본 연구에서는 호기성 혼합 균주의 기질로서 Laminaria japonica를 사용하여 수용성 당과 환원당을 생산하였다. Laminaria japonica는 대표적인 갈조류 바이오매스로 높은 생장율, 저렴한 가격 및 탄수화물 함량이 높은 특성을 가지고 있다. 본 실험에 적용된 당화공정은 호기성 혼합 균주를 배양 및 순응시켜서 안정화하였다. 수용성 당과 환원당의 생산이 각기 다른 수리학적 체류시간에서 관찰되었으며, 당화 효율은 수리학적 체류시간을 낮출수록 증가하였다. 또한, 연속식 공정을 이용하여 1 day의 수리학적 체류시간 조건에서 최대 당화 효율을 나타내었으며, 17.96 g/L/day의 가용화된 탄수화물과 4.30 g/L/day의 환원당을 수득하였다. 그러나 수리학적 체류시간을 0.5 day로 낮추었을 때 당화수율이 급격히 감소하는 현상을 확인할 수 있었다. 실험결과를 통하여 Laminaria japonica가 바이오리파이너리 공정을 통하여 유용물질을 생산하기에 매우 적합한 바이오매스임이 확인되었다. 결론적으로 혼합균주를 이용한 연속 생물학적 전처리 시스템이 바이오리파이너리 기술에 성공적으로 사용될 수 있음을 확인하였다.
최근 바이오 화학산업, 특히 바이오 리파이너리를 위한 발효공정에서 물질 분리를 위하여 분리막이 적용되고 있다. 이에 본 논문에서는 바이오 화학산업에서 분리막을 응용하고 있는 연구들을 살펴보고자 한다. 특히 전처리로서의 분리막 응용과 발효산물의 분리막 응용을 알아보았다. 다양한 바이오 물질의 분리 및 정제를 위한 공정에서 분리막의 사용을 알아보고 특히 리그노셀룰로스를 이용한 바이오 리파이너리에서의 분리막 사용을 강조하였다.
We present results of the optical design and fabrication of a light-guiding plate (LGP) for a photobioreactor by using sunlight and a linear Fresnel lens. LGP patterns were designed by optical simulations with an illumination design tool, LightTools, and fabricated by using a computerized numerical control machine. Optical characteristics of average deviation of illuminance distribution and light throughput efficiency were measured and compared with simulation results.

The non-isothermal and isothermal pyrolysis properties of H lignin and P lignin extracted from different biorefinery processes (such as supercritical water hydrolysis and fast pyrolysis) were studied using thermogravimetry analysis (TGA) and pyrolyzer-gas chromatography/mass spectrometry (Py-GC/MS). The lignins were characterized by ultimate/proximate analysis, FT-IR and GPC. Based on the thermogravimetry (TG) and derivative thermogravimetry (DTG) curves, the thermal decomposition stages were obtained and the pyrolysis products were analyzed at each thermal decomposition stage of non-isothermal pyrolysis. The isothermal pyrolysis of lignins was also carried out at 400, 500, and $600^{\circ}C$ to investigate the pyrolysis product distribution at each temperature. In non-isothermal pyrolysis, P lignin recovered from a fast pyrolysis process started to decompose and produced pyrolysis products at a lower temperature than H lignin recovered from a supercritical water hydrolysis process. In isothermal pyrolysis, guaiacyl and syringyl type were the major pyrolysis products at every temperature, while the amounts of p-hydroxyphenyl type and aromatic hydrocarbons increased with the pyrolysis temperature.
Trehalose is a non-conventional sugar with potent applications in the food, healthcare and biopharma industries. In this study, trehalose was synthesized from maltose using whole-cell Pseudomonas monteilii TBRC 1196 producing trehalose synthase (TreS) as the biocatalyst. The reaction condition was optimized using 1% Triton X-100 permeabilized cells. According to our central composite design (CCD) experiment, the optimal process was achieved at 35℃ and pH 8.0 for 24 h, resulting in the maximum trehalose yield of 51.60 g/g after 12 h using an initial cell loading of 94 g/l. Scale-up production in a lab-scale bioreactor led to the final trehalose concentration of 51.91 g/l with a yield of 51.60 g/g and productivity of 4.37 g/l/h together with 8.24 g/l glucose as a byproduct. A one-pot process integrating trehalose production and byproduct bioremoval showed 53.35% trehalose yield from 107.4 g/l after 15 h by permeabilized P. moteilii cells. The residual maltose and glucose were subsequently removed by Saccharomyces cerevisiae TBRC 12153, resulting in trehalose recovery of 99.23% with 24.85 g/l ethanol obtained as a co-product. The present work provides an integrated alternative process for trehalose production from maltose syrup in bio-industry.
