Isolation of Bacteria Producing a B-Cell-Specific Biological Response Modifier Found in Korean Fermented Soybean Paste

  • In a previous study, a biological response modifier (BRM) specifically enhancing the function of B-cells was isolated from Korean fermented soybean paste (Kfsp), but not from non-fermented soybeans. In this study, we attempted to isolate the bacteria producing the BRM from Kfsp (KfspBRM) by ELISA using anti-KfspBRM and by B-cell proliferation. Five bacteria whose culture supernatants showed the BRM activities were isolated, and one of them was identified as Bacillus licheniformis E1. The bacterial BRM (bBRM) originated from a slime layer of B. licheniformis El had a molecular weight of 1,594 kDa, and contained $33\%\;(w/w)$ of reduced sugar and $4.6\%\;(w/w)$ of protein content. The bBRM appeared to be a glycoprotein that is physically, structurally, and functionally similar to the KfspBRM, suggesting that the isolates including B. licheniformis El may produce the KfspBRM in the fermentation process of soybean paste. The mass production of the BRM by the bacterium may help to study B-cells in immunology, and the enrichment of the BRM in Kfsp may help patients in future who are medically in need of potentiation of B-cell proliferation and antibody production.

면역증강물질 강화 청국장 발효 (The Bacterial Biological Response Modifier Enriched Chungkookjang Fermentation)

  • 전통된장에서 B 세포만을 선택적으로 증가시키는 면역증강 물질을 생산하는 B. licheniformis E1을 분리한 바 있다(22). 이를 이용하여 면역증강 청국장을 제조하였으나 관능적인 측면에서 기호도가 좋지 않았고 아미노태 질소함량이 식품공전의 규격(280 mg%)에 적합하지 않았다. 이를 보완하기 위해 시판 청국장으로부터 발효미생물을 분리하였다. 단백질 분해효소 활성과 청국장 발효제조시 기호도에서도 가장 우수한 청국장 발효미생물이 선발되었고 동정한 결과 B. subtilis S2로 명명하였다. 청국장 발효에 있어서 최적 발효온도와 최적 발효시간을 조사한 결과 $40^{\circ}C$에서 48시간 동안 발효하였을 때의 기호도가 가장 우수하였다. 단일균주 B. licheniformis E1, B. subtilis S2 및 B. licheniformis E1과 B. subtilis S2의 혼합균주를 각각 접종하여 발효제조한 청국장과 시판 청국장을 대조구로 사용하여 청국장의 기호도, 이화학적 특성조사와 면역증강 활성 비교를 조사하였는데, B. licheniformis E1과 B. subtilis S2의 혼합균주를 접종하여 제조한 청국장에서 기호도가 가장 우수하였고 아미노태 질소함량과 조단백질 함량은 359 mg%와 45.6%로 식품공전의 규격에 적합하였다. 청국장으로부터 면역증강 물질을 분리하여 마우스 비장 림프구의 증식 및 억제 현상을 측정한 결과, 20,480 unit으로 림프구의 증식에 있어서도 가장 효과적인 것으로 확인되었다.

  • Ginsan, a polysaccharide extracted from Panax ginseng. was earlier scrutinized for a biological-response modifier. We further studied the protective and restorative activity of Ginsan against sublethal dose irradiation owing to increase production of endogenous hematopoietic growth factors such as IL-1. TNF-${\alpha}$. IL-6, GM-CSF. Which induce strong redox-emzyme elevation. Exposing to radiation induces reactive oxygen species (ROS). which play an important causative role in radiation damage. Xenorhabuds nematophila 세균 배양액 유래 미확인 생리활성 물질의 비티플러스 살충력 상승효과 (Enhancement of Bt-Plus Toxicity by Unidentified Biological Response Modifiers Derived from the Bacterial Culture Broth of Xenornabdus nematiphila)

  • 비티 포자와 Xenorhabdus nematophila (Xn)의 배양액을 혼합하여 비티플러스가 개발되었다. 높은 살충력에도 불구하고 비티플러스는 다양한 해충에 대한 넓은 살충범위를 보이지 않는 한계가 있다. 본 연구에서는 Xn 대사물질 첨가를 통한 파밤나방과 같은 비감수성 해충에 대한 비티플러스의 살충력 향상에 초점을 맞추었다. Xn의 주요 대사물질인 oxindole (OI)과 benzylideneacetone (BZA)는 비티의 살충력을 향상시킨다고 보고되었다. 본 연구에서 OI 또는 BZA의 첨가는 비티플러스의 살충력을 향상시켰다. 그러나 동결건조된 Xn 배양액의 첨가는 보다 낮은 농도의 OI 또는 BZA로도 충분히 비피플러스의 살충력을 상승시켰다. HPLC 분석에서 Xn 배양액에 최소 12개의 대사물질이 포함되어 있는 것을 확인하였다. 이러한 결과는 OI와 BZA 외에도 Xn 대사물질에 생리활성물질이 존재하는 것을 제시한다.

Effect of Silsosangami on Neutrophil Functions and Cyclo-oxygenase-2

  • Silsosangami(SSG) is a formula of oriental medicines as an effective biological response modifier for augmenting host homeostasis of body circulation. Also SSG has been known to have an anti-diabetic activity and anti-platelet aggregation activity. The present study was undertaken to examine the effects of a new SSG on murine macrophage and human neutrophil functions as well as on several enzymes relevant to the inflammatory process. The results of the present study indicate that SSG exerts anti-inflammatory effects related to the inhibition of neutrophil functions and of PGE2 production, which could be due to a decreased expression of and COX-2.

