Aerobic and Graduated Treadmill Exercise Decreases Blood Glucose Levels, Lipid Levels and Oxidative Stress in an Animal Model of Type 1 Diabetes Mellitus

  • Purpose: Exercise has been shown to be a simple and economical therapeutic modality that may be considered as an effective aid for diabetic mellitus. For example, exercise training increases insulin sensitivity in type 2 diabetes. But we found no reported of how exercise affect type 1 diabetes. This study investigated the impact of aerobic and graduated treadmill exercise regimens on body weight, glucose and insulin concentrations, lipid profiles, and oxidative stress indicators in rats with streptozotocin (STZ) induced diabetes. Glycosylated hemoglobin ($HbA_{1c}$) was determined as an indicator of glucose control during exercise. Methods: In our study, a total of 40 rats were used. Three groups of 10 rats each were given STZ to induce diabetes. The remaining 10 rats became the normal group. After 28 days we determined biochemical parameters such as glucose, glycosylated hemoglobin ($HbA_{1c}$), insulin concentration, serum total cholesterol (TC), triglycerides (TG), and high-density lipoprotein (HDL). Superoxide dismutase (SOD) and catalase activities were also measured. Results: Concentrations of blood glucose and $HbA_{1c}$ in the moderated exercise groups were significantly decreased after 28 days compared with the control group (p<0.05). There was a significant reduction in serum TC and TG in the experimental groups. The activity of SOD increased significantly by 17.70% and 48.25% respectively. Conclusion: These results indicate that physical training and exercise training affects body weight, fasting blood glucose, $HbA_{1c}$, insulin, lipid profiles, and antioxidant status in rats with streptozotocin-induced diabetes. We suggest that graduated treadmill exercise may have therapeutic, preventative, and protective effects against diabetes mellitusby improving glycemic control, oxidant defenses, and lipid metabolism.

  • Korean red ginseng has broad efficacious effects against hypertension, diabetes, nociception, and cancer, and it counteracts weakness. It has been reported that Korean red ginseng is able to normalize blood pressure, improve cholesterol and lower blood glucose levels. We have recently reported that Korean red ginseng extract (KRGE) significantly prevented rat carotid arterial thrombosis in vivo, and inhibited platelet aggregation ex vivo and in vitro in a dose-dependent manner. The purpose of this study was to examine the effects of KRGE on blood circulation in human by measuring ex vivo platelet aggregation, plasma coagulation and serum lipid profiles in healthy volunteers. Subjects were randomly divided into three groups (placebo-group, KRGE-low dose group, KRGE-high dose group). Administration of KRGE to subjects significantly inhibited ADP-induced platelet aggregations both in KRGE-low dose group from $72.79{\pm}20.53$ to $62.00{\pm}23.06%$ (p=0.0009), and in KRGE-high dose group from $75.14{\pm}21.86$ to $64.52{\pm}24.72%$ (p=0.0039), respectively. Administration of KRGE to subjects also significantly inhibited collagen-induced platelet aggregations both in KRGE-low dose group from $85.52{\pm}12.57$ to $79.62{\pm}20.47%$ (p=0.0916), and in KRGE-high dose group from $80.24{\pm}18.11$ to $70.31{\pm}25.93%$ (p=0.0565), respectively. Whereas, KRGE has no significant effects on coagulation system, such as prothrombin time (PT) and activated partial thromboplastin time (APTT), and serum lipid profiles, such as total cholesterol, low density lipoprotein cholesterol, high density lipoprotein cholesterol and triglyceride. KRGE also has no significant effects on hematological and serum biochemical profiles. These results suggest that KRGE has a potential to improve blood circulation through antiplatelet activity in human, and KRGE intake may be beneficial for the individuals with high risks of thrombotic and cardiovascular diseases.

