  • This paper represents the characteristics of evapotranspiration rate (EVTR) and graft-taking of watermelon grafted seedlings in a graft-taking enhancement system using fluorescent lamps as artificial lighting source. Four air temperature levels of 23, 25, 27 and 29C, three humidity levels of 85, 90 and 95%R.H. and two photosynthetic photon flux (PPF) levels of 30 and 50 ${\mu}$mol m$\^$-2/ S$\^$-1/ were provided to investigate the effects of air temperature, relative humidity and light intensity on EVTR and graft-taking of grafted seedlings. EVTR of grafted seedlings increased with increasing air temperature and the passage of time after grafting. Also EVTR increased with decreasing relative humidity. As relative humidity decreased and air temperature increased, vapor pressure deficit increased and thus EVTR increased. It is required to maintain a low level vapor pressure deficit for suppressing EVTR of grafted seedlings during first 1-2 days after grafting. Therefore, less EVTR at initial stage after grafting would be adequate for smooth joining of the scion and rootstock.

New records of Parasitidae mites (Acari; Mesostigmata) in Korea

  • Korean fauna mites in the family Parasitidae of Mesostigmata were reported as 6 genera, 22 species. Among those 20 species were recorded from Northern part of Korea by Athias-Henriot (1977; 1980), Daele (1975) and Tichomirov (1977), but two from Southern part of Korea by Choi (1994) and Korean zoological record (KSSZ, 1997). During the faunal study of soil predatory mites of diverse habitats in South Korea, four species of P. beta, P. fimetorum, P. insignis and Poecilochirus carabi are the new records to Korean peninsula. Taxonomic details and other biological characteristics are reported.

  • Mites of the genus Antennoseius of family Ascidae are free-living soil predator mostly observed on the open grass field. In Korea, only two species, Antennoseius imbricatus Ishikawa, 1969 and A. japonicus Ishikawa, 1979 were reported in 1990s. Recent series of soil acarine biodiversity survey in Gyeongbuk province during 2009-2015. We recovered a new record of Antennoseius avius (Karg, 1976) to Korean inventory from riparian grassland. Detailed description of the species as well as the identification key to the genus and species were provided.

  • Mite of the genus Hypoaspis of family Laelapidae are free-living soil predators mostly observed on the open grass field. Five species in the genus Hypoaspis were listed in Korean Catalog (NIBR, 2013) without detail information. Recent series of soil acarine biodiversity survey in Gyeongbuk and Gangwon provinces during 2009-2015 recovered a new record of Hypoaspis sardous (Canestrini, 1884) to Korean inventory from apple orchard and riparian grass land. Detailed description and morphological comparison with the related species, H. aculeifer which is well known biological control agent of horticultural crop pests were provided with identification keys.

  • Main tobacco diseases were surveyed on major flue-cured leaf tobacco fields throughout South Korea in 2006. Mosaic caused by potato virus Y and bacterial wilt caused by Ralstonia solanacearum were most severe during harvest season. During last ten years, the damage by tobacco mosaic virus was reduced but the incidence of bacterial wilt increased. These changes of the disease incidences coincide with release time of the tobacco cultivar resistant to the tobacco mosaic virus but susceptible to bacterial wilt pathogen. Wild fire(Pseudomonas syringae pv. tabaci strain ungulate) occurred severely at Kangwon province, though the symptom of wild fire with yellow halo was not observed.

Production, Purification, and Characterization of Taxol and 10-DABIII from a new Endophytic Fungus Gliocladium sp. Isolated from the Indian Yew Tree, Taxus baccata

  • We have isolated endophytic fungi from the Indian yew tree, Taxus baccata, and then screened for taxol production. Out of the 40 fungal cultures screened, one fungus Gliocladium sp. was found to produce taxol and 10-DABIII (10-deacetyl baccatin III). These compounds were purified by TLC and HPLC and characterized using UV-spectroscopy, ESI-MS, MS/MS, and proton NMR. One liter of Gliocladium sp. culture yielded $10\;{\mu}g$ of taxol and $65\;{\mu}g$ of 10-DABIII. The purified taxol from the fungus showed cytotoxicity towards cancer lines HL-60 (leukemia), A431 (epidermal carcinoma), and MCF-7 (breast cancer).

  • In the search for novel, natural melanogenesis inhibitors, a new sesquiterpene, inularin, was isolated from the flowers of Inula britannica, and the structure was determined using spectroscopic and chemical methods. The antimelanogenic effects of inularin on B16F10 melanoma cells and zebrafish embryos were evaluated. Inularin dose-dependently reduced melanocyte-stimulating hormone-induced melanin production and L-DOPA oxidation in B16F10 cells. Zebrafish embryos were used to confirm the antimelanogenic activity. Inularin significantly decreased the pigmentation of embryos compared with untreated controls.

  • Potassium phosphonate inhibited less mycelial growth of Phytophthora cactorum in vitro than metalaxyl-M. But the net protection rate in leaflet test and whole plant pot test was greater in potassium phosphonate than metalaxyl-M. P. ginseng appeared to have an induced resistance against P. cactorum with phosphonate around 50~100 ppm.

  • Microbial of enzymatical methods are suitable for production of rare monosaccharides. Using oxidation and reduction ability of Microorganisms, various rare ketoses and polyols can be produced, for example D-tagatose from galagtitol by Enterobacter agglomerans strain 221e. L-tagatose from galactitol by Klebsiella pheumonias strain 40b, L-psicose from allitol by Gluconobacter frateurii IFO 3254, D-talitol from d-tagatose by Aureobasidium pullulans strain 113B, allitol from D-psicose by Enterobacter agglomerans strain 221e and so on. We can produce various rare aldoses and ketoses using aldose isomerases, for example L-galactose from L-tagatose by D-arabnose isomerase, and L-ribose from L-ribulose by L-isomerase, and so on. D-Tagatose 3-epimerase of Pseudomonas sp. ST-24 is very useful for preparationof various rare ketoses, for example D-psicose from D-fructose, D-sorbose from D-tagatose, L-fructose, from L-psicose and so on. Using polyol dehydrogenases, aldose isomerases and D-tagatose 3-epimerase, we can design the suitable for production of a certain rare monosaccharide from a suitable substrate.

  • Polydnaviruses are a group of double-stranded DNA viruses and are symbiotically associated with some ichneumonoid wasps. As proviruses, the replication of polydnaviruses occurs in the female reproductive organ at the pupal stage. This study analyzed the effects of two developmental hormones, juvenile hormone (JH) and ecdysteroid, on the viral replication of Cotesia plutellae bracovirus (CpBV). All 23 CpBV segments identified contained a conserved excision/rejoining site ('AGCTTT') from their proviral segments. Using quantitative real-time PCR based on this excision/rejoining site marker, initiation of CpBV replication was determined to have occurred on day 4 on the pupal stage. Pyriproxyfen, a JH agonist, significantly inhibited adult emergence of C. plutellae, whereas RH5992, an ecdysteroid agonist, had no inhibitory effect. Although RH5992 had no effect dose on adult development, it significantly accelerated viral replication. The results of immunoblotting assays against viral coat proteins support the effects of the hormone agonists on viral replication.
