Rat의 intestine 각 부위에 수술적으로 투여 된 insulin 제제에 의한 혈당 변화 (Blood glucose change after surgical administration of insulin formula into rat intestinal regions)

  • The present study was carried out to examine the effect of insulin formula on blood glucose change in normal Sprague-Dawley male rats. Also, this study was performed to investigate the feasibility of oral insulin formula development. To administrate the insulin formula into intestine, the surgical technique, celiotomy, was performed in rats. Insulin formula was administrated at a dose of 24.5 IU/kg via duodenum, ileum, and colon of the rats, and the blood glucose level was measured. For the comparison, the vehicle without insulin was administrated into ileum via celiotomy. Also, this insulin formula was administrated into rats orally using sonde and the same parameter was treasured. The bloods of all groups were collected from tail veins using syringes at given time interval. Orally administrated group did not show the change of blood glucose level and control group slightly show the change of blood glucose level at 1 hour after celiotomy. All intestinally administrated groups showed the change of blood glucose level. Among the tested groups, ileac administration group and colonic administration group showed the significant change of blood glucose level. Particularly, ileac administration group showed the lowest blood glucose level. To calculate the bioavailability of intestinal and oral administration, insulin solution was injected subcutaneosly, common insulin injection route, into another normal rats. The bioavailability of ileac group was 8.3% when compared with subcutaneous injection, duodenal group was 1.8%, colonic group was 4.2%, and oral group was 0.2%, respectively.

정상 성인의 혈청 인슐린 농도에 따른 비만지수, 혈청 지질, 혈압 및 영양소 섭취상태 비교 (Comparisons of BMI, Blood Lipid Profile, Blood Pressure and Nutrient Intakes by Serum Insulin Concentrations)

  • This study was conducted to figure out the differences of the BMI, blood lipids, blood pressure and nutrient intakes by serum insulin concentration among adults in Korea. Ninety-three subjects(male : n=37, female : n=56) participated in this study. The subjects were grouped by serum insulin concentrations into two groups-hyperinsulinemia group(n=17) and normoinsulinemia group(n=76). Anthropometric measurements, blood lipid profiles, blood pressure, oral glucose tolerance test, and daily nutrient intakes were analyzed. And serum glucose and insulin secretion pattern as shown through and oral glucose tolerance test were performed. BMI (p<0.05) and WHR(p<0.01) were significantly higher in hyperinsulinemic group than in normoinsulinemia group. There was no differences in fasting blood glucose level between groups, but total glucose area (p<0.001) and insulin glucose resistance(p<0.001) were significantly higher in hyperinsulinemia group than in normoinsulinemia higher in hyperinsulinemia group, but HDL-cholesterol(p<0.05), and systolic blood pressure(p<0.05) were significantly higher in hyperinsulinemia group, but HDL-cholesterol(p<0.001) was significantly lower in hyperinsulinemia group. They showed significant differences in energy, carbohydrate, potassium, riboflavin, niacin and dietary fiber intakes(p<0.05), the intake of those nutrients were low in normoinsulinemia group. Further investigation is necessary to determine the effects of amounts and types of carbohydrate and dietary fiber on serum insulin concentrations.

실험적 당뇨쥐의 인슐린 치료시 타우린과 베타알라닌이 혈당 및 혈중 지질대사에 미치는 영향 (The Effects of Taurine and $\beta$-alanine on Blood Glucose and Blood Lipid Concentrations in Insulin-treated Diabetic Rats)

  • The purpose of this study was to determine the effects of taurine supplementation and taurine depletion on blood glucose and blood lipid concentrations in insulin-treated diabetic rats. Four groups of Sprague-Dawley male rats were fed the purified diet for 3 weeks ; nontaurine-supplemented diabetic rats(E0), nontaurine-supplemented diabetic rats with insulin treatment(E0+I), 1% taurine-supplemented diabetic rats with insulin treatment(E1+I) and taurine-depleted diabetic rats with insulin treatment(EA+I). Diabetes was induced by streptozotocin injection(50mg/kg B.W.). Isophane insulin was given subcutaneously into the abdominal wall of the diabetic rats(4 unit/rat/day). E1+I were supplemented with 1% taurine in drinking water. To induce taurine depletion, EA+I were treated with 5% $\beta$-alanine in drinking water. E1+I had significantly higher body weight compared to that of E0. The food intakes of E1+I and E0+I were significantly decreased compared to that of E0. There was no sigfniciant difference in food intake between E1+I and E0+I. The water intake of rats was significantly different among the groups ; E0>E0+I>E1+I>EA+I. The urine volume of E0 was significantly increased compared to those of insulin-treated goups. The blood glucose concentration of E0 was significantly increased compared to those of insulin-treated groups. In the oral glucose tolerance test(OGTT), E0+I and E1+I had significantly lower blood blucose concentrations compared to E0 after 30 min. Also EA+I had significantly lower bloodglucose concentrtion compared to E0 and E0+I. The plasma total cholesterol and LDL-cholesterol concentratons of EA+I were significantly incrased compared to those of other groups. Therefore, it may be suggested that tuarine supplementation is useful for insulin-dependent diabetes in order to prevent diabetic complications suchas cardiac vascular diseases.

