Experimental Production of Canine Pyometra by Inoculation with Escherichia coli into the Uterus (대양균 자궁내 접종에 의한 개의 자궁축농증 발증시험)

  • An attempt to reproduce pyometra by means of experimental uterine infection was conducted with the 10 mongrel hitches of the anestrus, proestrus, metestrus, and postpartum uterine involution stages. Organism used was hemoltic E. coli. origially isolated from uterine pus of a bitch that had the canine pyometra. One ml of broth culture containing $2{\times}10^6$ CFU was inoculated into the uterine horn with the uterine cervix ligated. Aか bitches were ovariohysterectomized at the 7th or 14th day after inoculation and the uterus was provided for macroscopic and microscopic observations. An experimental production of pyometra was successful in 7(70%) of 10 mongrel bitches, and the incidence of pyometra was seen 100% in the stage of anestrus, 100% in the stage of metestrus, 50.0% in the stage of pyomestrus, and 33.359 in the stage of uterine involution. Histologically, these uteri were characterized by the presence of cystic. endomeetrial hyperplasia(CEH) and acute endometritis superimposed on the cystic hyperplasia. And the pyometra with CEH could be seen 66.7% in metestrus 50% in anestrus, 33.3% in involution and in 0% in proestrus stage. Blood progesterone concentrations were showed as a normal range of the each stage of sexual cycle and any relationships between the progesterone concentrations and the incidence of pyometra were not observed.

  • The gestation period and parturition days were accurately predicted by measuring progesterone concentration in the plasma from 40 pregnant companion bitches. The mean length of the estrous cycle based on plasma progesterone concentrations were 8.14 ± 1.39 (Mean ± SD) days for proestrus, 9.19 ± 2.01 days for estrus, and 55.38 ± 1.96 days for diestrus phase, respectively. The gestation length from each based on the days was 65.61 ± 2.47 days from the first day of estrus after the first vaginal discharge, 63.21 ± 0.99 days from the day when plasma progesterone concentrations increase above 4.0 ng/ml, and 54.51 ± 3.51 days from the first day of diestrus, respectively. Therefore, the parturition day was estimated 65 days from the first day of estrus after the first vaginal discharge, 63 days from the day when plasma progesterone concentrations increase above 4.0 ng/ml, and 54 days from the first day of diestrus, respectively.

  • Pyometra, a common disorder in intact bitches, can lead to canine sepsis. Identification of biomarkers for sources of infection in the uterus using two-dimensional electrophoresis (2-DE)-mass spectrometry (MS) analysis may enable the discovery of novel diagnostic markers of sepsis. Toward this end, surgically resected uterus samples from four bitches (three pyometra and one healthy) were randomly selected for 2-DE-MS, which identified 32 differentially expressed proteins, including seven inflammatory proteins, five non-inflammatory proteins, and 20 functionally unknown proteins. Despite the limited information on canine uterus proteomics, we suggest the potential use of differentially expressed uterus proteins as candidate biomarkers to discover targets to attenuate inflammation in pyometra. Further identification of the functionally unknown proteins is warranted.

  • Serial ultrasonographic examinations were performed to establish a prediction table of parturition date in pregnant Maltese, Yorkshire Terrier, Shih-tzu and Miniature Schnauzer bitches. The inner chorionic cavity diameter and fetal head diameter in 45 pregnant bitches were converted retrospectively based on the day of parturition. The data of inner chorionic cavity diameter obtained from Day-44 to Day-25 and fetal head diameter obtained from Day-25 to Day-1 were used to compile a equations of prediction of parturition date. The 70 pregnant bitches with unknown mating time were examined to assess an accuracy of the equations established in this study. And these results were applied to the prediction of parturition date and compared to actual parturition date. The accuracy for parturition date within 0, $\pm$1, and $\pm$2 days interval using the equations of prediction of parturition date were 64.3%, 22.8% and 12.8%, respectively. The overall accuracy of prediction table of parturition day based on the ICCD and HD was 100% accurate within $\pm$2 days. Therefore, the prediction table seems to be a useful tool of the prediction of parturition day in practice.

  • The trunk and limb of the Jindo, Korean native dog was measured in the present study. One hundred and ninety five Jindoes were randomly selected from the 1,985 dogs in the Jindo County in August 1998, which were screened by the judging committee of the County and registered by inserting electronic chips in the withers. They were about a year and half old, and were white and fawn dogs. A total of 18 parts of the trunk and limb were measured using calipers and tapes. The average height at the withers was 50.67 cm in dogs and 47.48 cm in bitches. The average body length was 52.99 cm in dogs and 50.33 cm in bitches. And the average height : body length ratio was 104.70 in dogs and 106.08 in bitches. No differences were observed between the Jindoes of the two coat colors in these measurements. The results of this study would be useful for compiling the official standard for the adult Jindo.

Closed-Cervix Pyometra in Young Dogs: 2 Cases

  • Canine pyometra generally occurs in intact female dogs of more than four years of age. However, in rarely cases, pyometra can occur in young bitches. In the present report, two cases of pyometra in thirteen and eighteen months of age were presented. In both cases, absent of vaginal discharge represented that both patients had closed-cervix pyometra and diagnosis was somewhat complicated. In complete blood count, elevated level of leukocyte with left shift was found in both cases. Serum chemistry analysis showed elevation of alkaline phosphatase in both cases. Additionally, mild elevation of aspartate transferase and total protein were also found in $2^{nd}$ case. Radiographic and ultrasonographic findings show the enlargement of uterus and accumulation of fluid contents inside the uterine lumen. Ovariohysterectomy followed by antibiotics and anti-inflammatory drugs for post-surgery medication was performed for treatment. After a week, both patients fully recovered. To reduce mortality of pyometra in young intact female dogs, radiography or ultrasonography should be indicated immediately in the bitches showing severe infectious inflammation.

