Treatment Characteristics of Plating Wastewater Containing Freecyanide, Cyanide Complexes and Heavy Metals (II) - Effect of Aldehyde Compounds and Polysulfide - (도금폐수내 유리시안과 착염시안 및 중금속의 처리특성 (II) - aldehyde와 polysulfide첨가에 따른 영향 -)

  • The objective of this study is to investigate the effect of aldehyde compounds and ploysulfide as accelerating agents on removal of heavy metals and CN in plating wastewater. As a results of the experiments, the removal efficiency of cyanide using the formaldehyde type of aldehydes was the highest at pH 9. Next types were sodium formaldehyde bisulfite addut> paraldehyde> paraformaldehyde. Also, optimum pH and dosage for treating the residual heavy metals by using polysulfide were pH 9 and 30 mg/L, respectively. The removal efficiencies of cyanide, chromium, zinc and copper were above 96.7% at optimum condition.

  • As the International Human Epigenome Consortium (IHEC) launched officially at the 2010 Washington meeting, a giant step toward the conquest of unexplored regions of the human genome has begun. IHEC aims at the production of 1,000 reference epigenomes to the international scientific community for next 7-10 years. Seven member institutions, including South Korea, Korea National Institute of Health (KNIH), will produce 25-200 reference epigenomes individually, and the produced data will be publically available by using a data center. Epigenome data will cover from whole genome bisulfite sequencing, histone modification, and chromatin access information to miRNA-seq. The final goal of IHEC is the production of reference maps of human epigenomes for key cellular status relevant to health and disease.

  • A series of poly[3-methylene-3'-methylenesodiumbisulfite-bis(4-hydroxyphenyl) sulfone] (PMSBPS) was synthesized by the reaction of bis-(4-hydroxyphenyl)sulfone(BPS), formalin, and formaldehyde sodium bisulfite(FSB), and their effect on the staining of direct dye on nylon in the dyeing of nylon/cotton union fabric was investigated. PMSBPSes have good staining resist effect on nylon in the dyeing of nylon/cotton union. Prolonged reacting time between BPS and formalin is effective in improving the staining resist of final PMSBPS. Too many sulfonic acid groups than necessary in the PMSBPS increase the staining of nylon, presumably by increasing the solubility of PMSBPS. Staining resist effect of PMSBPS was evident in the measurement of color diffence of dyed goods.

  • The ginseng growth-promoting bacterium Pseudomonas fluorescens KGPP 207 synthesized indole-3- acetic acid (IAA) from L-tryptophan, indole-3-pyruvic acid (IPyA), and indole-3-acetaldehyde (IAAld), but not from indole-3-acetamide (lAM) and other intermediates of various IAA biosynthetic pathways in the experiment with indole compound supplemented cell suspensions. TLC, HPLC, and GC-MS analyses revealed the presence of IPyA, indole-3-ethanol, indole-3-lactic acid and its methyl ester, IAA and its methyl, and ethyl esters in the culture supernatant of the bacterium. IAAld was detected in the supernatant using sodium bisulfite and TLC. The results indicate that unlike gall-forming bacteria which can synthesize IAA by lAM, the indole-3-pyruvic acid pathway is the route for IAA biosynthesis in this beneficial strain of P. fluorescens.

  • Wool samples were treated by potassium tert-Butoxide(t-BuOK) in anhydrous tert-butanol to remove the bound surface lipid-layer, and the weight loss behaviors in protease solution and dyeabilities of the samples were studies. The C/N ratio of the surface of the t-BuOK-treated wool was shown to be 4.3 from XPS analysis. From SEM pictures any remarkable change in the shape of surface curticle during the proteasw treatment was not observed regardless of prior t-BuOK treatment. Dyeing rate and equilibrium adsorption of Orange II, a typical levelling type acid dye, on wool were not changed by protease or t-BuOK treatment, but those of Milling Cyanine 5R, a typical milling type acid dye, on wool were greatly enhanced by t-BuOK treatment in spite that, from alkali and urea-bisulfite solubilities, no damage on the inner part of wool fiber was expected by t-BuOK treatment.

  • DNA methylation in Streptomyces griseus IFO 13350 was analyzed by high-performance liquid chromatographic analysis and bisulfite-based analysis to reveal two methylation sites, 5'-$GC^{5m}$ CGGC-3' and 5'-$GAG^{5m}$ CTC-3'. The methylation was reconstituted in Escherichia coli by simultaneous expression of S. griseus SGR4675 and S. achromogenes M.SacI. The E. coli cells produced plasmids that mimicked the methylation profile of S. griseus DNA, which was readily introduced into S. griseus. The results of this study raise the possibility of a promising approach to establish efficient transformation in several streptomycetes.

Estimation of the Prosthetic Group of Laccase Secreted from Pleurotus ostreatus (Pleurotus ostreatus에서 분비된 Laccase의 보결단 추정)

  • Extracellular laccase secreted from Pleurotus ostreatus was activated by $Cu^{2+}$ and $Cu^{+}$ . The enzyme was strongly inactivated by 8-hydroxyquinoline, potassium cyanide, sodium azide, sodium bisulfite and 2-mercaptoethanol. The two ionogenic groups, which have pKa values of 5.60-5.70 and 6.70-6.85 respectively, were found to relate with the active site of this enzyme. The oxidation reactions were brought about by initial single electron transfer process on the active site. The enzyme was found to be a metalloprotein which had about 3.9 cupric ions per molecule of protein as a prosthetic group. The enzyme showed a strong peak at 605 nm and a weak shoulder at 330 nm in UV-Visible absorption spectrum. Both signals disappeated upon treatment of the enzyme with 4 electron equivalent ascorbate. These results indicate that type I Cu peak and type III Cu shoulder are present in laccase.

Differential Inheritance Modes of DNA Methylation between Euchromatic and Heterochromatic DNA Sequences in Ageing Fetal Bovine Fibroblasts

  • To elucidate overall changes in DNA methylation that occurs by inappropriate epigenetic control during ageing, we compared fetal bovine fibroblasts and their aged neomycin-resistant versions using bisulfite-PCR technology. Reduction in DNA methylation was observed in euchromatic repeats (18S-rRNA/art2) and promoter regions of sing1e-copy genes (the cytokeratin/-lactoglobulin/interleukin-13 genes). Contrastingly, a stable maintenance of DNA methylation was revealed in various heterochromatic sequences (satellite I/IIalphoid and Bov-B). The differential inheritance modes of DNA methylation was confirmed through the analysis of individual neomycin-resistant clones. These global, multi-loci analyses provide evidence on the tendency of differential epigenetic modification between genomic DNA regions during ageing.

Multi-omics techniques for the genetic and epigenetic analysis of rare diseases

  • Until now, rare disease studies have mainly been carried out by detecting simple variants such as single nucleotide substitutions and short insertions and deletions in protein-coding regions of disease-associated gene panels using diagnostic next-generation sequencing in association with patient phenotypes. However, several recent studies reported that the detection rate hardly exceeds 50% even when whole-exome sequencing is applied. Therefore, the necessity of introducing whole-genome sequencing is emerging to discover more diverse genomic variants and examine their association with rare diseases. When no diagnosis is provided by whole-genome sequencing, additional omics techniques such as RNA-seq also can be considered to further interrogate causal variants. This paper will introduce a description of these multi-omics techniques and their applications in rare disease studies.