Acidic endo-polygalacturonases are the major part of pectinase preparations and extensively applied in the clarification of fruits juice, vegetables extracts, and wines. However, most of the reported fungal endo-polygalacturonases are active and stable under narrow pH range and low temperatures. In this study, an acidic endo-polygalacturonase (EPG4) was purified and characterized from a mutant strain of Penicillium oxalicum. The N-terminal amino acid sequence of EPG4 (ATTCTFSGSNGAASASKSQT) was different from those of reported endo-polygalacturonases. EPG4 displayed optimal pH and temperature at 5.0 and 60-70℃ towards polygalacturonic acid (PGA), respectively, and was notably stable at pH 2.2-7.0. When tested against pectins, EPG4 showed enzyme activity over a broad acidic pH range (>15.0% activity at pH 2.2-6.0 towards citrus pectin; and >26.6% activity at pH 2.2-7.0 towards apple pectin). The Km and Vmax values were determined as 1.27 mg/ml and 5,504.6 U/mg, respectively. The enzyme hydrolyzed PGA in endo-manner, releasing oligo-galacturonates from PGA, as determined by TLC. Addition of EPG4 (3.6 U/ml) significantly reduced the viscosity (by 42.4%) and increased the light transmittance (by 29.5%) of the papaya pulp, and increased the recovery (by 24.4%) of the papaya extraction. All of these properties make the enzyme a potential application in the beverage industry.
The new movement towards green chemistry and renewable feedstocks makes microbial production of chemicals more competitive. Among the numerous chemicals, organic acids are more attractive targets for process development efforts in the renewable-based biorefinery industry. However, most of the production costs in microbial processes are higher than that in chemical processes, among which over 60% are generated by separation processes. Therefore, the research of separation and purification processes is important for a promising biorefinery industry. This review highlights the progress of recovery processes in the separation and purification of organic acids, including their advantages and disadvantages, current situation, and future prospects in terms of recovery yields and industrial application.
본 연구에서는 바이오디젤 생산의 부산물인 글리세롤로부터 수증기 개질(Steam Reforming, SR) 반응을 통해 수소를 생산하는 공정의 모델링과 모사 및 최적화를 수행했다. 글리세롤을 이용한 수소 생산 방법은 기존의 수소 생산방법인 메탄의 수증기 개질법(Steam Methane Reforming, SMR)을 대체할 수 있는 새로운 방법으로 세계 여러 곳에서 연구가 진행 중이다. 글리세롤과 수증기의 기체 혼합물을 고온의 반응기 내에서 개질시켜 합성가스(CO, $H_2$)를 생산하고, 합성가스에 포함된 일산화탄소를 수성 가스 전화 반응(Water-Gas Shift, WGS)을 통해 수증기와 반응시켜 수소를 생성하고, 최종적으로 Pressure Swing Adsorption (PSA) 공정을 통하여 이산화탄소와 수소를 분리하여 정제된 수소를 얻는다. 공정시뮬레이션 프로그램인 UniSim을 이용하여 시뮬레이션을 진행하였으며, 열효율 개선을 실시하여 운전 비용을 절감하고자 하였다. 기존 연구인 미국 DOE와 독일 Linde의 글리세롤 이용 수소 생산공정과 수율 비교를 진행하였고, 수소 에너지 인프라 구축에 기여하기 위한 최적의 생산방법을 제안하였다.
Enzymatic hydrolysis of lignocellulosic biomass into fermentable sugars is a key step in the conversion of agricultural by-products to biofuels and value-added chemicals. Utilization of a robust microorganism for on-site production of biomass-degrading enzymes has gained increasing interest as an economical approach for supplying enzymes to biorefinery processes. In this study, production of multi-polysaccharide-degrading enzymes from Aspergillus aculeatus BCC199 by solid-state fermentation was improved through the statistical design approach. Among the operational parameters, yeast extract and soybean meal as well as the nonionic surfactant Tween 20 and initial pH were found as key parameters for maximizing production of cellulolytic and hemicellulolytic enzymes. Under the optimized condition, the production of FPase, endoglucanase, ${\beta}$-glucosidase, xylanase, and ${\beta}$-xylosidase was achieved at 23, 663, 88, 1,633, and 90 units/g of dry substrate, respectively. The multi-enzyme extract was highly efficient in the saccharification of alkaline-pretreated rice straw, corn cob, and corn stover. In comparison with commercial cellulase preparations, the BCC199 enzyme mixture was able to produce remarkable yields of glucose and xylose, as it contained higher relative activities of ${\beta}$-glucosidase and core hemicellulases (xylanase and ${\beta}$-xylosidase). These results suggested that the crude enzyme extract from A. aculeatus BCC199 possesses balanced cellulolytic and xylanolytic activities required for the efficient saccharification of lignocellulosic biomass feedstocks, and supplementation of external ${\beta}$-glucosidase or xylanase was dispensable. The work thus demonstrates the high potential of A. aculeatus BCC199 as a promising producer of lignocellulose-degrading enzymes for the biomass conversion industry.
