Preparation and Characterization of Silk and Mulberry/Silk Papers

  • Papers were prepared from cut cocoons and mulberry branches, which are byproducts from sericulture industry. The long filament of silk should be cut into appropriate length in order to prepare paper and this was achieved by chemical method. By a mixture of sodium hydroxide and sodium carbonate solution, the silk filaments were cut into short fibers (less than 1 mm in length). Since the short silk fibers (sSf) could not bind each other by itself, starch and poly(ethylene oxide)(PEO) were added as a bonding agent. When starch and PEO were used in a ratio of 3:7, the silk papers had optimum mechanical properties for paper. Fibers from the skin of mulberry branches (MBF) were added to sSf to enhance the mechanical properties of pure silk paper. Bleaching of MBF was performed with a mixture of hydrogen peroxide and sodium silicate. The mechanical properties were greatly enhanced and the optimum blend ratio of MBF and sSf were 7:3. The mulberry/silk paper has good absorption property against formaldehyde, and therefore, the paper could be applied as a wall paper for preventing the sick house syndrome.

  • Maltosyl(G2)-erythritol, produced by the transglycosylation reaction of erythritol with maltotriose by Bacillus stearothermophilus maltogenic amylase, was not utilized either as a substrate for lactic acid production or for water-insoluble glucan synthesis. An inhibition assay of dextransucrase and mutansucrase showed that the dental caries suppression effect of G2-erythritol was greater than that of erythritol.

Quantitative Analysis for the Quality Evaluation of Sinomenine, Magnoflorine and Syringaresinol in Sinomenium acutum (방기의 품질 평가를 위한 Sinomenine, Magnoflorine, Syringaresinol의 함량 분석)

  • Sinomenium acutum has been used for the treatment of rheumatoid arthritis, arrhythmia, and pain. We evaluated the quality of S. acutum extract by quantitatively analyzing its components such as sinomenine, magnoflorine and syringaresinol with the simultaneous determination method using HPLC-DAD. A total of 53 samples collected from different localities were evaluated with this quality evaluation method. Sinomenine, magnoflorine and syringaresinol from tested samples ranged from 0.0649~9.1050%, 0.7460~10.7590% and 0.0010~0.2441%, respectively. In the current study, we were able to exhibit the diverse quality of S. acutum samples collected from various locations using the simultaneous determination method.

  • Truncated form of UBP1, an ubiquitin-specific protease of Saccharomyces cerevisiae, was overexpressed in Escherichia coli. The hexahistidine residue $(His_6)$ was fused to the N-terminus of truncated UBP1 and the corresponding recombinant protein was purified with high yield by immobilized metal affinity chromatography. The truncated form of UBP1 protein was functional to cleave ubiquitinated human growth hormone as substrate. Effects of pH and temperature were investigated in order to optimize deubiquitinating reactions for the truncated UBP1. Optimum temperature and pH for the cleavage reaction were $40^{\circ}C$ and pH 8.0, respectively.

Neuroprotective Compounds Isolated from the Methanolic Extract of Lonicera japonica

  • A $CH_3Cl$ extract from the flower of Lonicera japonica (Lauraceae) significantly protected primary cultures of rat cortical cells injured by the excitotoxic amino acid, L-glutamate. Loganin (1), secoxyloganin (2), caffeic acid (3) rutin (4), hyperoside (5), quercetin-3-O-glucoside (6), lonicerin (7), kaempferol-3-O-rutinoside (8), luteolin-7-O-b-D-glucopyranoside (9), quercetin (10) and luteolin (11) were isolated by bioactivity-guided fractionation from the $CH_3Cl$ fraction and further separated using chromatographic techniques. Caffeic acid, lonicerin, kaempferol-3-O-rutinoside, quercetin and luteolin had significant neuroprotective activities against glutamate-induced neurotoxicity in primary cultures of rat cortical cells at concentrations ranging from $0.1{\mu}M$ to $10.0\;{\mu}M$.