  • The semen from four male dogs which had been proven to be fertile in the past were frozen in a deep freezer at $-60^{\circ}C$ by simple freezing method using methanol and preserved at the same teperature for from 7 to 10 days. The semen were inseminated to 7 female dogs in estrus to find out the usability of this freezing method in artificial insemination for dogs. In addition, post-thaw motility and viability of sperm from two male dogs which had been fertile were also evaluated to investigate individual difference. Successful pregnancy was obtained by artificial insemination with canine semen frozen at $-60^{\circ}C$ by simple freezing method using methanol, namely, 3 bitches among 7 bitches which had been inseminated delivered puppies(42.8%). The average litter size of the whelping dogs were 4.3 puppies. The average post-thaw motility of canine sperm in the cases of conception was showen higher than those of non-conception(65.0% vs. 42.5%), along with the, same result in the average post-thaw viability between the two groups(53.3% vs, 27.5%). Individual difference of post-thaw motility and viability was obtained between two fertile dogs(p<0.05).

Serial Ultrasonographic Appearance of Postpartum Uterine Involution in Cocker Spaniel Bitches (Cocker Spaniel 개에서 분만 후 자궁수복의 연속적 초음파상)

  • This study was undertaken to determine the normal appearance of the postpartum uterine involution. Postpartum changes in uterine shape, architecture, echogenicity and diameter were monitored with ultrasonography in 8 Cocker spaniel bitches. The excretory period of vaginal discharges in 8 normal bitches of uterine involution was finished completely at 23.20$\pm$2.77 days (Mean$\pm$SD) postpartum. The short axis shape of the uterus was varied from polygonal to circular. This lasted until 1600$\pm$2.12 days Postpartum, during which time the short axis uterine shape gradually changed to circular. Also, the long axis shape of the uterus was created a beaded appearance of the horns until 25.60$\pm$2.51 days postpartum. The uterine diameter was decreased not only in the placental sites from 24.20$\pm$2.06mm at 1 day to 13.18$\pm$0.84mm at 7 days postpartum, but also in the interplacental sites 14.26$\pm$2.22mm at 1 day, 9.81$\pm$0.7mm at 7 days postpartum. There was a general trend of decreasing uterine diameter, which occurred more rapidly at the placental sites. In conclusion, normal postpartum uterine involution in Cocker spainel bitches appeared to be completed around 68 days postpartum by gross findings such as vaginal discharges, and by ultrasongraphic findings, uterine shape and echogenicity.

Serial ultrasonographic appearance of postpartum uterine involution in Korea Jin-do dogs (진도개에서 분만후 자궁수복의 연속적 초음파상)

  • This study was undertaken to determine the normal appearance of the postpartum uterine involution. Postpartum changes in uterine shape, architecture, echogenicity and diameter were monitored with ultrasonography in 11 Korea Jin-do dogs. Serial ultrasonographic examination was done daily during the first week, 3 days interval from 8 to 30 days, and weekly from 31 to 100 days postpartum, respectively. Of 11 postpartum bitches, 10 bitches (90.9%) had normal involution and 1 bitch (9.1%) had subinvolution of the placental sites (SIPS) by gross findings, vaginal discharges, and by ultrasographic findings, uterine shape and echogenicity. The excretory period of vaginal discharges in 10 normal bitches of uterine involution was finished completely at $20.2{\pm}4.6$ days (Mean${\pm}$SD) postpartum, but in 1 SIPS at 50 days postpartum. The short axis shape of the uterus was initially often flaccid-appearing. It varied from circular to crescent shaped to polygonal. This lasted until $16.5{\pm}3.7$ days postpartum, during which time the short axis uterine shape gradually changed to circular. Also, the long axis shape of the uterus was created a beaded appearance of the horns until $29.9{\pm}3.2$ days postpartum. After 30 days, it was appeared as tubular shape without distinguished between placental and interplacental sites. The ultrasonographic image of the postpartum uterus consisted of four echogenicity distinct layers. Uterine wall was represented the very hyperechoic serosa, hypoechoic myometrium, hyper echoic endometrium and anechoic structures of fluid in the uterine cavity until 7 days postpartum. The individual uterine layers were most prominent during the first week postpartum, and they became progressively less distinct throughout the course of uterine involution. Anechoic structures of fluid in the uterine cavity was detected until $25.0{\pm}6.4$ days postpartum, after which time it was not reliably detected. The uterine diameter was decreased not only in the placental sites frorm $24.1{\pm}2.5mm$ at 1 day to $15.4{\pm}1.4mm$ at 7 days postpartum, but also in the interplacental sites $14.9{\pm}1.5mm$ at 1 day, $10.6{\pm}0.8mm$ at 7 days postpartum. There was a general trend of decreasing uterine diameter, which occurred more rapidly at the placental sites. At 31 days pastpartum, these diameter reached almost same size, after that time, they could be not distinguished between placental and interplacental sites. At 87 days postpartum, the uterine diameter was $5.6{\pm}0.6mm$ both placental and interplacental sites, and the uterine horns were uniform hypoechoic, tubular structures without enlargement. Therefore, complete involution of the uterus occurred at that time. It was concluded that normal postpartum uterine involution in Korea Jin-do bitches appeared to be completed around 87 days postpartum by gross findings such as vaginal discharges, and by ultrasonographic findings, uterine shape and echogenicity. Also ultrasonographic characteristics of the postpartum uterine involution were described. Therefore, these result suggest that ultrasonographic assessment is a reliable method for diagnosing the SIPS and uterine dysfunction, such as pyometra in bitch.
