  • The present report aimed at evaluating the effect of bisphenol A(BPA) and diethylstilbestrol(DES) on Leydig or Sertoli cell-lines. To identify the differences in the susceptibility to BPA upon different cell-types, assay of the cell viability was done on TM3(Leydig cells) and TM4(Sertoli cells) cell-lines. The result indicates that Sertoli cells are more sensitive to low dose of BPA than Leydig cells. Also, the BPA- or DES-treated Sertoli cells showed a reduction of phospholipase D(PLD) activity identically. According to the confirmation of the mRNA expression of fas receptor and fas ligand in the BPA-treated cells, fas/fasL system activated by BPA will deliver the apoptosis signal onto Sertoli TM4 cells. However, Fas/FasL system was not activated in the DES-treated cells unlike the BPA-treated cells.

  • Bisphenol A (BPA) and di-2-ethyhexyl phthalate (DEHP) is the considerably hazardous materials, endocrine disrupting chemicals (EDCs), which is accumulated to the human's fat and tissue. We studied the relationship between the concentrations of BPA and DEHP in the precocious puberty and the normal controls using GC/MSD. BPA concentrations of the precocious puberty are lower than that of normal controls. But DEHP concentrations of the precocious puberty are higher than that of normal controls. From these results, BPA is not related to the precocious puberty. Otherwise, DEHP is somewhat related to the precocious puberty and the concentrations of the precocious puberty are approximately 45% higher than that of normal controls.

  • In this study, gametotoxicity and embryotoxicity experiments using Hemicentrotus pulcherrimus were carried out to investigate the ecotoxicological effects of bisphenol A (BPA). We examined the effects of BPA on fertilization and normal embryogenesis at various concentrations (0, 300, 500, 800, 1000, and 1500 ppb). The results demonstrated that the fertilization rates were not changed. The normal embryogenesis rates were gradually decreased in a dose-dependent manner, and were significantly lowered following 800 ppb BPA treatment ($EC_{50}$=1056.1 ppb, 95% Cl=981.8~1163.9 ppb). The observed effective concentration and the lowest observed effective concentration of the normal embryogenesis rate were 500 ppb and 800 ppb, respectively. The embryos showed retarded development at each tested concentration, indicating the fact the embryonic development was delayed due to the increasing concentrations of BPA. Furthermore, we examined the expression of glutathione S-transferase (GST) mRNA at various concentrations of BPA in H. pulcherrimus. Interestingly, it was found that the expression level of GST mRNA was significantly increased in the experimental group exposed to BPA. Based on these results, we suggested that BPA at greater than 800 ppb has a toxic effect during the early embryonic stages of H. pulcherrimus, and GST mRNA may be used as a biomarker for risk assessment of BPA contamination.

Synthesis and Characterization of Branched Sulfonated Poly(Ether Sulfone-ketone) Copolymer and Organic-inorganic Nano Composite Membranes

  • Branched sulfonated poly(ether sulfone-ketone) copolymer was prepared with bisphenol A, 4,4-difluorobenzophenone, sulfonated chlorophenyl sulfone (40mole% of bisphenol A) and THPE (1,1,1-tris-p-hydroxyphenylethane). THPE was used 0.4 mol% of bisphenol A to synthesize branched copolymers. Organic-inorganic nano composite membranes were prepared with copolymer and a series of $SiO_2$ nanoparticles (20 nm, 4, 7 and 10 wt%). The composite membranes were cast from dimethylsulfoxide solutions. The films were converted from the salt to acid forms with dilute hydrochloric acid. The membranes were studied by differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). Sorption experiments were conducted to observe the interaction of sulfonated polymers with water and methanol. Branched copolymer and nano composite membranes exhibit proton conductivities from $1.12{\times}10^{-3}$ to $6.04{\times}10^{-3}\;S/cm^2$, water uptake from 52.9 to 62.4%, IEC from 0.81 to 1.21 meq/g and methanol diffusion coefficients from $1.2{\times}10^{-7}$ to $1.5{\times}10^{-7}\;cm^2/S$.

Relationship between Bisphenol A Exposure and Obesity in Korean Adults from the Second Stage of KoNEHS (2012-2014) (한국 성인의 비스페놀 A 노출과 비만과의 관련성 연구: 제2기 국민환경보건기초조사(2012-2014))

  • Objectives: Bisphenol A (BPA) has been extensively used in a variety of consumer products, resulting in widespread non-occupational human exposure. It is often detected in the human body. Studies have reported many health effects associated with endocrine and metabolic disruptions, including obesity, diabetes, hypertension, and cardiovascular diseases. This study was performed to explain the relationship between BPA exposure and obesity in the Korean adult population. Methods: The second stage of the Korean National Environmental Health Survey (KoNHES) was conducted from 2012 to 2014 with 6,478 persons participating. Using the results of the survey, we analyzed the exposure levels for BPA and the influence on obesity of BPA. Results: In model 1, the volume-based measure concentration of BPA, total, female and the 30s to 60s age group were positively related with BMI. In model 2, creatinine adjusted as a covariate and positive associations for BPA with BMI were observed in the female group and was marginally significantly associated in low body weight group. In model 3, creatinine adjusted (g/g-creatinine), BPA exposure, and BMI were positively related with sex, in females, and there was a marginally significant association with the low body weight group in the BMI categories. BMI was significantly associated with BPA in the female group in all three models. Conclusion: This study added further evidence that exposure to EDCs, include bisphenol A, is related with obesity among the general population. Given the environmental health concerns over BPA, it is necessary to develop comprehensive measures to reduce BPA exposure.

  • We investigated the effects of bisphenol A (BPA), endocrine disruptor, on the mixed lymphocyte reaction and TNF-$\alpha$ production of antigen presenting cells in mice. Cells from mouse (C57BL/6) bone marrow were cultured with GM-CSF for 8 days and mature dendritic cells (DCs) were prepared. These DCs proliferation in response to Balb/c splenocytes was measured at 72 h of culture with BPA by tritiated thymidine incorporation ([3H]TdR) and [3H]TdR incorporation was determined by scintilation counting.   • Bisphenol A [2, 2 bis (4-hydoxyphenyl) propane; BPA] is a widely used endocrine disruptors and has estrogenic: activities. Although interests on biological effect of BPA are rising, evidences of its effect on immune system are lacking. We investigated that the effect of BPA on immune parameters to postulate the mechanism, and BPA interruptions between neuroendocrine and immune system. BPA was administrated to mice by p.o. (as a drinking water) dose on 0.015, 1.5 and 30 mg/ml for 4 weeks. The BPA treatment did not result in any change in body weight, spleen weight and distribution of lymphocyte subpopulation collected from spleen. BPA induced prolactin production in spleen, and exposure of SPA increased the activity of splenocyte proliferation in response to Con A (p<0.001). The production of a strong Th-1 type cytokine ($IFN-{\gamma}$) was induced while Th-2 type (IL-4) was suppressed by SPA treatment. These were consistent with RT-PCR results of transcription factor GATA-3 and IRF-1. These findings suggested that stimulation of prolactin production by estrogenic effects of SPA would affect cytokine profiles, and lead to imbalanced cellular immune response. In addition, we could speculate that prolactin and cytokine is important mediator involved in network between neuroendocrine and immune system by BPA.
