Regulatory Effect of Cannabidiol (CBD) on Decreased β-Catenin Expression in Alopecia Models by Testosterone and PMA Treatment in Dermal Papilla Cells

  • Objectives: The hair follicle is composed of more than 20 kinds of cells, and mesoderm derived dermal papilla cells and keratinocytes cooperatively contribute hair growth via Wnt/β-catenin signaling pathway. We are to investigate β-catenin expression and regulatory mechanism by CBD in alopecia hair tissues and dermal papilla cells. Methods: We performed structural and anatomical analyses on alopecia patients derived hair tissues using microscopes. Pharmacological effect of CBD was evaluated by β-catenin expression using RT-PCR and immunostaining experiment. Results: Morphological deformation and loss of cell numbers in hair shaft were observed in alopecia hair tissues. IHC experiment showed that loss of β-catenin expression was shown in inner shaft of the alopecia hair tissues, indicating that β-catenin expression is a key regulatory function during alopecia progression. Consistently, β-catenin expression was decreased in testosterone or PMA treated dermal papilla cells, suggesting that those treatments are referred as a model on molecular mechanism of alopecia using dermal papilla cells. RT-PCR and immunostaining experiments showed that β-catenin expression was decreased in RNA level, as well as decreased β-catenin protein might be resulted from ubiquitination. However, CBD treatment has no changes in gene expression including β-catenin, but the decreased β-catenin expression by testosterone or PMA was restored by CBD pretreatment, suggesting that potential regulatory effect on alopecia induction of testosterone and PMA. Conclusion: CBD might have a modulating function on alopecia caused by hormonal or excess of signaling pathway, and be a promising application for on alopecia treatment.

Conceptus-derived cytokines interleukin-1β and interferon-γ induce the expression of acute phase protein serum amyloid A3 in endometrial epithelia at the time of conceptus implantation in pigs

  • Objective: Serum amyloid A3 (SAA3), an acute phase response protein, plays important roles in opsonization, antimicrobial activity, chemotactic activity, and immunomodulation, but its expression, regulation, and function at the maternal-conceptus interface in pigs are not fully understood. Therefore, we determined the expression of SAA3 in the endometrium throughout the estrous cycle and at the maternal-conceptus interface during pregnancy. Methods: Endometrial tissues from pigs at various stages of the estrous cycle and pregnancy and with conceptuses derived from somatic cell nuclear transfer (SCNT), conceptus tissues during early pregnancy, and chorioallantoic tissues during mid- to late pregnancy were obtained and the expression of SAA3 was analyzed. The effects of the steroid hormones, interleukin-1β (IL1B), and interferon-γ (IFNG) on the expression of SAA3 were determined in endometrial explant cultures. Results: SAA3 was expressed in the endometrium during the estrous cycle and pregnancy, with the highest level on day 12 of pregnancy. The expression of SAA3 in the endometrium was significantly higher on day 12 of pregnancy than during the estrous cycle. Early-stage conceptuses and chorioallantoic tissues during mid to late pregnancy also expressed SAA3. The expression of SAA3 was primarily localized to luminal epithelial cells in the endometrium. In endometrial explant cultures, the expression of SAA3 was induced by increasing doses of IL1B and IFNG. Furthermore, the expression of SAA3 decreased significantly in the endometria of pigs carrying conceptuses derived from SCNT on day 12 of pregnancy. Conclusion: These results suggest that the expression of SAA3 in the endometrium during the implantation period increases in response to conceptus-derived IL1B and IFNG. The failure of those appropriate interactions between the implanting conceptus and the endometrium leads to dysregulation of endometrial SAA3 expression, which could result in pregnancy failure. In addition, SAA3 could be a specific endometrial epithelial marker for conceptus implantation in pigs.