Neuroprotective Activity of So Cheong Ryong Tang with Steaming Process (증숙 과정을 통한 소청룡탕의 뇌신경세포 보호 활성 변화에 관한 연구)

  • So Cheong Ryong Tang (SCRT) has been used as a traditional herbal medicine for the treatment of the bronchial asthma. In this study, the effect of steaming process on changes in 11 major compounds (homogentisic acid, ephedrine HCl, paeoniflorin, cinnamic acid, cinnam aldehyde, glycyrrhizin, 6-gingerol, schizandrin, methyleugenol, gomisin A and gomisin N) content and neuroprotective activity of SCRT were evaluated. Major compound content was slightly different with steaming produce. The contents of paeoniflorin, cinnamic acid, cinnam aldehyde, 6-gingerol and methyleugenol were decreased and homogentisic acid, ephedrine HCl, glycyrrhizin, schizandrin and gomisin A were increased by steaming process. The neuroprotective activity of steaming SCRT was determined in mouse hippocampal HT22 cells by MTT assay. As a result, neuroprotective activity of a steaming SCRT was higher than that of a non steaming SCRT. This study demonstrated that neuroprotective activity of SCRT can be improved through steaming process.

Neuroprotectuve Effect of Homosyringaldehyde Isolated from Cynanchum panuculatum against Glutamate-Induced Neurotoxicity (서장경으로부터 분리한 Homosyringaldehyde의 뇌신경세포 보호 활성)

  • Homosyringaldehyde was isolated and identified from the 80% methanol extract of roots of Cynanchum paniculatum. C. paniculatum has been widely used for the treatment of various diseases such as neurasthenia, insomnia, dysmenorrheal and toothache. This compound exerted significant neuroprotective activities against glutamate-induced neurotoxicity in hippocampal HT22 cell line by 37.53% (at the concentration of $100{\mu}M$). We investigated mode of action of this compound. Homosyringaldehyde ($100{\mu}M$) significantly decreased the ROS level and $Ca^{2+}$ concentration in the oxidative stress induced HT22 cells by oxidative glutamate toxicity. Thus, our results suggest that homosyringaldehyde significantly protect HT22 cells against glutamate-induced oxidative stress, via antioxidative activities. As the results, we suggest that homosyringaldehyde may be useful in the treatment of neurogenerative disorders.

Enhancement of Bioactivity of Sosihotang with Steaming Process (증숙에 따른 소시호탕의 활성 변화)

  • Sosihotang is a herbal medicine traditionally used to treat chronic liver disease. This study was performed to investigate the effect of steaming Sosihotang at a $121^{\circ}C$ on its major compounds and neuroprotective activity. We evaluated quantitative changes of 6 major compounds in Soshihotang, saikosaponin A, homogentisic acid, baicalin, ginsenoside $Rg_3$, glycyrrhizin and 6-gingerol using HPLC-DAD. After steaming produce, saikosaponin A, homogentisic acid, baicalin, glycyrrhizin and 6-gingerol in Sosihotang were decreased and ginsenoside $Rg_3$ was increased. The neuroprotective activity of steamed Sosihotang was determined by MTT assay, comparing to Sosihotang. The steamed sosihotang showed a higher neuroprotective activity than Sosihotang. In conclusion, steam produce could improve neuroprotective activity of Soshihotang.

  • Aucklandia lappa Decne (Compositae) has been used for treatment of abdominal pain, vomiting, diarrhea, chronic inflammation, and antibacterial effect. The quality of these herbs has been affected by many factors such as collection time, place, temperature, cultivation environment and manufacturing process. We used costunolide and dehydrocostuslactone as marker compounds for quality evaluation of rhizome of Aucklandia lappa. 66 samples of Aucklandia lappa were collected from those habitats in Korea and China. The developed HPLC-DAD method was applied to investigate for quality control of Aucklandia lappa samples. The average contents of the costunolide and dehydrocostuslactone were 2.3895% and 0.9258%, respectively. The principal component analysis (PCA) exhibited that classification of Aucklandia lappa according to origin not separated. Results of this study may be satisfactory applied to determination of content criteria of Aucklandia lappa.

  • In this study, a model reduction technique was used to develop a precise noise and vibration prediction model for the individual components of a driveline system. The dynamic reduced-order model generated by the Craig-Bampton method was applied to perform dynamic analysis of an electric agricultural power cart. The natural frequency and acceleration response results were analyzed according to the different number of dominant sub-structural modes contained in the reduced-order models. Through the analysis results, it was confirmed that a sufficient number of dominant sub-structures to satisfy the operating conditions should be selected to construct an optimal reduced-order model.
