with round or oval nuclei. These epithelial cells have numerous microvilli and also contained electron-dense, round secretary granules of uniformly size and electron-lucent vesicles of various size. Spermatozoa are seen as the cross-sections of various regions of heads and tails in glandular tubules. Also spermatozoa arranged longitudinally parallel within the glandular tubules.
Objective: To compare black rice (Oryza sativa L) extract with three different staining methods for human sperm head assessment. Methods: Semen samples were collected from 34 volunteers. Four smears of each ejaculate were prepared for staining using the rapid Papanicolaou (PAP) stain, SpermBlue, DipQuick, and black rice extract. The percentage of defective sperm heads (mean${\pm}$standard deviation) was compared. Results: Black glutinous rice extract, a natural dye, was used instead of hematoxylin to stain the nuclei of the sperm heads. The percentage of defective sperm heads showed a significant difference between black rice extract and DipQuick (p= 0.000). In contrast, black rice extract and rapid PAP showed no statistically significant difference (p= 0.974). A strong correlation (r = 0.761) was found between the findings obtained using rapid PAP and black rice extract. In contrast, a weak correlation (r = 0.248) was obtained between DipQuick and black rice extract for the percentage of defective sperm heads. Conclusion: The results showed good agreement and a strong correlation between the rapid PAP and black rice extract stains. The advantages of black rice extract as a novel substitute for hematoxylin for nuclear staining include ease of preparation, local availability, and favorable nuclear staining properties. Further studies could also focus on comparing staining techniques in clinical samples.