Circular RNA expression profiles in the porcine liver of two distinct phenotype pig breeds

  • Objective: An experiment was conducted to identify and characterize the circular RNA expression and metabolic characteristics in the liver of Jinhua pigs and Landrace pigs. Methods: Three Jinhua pigs and three Landrace pigs respectively at 70-day were slaughtered to collect the liver tissue samples. Immediately after slaughter, blood samples were taken to detect serum biochemical indicators. Total RNA extracted from liver tissue samples were used to prepare the library and then sequence on HiSeq 2500. Bioinformatic methods were employed to analyze sequence data to identify the circRNAs and predict the potential roles of differentially expressed circRNAs between the two breeds. Results: Significant differences in physiological and biochemical traits were observed between growing Jinhua and Landrace pigs. We identified 84,864 circRNA candidates in two breeds and 366 circRNAs were detected as significantly differentially expressed. Their host genes are involved in lipid biosynthetic and metabolic processes according to the gene ontology analysis and associated with metabolic pathways. Conclusion: Our research represents the first description of circRNA profiles in the porcine liver from two divergent phenotype pigs. The predicted miRNA-circRNA interaction provides important basis for miRNA-circRNA relationships in the porcine liver. These data expand the repertories of porcine circRNA and are conducive to understanding the possible molecular mechanisms involved in miRNA and circRNA. Our study provides basic data for further research of the biological functions of circRNAs in the porcine liver.

Serum biochemical profiles of repeat breeder holstein friesian cows

  • The fertility of dairy cows has been declining worldwide. The number of services per conception has increased, and repeat breeder (RB) cows are considered important in the dairy industry. However, there has been little research on RB cows in Korea. The objective of this study was to investigate the relationship between serum biochemical profiles and RB Holstein cattle in Korea and compare the results with those of studies conducted abroad. In addition, we investigated hidden factors that are needed for RB cattle to become pregnant. Overall, 34 Korean Holstein Friesian cows were divided into three groups: pregnant with normal cycling (PNC), pregnant with repeat breeder (PRB), and non-pregnant with repeat breeder (NRB). Blood samples were collected from the jugular vein, and serum parameters (ALT, AST, ALP, GGT, LD, CK, TP, ALB, GLOB, TBIL, BUN, CRE, GLU, TC, TG, NEFA, CA, MG, and IP) were analyzed. GGT levels were significantly different among groups (P=0.01). The concentration of GGT was the highest in the NRB and the lowest in the PNC. In contrast to the findings of other studies, there were no differences in GLU, TP, TC, and BUN levels. This study is the first to investigate serum biochemistry in RB cattle and provides results that differ from those of previous studies; these findings would help establish a novel approach to improve fertility of RB cattle.

Characterization of Aeromonas hydrophila Isolated from Rainbow Trouts in Korea

  • Eight strains of Aeromonas hydrophila isolated from diseased trout in Korea were characterized and compared with an American type strain by various methods including biochemical and physiological tests, PCR, randomly amplified polymorphic DNA (RAPD), plasmid profiling, and gel electrophoresis of total, membrane, and extracellular proteins. Virulence factors such as surface array proteins, cytotoxin, hemolysin, haemagglutinin, and protease were also investigated. The Korean strains showed heterogeneity in Iysine decarboxylase production, utilization of various carbon sources, and production of acetoin. Five strains had the same profiles of total and membrane proteins. Six strains haemagglutinated with trout red blood cells (RBCs) which was inhibited by fucose, galactose, and mannose, except for No. 1 where haemagglutination was inhibited by only galactose and mannose, but not by fucose. Four isolates haemagglutinated with human RBCs which was inhibited by fucose and mannose yet not by galactose. The type strain haemagglutinated only with trout RBCs which was inhibited by fucose, galactose, and mannose. Every isolate secreted protease, hemolysin, cytotoxin, and siderophore, but no enterotoxin. Results showed that the Korean isolates, except for No.7, had very different biochemical and molecular characteristics from those of the American type strain.

