Effect of S100A8 and S100A9 on expressions of cytokine and skin barrier protein in human keratinocytes

  • Atopic dermatitis (AD ) is an inflammatory skin disorder caused by immunological dysregulation and genetic factors. Whether the expression levels of cytokine and skin barrier protein were altered by S100 calcium binding protein A8 (S100A8) and S100A9 in human keratinocytic HaCaT cells was examined in the present study. Alterations of cytokine expression were examined by ELI SA following treatment with S100A8/9 and various signal protein-specific inhibitors. Activation of the mitogen activated protein kinase (MAPK) pathway and nuclear factor (NF)-κB was evaluated by using western blotting and an NF-κB activity test, respectively. The expression levels of interleukin (IL )-6, IL- 8 and monocyte chemoattractant protein-1 increased following treatment with S100A8 and S100A9, and the increase was significantly blocked by specific signaling pathway inhibitors, including toll-like receptor 4 inhibitor (TLR 4i), rottlerin, PD98059, SB203580 and BAY-11-7085. Extracellular signal-regulated kinase (ER K) and p38 MAPK pathways were activated in a time-dependent manner following treatment with S100A8 and S100A9. Phosphorylation of ER K and p38 MAPK were blocked by TLR 4i and rottlerin. S100A8 and S100A9 induced translocation of NF-κB in a time-dependent manner, and the activation of NF-κB was inhibited by TLR 4i, rottlerin, PD98059 and SB203580. In addition, S100A8 and S100A9 decreased the expression of skin barrier proteins, filaggrin and loricrin. These results may help to elucidate the pathogenic mechanisms of AD and develop clinical strategies for controlling AD.

Anti-stress Effect of Scutellatia baicalensis in SD Rats and ICR Mice

  • The aim of this study is to investigate anti-stress effect of Scutellaria baicalensis(SB). The experiments were performed with the use of young (9 weeks of age) male rats of SD strain and the male ICR mice (20-25 g) at the time of first treatment with SB. Animals of the normal group were not exposed to any stress and the control group were exposed to stress. The rats of the Ginseng, Diazepam(BZ) and SB supplementary group were orally administered once a day 100 mg of red ginseng extract, 5 mg of BZ or 100 mg of SB extract/kg body weight and they were exposed to stress. The mice of the Ginseng, BZ and SB supplementary group were given water containing 200 mg of red ginseng extract, 10 mg of BZ or SB extract/100 ml potable water and exposed to stress. Animals were given supplements for 7 days without stress, and then were given supplement for 5 days with restraining and electroshock stress. We recorded stress related behavioral changes of the experimental animals by stressing them using the Etho-vision system and measured levels of blood corticosterone and IL-2. SB supplementation partially blocked the stress effect on locomotion in the rats and mice, and also partially blocked stress-induced behavioral changes such as freezing, burrowing, grooming, smelling, and rearing behavior in the rats and smelling, grooming, tailing, and rearing in the mice. in elevated plus maze test, the staying time of the stressed rats and mice in the open area decreased while it increased in the closed area. But these changes also partially were blocked by SB-supplementation. SB-supplementation decreased levels of the blood corticosterone which was increased by stress in the rats but did not significantly increase levels of blood interleukin 2 which was decreased by stress in mice.

  • In this paper, a nonhierarchial integrated information network with circuit switched and packet switched traffic is considered. It is assumed that circuit switched traffic is allowed to attempt an alternate path if the direct path is blocked, whereas packet switched traffic is not. The network uses a movable boundary scheme to allocate bandwidth dynamically. To analyze the performance of this type of network, EEBP(End to End Blocking Probability) is selected as a measure for circuit switched traffic and average time delay for packet switched traffic, respectively. EEBP and average time delay are derived analytically. Using the two proposed measure, the performance of the network under various bandwidth allocations and arrival patterns are observed. Moreover, the arrival rate of one link for circuit switched traffic is obtained from an approximation formula. Simulation results reveal the validity of the proposed approximation method.

  • The insertion of the crowbar system in the doubly fed induction generator rotor circuit for a short period of time during grid disturbance enables a more efficient way of limiting transient rotor current and hence protecting the rotor side converter (RSC) and the DC - link capacitor. When crowbar is activated at fault occurrence and clearance time, RSC is blocked while DC -link capacitor and the grid side converter (GSC) can be controlled to provide reactive power support at the PCC and improve the voltage which helps to comply with grid codes. In this paper, control strategies for crowbar system to limit the rotor current during fault is presented with RSC and GSC controllers are modified to control PCC voltage during disturbance to enhance DFIG wind farm to comply with some strict grid codes. Model simulated on MATLAB/Simulink verify the study through simulation results presented.

Association of Anti-apoptotic Mechanism Due to House Dust Mite in Neutrophils with Protein Synthesis and Bad

  • House dust mite is an essential allergen in the pathogenesis of allergic diseases. Abnormal regulation of neutrophil apoptosis is an important pathogenic process in allergic diseases. In the present study, we investigated the effects of house dust mites on spontaneous apoptosis of neutrophils and its associated mechanisms. Extract of Dermatophagoides pteronissinus (DP) inhibited neutrophil apoptosis in a time-dependent manner. Cycloheximide (CHX), an inhibitor of translation, increased apoptosis of DP-treated neutrophils as well as control cells. The pro-apoptotic effect of CHX was blocked by DP in neutrophils. In addition, DP increased the phosphorylation of Bad in a time-dependent manner, indicating that it exerted an inhibitory effect on the function of Bad. These results suggest that DP has anti-apoptotic effects of neutrophils and may regulate protein synthesis and activation of Bad. Moreover, these findings may shed light on elucidation of allergy pathogenesis due to house dust mites.

