  • The electron beam irradiation was applied as a pretreatment of the enzymatic hydrolysis of yellow poplar with doses of 0$\sim$450 kGy. The higher irradiation dose resulted in the more degradation of hardwood biomass not only from carbohydrates but also from lignin. This changes originated from the irradiation resulted in the better response to enzymatic hydrolysis with commercial cellulases (Celluclast 1.5L and Novozym 342). The more improvement on enzymatic hydrolysis by the irradiation was found in the xylan than in the cellulose of yellow poplar.

투고증발을 이용한 섬유성바이오매스의 동시당화 및 추출발효 (Simultaneous Saccharification and Pervaporative Fermentation of Cellulosic Biomass)

  • Application of pervaporative extraction of ethanol to simultaneous saccharification and fermentation(SSF) of cellulose was investigated. From batch experiments, optimum cellulose substrate and enzyme loadings were found to be 10% and 15 IFPU/g cellulose, respectively. The cellulose conversion was lowered in fed-batch system due to the ethanol accumulation. The activity of the yeast Saccharomyces uvarum used in this study was significantly reduced at ethanol concentrations above around 40 g/L. From pervaporation experiments using PDMS membrane, ethanol was efficiently separated at 38$^\circ C$ and 10 mmHg of a down stream pressure. The pervaporation unit with 240 cm$^2$ of surface area was combined into the SSF reactor. The continuous removal of ethanol by pervaporation during SSF resulted in an improved cellulose conversion. Within the scope of this experiment, ethanol yields in the pervaporative SSF and simple SSF were 68.3% and 56.6%, respectively. The permeate flux for SSF broth pervaporation was about one-half that for the pervaporation of aqueous ethanol solution. Accordingly, the development of a membrane with higher ethanol selectivity and flux will increase the feasibility of this technology.

  • Ethanol fermentations were carried out using simultaneous saccharification and fermentation (SSF) and separated hydrolysis and fermentation (SHF) processes with monosaccharides from seaweed, Saccharina japonica (sea tangle, Dasima) as the biomass. The pretreatment was carried out by thermal acid hydrolysis with $H_2SO_4$ or HCl. Optimal pretreatment condition was determined at 10% (w/v) seaweed slurry with 37.5 mM $H_2SO_4$ at $121^{\circ}C$ for 60 min. To increase the yield of saccharfication, isolated marine bacteria Bacillus sp. JS-1 was used and 48 g/L of reducing sugar were produced. Ethanol fermentation was performed using SSF and SHF process with Pachysolen tannophilus KCTC 7937. The ethanol concentration was 6.5 g/L by SSF and 6.0 g/L by SHF.

  • Dissolving part of xylan and lignin in lignocellulosic biomass by base can be used as pretreatment technique. Cork oak was pretreated with sodium hydroxide solution and the pretreatment effects were evaluated with two critical factors - NaOH concentration and pretreatment temperature. Some of xylan and lignin were removed by base pretreatment. At $90^{\circ}C$ and 13% NaOH pretreatment, 22.0% of lignin and 78.8% of xylan removed by base treatment. Enzymatic hydrolysis of cork oak which was pretreated at higher temperature or concentration was further improved. After pretreatment of cork oak with 13% NaOH at $90^{\circ}C$, the conversion rate of cellulose to fermentable sugars were reached up to 91.3%. At ethanol fermentation with enzymatic hydrolysate from different pretreatment conditions, all enzymatic saccharification liquids were well fermented by Saccharomyces cerevisiae.

Butanol 생산을 위한 동시 당화 발효법의 최적화 (Optimization of Simultaneous Saccharification and Fermentation of Rice Straw to Produce Butanol)

