Growth characteristics of bloom forming Mallomonas elongata (Synurophyceae) based on silicate and light intensity

  • A dominant planktonic bloom-forming species, Mallomonas elongata was isolated from a small shallow eutrophic pond. The growth characteristics of this species on variable silicate concentrations and light intensities were investigated in laboratory unialgal cultures. In culture condition of $15^{\circ}C$, the maximum population growth and the highest growth rate of M. elongata occurred at a light intensity of $80\;{\mu}mol\;m^{-2}s^{-1}$, and in culture condition of $18^{\circ}C$, it exhibited the maximum population growth and the highest growth rates at a light intensity of $50\;{\mu}mol\;m^{-2}s^{-1}$. Silicate concentration had no effect on the population growth and growth rate of M. elongata.

Long-term Preservation of Bloom-forming Cyanobacteria by Cryopreservation

  • Long-term preservation of bloom-forming cyanobacteria was evaluated using cryopreservation and freeze-drying of nine strains belonging to four genera and seven species. All test strains, except Aphanizomenon flos-aquae NIER- 10028, showed partial or complete survival following cryopreservation and freeze-drying. Frozen and freeze-dried strains were preserved for more than two years and were revived monthly. Most strains showed higher post-thaw viability after cryopreservation, especially without cryoprotectant compared to freeze-drying. Microcystis aeruginosa NIER-10010, M. viridis NIER-10020, M. ichthyoblabe NIER-10023, M. novacekii NIER-10029 and Oscillatoria sancta NIER-10027 were revived after 2.5 years of cryopreservation. These results suggest that cryopreservation may be an easy and timesaving long-term preservation method for bloom-forming cyanobacteria.

녹조현상 원인조류들의 염소처리에 의한 소독부산물 생성 및 microcystins 유출 (Effect of Chlorination on Disinfection Byproducts Production and Release of Microcystins from Bloom-forming Algae)

  • The effect of chlorination on disinfection byproducts (DBPs) production from bloom-forming freshwater algae including 7 strains of cyanobacteria and 6 strains of diatoms was investigated. The release and degradation of hepatotoxin (microcystins) by the chlorination on Microcystis under differential condition of the chlorination time and dose were also investigated. The disinfection byproducts formation potentials (DBPFP) of cyanobacterial species and diatoms were ranged from 0.017 to $0.070{\mu}mol\;DBPs/mg$ C and from 0.129 to $0.708{\mu}mol\;DBPs/mg$ C respectively. Among three major groups of DBPs, haloacetonitrils (HANs) was major product in most test strains except Aphanizomenon sp. and Oscillatoria sp. Haloacetic acids (HAAs) was less than 5 % of total DBPs. Chloroform and dichloroacetonitril (DCAN) were dominant compounds in trihalomethanes (THMs) and HANs respectively. After 4 hours chlorination of toxic Microcystis aeruginosa under the dose range of 0.5 to $10mg\;Cl_2/L$, the concentration of intracellular microcystins decreased, but dissolved dissolved microcystins concentration increased with the treatment of more than $3mg\;Cl_2/L$. However the total amount of microcystins was almost constant even at $10mg\;Cl_2/L$ of chlorination. To conclude, our results indicate that the chlorination causes algal cell lysis and release of intracellular microcystins in the intact form to surrounding waters.

수종 담수적조 원인종들의 형광특성과 적용연구 (The study on the Fluorescence Characteristics of Several Freshwater Bloom Forming Algal Species and Its Application)

  • The freshwater blooms mainly blue-green algal blooms occur frequently in the lower Naktong River in summer, which provoke many socio-economical problems; therefore, the early detection of bloom events are demanding through the quantitative and qualitative analyses of blue green algal species. The in vivo fluorescence properties of cultured strains of Microcystis aeruginosa, M. viridis, M. wesenbergii, M. ichthyoblabe, Anabaena cylindrica, A. flos-aquae, and Synedra sp. were investigated. Wild phytoplankton communities of the lower Naktong River were also monitored at four stations in terms of their standing stocks, biomass and fluorescence properties compared with its absorption spectram. The 77K fluorescence emission spectra of each cultured strains normalized at 620 nm was very specific and enabled to detect of blue green algal biomass qualitatively and quantitatively. The relative chlorophyll a concentration determined by chlorophyll fluorescence analysis method showed significant relationship with chlorophyll a concentration determined by solvent extraction method ($R^2$ = 0.906), and the blue-green algal cell number determined by microscopic observation ($R^2$ = 0.588), which gives insight into applications to early detection of blue green algal bloom.

Re-evaluation of green tide-forming species in the Yellow Sea

  • Green tides occur every year in the Yellow Sea (YS), and numerous investigations are proceeding on various aspects of the phenomenon. We have identified bloom-forming species collected from diverse locations in the YS using morphological traits and the chloroplast gene for the large subunit of ribulose-1,5-bisphosphate carboxylase (rbcL). Morphological and rbcL sequence data analyses characterized the blooming species on both sides of the YS as belonging to the Ulva linza-procera-prolifera (LPP) complex clade or U. prolifera of earlier reports. However, U. procera within the LPP complex must be regarded as synonym of U. linza. Moreover, U. prolifera in free-floating samples collected from the Qingdao coast in 2009 was clearly in a distinct clade from that of the blooming species. Therefore, U. linza is the main green tide alga in the YS and has the procera-morphology. The green drift mats in the southeastern part of the YS (southwest sea of Korea) consisted predominantly of U. linza and rarely of U. compressa or U. prolifera.