  • Objectives Although antipsychotic drug clozapine has superior efficacy, this is hampered by metabolic side effects such as weight gain and diabetes. Recent studies demonstrate that clozapine induces insulin resistance. However, the identity and location of insulin resistance induced by clozapine has not been clarified. In this study, the effect of clozapine on central insulin response was investigated in rats. Methods Male Sprague-Dawley rats received intraperitoneal injection of clozapine or vehicle, which was followed by intracerebroventricular injection of insulin or its vehicle. The effects of clozapine on insulin-induced changes in blood glucose level and Akt phosphorylation in hypothalamus were investigated. Results Intraperitoneal injection of clozapine (20 mg/kg) increased blood glucose in rats. Intracerebroventricular injection of insulin reduced blood glucose in rats, which was blunted by pretreatment of clozapine. Accompanied with the antagonistic effect of clozapine to central insulin action in terms of blood glucose, clozapine inhibited the insulin-induced phosphorylation of Akt at Ser473 in rat hypothalamus. Conclusion Administration of clozapine inhibited the central insulin-induced changes in blood glucose and Akt phosphorylation in rat hypothalamus. These findings suggest that hypothalamus could be the site of action for the clozapine-induced insulin resistance.

정상인과 당뇨병 환자에 있어서 Maltitol 경구 투여가 혈당과 혈액내 insulin농도 변화에 미치는 영향 (Effect of Maltitol on Blood Glucose and Insulin Responses in Normal and Diabetic Subjects)

  • This study was an attempt to investigate the usefulness of maltitol as an alternative sweetener. The acute effects of oral ingestion of 50g of maltitol or glucose on blood glucose and insulin levels following test dose were investigated by using five healthy normal subjects and ten diabetic patients. The data demonstrated marked differences between the utilization of maltitol and of glucose in both groups. Blood glucose and insulin responses to glucose were significantly greater than to maltitol in normal subjects(p<0.05). In diabetic patients, the peaks of the mean increment in blood glucose concentration after glucose and maltitol were reached at 60 minutes with mean values of 135mg/dl and 49mg/dl, respectively, and these differences were statistically significant(p<0.001). As for blood insulin responses in diabetic patients, the peak of the mean increment after glucose was 25.03$\mu$U/ml at 120 minutes. In contrast insulin responses to maltitol were significantly lower than to glucose(p<0.05), and the peak value was 7.98$\mu$u/ml at 60min. From these results it can be concluded that ingestion of maltitol resulted in significantly lower blood glucose and insulin increments than did glucose in both normal and diabetic patients.

비만 남자 대학생의 비만 유형에 따른 혈중 지질, 인슐린 농도 및 영양소섭취량 비교 연구 (Comparison of the Blood Lipid , Insulin and Nutrients Intake by Fat Distribution of Obese Male in Korea)

  • This study was intended to figure out the difference of the blood lipids, insulin and nutrients intake by fat distribution among the obese young male in Korean. Twenty-eight male college students participated in this study, whose to find out body fat distributions were classified on the basis of Waist /hip ratio (WHR) into three groups-upper body type (UBTM), intermediate body type(IBTM) and lower body type (LBTM). To conduct this study, anthropometric measurements , blood pressure, serum lipids and insulin levels, and daily nutrients intake were analized . The results are as follows : The three body type groups showed significant differences each other in waist circumference(p<0.05), WTR(p<0.001) and body fat percentage (p<0.05). Also they showed significant difference each other in the daily construction in the daily energy intake. But they did not show significant differences in the blood pressure, serum lipids and insulin levels. From the above results, we could concluded that the body fat distribution in obese the young men could affect nutrients intake pattern.