  • 섬유소 폐기물인 볏짚으로부터 butanol를 생산하기 위하여 Clostridium acetobutylicum KCTC1037 와 cellulase(from Trichoderma viride)을 발효액에 동시에 첨가하여 발효시키는 동시당화 발효법(Simultaneous saccharification and fermentation, SSF)을 수행하였다. Alkali 처리한 볏짚을 발효기질로 사용한 결과 그 농도를 25%로 사용하였을 때 최고 150mM의 butanol이 생산되었고, 15% 볏짚을 사용하였을 때는 97mM의 butanol이 생산되었다. 그러나 ball milled 볏짚의 경우 발효산물 중 대부분이 acetate와 butyrate로 주로 산이 생산되었으며 따라서 solventogenesis는 거의 일어나지 않았다. 또한 그 농도별 실험에서 보면 8%의 ball milled 볏짚 사용시 66mM의 butanol이 생산된 반면 그 이상의 농도에서는 butanol 생산량이 점차 감소하는 추세를 보였으며 acetate, butyrate 같은 산은 계속 증가 추세를 보였다. 이것으로 보아 ball milled 볏짚에는 butanol 발효 과정 중 acidogenesis에 서 solventogenesis로의 전이 (shift)를 방해하는 어떤 인자가 있으리라고 추측되었으며 alkali 처리방법에 의해서 이 방해자는 제거될 수 있는 것으로 관찰되었다.

Optimization of ${\beta}$-Glucosidase Production by a Strain of Stereum hirsutum and Its Application in Enzymatic Saccharification

  • A high ${\beta}$-glucosidase (BGL)-producing strain, Stereum hirsutum, was identified and isolated and showed a maximum BGL activity (10.4 U/ml) when cultured with Avicel and tryptone as the carbon and nitrogen sources, respectively. In comparison with other BGLs, BGL obtained from S. hirsutum showed a higher level of activity to cellobiose ($V_{max}$ = 172 U/mg, and $k_{cat}$ = 281/s). Under the optimum conditions (600 rpm, $30^{\circ}C$, and pH 6.0), the maximum BGL activity of 10.4 U/ml with the overall productivity of 74.5 U/l/h was observed. BGL production was scaled up from a laboratory scale (7-L fermenter) to a pilot scale (70-L fermenter). When S. hirsutum was cultured in fed-batch culture with rice straw as the carbon source in a 70-L fermenter, a comparable productivity of 78.6 U/l/h was obtained. Furthermore, S. hirsutum showed high levels of activity of other lignocellulases (cellobiohydrolase, endoglucanase, xylanase, and laccase) that are involved in the saccharification of biomasses. Application of S. hirsutum lignocellulases in the hydrolysis of Pinus densiflora and Catalpa ovata showed saccharification yields of 49.7% and 43.0%, respectively, which were higher than the yield obtained using commercial enzymes.

흰 코뿔소 배설물로부터 분리한 Bacillus pumilus H10-1의 Xylanase 활성 (Xylanase Activity of Bacillus pumilus H10-1 Isolated from Ceratotherium simum Feces)

  • Xylanase have been used to convert the polymetric xylan into fermentable sugars from the production of ethanol and xylitol from plant biomass. The aim of this study was to isolate and identify xylanolytic bacterium from herbivore feces and was to used the xylanase for enzymatic hydrolysis of biomass. Xylanolytic strains were isolated from 59 different feces of herbivores from Seoul Grand Park located in Gwacheon Gyeonggi-do. The xylanolytic strains were selected by congo red staining and DNS method. Total 67 strains isolated from the herbivores feces were tested for xylanase activity. Among the strains, H10-1, which has the highest xylanase activity, was isolated from feces of Ceratotherium simum. The H10-1 strain was identified as Bacillus pumilus based on its morphological/biochemical characteristics and partial 16S rDNA gene sequences. Culture conditions of B. pumilus H10-1 such as initial medium pH, incubation temperature and incubation time were optimized for maximum xylanase production. And also xylanase produced by B. pumilus H10-1 was applied for the saccharification of Miscanthus sacchariflorus cv. 'Geodae 1', which was pretreated with 1.5M NaOH. The optimized culture conditions of B. pumilus H10-1 were pH 9, $30^{\circ}C$ incubation temperature, and 7 day incubation time, respectively. This xylanase activity under the optimized conditions was $20.4{\pm}3.3IU$. The crude xylanase produced by B. pumilus H10-1 was used for the saccharification of xylan derived from pretreated 'Geodae 1'. The saccharification conditions were $50^{\circ}C$, 200 rpm, and 5 days. Saccharification efficiency of pretreated 'Geodae 1' by B. pumilus H10-1 was 8.2%.

