Dietary inclusion effect of yacon, ginger, and blueberry on growth, body composition, and disease resistance of juvenile black rockfish (Sebastes schlegeli) against Vibrio anguillarum

  • Background: To minimize the use of antibiotics and to obtain a more sustainable fish culture and aquaculture industry, development of alternative natural source of immunostimulant to replace antibiotic in aquafeed is highly needed. Objective: Dietary inclusion effect of yacon (YC), ginger (GG), and blueberry (BB) on growth, body composition, and disease resistance of black rockfish against Vibrio anguillarum was compared to ethoxyquin (EQ). Methods: Four hundred eighty juvenile (an initial weight of 4.2 g) fish were randomly distributed into 12 of 50 L flowthrough tanks (forty fish per tank). Four experimental diets were prepared; the control (Con) diet with 0.01% EQ inclusion, and YC, GG, and BB diets at 1% each additive inclusion. Each additive was included into the experimental diets at the expense of wheat flour. Each diet was assigned to triplicate tanks of fish and hand-fed to satiation twice daily for 8 weeks. At the end of 8-week feeding trial, 20 fish from each tank fish were artificially infected by intraperitoneal injection with 0.1 mL of culture suspension of pathogenic V. anguillarum containing 3.3 × 106 cfu/mL respectively. Fish were monitored for the following 8 days after V. anguillarum infection and dead fish were removed every 6 h for the first 4 days and 12 h for the rest of the study. Results: Weight gain, specific growth rate (SGR), and feed efficiency ratio (FER) of fish fed the YC diet was higher than those of fish fed all other diets. However, feed consumption, protein efficiency ratio, and protein retention was not affected by dietary additive. Moisture, crude protein, and crude lipid content of the whole body of fish were affected by dietary additive. Analysis of the Kaplan-Meier survival curves showed that survival of fish fed the YC, BB, and GG diets was higher than the Con diet. Conclusion: Oral administration of YC can improve not only weight gain, SGR, and FER of black rockfish, but lower mortality of rockfish at occurrence of V. anguillarum.

  • Experiments were performed to investigate changes in metabolic rate (MO2), critical oxygen saturation (Scrit) and hematological parameters of black rockfish, Sebastes schlegeli exposed to hypoxia at 15, 20 and 25℃. The MO2 was measured at an interval of 10 min using intermittent-flow respirometry. The normoxic standard metabolic rate (SMR) was 116.5±5.5, 188.6±4.2 and 237.4±6.8 mg O2/kg/hr, and Scrit was 22.1±1.2, 30.6±1.5 and 41.9±1.4% air saturation at 15, 20 and 25℃, respectively. Q10 values were 2.62 between 15 and 20℃, 1.58 between 20 and 25℃, and 2.04 over the full temperature range. In the investigation of blood (hematocrit and hemoglobin) and biochemical parameters (plasma cortisol, glucose, electrolyte and osmolality), the rockfish were subjected to Scrit for each temperature during 4 hr. All of hematological parameters of the rockfish exposed to hypoxic water were significantly higher than those of normoxic control. Moreover, blood and biochemical parameters of the rockfish maintained to normoxic water showed the tendency of increase with temperature, and were significantly higher at 25C. As a result of this experiment, it was found that physiological stress due to hypoxia increased at high temperature.

  • The single anatomical attribute that has the greatest influence on acoustic scattering from fish is the presence or absence of a swimbladder. This study examined the effect of removing the gas from the swimbladder on the broadband backscattering characteristics of six species of swimbladdered fish: striped beakperch Oplegnathus fasciatus, black scraper Thamnaconus modestus, dark-banded rockfish Sebastes inermis, goldeye rockfish Sebastes thompsoni, black rockfish Sebastes schlegeli and panther puffer Takifugu pardalis. Before and after removing the gas from the swimbladder, the species-specific, frequency-dependent backscattered echo signals from anaesthetized individuals of each fish species were measured at approximately 1° intervals spanning a 90° aspect angle range from -45° (head down) to +45° (head up) using a broadband echo sounder operating at 100-200 kHz. The relationship between the wavelength-normalized backscattering cross section (σ/λ2) and fish length (L/λ) was calculated for each species. The average σ/λ2 value for the six fish species at a L/λ range of 19.79-25.85, with a mean of 22.89, was reduced by approximately 52.3% when the gas was removed.

