Effect of PHA and conditioned medium on blastogenesis and rosette formation of bovine circulating blood lymphocytes (PHA 및 conditioned medium 이 소의 순환혈액 림프구의 유약화와 rosette 형성에 미치는 영향)

  • This study was planned to estimate the activity of bovine circulating blood lymphocytes using phytohemagglutinin-M(PHA) known as T cell mitogen. Bovine circulating blood mononuclear cells(MNCs) was separated, and cultured with or without macrophage($PHA^+/M{\phi}^+$ or $PHA^+/M{\phi}^-$) in conditioned medium which stimulated with various concentration of PHA(0, 5, 10, 15 and $20{\mu}g/ml$ in medium), and then investigated the blastogenic response and rosette formation of lymphocytes. Blastogenic rate(BR) was especially increased in PHA concentration(10 and $15{\mu}g/ml$) of $PHA^+/M{\phi}^+$ group and their BR were $41.5{\pm}6.8%$ and $44.4{\pm}8.9%$, respectively and BR in PHA concentration(15 and $20{\mu}g/ml$) of $PHA^+/M{\phi}^-$ group was $32.8{\pm}6.2%$ and $31.4{\pm}4.6%$, respectively. BR of lymphocytes was more increased in $PHA^+/M{\phi}^+$ than $PHA^+/M{\phi}^-$ group when these cells were stimulated by PHA. Rosette forming rate(RFR) of lymphocytes to SRBC highly increased when SRBC was treated with AET and/or dextran, respectively. On the orther hand, RFR significantly increased more in $PHA^+/M{\phi}^+$ and $PHA^+/M{\phi}^-$ group than in control group, but when compared with two groups, statistical significancy was recognized only in PHA concentration($15{\mu}g/ml$, p<0.026) of $PHA^+/M{\phi}^+$ group.

  • By inserting Boa-tang into culture median of lymphocytes from spleen of rat and lymphocytes in human peripheral blood The results were as follow. 1. In case of spleen lymphocytes of rat, cultures of lymphocytes did not significantly increased which were inserted Boa-tang $100{\mu}g/ml$, and which were inserted $10{\mu}g/ml$ at 3rd day 2. In case of spleen lymphocytes of rat, cultures of lymphocytes were significantly increased which were inserted Boa-tang $10{\mu}g/ml$ with Con A $2.5{\mu}g/ml$ at 2nd, 3rd day, and Boa-tang $1{\mu}g/ml$ with ConA $2.5{\mu}g/ml$ at 3rd day than inserted ConA $2.5{\mu}g/ml$ 3. In case of lymphocytes in human peripheral blood, cultures of lymphocytes were significantly increased which were inserted Boa-tang $100{\mu}g/ml$ and $10{\mu}g/ml$ at 2nd day to 5st day, and which were inserted Boa-tang $1000{\mu}g/ml$ at 2nd day to 5th day, and which were inserted $1{\mu}g/ml$ at 2nd day By looking at the following results, in case of rat cultures of lymphocytes were significantly increased which were inserted the lower density, but in case of human cultures of lymphocytes were significantly increased which were inserted the higher density.

  • Cytogenetic and hematological analysis was performed in peripheral blood of cattle in the vicinity of Uljin nuclear power station and control area. The frequency of micronuclei(MN) in peripheral blood lymphocytes from cattle was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN ratio in lymphocytes of cattle from Uljin nuclear power station and control area were 8.90/1,000 and 9.60/1,000, respectively. There were no significant differences in MN frequencies and hematological values in cattle between Uljin and control area.