  • 백합나무를 원료로 바이오 에탄올을 생산하기 위하여 알칼리 가수분해 처리 후 잔재물을 상업용 혼합 셀룰라아제(Celluclast 1.5L과 Novozym 342)를 사용하여 효소당화 후, 발효하여 바이오 에탄올을 생산하였다. 알칼리 가수분해 후 51.1%의 목재 성분이 회수 되었으며, 이중 셀룰로오스가 82.2%, xylan이 17.6%와 리그닌 2.0%의 조성을 보였다. 백합나무의 알칼리 가수분해과정에서 셀룰로오스 96.9%, xylan 38.0%, 리그닌 5.7%가 잔류하였다. 알칼리 가수분해 잔류물을 상업용 혼합 셀룰라아제에 의한 효소 당화결과, 셀룰로오스의 glucose 전환율은 87.0%였으며 xylan의 xylose로의 전환율은 87.2%였다. 분해된 단당류를 발효효모를 사용하여 바이오 에탄올을 생산하였는데 Saccharomycess cerevisiae 균주는 대부분의 glucose를 발효에 사용하였고, 0.4-1.4%의 소량의 glucose만을 잔류 시킨데 대하여, xylose의 경우는 92.1-99.5%가 잔류하여 이 균주는 발효과정에서 xylose를 거의 사용하지 않았다. 24시간 발효에서 에탄올의 농도는 57.2 g/L수준이었지만 발효 균주에 의한 에탄올 소비로 인하여 48시간 및 72시간 발효에서 에탄올 농도가 각각 56.2 g/L와 54.3 g/L로 점차 감소하였다.

Engineering of Sulfolobus acidocaldarius for Hemicellulosic Biomass Utilization

  • The saccharification of cellulose and hemicellulose is essential for utilizing lignocellulosic biomass as a biofuel. While cellulose is composed of glucose only, hemicelluloses are composed of diverse sugars such as xylose, arabinose, glucose, and galactose. Sulfolobus acidocaldarius is a good potential candidate for biofuel production using hemicellulose as this archaeon simultaneously utilizes various sugars. However, S. acidocaldarius has to be manipulated because the enzyme that breaks down hemicellulose is not present in this species. Here, we engineered S. acidocaldarius to utilize xylan as a carbon source by introducing xylanase and β-xylosidase. Heterologous expression of β-xylosidase enhanced the organism's degradability and utilization of xylooligosaccharides (XOS), but the mutant still failed to grow when xylan was provided as a carbon source. S. acidocaldarius exhibited the ability to degrade xylan into XOS when xylanase was introduced, but no further degradation proceeded after this sole reaction. Following cell growth and enzyme reaction, S. acidocaldarius successfully utilized xylan in the synergy between xylanase and β-xylosidase.

Towards a Miniaturized Culture Screening for Cellulolytic Fungi and Their Agricultural Lignocellulosic Degradation

  • The substantial use of fungal enzymes to degrade lignocellulosic plant biomass has widely been attributed to the extensive requirement of powerful enzyme-producing fungal strains. In this study, a two-step screening procedure for finding cellulolytic fungi, involving a miniaturized culture method with shake-flask fermentation, was proposed and demonstrated. We isolated 297 fungal strains from several cellulose-containing samples found in two different locations in Thailand. By using this screening strategy, we then selected 9 fungal strains based on their potential for cellulase production. Through sequence-based identification of these fungal isolates, 4 species in 4 genera were identified: Aspergillus terreus (3 strains: AG466, AG438 and AG499), Penicillium oxalicum (4 strains: AG452, AG496, AG498 and AG559), Talaromyces siamensis (1 strain: AG548) and Trichoderma afroharzianum (1 strain: AG500). After examining their lignocellulose degradation capacity, our data showed that P. oxalicum AG452 exhibited the highest glucose yield after saccharification of pretreated sugarcane trash, cassava pulp and coffee silverskin. In addition, Ta. siamensis AG548 produced the highest glucose yield after hydrolysis of pretreated sugarcane bagasse. Our study demonstrated that the proposed two-step screening strategy can be further applied for discovering potential cellulolytic fungi isolated from various environmental samples. Meanwhile, the fungal strains isolated in this study will prove useful in the bioconversion of agricultural lignocellulosic residues into valuable biotechnological products.