Roles of SUMO in Plants

  • The covalent conjugation of SUMO(Small Ubiquitin-related MOdifier) protein to its substrates regulates numerous cellular processes, including protein stability and activity in eukaryotes as well as in plants. In this present review, we summarize biochemical aspects of SUMO conjugation and deconjugation and the functions of SUMO and sumoylation-related proteins in Arabidopsis and other plants. In particular, we provide an overview of the roles of the SUMO in widely different biological processes including the ABA response, floral induction, pathogen defense, abiotic stresses and hormone signaling. Furthermore, we explore the possible roles of SUMO in embryo and seed development.

단삼사물탕이 조혈작용에 미치는 영향 (The Effect of DanSamSaMul-Tang on Hematopoiesis)

  • Objectives : Korean traditional medicinal herbs and some formulas have been used to improve the function of the hematopoietic system. The present research is to examine the effect of DanSamSaMul- Tang(DSSMT) as a biological response modifier related to hemato-potentiating function using bone marrow cell as a testing system. Methods : Mice with leukopenia and thrombocytopenia induced by CTX were treated with DSSMT Extracts. Then we observed cytokines mRNA expression level, the number of CFU-GEMM and BFU-E, and the expression of TPO and SCF in bone marrow cells of mice. Results : In the DSSMT group, the gene expressions of hematopoietic cytokines were increased significantly and the number of CFU-GEMM and BFU-E of this group was significantly increased compared to the group treated only with EPO and IL-3. The expressions of TPO and SCF in the group treated with DSSMT were higher than those of the control group. Conclusions : We showed that DSSMT has a good effect on the hematopoietic system. This could also be used as important research data for Korean oriental medicine about hemato-potentiating.

Potential Health-Promoting Benefits of Paraprobiotics, Inactivated Probiotic Cells

  • Viability plays an important role in the beneficial microbes (probiotics) to produce health benefits. However, this idea has been changed after the invention of the term "paraprobiotics," indicating that non-viable microbes could produce health benefits similar to those produced by live probiotics. Occasionally, it might be dangerous to administer live probiotics to people with weak immunity. In such cases, ingestion of paraprobiotics could be a potential alternative. The definition of paraprobiotics refers to the use of inactivated (non-viable) microbial cells or cell fractions to provide health benefits to the consumer. Paraprobiotics have attracted much attention because of their long shelf life, safety, and beneficial effects, such as modulation of immunity, modification of biological responses, reduction of cholesterol, anti-inflammatory, and antiproliferative properties. These features indicate that paraprobiotics may play a vital role in improving the health of the consumer by enhancing particular physiological functions, even though the exact underlying mechanisms have not yet been completely elucidated. In this mini-review, we briefly discuss the historical backgrounds of paraprobiotics and evidence of their health-promoting effects, prophylactic, and therapeutic properties.

천연물로부터 항암면역증강물질 탐색연구 (Screening of Antineoplastic Immunomodulator from Herbal Medicines)

  • Currently, cancer is the primary cause of death and 50% of cancer patients are incurable by surgery, radiotherapy and chemotherapy. Therefore, immunotherpy is interested as the fourth remedy. Biological response modifier (BRM), such as organometallic compounds, glycoproteins, polysaccharides and other natural products. Is the one which can enhance the immune response against cancer cell. To develop new BRM from natural sources, we investigated 63 species Korean traditional medicines by observing the mitogenic activity to splenocytes, generation of activated killer cells and activation of macrophages. Finally, we selected 9 species including Angelicae gigantis Radix, Mori Cortex Radicis, Arisaematis Tuber, Salviae Radix, Cnidii Rhizoma, Ligusti Fructus, Pasoraliae Semen, Loranthi Ramulus, Ginseng Radix. Bioassay-guided fractionation and purification is undergoing.

고춧가루 첨가 식이가 흰주의 체액성 면역기능에 미치는 영향 (Effect of Dietary Hot Red Pepper Powder on Humoral Immune Response in Rat)

  • 고추가루 첨가 식이가 체액성 면역기능에 미치는 영향을 조사하였다. 실험동무로 흰쥐를 사용하여 대조군과 2%, 5%, 10% 고추가루 첨가 식이군을 설정한 후 27일간 실험식이로 사육하였다. 실험종료 10일 전에 각군의 흰쥐를 면양적혈구로 세차례 면역시킨 후 항체생성 세포수 및 혈청 중 항체 농도 등 체액성 면역기능을 중심으로 조사하였다. 그 결과, 대조군에 비해 고추가루 첨가 식이군의 항체 생성 세포수, 적혈구 응집반응 및 혈청중 항체 농도가 높은 것으로 나타났으며, 또한 이때 고추가루 첨가식이군의 각 장기조직 중 AsA 함량은 대조군에 비해 높은 것으로 나타났다. 이들 결과로 보아, 고추성분은 체액성 면역기능의 활성화에 기여하는 물질을 함유하고 있을 뿐만이 아니라, AsA의 공급효과가 뛰어난 것이 확인되었다.