  • With an effort to clarify the nature of the azurphil granules which occasionally occur in the circulating lymphocytes, these granules were investigated by examining smear of the peripheral blood of the chikens in various stage of the individual growth and after injection of methylene blue and gentian violet. In addition, the fine structure of these granules were also investigated. The results were: 1. These granules were first occurred in the lymphocytes just after their hatching (0.004%). The proportion of lymphocytes containing these granules were increased with their growth and in adult chicken its occurrence was higher than mammals. 2. Marked variations in its fine structure, particularly in its size and cotents, were noted but they were believed to belong to categories of lysosome of de Duve. 3. Lymphocytes containing azurophil granule were increased after injection of the non-immunogenic substances, such as gentian violet and methylene blue. 4. From the above results, chicken is bettor animal to study theme granules because of its higher occurrence. They are believed to have intimate relationship with bodily cellular reaction against the foreign materials because they are increased after non-immunogenic stimuli.

  • Cytogenetic and hematological analysis was performed in bovine peripheral blood from the regions around Wolsong nuclear power plant and control area. The frequency of micronuclei (MN) in peripheral blood lymphocytes from cattle was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dare of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN rates in lymphocytes of cattle from Wolsong nuclear power plant and control area were 9.87/1,000 and 9.60/1,000, respectively. There were no significant differences in MN frequencies and hematological values in cattle between Wolsong and control area. The study indicates that the MN assay is a rapid, sensitive and accurate method that can be used to monitor a large population exposed to radiation.

  • Objectives: The purpose of this study was to verify the effect of Palmiboshinwhan (PMBSW) in methotrexate (MTX)-induced immunosuppressed SD rats. Methods: The test articles were once a day dosed for 14 days by gastric gavage from 2 days after last MTX-dosing, and changes in body weight, spleen weight and total blood leukocyte numbers were observed with total lymphocyte numbers, B and T lymphocyte percentages, CD3+CD4+, CD3+/CD8+, CD4+/CD8+ T lymphocyte percentages in the blood and spleen, the serum interleukin (IL)-2 levels and the productivity of IL-2 of splenic cells. Result & Conclusion: It is concluded that PMBSW has relatively good immunostimulating effect in the MTX-induced immunosuppressed SD rats. Theefficient dosage was considered above 500mg/kg. In addition, it is considered that the immunostimulating effect of PMBSW was mediated to both the B and T lymphocytes. The more favorable effects were detected in T lymphocytes rather than B lymphocytes, and PMBSW showedrelatively good stimulating potential against CD4+ T lymphocytes but not any stimulating effect against CD8+ T lymphocytes in the present study.

  • We have previously demonstrated that intracellular expression of an RNA aptamer termed RRE40, which was selected in vitro to bind HIV Rev 10-fold much tighter than wild-type RRE, efficiently protected human CD4+ T cell line, CEM, from HIV-1. In this study, to evaluate the efficacy of the RRE40 RNA in clinical settings, polyclonal CD4+ peripheral blood lymphocytes (PBLs) were transduced with retroviral vectors expressing RRE40 decoy RNA and then challenged with clinical isolates of HIV-1. In contrast to the control cells transduced with vectors expressing control tRNA, intracellular expression of RRE40 RNA more effectively inhibited HIV-1 replication in CD4+ PBLs. However, transient and diminished inhibition, rather than complete inhibition, of HIV-1 replication in PBLs expressing RRE40 decoys have been observed. These results suggest that RRE40 decoy RNA would be useful to inhibit HIV-1 replication in cells. However, development of more efficient gene transfer protocols and/or more effective decoy RNAs would be needed to apply RNA decoy to modulate HIV-1 patient.

Effects of Gossypol Injection into the Stroma of Testis on Blood Constituents in Mice (생쥐 정소실질내 Gossypol 투여가 혈액성상에 미치는 영향)

  • We have previously shown that gossypol injection into the stroma of testis was found to effectively inhibit the process of spermatogenesis. In this study, properties and chemistry of blood were investigated weekly in ICR mouse after the injection of gossypol into the testicular stroma(5, 10 or 15mg per kg body weight). There were no significant differences in red blood cell(RBC), hematocrit, white blood cell(WBC), basophils, eosinophils and monocytes during first 4 weeks after injection of gossypol between treatments, but neutrophils increased and lymphocytes decreased, respectively(P<0.05). Total content of protein, albumin and globulin were not different, compared with control. However, the concentration of glucose after injection of gossypol increased significantly(P<0.05). In conculsion, the results of this study indicated that injection of gossypol into stroma of testis might affect both properties and chemistry of blood in mice.

