Preparation of Blood Glue from Porcine Plasma Protein and Cross-linking Reaction of Plasma Protein with Formaldehyde

  • Blood glue was prepared to reutilize porcine blood. Plasma proteins after lyophilization were treated by addition of wood flour, sodium hydroxide, sodium silicate, and hydrated lime to make blood glue with a suitable adhesivity. Characteristics of the prepared blood glue was monitored by measuring the viscosity with time, and the relationship between degree of hydrolysis of plasma proteins by addition of various amounts of sodium hydroxide and adhesivity was studied. To prevent the emission of formaldehyde during manufacturing of plywood by blood glue, the cross-linking reaction of plasma protein with formaldehyde was also examined. Fourier transform infrared, circular dichroism, and fluorescence spectroscopy study showed that blood plasma proteins react with formaldehyde, resulting in removal of formaldehyde by cross-linking reaction.

흉골 출혈에 대한 fibrin glue의 지혈 효과 (Hemostatic effect of fibrin glue for sternal marrow bleeding.)

  • Bleeding from bone marrow after sternotomy for open cardiac surgery can be sometimes difficult to control and even lead to reoperation for hemostasis. A clinical comparative study was carried out to demonstrate the hemostatic effect of fibrin glue [Beriplast] for sternal marrow bleeding after sternotomy for open heart surgery. Postoperative blood loss was measured in two patient groups, group A included 19 patients operated upon from June to October 1987 and the fibrin glue was applied to the sternal marrow together with collagen fleece and group B consisted of 22 patients from January to May 1987 and only collagen fleece was applied without fibrin glue. There was no difference between two groups in age and sex distributions, coagulation state, method of extracorporeal circulation and operative management. The blood loss one hour after operation was 2.04 ml/hr/kg in group A and 3.55 ml/hr/kg in group B [P<0.001]. The most significant difference was observed during the first 4 hours after surgery with 1.34 ml/hr/kg versus 2.05 ml/hr/kg. over the following 20 hours the amount of drainage from the chest tubes was identical in both groups. Fibrin glue reduces blood loss after open heart surgery by local hemostasis at sternum. Our study has shown that local application of fibrin glue to sternal marrow is an effective method of controlling the sternal bleedings. No side effect or complication of fibrin glue was noted.

Comparison of the Effects of Deer Antler, Old Antler, and Antler Glue on Osteoporosis in Ovariectomized Rats

  • Background: Examination of the effects of deer antler, old antler, and antler glue on postmenopausal osteoporosis in an ovariectomized Sprague-Dawley rat model. Methods: The study involved 7 experimental groups; SHAM (sham-operated rats), OVX (ovariectomized rats), E2 (ovariectomized rats with estradiol $10{\mu}g/kg$ daily, orally), DA (ovariectomized rats with deer antler extract 5.83 mg/kg), OA (ovariectomized rats with old antler extract 3.8 mg/kg), low-AG (ovariectomized rats with low dose of antler glue powder 12.5 mg/kg), high-AG (ovariectomized rats with high dose of antler glue powder 37.5 mg/kg). After 6 weeks of treatment, body weight, blood calcium, phosphorus, estradiol, liver [alkaline phosphatase (ALP), aspartate transaminase (AST), alanine transaminase (ALT)] and kidney [blood urea nitrogen (BUN)/creatinine ratio] function, and femoral bone mineral density (BMD) were measured. Results: The body weights of DA, OA, low-AG, and high-AG groups did not significantly differ from OVX group. Blood estradiol levels were significantly increased in the DA, low-AG, and high-AG groups compared to the OVX group. Blood calcium, phosphorus, ALP, AST, and ALT levels and BUN/creatinine ratio did not show significant changes in the DA, OA, low-AG, and high-AG groups. BMDs of the femur, and femoral head and neck were significantly increased in the low-AG group. In the OA group, the BMD of the femoral head and neck was significantly increased. Conclusion: Treatment with deer antler, or antler glue for 6 weeks was effective for increasing estradiol and femoral BMD in ovariectomized rats, suggesting that this may be of therapeutic benefit for osteoporosis.