Effect of Antioxidants on Physio-biochemical and Hematological Parameters in Broiler Chicken at High Altitude

  • The present study was carried out on broilers to study the effect of oral administration of vitamin E and selenium (E-care Se) on growth performance, haematological and biochemical parameters for a period of 42 days (6 weeks). A total of 90 oneday-old broiler chicks were divided into three equal groups: $T_1$, $T_2$ and $T_3$. Group T1 was maintained as control and was fed only with the basal diet throughout the experimental period. Two experimental diets, $T_2$ and $T_3$, were formulated to contain an additional 100 g (150 IU vitamin E/kg+0.5 mg Se/kg) and 200 g (300 IU vitamin E/kg+1.0 mg Se/kg) of E-care Se which was the source of vitamin E and selenium. Body weight was significantly (p<0.05) higher in antioxidant-treated groups compared to the control group. There were no significant differences in feed conversion ratio (FCR). Blood samples were collected from the jugular vein for haematological (TEC, Hb, PCV and ESR) and biochemical (GOT and GPT) study. Body weight was increased significantly in both treated groups compared with the control group and highest body weights were recorded in group $T_2$. TEC, PCV and Hb content increased significantly (p<0.01) in the treated groups as compared to the control group, but ESR, GOT and GPT values decreased significantly (p<0.01) in both treated groups as compared to the control group. The result reveals that use of antioxidants (vitamin E and selenium) is an effective way of getting the best result in terms of body weight gain and haemato-biochemical profiles in broiler birds at high altitude.

Effect of fermented biogas residue on growth performance, serum biochemical parameters, and meat quality in pigs

  • Objective: This study investigated the effect of fermented biogas residue (FBR) of wheat on the performance, serum biochemical parameters, and meat quality in pigs. Methods: We selected 128 pigs (the mean initial body weight was $40.24{\pm}3.08kg$) and randomly allocated them to 4 groups (1 control group and 3 treatment groups) with 4 replicates per group and 8 pigs per pen in a randomized complete block design based on initial body weight and sex. The control group received a corn-soybean meal-based diet, the treatment group fed diets containing 5%, 10%, and 15% FBR, respectively (abbreviated as FBR5, FBR10, and FBR15, respectively). Every group received equivalent-energy and nitrogen diets. The test lasted 60 days and was divided into early and late stages. Blood and carcass samples were obtained on 60 d. Meat quality was collected from two pigs per pen. Results: During the late stage, the average daily feed intake and average daily gain of the treatment groups was greater than that of the control group (p<0.05). During the entire experiment, the average daily gain of the treatment groups was higher than that of the control group (p<0.05). Fermented biomass residue did not significantly affect serum biochemical parameters or meat quality, but did affect amino acid profiles in pork. The contents of Asp, Arg, Tyr, Phe, Leu, Thr, Ser, Lys, Pro, Ala, essential amino acids, non-essential amino acids, and total amino acids in pork of FBR5 and FBR10 were greater than those of the control group (p<0.05). Conclusion: These combined results suggest that feeding FBR could increase the average daily gain and average daily feed intake in pigs and the content of several flavor-promoting amino acids.

다낭성 난소 증후군 환자에서 Isoflavone 섭취에 따른 혈중 생화학적 지표의 변화 (The Effect of Isoflavone Supplementation on Plasma Biochemical Parameters of Women with Polycystic Ovary Syndrome)

  • Objective: This study was performed to evaluate the effect of isoflavone supplementation on hormone levels, lipid profiles and total antioxidant status in patients with polycystic ovary syndrome. Methods: Total 11 women with polycystic ovary syndrome were supplemented daily with 150mg of isoflavone for 6 months. Blood samples were collected 0, 3, and 6 months after supplement of isoflavone for analysis of LH, FSH, E2, testosterone, free testosterone, SHBG levels, serum lipid profiles, and total antioxidant status (TAS). Results: After 6 months isoflavone supplementation, the hormone levels did not change significantly. Serum lipid profiles did not show any significant change in total cholesterol, LDL-cholesterol, triglyceride, lipoprotein(a), and free fatty acid levels. However, there was significant increase in HDL-cholesterol (p<0.05) for 3 months. Total antioxidant status was increased significantly after isoflavone supplementation for 6 months (p<0.05). Conclusion: Isoflavone supplementation showed positive effects on the HDL-cholesterol and total antioxidant status. it is implicated that isoflavone supplementation will may have a effect on cardiovascular disease in patients with polycystic ovary syndrome.