Matrix metalloproteinases: expression and regulation in the endometrium during the estrous cycle and at the maternal-conceptus interface during pregnancy in pigs

  • Objective: Matrix metalloproteinases (MMPs) are a family of endoproteases produced by various tissues and cells and play important roles in angiogenesis, tissue repair, immune response, and endometrial remodeling. However, the expression and function of MMPs in the pig endometrium during the estrous cycle and pregnancy have not been fully elucidated. Thus, we determined the expression, localization, and regulation of MMP2, MMP8, MMP9, MMP12, and MMP13 in the endometrium throughout the estrous cycle and at the maternal-conceptus interface during pregnancy in pigs. Methods: Endometrial tissues during the estrous cycle and pregnancy and conceptus and chorioallantoic tissues during pregnancy were obtained and the expression of MMPs was analyzed. The effects of steroid hormones and cytokines on the expression of MMPs were determined in endometrial explant cultures. Results: Expression levels of MMP12 and MMP13 changed during the estrous cycle, while expression of MMP2, MMP9, MMP12, and MMP13 changed during pregnancy. Expression of MMP2, MMP8, and MMP13 mRNAs was cell type-specific at the maternal-conceptus interface. Gelatin zymography showed that enzymatically active MMP2 was present in endometrial tissues. In endometrial explant cultures, estradiol-17β induced the expression of MMP8 and MMP12, progesterone decreased the expression of MMP12, interleukin-1β increased the expression of MMP2, MMP8, MMP9, and MMP13, and interferon-γ increased the expression of MMP2. Conclusion: These results suggest that MMPs expressed in response to steroids and cytokines play an important role in the establishment and maintenance of pregnancy by regulating endometrial remodeling and processing bioactive molecules in pigs.

탁도 변화가 붕어 조직의 항산화효소 활성에 미치는 영향 (Effect of Turbidity Changes on Antioxidant Enzyme Activity of Carassius auratus Tissues)

  • 본 연구에서는 붕어를 이용하여 탁도 변화에 따른 여러 조직의 항산화효소 활성을 조사하였다. 탁도 50, 100, 150 NTU에서 사육기간에 따른 붕어 조직의 항산화효소 활성의 변화는 50과 100 NTU에서는 대조구에 비해 큰 변화가 없었으며, 고탁도인 150 NTU에서 비교적 두드러진 차이를 나타냈다. 붕어 조직의 항산화능을 DPPH 소거 활성도에서 보면 아가미, 간 조직에서 높은 활성을 보여 고탁도의 사육 조건에서 장기간 사육될 때 비효소적 항산화작용이 일어나는 것으로 생각된다. 효소적 항산화작용은 50, 100, 150NTU로 탁도가 높아짐에 따라 항산화효소의 활성이 증가하였으며 SOD, CAT및 GST의 활성은 표피를 제외한 모든 조직에서 사육 후 20일까지는 증가한 후 사육 후기로 갈수록 감소하는 경향을 나타냈다. 그러나 GR과 GPX의 활성은 거의 모든 조직에서 활성이 매우 낮으므로 어류의 항산화과정에서 이들 효소의 역할은 매우 적을 것으로 생각된다.

안동.임하호에 서식하는 수종 어류의 조직 내 중금속 함량 (Heavy Metal Contents in Tissues of Fishes in Andong and Imha Reservoirs)

  • 안동호와 임하호에 서식하는 끄리(O. bidens), 잉어(C. carpio), 치리(H. eigenmanni), 백조어(C. brevicauda)의 부위에 따른 중금속 축적량을 비교하여 수환경 내 중금속이 어류에 미치는 영향을 알아보고자 하였다. 안동호와 임하호에 서식하는 어류의 중금속 함량은 안동호에 서식하는 어류에서 높은 함량을 보였으며, 중금속에 따라 함량의 차이도 컸다. 안동호와 임하호에 서식하는 어류에서 Cr, Cu, Zn 및 Pb은 뼈 조직에서 높은 함량을 보였고, Cd는 신장 조직에서 높은 함량을 보였다. 부위별 중금속함량은 Pb와 Zn을 제외한 전 중금속에서 높은 차이를 보였으며, 중금속별로는 Zn이 가장 많았고 Cd가 가장 낮았다. 또한 Cr은 안동호의 어류에서 임하호 보다 2배의 높은 함량의 차이를 보였다. 본 연구의 결과 안동호에 서식하는 어류가 임하호에 서식하는 어류 보다 중금속 함량이 높았으며, 이는 안동호의 수질과 저니에서 중금속 함량이 높은 것으로 보아 수환경의 수질과 저니에 포함된 유기물을 어류가 먹이로 섭취함으로써 어류에서도 높은 함량을 보인 것으로 생각된다.