Residues of Fluoroquinolones in Olive Flounder (Paralichthys olivaceus) and Black Rockfish (Sebastes schlegell) Following Oral Administration (Fluoroquinolone계 항생제의 경구투여에 따른 양식넙치 및 조피볼락의 체내 잔류량의 변화)

  • The residues of fluoroquinolones in Olive flounder (Paralichthys olivaceus) and Black rockfish (Sebastes schlegeli) were investigated after oral doses of 20 mg/kg for 30 days. Blood samples were taken at 5 hours, 10 hours, 1 day, 2 days, 3 days, 5 days, 7 days, 9 days, 13 days, 20 days and 30 days after treatment. The concentrations of fluoroquinolones were detected by high performance liquid chromatography using a UV detector. The recovery rates of fluoroquinolones in fish samples ranged from 92.0-99.7%, 93.4-97.5% and 93.0-97.9% for treatment of 0.1, 0.5, and $1.0{\mu}g/g$, respectively. Ciprofloxacin, enrofloxacin, norfloxacin and sarafloxacin were detected in the blood of the olive flounder at 10 hours, 10 hours, 1 day and 10 hours after treatment with $6.43{\mu}g/g$, $6.07{\mu}g/g$, $11.83{\mu}g/g$ and $11.02{\mu}g/g$ as maximum level, respectively. Ciprofloxacin, enrofloxacin, norfloxacin and sarafloxacin were detected in the blood of black rockfish 1 day, 1 day, 1 day and 10 hours after treatment with $8.26{\mu}g/g$, $8.03{\mu}g/g$, $7.29{\mu}g/g$ and $9.38{\mu}g/g$ as maximum level, respectively. However, ciprofloxacin, enrofloxacin, norfloxacin and sarafloxacin were not detected in olive flounder samples at 20, 30, 20 and 20 days after treatment in any experiments (

Movement Ranges and Routes of Black Rockfish Sebastes schlegeli in Summer and Autumn from Acoustic Telemetry

  • The movement range and diurnal behavior of 17 wild black rockfish (Sebastes schlegeli) were monitored within the marine ranching area of Tongyeong from 27 July to 7 November, 2005 using acoustic telemetry. Coded transmitters were surgically implanted into the abdomen of fish. Two of ten fish released in summer moved about 2 km away from the capture point for 1 day. The others stayed within a 500-m radius of the release point for 1 week after release. The seven fish released in autumn stayed within 500 m of the release point. Most fish were more active and had a greater range of horizontal diurnal movement at night than during the day. Additionally, the range of horizontal diurnal variation was greater in summer than in autumn. While fish released in summer were more vertically active at night than during the day, those released in autumn preferred to move vertically during the day.

Characterization of Vibrio harveyi, the Causal Agent of Vibriosis in Cultured Marine Fishes in Korea

  • An epizootic causing mortality among cultured marine finfishes occurred in 1999 in the province of Kyoungsang, Korea. The disease was characterized by the presence of enterocele, abdominal swelling, and gastroenteritis. The causative bacteria were isolated from olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), turbot (Scophthalmus maximus) and the rearing water. These bacteria showed swarming activity on agar plates and yellowish or greenish colonies on thiosulfate-citrate-bile salts-sucrose (TCBS) agar plates, but no luminescence. The pathogen was identified as Vibrio harveyi based on morphological and biochemical characteristics and the sequence of l6S rDNA. The lethal doses (LD$_{50}$) of olive flounder and black rockfish were estimated to be $1.24\times10^6-1.36\times10^8$ and $3.24\times10^5-5.8\times10^7$ CFU/fish respectively following intraperitoneal injection.

An Economic Analysis of Black Rockfish, Sebastes schlegeli in the Marine Floating Cage Culture (조피볼락 해상가두리 양식업의 지역별·규모별 경제성 분석)

  • This study is aimed to analyze the economic performance of black rockfish aquaculture by standardizing cost structures by region and farming size. The result of survey on farming and sale condition in each region, stocking density, survival rate, juvenile price, and unit production was the highest at Yeosu and Tongyeong, Heuksando showed the lowest. While rearing period was the longest at Tongyeong, the shortest region was Yeosu and Wando. In farming cost structure by region, amount of feeing was the highest at Tongyeong and Yeosu, and the lowest was Heuksando. Cost of medicine was high in Wando and Taean region, Yeosu and Hecksando was low. In case of farm size, feed cost ratio was high in the order of medium(0.75ha), small(0.25ha) and large(1.25ha) size. Standard production cost at every farm size of Heuksando showed the lowest among these regions. Taean and Yeosu was middle, and Tongyeong and Wando was the highest. According to the income, profit rate and investment return of farm size in all regions, as the bigger farm size, the higher income and profit rate was revealed. However, in case of Wando, Taean, and Heuksando which regions has high investment return, medium farm size was higher than large size. The result of economic analysis according to various factors, economic feasibility of black rockfish aquaculture in marine floating cage was showed significant changes by rearing and market condition.

