Biocontrol of Blue Stain in Pine Wood with Lyophilized Mycelium of Ophiostoma quercus Albino Strain

  • Mycelium of Ophiostoma quercus albino strain cultured in liquid culture media was harvested, lyophilized, and stored for examining biocontrol efficacy against wood discoloration by staining fungi in the laboratory and field conditions. Dry weight of mycelium grown in brown sugar yeast extract broth(BYB) showed 3.8 times higher than that grown in potato dextrose broth(PDB). The optimum culture period in BYB was 4 weeks. In vitality test of the albino strain, the lyophilized mycelium stored in liquid nitrogen($-196^{\circ}C$) or in a refrigerator($4^{\circ}C$) kept the vitality until 13 months after storage; however, the mycelium stored at room temperature lost the vitality completely after 13 months. The mycelium stored in liquid nitrogen or in a refrigerator protected wood chips from the discoloration by pretreating mycelial suspension on pine wood chips. The mycelium stored at room temperature for 7 months also showed complete protection. These results suggest that the lyophilized mycelium have a biocontrol efficacy only if it keeps the least vitality. In the field conditions, both albino strain and $Woodguard^{(R)}$(commercial chemical protectant) showed significant differences(p=0.05) in discoloration rate as compared to the non-treated control when these were treated on the wood logs of Pinus rigida. The albino strain showed better protection than $Woodguard^{(R)}$. Isolation frequency of blue stain fungi from the chips of wood logs treated with the albino strain was 0% at three months after treatment, while that treated with $Woodguard^{(R)}$ was 76.7%. In another experiment, pre-treatment of mycelial suspension on the cut surface of wood logs also showed significant protection from wood discoloration. Spraying of both albino strain on the cut surface and insecticides on the bark also showed relatively good control effects as compared to insecticide alone on the bark or nontreated control.

  • The purpose of the current study was to evaluate the effect of the zinc-deficient diet on the connectine tissue of oral mucosa in the albino rats. The zinc dificient diets were supplied to the rats for 4 weeks. The results were as follows: Most of the papillas of lamina prmina propria were shortened. The abnormal distribution and orientation of the fibrous elements were observed. The stain ability of fihrous elements in the Mallory azan stain was decreased. The amorphous intercellular substance showed slight metachromasia in the toluichine blue stain and low PAS reactinity.

  • We investigated the possibility of systemizing data for the colour gradation of classical ceramics by build-ing program and D-basing each data information in pottery field that has been fall behind in forming data be-cause of many variable and lacking of data system. In this study we manufactured $CoAl_2O_4$ stain that has stable spinel structure at high temperature and then mixed base glaze with stain for the additive per-centage of 0.5 , 1, 5, 10, 15 wt% In thie colouring of Lime base glaze with the wide ranged melting tem-perature the colour development of cobalt-blue sintered at $1250^{\circ}C$ is better than that of $1150^{\circ}C$ $1350^{\circ}C$ The experimental conditions and data are D-based simulated programming of computer. The result of -based data appeared to be more smoothed fusion-adherence in the Matt glaze range of Lime base glaze, The variation of colouring is not found when additive content of glaze is more than 5 wt% and the stable colour development of stain was shown at the content that RO, $R_2O$ percentage is high (the content that R2O3 per-centage is low)

  • Background : cis-Diamminedichloroplatinum(Ⅱ)(cis-platin) has been exhibited similar to bifunctional alkylating agents in the cell and known as an effective anticancer drug. Although this agent is very beneficial to the cancer patients, it may also damage to the normal cell. Many side effects were developed. Objectives : This experiments was undertaken to pursue the effect of cis-Platin on the mucous variation of the trachea. Materials and Methods : The male of Sprague-Dawley strain were used as and experimental animals. The experimental animals were killed at 1, 3 days and 1, 2 and 3 weeks after the third injection of cis-Platin was administered 1.5mg/kg to intraperitoneal injection at once a week for three weeks. The trachea was fixed to neutral formaline and stained with alcian blue(pH 1.0)-PAS, alcian blue(pH 2.5)-PAS double stains and these preparation observed with light microscope. Results : 1. In the trachea stained with alcian blue(pH 2.5)-PAS double stain, the epithelial cells were constricted in the 1 week, In the 1st day, 3rd day and 1st week, acidic mucopolysaccharide was increased but in the 2nd week, neutral mucopolysaccharide was increase. 2. In the trachea stained with alcian blue(pH 1.0)-PAS double stain, the 1st and 3rd day exhibited sulfa mucopolysaccharide with moderately or weakly positive reaction. In the 1st week the sulfa mucopolysaccharide with strongly positive reaction was increased. In the 2nd and 3rd week, the sulfa mucopolysaccharide with weakly positive and non-sulfa or neutral mucopolsaccharide with negative reaction were modified. Conclusion : It is consequently suggested that cis-Platin induces reversible toxic damage to tracheal cells including goblet cell.

Frequency of Blue Staining Fungi isolated from Pine Trees of Experimental Forests in Kangwon National University and Its Resistance to Fungicide, Woodguard

  • This study was performed to investigate the frequency of blue staining fungal species collected from pine trees, Experimental Forests of Kangwon National University in Korea based on their morphological characteristics. In addition the tolerance to fungicide, Woodguard, was assessed to get basic knowledges for preventing blue stain of wood. Totally Leptographium-type fungi were dominated by 79.3% among Ophiostomatoid fungi associated with scolityd bark beetles in pine trees. Leptographium-type Ds-isolates which have unusual morphology were collected as frequency of 17.0%. The most distinct differeneces of these Ds-isolates from L. procerum were the presence of roughened hyphae and flask-shaped conidiophores that have never been mentioned formerly for L. procerum, but since these Ds-isolates formed black concentric rings being a property of L. procerum, the Df-isolates were characterized as Leptographium-type fungi, which are the most common species with the highest frequency by 33.2% in this particular area. According to our experimental results, Leptographium-type Ds- and Df-isolates were very resistant to fungicide, Woodguard, therefore it was suggested that a new method for wood protection from the blue staining fungi should be developed. Exact identification of blue staining isolates collected from pine trees is keep going.