Identification and Expression of Equine MER-Derived miRNAs

  • MicroRNAs (miRNAs) are single-stranded, small RNAs (21-23 nucleotides) that function in gene silencing and translational inhibition via the RNA interference mechanism. Most miRNAs originate from host genomic regions, such as intergenic regions, introns, exons, and transposable elements (TEs). Here, we focused on the palindromic structure of medium reiteration frequencies (MERs), which are similar to precursor miRNAs. Five MER consensus sequences (MER5A1, MER53, MER81, MER91C, and MER117) were matched with paralogous transcripts predicted to be precursor miRNAs in the horse genome (equCab2) and located in either intergenic regions or introns. The MER5A1, MER53, and MER91C sequences obtained from RepeatMasker were matched with the eca-miR-544b, eca-miR-1302, and eca-miR-652 precursor sequences derived from Ensembl transcript database, respectively. Each precursor form was anticipated to yield two mature forms, and we confirmed miRNA expression in six different tissues (cerebrum, cerebellum, lung, spleen, adrenal gland, and duodenum) of one thoroughbred horse. MER5A1-derived miRNAs generally showed significantly higher expression in the lung than in other tissues. MER91C-derived miRNA-5p also showed significantly higher expression in the duodenum than in other tissues (cerebellum, lung, spleen, and adrenal gland). The MER117-overlapped expressed sequence tag generated polycistronic miRNAs, which showed higher expression in the duodenum than other tissues. These data indicate that horse MER transposons encode miRNAs that are expressed in several tissues and are thought to have biological functions.

Ecological health assessments using multiple parameters of fish blood tissues to community along with water chemistry in urban streams

  • The objectives of this study were to identify multi-level stressors from blood biomarkers to community-level bioindicators and diagnose the stream ecosystem health in polluted streams. Blood chemistry such as total protein ($T_{Pro}$), blood urea nitrogen ($B_{UN}$), total cholesterol ($T_{Cho}$) and $A_{lb}$umin ($A_{lb}$) were analyzed from sentinel fish tissues; the functions of kidney, gill and liver were significantly decreased in the impacted zone ($I_z$), compared to the control zone ($C_z$). Histopathological analysis showed that fish liver tissues were normal in the $C_z$. Fish liver tissues in the $I_z$, however, showed large cell necrosis and degeneration and also had moderate lobular inflammation and inflammatory cell infiltration of lymphocytic histocytes. Species biotic index (SBI) at species level and stream health assessment (SHA) at community level indicated that chemical impacts were evident in the $I_z$ (ecological health; poor - very poor), and this was matched with the blood tissue analysis and histopathological analysis. The impairments of the streams were supported by water chemistry analysis (nitrogen, phosphorus). Tolerance guild analysis and trophic guild analysis of fish were showed significant differences (P < 0.01) between $C_z$ and $I_z$. Overall, multiple parameter analysis from biomarker level (blood tissues) to bioindicator level (community health) showed significantly greater impacts in the $I_z$ than $C_z$. This approach may be effective as a monitoring tool in identifying the multilateral and forthcoming problems related to chemical pollution and habitat degradation of stream ecosystems.

Long Terminal Repeat of an Endogenous Retrovirus HERV-K Family from Human Liver and Kidney cDNA

  • Long terminal repeat (LTR) of human endogenous retrovirus K family (HERV-K) has been found to be coexpressed with sequences of closely located genes. We examined the transcribed HERV-K LTR elements in human liver and kidney tissues. Using the cDNA synthesized from mRNA of human liver and kidney, we performed PCR amplification and identified six HERV-K LTR elements. Those LTR elements showed a high degree of sequence similarity (93.3∼96.6%) with human-specific LTR. A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-K LTR elements (Liv-1, 2, 3 and Kid-1, 2, 3) were belonged to group I. Our data suggests that HERV-K LTR elements are active on human liver and kidney tissues and may represent a source of genetic variation connected to human disease.