Antimicrobial Effects of Chemical Disinfectants on Fish Pathogenic Bacteria

  • This study was to examine the potential disinfection efficiencies of 10 compounds by determining their antimicrobial capacity and ichthyotoxicity. Antimicrobial effects against Vibrio sp., Edwadsiella tarda, Streptococcus sp., and Staphylococcus sp. were tested using 10 different disinfectants; hydrogen peroxide, sodium hypochlorite, chlorine dioxide, povidon iodine, formaldehyde, glutaraldehyde, quaternary ammonium compounds (QACs), didecyl dimethyl ammonium chloride (DDAC), ortho-dichlorobenzen, and copper sulfate. Chlorine dioxide ($ClO_2$) containing 5% $ClO_2$ and copper sulfate had no effects on bactericidal activity, while the other disinfectants resulted in 99.99% bactericidal activity against 4 strains of fish pathogenic bacteria. The ichthyotoxicity of the 10 disinfectants was investigated using 3 kinds of fish species; flounder (Paralichthys olivaceus), rockfish (Sebastes pachycephalus), and black sea bream (Acanthopagrus schlegelii). Median lethal concentration ($LC_{50}$) values of the 10 disinfectants were estimated to determine toxicity ranges of the doses within 24 hr. Among test disinfectant solutions, hydrogen peroxide showed the highest $LC_50$ in flounder (201.3), rockfish (269.7), and black sea bream (139.3 ppm). DDAC revealed the lowest $LC_{50}$ in flounder (2.1), rockfish (1.0), and black sea bream (1.5 ppm). These results suggest that DDAC, quaternary ammonium compounds, glutaraldehyde, and sodium hypochlorite are effective disinfectants for fish and bacterial species examined in this study.

  • Age and growth of black rockfish, Sebastes schlegeli sampled from the Tongyeong marine ranching area in Korea waters were determined from 1,173 otoliths from July, 2001 to May, 2004. Examination of outer margins of the otolith showed that the opaque zone was formed once a year. Marginal increment of the otolith formed annual ring from July. The von Bertalanffy growth curve had the growth parameters estimated from non-linear regression were $L_t=48.45(1-e^{-0.2139(t+0.4313)})$, $W_t=1,837.93(1-e^{-0.2139(t+0.4313)})^{3.02}$ 3.02 for females and, $L_t=49.32(1-e^{-0.1775(t+0.7403)})$, $W_t=1,887.83(1-e^{-0.1775(t+0.7403)})^3$ for males, where t is age (year) and Lt is body length (mm) at age t. Growth at the age of male and female shows significant difference (p<0.01).

A Study on the Selectivity of the Mesh type Escape Device and the Applicability in a Set Net (망목형 탈출장치의 선택성과 정치망에 적용 가능성)

  • This thesis is the fundamental study on the adaptation of escape device for reducing small fishes in set-net. The escape devices for experiments were made the mesh-type devices with three different mesh sizes (60.6, 75.8 and 120.0mm). The experiments of size selectivity on escape devices were conducted by using two kinds of species as black rockfish (Sebastes schlegeli) and sea perch (Lateolabrax maculatusi) in the experimental tank. The size selectivity curve was fitted by using a logistic function and the parameters of selectivity curve were estimated by a maximum likelihood method. In the results; 50% selection ranges for the mesh-type escape devices with three different mesh sizes were; a black rockfish was 18.99 in mesh size 60.6mm and 21.96 in mesh size 75.8mm (120mm could not estimate). A sea perch was 22.02 in mesh 60.6mm and 24.46 in mesh size 75.8mm (120mm could not estimate). The 50% selection range of a black rockfish was wilder than a sea perch about 1.1~.2 time. Therefore, the small fishes are able to reduce by using the mesh type escape device. However, the optimum mesh size should be decided to consider the size of target species and economics of catches.