Reference Values for Peripheral Blood Lymphocyte Subsets in a Healthy Korean Population

  • Flow cytometric immunophenotyping of peripheral blood lymphocyte subsets is a powerful tool for evaluating cellular immunity and monitoring immune-mediated diseases. The numbers and proportions of blood lymphocyte subsets are influenced by factors such as gender, age, ethnicity, and lifestyle. This study aimed to establish reference ranges for peripheral blood lymphocyte subsets in a healthy Korean population. Blood samples from 294 healthy adults were collected. Lymphocyte subsets were analyzed using a single-platform method with a flow cytometer; white blood cells and lymphocytes were analyzed using an automated hematology analyzer. The mean value of the white blood cell count was $5,665cells/{\mu}l$, and the mean values of the subtype counts (percentages) were as follows: lymphocytes, $1,928cells/{\mu}l$ (35.08%); $CD3^+$ cells, $1,305cells/{\mu}l$ (67.53%); $CD3^+CD4^+$ cells, $787cells/{\mu}l$ (40.55%); $CD3^+CD8^+$ cells, $479cells/{\mu}l$ (25.23%); $CD3^-CD19^+$ cells, $203cells/{\mu}l$ (10.43%); and $CD3^-CD56^+$ cells, $300cells/{\mu}l$ (15.63%). Additionally, the $CD4^+/CD8^+$ ratio was 1.81. In this study, gender and age significantly influenced blood lymphocyte subsets. Our results demonstrate that, as with other populations, a healthy Korean population has its own, region-specific, lymphocyte subset reference ranges.

Inhibitory Mechanism of Propranolol on the Effects of VIP in Peripheral Blood T-lymphocytes of Rat (흰쥐 말초혈액 T-림프구에서 Vasoactive Intestinal Polypeptide의 효과에 대한 Propranolol의 억제 기전)

  • Vasoactive intestinal polypeptide(VIP) and ${\beta}-adrenergic$ agonists have immunomodultory effects on the peripheral blood T-lymphocytes of rat through their own receptors. Both of them utilize the same signal transduction pathway. That is, the stimulatory guanine nucleotide binding protein(G protein) mediates the receptor-adenylyl cyclase coupling, producing intracellular increase of cyclic adenosine monophosphate(cAMP). In the previous experiment, propranolol, a ${\beta}-adrenergic$ receptor blocker, inhibited the VIP-induced protein phosphorylation in lymphocytes. However, propranolol could not block the effect induced by forskolin. Therefore, this study was designed to elucidate the mechanism of the inhibitory action of propranolol on the effects of VIP. Using peripheral blood lymphocytes of rats, the effect of propranolol on the receptor binding characteristics of VIP was observed. And the effects of propranolol were compared to the effects of timolol on the cAMP increase induced by isoproterenol, VIP or forskolin. The results obtained are as follows. 1) Receptor binding study showed no significant differences in the affinity or density of VIP receptor between the control and propranolol-pretreated groups. 2) VIP-induced increase of cAMP was inhibited by propranolol, but not by timolol. 3) Both propranolol and timolol suppressed the isoproterenol-induced cAMP increase. 4) Propranolol also inhibited the histamine-induced cAMP increase. 5) Propranolol did not inhibit the increase of cAMP stimulated by forskolin. 6) Lidocaine did not block the VIP-induced cAMP increase. These results show that the inhibitory mechanism of propranolol is not related to ${\beta}-adrenergic$ receptor or its membrane stabilizing effect, and it is suggested that propranolol can block the effects of VIP by inhibiting the intermediate step between the VIP receptor and adenylyl cyclase.