$^{32}$P-postlabelling법을 이용한 유기용제 작업장 근로자의 유전독성 평가 (An Assessment of Genotoxicity on Organic Solvent Workers by $^{32}$P-postlabelling Method)

  • To evaluate the genotoxicities of workers exposed to glue and glue cleaning solution, ambient air monitoring of working place, animal study and human monitoring were carried out. By GC-MS analysis, air samples collected from shoesmaking plant were found to be toluene, xylene, cyclohexane, n-hexane, methyl ethyl ketone, trichloroethylene, butylacetate, isopropyl alcohol. Glue and glue cleaning solution from shoesmaking plant were applicated topically to the CD-1 mice. DNA was isolated from skin 24 hr following the application and analysed for DNA-adducts using the nuclease $P_1$version of $^{32}$P-postlabelling assay. RAL (Relative Adduct Labelling, adducts$10^8$ nucleotides) was significantly increased in a dose-dependent manner in the glue cleaning solution treated mice skin. Peripheral blood DNA-adducts of workers exposed to glue and glue cleaning solution were also analysed by the same method, but there were not significant differences in the peripheral blood DNA-adducts level between exposed and control workers. In addition, glue cleaning solution from shoes factory was evaluated for mutagenicity in the Salmonella plate incorporation assay using strains TA 100 and TA 1535 in the presence and absence of Arochlor 1254-induced rat liver S$_{9}$. There was evident mutagenicity for cleaning solution in TA 100 regardless of $S_9$, but TA 1535 showed positive only in the absence of $S_9$when predicted by Stead model of mutagenicity prediction (p=0.0000). The urine concentrates from workers and controls were also assayed for mutagenicity towards strain TA 100 of Salmonella typhimurium in the presence of $S_9$ using Kado's microsuspension assay, but their mutagenic activities were not found to be significant. These data suggest that shoesmaking workers are exposed to genotoxic compounds and need to be monitored by testing the mutagenicity of human urines. However, $^{32}$P-postlabelling application requires further validation for the routine monitoring of human exposure.osure.

비디오흉강경을 이용한 자연기흉의 치료시 국소적 Fibrin Glue 도포 (Surgical Treatment of Spontaneous Pneumothorax by Thoracoscopic Wedge Resection with Fibrin Glue)

  • Background ; To evaluate the efficacy of Fibrin glue to decrease recurrence in video-assisted thoracoscopic surgery(VATS) for a treatment of spontaneous penumothorax. Material and Method : All medical records of 17 patients who underwent a thoracoscopic wedge resections of bullae with stapling device with Fibrin glue in our institute between May 1998 and December 1999 were reviewed. variables analyzed include affected sites primary indication of VATS. duration from admission to discharge duration of postoperative stay duration of chest tube drainage recurrence and complication. There were 16 men and 1 woman. Result : There was no evidence of hemodynamic instability or arterial blood gas abnormalities encountered during the procedure. Mean age at the time of the VATS was 26.9 years (range 15 to 61 years) The mean duration from admission to discharge was 7.8 days and mean postoperative stay was 5.1days mean chest tube indwelling period was 4..0 days. There was no recurrence of pneumothorx. Conclusion : Thoracoscopic wedge resections with introduction of fibrin glue are safe and effective and requires only a short hospital stay. We believe that this thoracoscopic technique will further simplify the surgical treatment of pneumothorax.

개에서 자가 fibrin glue가 골 결손치유에 미치는 영향 (Effects of bone healing capacity by autologous fibrin glue in experimental bone defect dogs)

  • This study was investigated of the bone healing capacity by autologous fibrin glue in experimental bone defect dogs. The autologous fibrin glue manufactured just before the experiment was mixed with the concentrated fibrinogen from whole blood of the experimental dog and bovine thrombin. The experimental group was constituted with seven dogs. The experimental osteotomy was performed 5 mm length in bilateral region of proximal diaphyseal fibulae. The defected regions of experimental group were filled with the autologous fibrin glue by duploject. The experimental regions had been radiographed biweekly for 16 weeks to observe new bone formation and union. Bone alkaline phophatase (BALP) in all groups was evaluated biweekly till the end of the experiment to determine osteoblast activities. New bone formation had been observed in five regions of three dogs at four weeks after the experimental treatment and in two regions of one dog at ten weeks. The other seven regions of the experimental group and control group were not observed new bone formation until the end of the experiment. BALP value in four dogs observed new bone formation was increased to 97.10 IU/L (453.96%) at two weeks after the experimental treatment. The results of this experiment were suggested that the autologous fibrin glue was moderately effective in new bone formation in dogs.