Secreted decoy of insulin receptor is required for blood-brain and blood-retina barrier integrity in Drosophila

  • Glial cells play important roles during neurogenesis and in maintaining complex functions of the nervous system. Here, we report the characterization of a gene, Sdr, which contains a putative insulin-like growth factor receptor domain and is required to maintain critical nervous system functions in Drosophila. Sdr is expressed in glial cells during embryonic and larval stages of development, but its role in adult flies is poorly understood. As insulin signaling is important throughout the lifespan in human, we investigated the Sdr's role in adult flies. Our results demonstrate that Sdr is expressed on surface glial cells that surround the nervous system. Mutation of Sdr did not affect development but caused defects in locomotion and lifespan. Sdr mutants also showed increasingly severe defects in the blood-brain- and blood-retina-barriers as they aged. Therefore, we suggest a novel role of Sdr in maintaining the integrity of the blood-brain- and blood-retina-barriers in adult flies.

체위변경이 혈장 Insulin농도에 미치는 영향 (Effect of Postural Change on Plasma Insulin Concentration in Normal Volunteer)

  • The concentrations of some blood constituents are known to be influenced by the postural change. The blood glucose and insulin concentrations were measured, first, in the supine, and then (30 minutes later) in the erect positions under the Lasting state. The effects of a duretic, furosemide, were also studied under the same condition for 5 consecutive days. The materials were 5 healthy volunteers aging 20-29 years old with out any diabetic past, or family histories. The blood glucose was measured by the Nelson's method, and plasma insulin by the radioimmunoassay method. Following are the results; 1. The plasma insulin concentration in the erect position is slightly higher than in the supine position, however, the increase is statistically insignificant because of the notable individual variations in the values of the supine position. 2. Four cut of 5 cases show the increase of about 80% of plasma insulin in the erect position, which is statistically significant if analyzed on the basis of frequency distribution. 3. The blood glucose concentration showed no postural changes. 4. The increase of the plasma insulin concentration in the erect position seems to be the result of limited extra vasation of insulin in the lower extremities.

Blood glucose levels, insulin concentrations, and insulin resistance in healthy women and women with premenstrual syndrome: a comparative study

  • Objective: To compare the blood glucose levels, insulin concentrations, and insulin resistance during the two phases of the menstrual cycle between healthy women and patients with premenstrual syndrome (PMS). Methods: From January of 2011 to the August of 2012, a descriptive cross-sectional study was performed among students in the School of Medicine of Jahrom University of Medical Sciences. We included 30 students with the most severe symptoms of PMS and 30 age frequency-matched healthy controls. We analyzed the serum concentrations of glucose, insulin, and insulin resistance by using the glucose oxidase method, radioimmunometric assay, and homeostasis model assessment of insulin resistance equation, respectively. Results: No significant differences between the demographic data of the control and PMS groups were observed. The mean concentrations of glucose of the two study groups were significantly different during the follicular and luteal phases (p=0.011 vs. p<0.0001, respectively). The amounts of homeostasis model assessment of insulin resistance of the two study groups were significantly different in the luteal phase (p=0.0005). Conclusion: The level of blood glucose and insulin resistance was lower during the two phases of the menstrual cycle of the PMS group than that of the controls.

Effects of Multi-Extracts of Mori Folium and of Exercise on Plasma Insulin and Glucose Levels in Streptozotocin-Induced Diabetic Rats

  • This research was conducted to study the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on plasma insulin and glucose levels in streptozotocin (STZ)-induced diabetic rats. Eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for 10 weeks. The animal groups consisted of a normal-control group, a STZ-control group, 3 STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 along with exercise. In the normal-control group, glucose tolerance tests resulted in the peak blood glucose level being achieved in 15 minutes and a fasting blood glucose level being achieved in 60 minutes. In the STZ-control group, the peak blood glucose level was reached after 60 minutes and, even after 90 minutes, blood glucose shown at a significantly higher level compared to the fasting levels. In the groups supplemented with MF extracts, the blood glucose level peaked after 30 minutes of glucose challenge, and returned to the fasting level after 90 minutes; the MF-360 and MF-360+exercise groups showed the best levels of glucose tolerance. Blood glucose levels in the STZ-induced diabetic groups were significantly higher compared to the normal-control group. However, after 7 weeks of supplementation with MF extracts, a significant lowering of blood glucose levels was observed in all groups supplemented with the MF extract. The best effect was observed in the group given MF extract combined with exercise. Compared to the normal-control group, blood insulin levels were significantly lower in all STZ-induced diabetic groups; however, a significantly higher level of insulin was observed in the groups given MF extracts compared to the STZ-control group. This study shows that the supplementation of MF extracts in STZ-induced diabetic rats resulted in increased blood insulin levels and lower blood glucose levels.