Newly recorded unarmored dinoflagellates in the family Kareniaceae(Gymnodiniales, Dinophyceae) in brackish and coastal waters of Korea

  • Unarmored dinoflagellates, in the family Kareniaceae, include harmful or toxic bloom-forming species, which are associated with massive fish kills and mortalities of marine organisms worldwide. The occurrence and distribution of the toxigenic species in the family Kareniaceae were investigated in the brackish and coastal waters of Korea between July 2018 and October 2020. During the survey, we collected seven newly recorded species; Karenia papilionacea, Karlodinium digitatum, Karl. veneficum, Karl. zhouanum, Takayama acrotrocha, T. helix, and T. tasmanica. A total of fifteen strains of the seven taxa were successfully established as clonal cultures and examined using LM, SEM, and molecular phylogeny inferred from LSU rDNA sequences. Herein, we present the taxonomic information, morphological features, and molecular phylogenetic positions of the unrecorded dinoflagellate species collected from Korean coastal waters.

  • 작은 부영양 저수지에서 수화를 형성하는 Mallomonas elongata의 계절적 변동(2004년 10월-2005년 9월)과 온도와 pH에 대한 증식 특성을 실험실 배양을 통해 조사하였다. 수온 12-$18^{\circ}C$, pH8.4-9.5의 범위를 나타낸 3월 말에서 4월 초의 짧은 기간 동안 M. elongata의 심한 bloom (최고 17,600 cells $mL^{-1}$이 발생하였다. 이 저수지로부터 분리한 M. elongata의 batch culture를 통한 다양한 온도에 대한 성장 반응은 bloom을 형성 하였을 때의 저수지 수온과 유사한 $15^{\circ}C$ 에서 최대성장률을 나타내었다. 반면 pH에 대한 증식 특성은 bloom 형성기의 저수지 pH 범위 보다 낮은 pH 6에서 최대 성장률을 나타내었다.

Taxonomy of Ulva causing blooms from Jeju Island, Korea with new species, U. pseudo-ohnoi sp. nov. (Ulvales, Chlorophyta)

  • Several species classified to the genus Ulva are primarily responsible for causing green tides all over the world. For almost two decades, green tides have been resulted in numerous ecological problems along the eastern coast of Jeju Island, Korea. In order to characterize the species of Ulva responsible for causing the massive blooms on Jeju Island, we conducted DNA barcoding of tufA and rbcL sequences on 183 specimens of Ulva from eight sites on Jeju Island. The concatenated analysis identified five bloom-forming species: U. australis, U. lactuca, U. laetevirens, U. ohnoi and a novel species, U. pseudo-ohnoi sp. nov. Among them, U. australis, U. lactuca, and U. laetevirens caused to the blooms coming mainly from the substratum. U. ohnoi and U. pseudo-ohnoi sp. nov. were causative the free-floating blooms. Four species, except U. australis, are characterized by marginal teeth. A novel species, U. pseudo-ohnoi sp. nov., is clearly diverged from the U. lactuca, U. laetevirens, and U. ohnoi clade in the concatenated maximum likelihood analysis. Accurate species delimitation will contribute to a management of massive Ulva blooms based on this more comprehensive knowledge.

북한강 수계 조류대발생 원인종 남조 Anabaena의 분자계통학적 검토 (Molecular Identification of the Bloom-forming Cyanobacterium Anabaena from North Han River System in Summer 2012)

  • 2012년 북한강수계 전역에 걸쳐 대발생을 일으킨 남조 Anabaena의 분자생물학적 특성을 검토하였다. 시료는 2012년 7월 13일에 남조 Anabaena 밀도가 높았던 3개 호소- 의암호, 청평호, 팔당호에서 각각 채집하였다. 분석은 선행연구 문헌을 근거로 하는 형태학적 분석, 16S rRNA 염기서열 분석을 통한 분자계통학적 분석을 동시에 실시하였다. 결과를 종합하면 북한강수계 3개 호소에서 조류 대발생을 일으킨 남조 Anabaena는 모두 동일종 Anabaena crassa (Lemmermann) Komark.-Legn. & Cronberg이며, 동시에 출현하고 본 종과 형태 및 분자생물학적으로 매우 유사한 A. circinalis는 환경요인에 따라 형태변이가 쉽게 일으키는 동일종으로 밝혀졌다.

Seasonal Changes in Cyanobacterial Diversity of a Temperate Freshwater Paldang Reservoir (Korea) Explored by using Pyrosequencing

  • The incidence of freshwater algal bloom has been increasing globally in recent years and poses a major threat to environmental health. Cyanobacteria are the major component of the bloom forming community that must be monitored frequently. Their morphological identities, however, have remained elusive, due to their small size in cells and morphological resemblances among species. We have analyzed molecular diversity and seasonal changes of cyanobacteria in Paldang Reservoir, Korea, using morphological and 16S rRNA pyrosequencing methods. Samples were collected at monthly intervals from the reservoir March-December 2012. In total, 40 phylotypes of cyanobacteria were identified after comparing 49,131 pyrosequence reads. Cyanobacterial genera such as Anabaena, Aphanizomenon, Microcystis and Synechocystis were predominantly present in samples. However, the majority of cyanobacterial sequences (65.9%) identified in this study were of uncultured origins, not detected morphologically. Relative abundance of cyanobacterial sequences was observed as high in August, with no occurrence in March and December. These results suggested that pyrosequencing approach may reveal cyanobacterial diversity undetected morphologically, and may be used as reference for studying and monitoring cyanobacterial communities in aquatic environments.