사망사고 관련된 유해화학 물질 남용자들의 혈액 중 톨루엔과 부탄의 분석 (Analysis of Blood Toluene and Butane in Death Cases of Inhalant Abusers)

  • 혈액 중에서 톨루엔을 HS-SPME 법을 이용하여 GC/MSD로 분석하였으며, 부탄가스류의 분석은 HS 법을 이용하여 CC/FID로 분석하였다. 우리 나라의 경우 지난 3년 동안 청소년들이 본드류나 부탄가스류 등을 흡입한 상태에서 사망한 경우가 총 75건이 발생하였다. 본드류를 흡입하다가 사망한 사람은 27명이었으며, 그 중에서 9명은 톨루엔 중독상태에서 자제력의 상실 등으로 인하여 고층에서 떨어지는 추락사를 하였으며, 이때의 혈중 농도 분포는 $1.3{\sim}21.6{\mu}g/mL$으로 상당히 높은 농도였다. 또한 부탄가스를 흡입하다가 질식 사망 또는 흡입 사망한 사람은 54명이었으며, 이 중에는 본드류와 부탄가스를 동시에 흡입하다가 사망한 6명도 포함되었다.

  • 도축동물의 대부분 폐기되는 혈액으로부터 기능성 소재를 발굴하고 수질오염을 방지하기 위해서 소, 돼지 도축혈액으로부터 간단하고 신속한 방법으로 혈장 단백질을 분리하는 방법을 개발했고 그것을 혈분접착제로 응용하였다. 혈액을 원심분리한후 혈장단백질은 2% TCA 또는 0.6N HCl을 최종농도가 되게 첨가하여 침전시킴으로써 얻어졌다. 소, 돼지 혈장단백질을 비교 분석하기 위하여 SDA-PAGE를 하였고 얻어진 혈장단백질을 이용하여 혈분접착제로서 합판제조에 사용하여 다른 합성접착제와 접착성 시험을 한 결과 다른 합성 접착제와 비교해서 만족한 결과를 얻었다.

급성심근경색 후 발생한 좌심실벽 파열에서 소심낭과 Fibrin Glue 압박을 이용한 치험 - 1예 보고 - (Repair of Left Ventricular Free Wall Rupture after Acute Myocardial Infarction: Application of Pericardial Patch Covering and Fibrin Glue Compression A case report)

  • 배경: 급성심근경색 후 좌심실벽 파열은 높은 사망률을 보이는 심각한 합병증으로 보통 응급 수술이 유일한 치료법이다. 지속적인 흉통과 실신을 주소로 내원한 76세 여자 환자로 심초음파에서 심낭 삼출 및 좌심실 측하부의 수축 저하 소견을 보였고 관상동맥조영술에서 첫 사선지의 완전 폐쇄소견이 관찰되었다. 폐쇄된 사선지에 관상동맥성형술 및 스텐트 삽입, 그리고 대동맥내 풍선펌프 삽입 후 응급수술을 시행하였다. 체외순환 및 심정지하에 관상동맥우회술을 시행하고 좌심실벽 파열부위는 소 심낭으로 덮고 인조사로 연속 봉합하였으며 소 심낭과 심장외막 사이의 공간은 fibrin glue로 채운 후 지혈될 때까지 압박하였다. 급성심근경색 후 발생한 좌심실벽 파열을 치험하였기에 보고한다.

합판용 페놀수지 접착제에 대한 TCA침전 혈액분말의 첨가효과 (Adding Effect of TCA-Precipitated Blood Powder to the Phenol Formaldehyde Resin for Plywood)

  • To utilize the waste materials and develope wood adhesive from isolated bloods of slaughtered cow and pig and also to prevent water pollution, simple and rapid method of isolation and purification of plasma proteins from pig bloods with trichloroacetic acid(TCA) treatment was developed. Adding of TCA-precipitated blood powder to the phenol formaldehyde resin(PF) improved dry and wet strength of plywood and resulted in fast hot pressing times.