Neuroprotection of Lithium is Associated with Inhibition of Bax Expression and Caspase 8 Activation

  • Neuroprotective properties of lithium were investigated by using in vivo NMDA excitotoxicity model. The appearance of TUNEL positive cells was prominent within 24 h of NMDA (70 mg/kg, i.p.) injection in the regions of the cortex, hippocampal formation, and thalamus of mouse cerebrum. NMDA treatment resulted in the extensive enhancement of Bax immunoreactivity in the cortical and hippocampal regions. NMDA also increased the immunoreactivity of caspase 8 in the similar regions of the mouse cerebrum. However, the increased immunoreactivity of Bax and caspase 8 were dramatically attenuated by chronic lithium pretreatment (lithium chloride, 300 mg/kg/d, i.p. for $7{\sim}10$ days). At the same time, lithium ion blocked the appearance of TUNEL positive cells, and the morphological assessment indicated an effective neuroprotection by lithium against NMDA excitotoxicity. Although the exact action mechanism of lithium is not straightforward at this time, we propose that the inhibition of Bax and caspase cascade is involved in the neuroprotective action of lithium.

A Study on Securing Safety for High Rise Building Fires - Applying Active Fire Escape Systems -

  • Under the Korean Enforcement Decree of the Building Act, all high story apartment houses more than 5 stories high are mandated to install a fire evacuation system to ensure safe evacuation from fire accidents and providing quick and easy bidirectional escape route when main entrance is blocked by flame or toxic smoke. However, the current fire evacuation system shows a lack of understanding from residents and thus is widely ignored for having insufficient safety functions, especially vis-à-vis fire emergencies. Studies have found that an alternative evacuation method, the escapable fire evacuation system, has been analyzed for safety evaluation compared with the conventional passive fire escape system and can bring efficient and safer solutions, providing high rise residents escape from fire accidents. Evaluation for safety evacuation has been performed by the Fire Dynamics Simulation and applying Pathfinder simulation. This resulted in providing appropriate escape routes within the safety escape time and allowed for people in high rise building fires to get to safety.

  • In this Paper, an adaptive microphone array system with self-delay estimator is proposed. By showing that the adaptive blocking matrix (ABM) of the generalized sidelobe canceller (GSC) can estimate the relative time delay between each sensor, the proposed system utilizes the ABM not only for blocking target components in the blocked signal path, but also for estimating the relative time delay. Therefore, the proposed system requires only the GSC structure while maintaining the system performance similar to the conventional system using an additional time delay estimator as a preprocessor. Simulation results show that the performance of the proposed system is identical to the conventional system that uses an additional time delay estimation module.

Reactive Oxygen Species Mediates Lysophosphatidic Acid-induced Migration of SKOV-3 Ovarian Cancer Cells (SKOV-3 난소암 세포주에서 lysophosphatidic acid 유도 세포의 이동에 있어 활성산소의 역할)

  • Cell motility plays an essential role in many physiological responses, such as development, immune reaction, and angiogenesis. In the present study, we showed that lysophosphatidic acid (LPA) modulates cancer cell migration by regulation of generation of reactive oxygen species (ROS). Stimulation of SKOV-3 ovarian cancer cells with LPA strongly promoted migration. but this migration was completely blocked by pharmacological inhibition of phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway. Inhibition of the ERK pathway had no effect on migration. Stimulation of SKOV-3 ovarian cancer cells with LPA significantly induced the generation of ROS in a time-dependent manner. LPA-induced generation of ROS was significantly blocked by pharmacological inhibition of PI3K or Akt, but inhibition of the ERK signaling pathway had little effect. LPA-induced generation of ROS was blocked by pretreatment of SKOV-3 ovarian cancer cells with an NADPH oxidase inhibitor, whereas inhibition of xanthine oxidase, cyclooxygenase, or mitochondrial respiratory chain complex I had no effect. Scavenging of ROS by N-acetylcysteine completely blocked LPA-induced migration of SKOV-3 ovarian cancer cells. Inhibition of NADPH oxidase blocked LPA-induced migration whereas inhibition of xanthine oxidase, cyclooxygenase, or mitochondrial respiratory chain complex I did not affect LPA-induced migration of SKOV-3 ovarian cancer cells. Given these results, we suggest that LPA induces ROS generation through the PI3K/Akt/NADPH oxidase signaling axis, thereby regulating cancer cell migration.

Fixed Time Synchronous IPC in Zephyr Kernel (Zephyr 커널에서 고정 시간 동기식 IPC 구현)

  • Linux Foundation has announced a real-time kernel, called Zephyr, for IoT applications recently. Zephyr kernel provides synchronous and asynchronous IPC for data communication between threads. Synchronous IPC is useful for programming multi-threads that need to be executed synchronously, since the sender thread is blocked until the data is delivered to the receiver thread and the completion of data transfer can be known to two threads. In general, 'IPC execution time' is defined as the time duration between the sender thread sends data and the receiver thread receives the data sent. Especially, it is important that 'IPC execution time' in the synchronous IPC should be fixed in real-time kernel like Zephyr. However, we have found that the execution time of the synchronous IPC in Zephyr kernel increases in proportion to the number of threads executing in the kernel. In this paper, we propose a method to implement a fixed time synchronous IPC in Zephyr kernel using Direct Thread Switching(DTS) technique. Using the technique, the receiver thread executes directly after the sender thread sends a data during the remaining time slice of the sender thread and we can archive a fixed IPC execution time even when the number of threads executing in the kernel increases. In this paper, we implemented synchronous IPC using DTS in the Zephyr kernel and found the IPC execution time of the IPC is always 389 cycle that is relatively small and fixed.