인삼 부산물의 첨가 급여가 고온 스트레스 하 육계의 생산성, 혈액조성, 장기발달 및 스트레스 지표에 미치는 영향 (Effects of Ginseng By-Products Supplementation on Performance, Blood Biochemical Profiles, Organ Development, and Stress Parameter in Broiler under Heat Stress Condition)

  • 본 연구는 인삼 부산물 첨가 급여가 고온에서 사육된 육계의 생산성, 장기발달, 혈액생화학적 특성, 면역반응 및 스트레스 지표에 미치는 영향을 구명하기 위해 실시하였다. 총 100수의 1일령 수컷 병아리(Ross 308)를 공시하여 5주간 육계용 실험 케이지에서 사육하였다. 첫 일주일간은 동일한 초기사료를 급여하였고, 2주차부터 대조구(CON), 진생베리 0.5% 첨가(GB1), 진생베리 1.0% 첨가(GB2), 인삼 잎·줄기 0.5% 첨가(GLS1), 인삼 잎·줄기 1.0% 첨가(GLS2), 총 5개의 시험구에 임의배치 하였다. 환경 온도는 매일 오전 9시부터 오후 5시까지 32±1℃, 이후 시간은 28±1℃를 유지하였고, 사료와 물은 자유채식 시켰다. 생산성은 처리구들 간에 유의한 차이를 보이지 않았으나, 사육후기(21~35일)와 전 기간(7~35일)에는 증체량이 모든 첨가급여구들에서 CON보다 높은 경향을 보였다. 혈액 생화학적 특성은 빌리루빈 수치를 제외하고는 시험구들 간에 유의한 차이가 없었다. 혈청 면역글로불린과 코르티코스테론 수치는 시험구들 간에 유의차가 나타나지 않았으나, 코르티코스테론 함량이 CON보다 첨가급여구들에서 첨가수준에 따라 감소하는 경향이 나타났다. 결론적으로, 인삼 부산물의 사료 내 첨가급여는 육계의 생산성 및 각종 생리지표에 부정적인 영향을 미치지 않으면서 성장기 육계의 고온 스트레스 저감에 긍정적인 효과가 기대되어 향후 지속적인 관심과 후속 연구가 필요할 것으로 사료된다.

일 대학교 교직원의 생활습관과 체질량지수 및 생화학적 지수에 관한 연구 (One University Staff Members' Life Styles, Body Mass Indices, Lipid Profiles and Plasma Glucose Levels)

  • Purpose: To analyze the differences of the Body Mass Index (BMI), blood biochemical indices (TC, TG, HDL, PP2) among college faculty members depending on their life styles and thereupon, provide for some basic data useful for healthcare education. Methods: 163 faculty members were sampled for a questionnaire survey and a medical checkup, both conducted from Jan. 20 to Jan. 26, 2010. Results: First, such blood biochemical indicesas BMI, TG, HDL and PP2 differed significantly depending on gender, while TC, TG and HDL differed significantly depending on age. Second, BMI differed significantly depending on drinking. Third, smokers showed significantly lower BMI and PP2 than non-smokers. Among the smokers, those smoking for 1-10 years showed a significantly lower level of TG. Fourth, those exercising as hard as sweating 3 times a week showed significantly lower BMI, TC and TG. Fifth, BMI was correlated positively with TC and TG, while being correlated negatively with HDL. On the other hand, TC was correlated positively with the TG which was correlated negatively with the HDL which was correlated positively with PP2. Conclusion: In order to prevent chronic diseases and live a healthy life, it must be necessary to control drinking, stop smoking and exercise regularly.