자외선 조사가 백서구강점막에 미치는 영향에 관한 실험적 연구 (EFFECT OF ULTRAVIOLET RADIATION ON THE ORAL MUCOSA OF THE RATS)

  • 최근 치과영역에서 자외선을 이용하는 치료방법이 많이 개발되고 있다. 자외선은 생체에 대하여 가역적 내지 불가역적인 생물학적 손상을 입히는 것으로 알려져 있다. 이에 저자는 Nuva-Lite를 이용하여 near-ultraviolet가 백서구강점막에 미치는 바를 조직학적 및 조직화학적으로 관찰하였다. 백서구강점막을 ether 마취하에 절취한후 Carnoy여과 10% 중성완충포르말린으로 고정한 후 파라핀으로 진공포매 하였다. 절편은 hematoxylin-eosin염색, Feulgen반응, methyl green-pyronin염색, alloxan Schiff반응, -SH기염색, PAS반응, colloidal iron반응, toluidine blue 염색을 실시하여 다음과 같은 결과를 얻었다. 1. 자외선 조사로 인하여 24시간 내지 3일에 극세포층에서 경미한 세포내 부종이 보였고, 6일 내지 10일에서 rete peg이 증대되었다. 2. 자외선 조사후 6시간 내지 3일에서 Feulgen반응에 양성인 DNA가 감소되는 경향이었다. 3. 자외선 조사후 -SH기 및 ${\alpha}$-amino산 반응이 상피의 상층에서 증가되는 경향이었다.

새로운 치면세균막 착색제 개발을 위한 식용색소의 생체적합성 연구 (BIOCOMPATIBILITY OF FOOD COLORING AGENTS TO DEVELOP NEW DENTAL PLAQUE DISCLOSANTS)

  • The purpose of study was to develop new dental plaque disclosants which could replace erythrosine. Three food coloring agents(Red No.40, Blue No.1, and Mixed Green), erythrosine and fluorescein were tested for their color difference, antibacterial property, and biocompatibility. Color difference of Red No.40 was greater than that of erythrosine as concentration of solution increased. Color differences of Blue No.1 and Mixed Green were smaller than that of red dyes. Erythrosine showed obvious antibacterial property, but food coloring agents showed almost no antibacterial property. The taste and sensation of erythrosine was the worst, and the taste of Red No.40 and the sensation of Mixed Green were the most tolerable. Erythrosine stained dental plaque and oral soft tissue most deeply and long, and Blue No.1 was the next in the depth and longevity of stain.

생쥐, 쥐 및 돼지의 난소내 난포의 폐쇄에 관한 조직화학적 연구 (Histochemical Study of the Atresia of Ovarian Follicles)

  • In order to study the mechanism of follicle growth and maturation, and also to supplement the criteria identifying the follicle state of normal of atretic, the histochemical investigation on the ovarian follicles according to the ovarian cycle of mouse, rat and pig has been done. The intercellular space of granulosa cells, especailly Call-Exner body, and follicular fluid in the antrum showed positive to PAS, and blue stain by trichrome dye. The resutls suggest that the mucous polysaccharide was synthesized by the granulosa cells, and secreted into the antrum through Call-Exner body so as to be the components of the follicular fluid as the follicles proceeded to growth and maturation. The further the follicles proceeded to atresia the more densely their theca externa were stained blue by follicles proceeded to atresia the more densely their theca externa were stained blue by trichrome dye, and the more densely the granulosa cells were stained red by oil red 0 dye. Therefore, these staining methods can be applied to the criteria identifying the follicle atresia.

뽕나무 오갈병 마이코플라스마의 몇가지 초본식물에의 전염과 조직화학적 검정 (Transmission and Histochemical Detection of Mulberry Dwarf Mycoplasma in Several Herbaceous Plants)

  • 뽕나무 오갈병 마이코플라스마가 매개곤충인 마름무늬매미충에 의하여 5가지 초본식물(일일초, 화이트클로우버, 라디노클로우버, 레드클로우버, 자운영)에 전염되었음이 병징발현과 광학 및 전자현미경적 방법에 의하여 확인되었다. 전염된 식물에서는 마름무늬매미충이 적어도 25일 이상 생존하였고, 산란이 확인되었다. 이 병의 잠복기간은 일일초에서 $25\~30$일, 클로우버류와 자운영에서는 $35\~40$일로 나타났다. 감염된 식물의 공통된 병징은 잎의 변색으로, 일일초는 엽맥투화 및 황화, 화이트 및 라디노클로우버는 갈색, 레드클로우버는 적색, 자운영의 잎은 광색으로 각각 나타났다. 한편 클로우버류와 자운영에서는 상기한 잎의 변색과 함께 식물체에 위축병상이 나타났다. Dienes 염색에 의한 광학현미경적 진단방법은, 모든 이병식물의 줄기 사부가 특이하게 염색디어 뽕나무 오갈병 마이코플라스마의 검출에 신빙성이 있고, 사용하기에 간편한 것으로 나타났다. Toluidine blue 염색에 의한 광학현미경적 방법과 전자현미경 관찰로, 이병식물조직내에서 마이코플라스마의 존재가 확인됨으로써 이 병의 전염이 입증되었다.
