Stabilization of Quinonoid Intermediate E-Q by Glu32 of D-Amino Acid Transaminase

  • The stable anchorage of pyridoxal 5'-phosphate (PLP) in the active site of D-amino acid transaminase (D-AT) is crucial for the enzyme catalysis. The three-dimensional structure of D-AT revealed that Glu32 is one of the active site groups that may playa role in PLP binding. To prove the role of Glu32 in PLP stability, we firstly checked the rate of the potential rate-limiting step. The kinetic analysis showed that the rate of the ${\alpha}$-deprotonation step reduced to 26-folds in E32A mutant enzyme. Spectral analyses of the reaction of D-AT with D-serine revealed that the E32A mutant enzyme failed to stabilize the key enzyme-substrate intermediate, namely a quinonoid intermediate (E-Q). Finally, analysis of circular dichroism (CD) on the wild-type and E32A mutant enzymes showed that the optical activity of PLP in the enzyme active site was lost by the removal of the carboxylic group, proving that Glu32 is indeed involved in the cofactor anchorage. The results suggested that the electrostatic interaction network through the groups from PLP, Glu32, His47, and Arg50, which was observed from the three-dimensional structure of the enzyme, plays a crucial role in the stable anchorage of the cofactor to give necessary torsion to the plane of the cofactor-substrate complex.

화학.생물학적 황화수소 제거 공정에 있어서 고정화 세포를 이용한 철산화 속도 증진 (Enhancement of Iron Oxidation Rate by Immobilized Cells in Chemo-biological Process for $H_2S$ Removal)

  • 본 연구는 황화수소 가스 제거를 위한 화학 . 생물학적 복합공정의 전체 효율을 결정하는 생물반응기에 의한 Fe(II) 산화 속도를 증진시키고자 배지 최적화와 함께 고정화 세포 반응기 시스템을 개발하는 것을 목표로 하였다. 고정화 담체로는 celite beads를 선정하였고 반응기는 airlift type의 반응기를 사용하였다. 먼저, 철침전물(jarosite) 최소화 배지 개발을 위한 연구를 수행하였고, 그 결과 기존에 사용되어졌던 9K 배지 사용시 질소원 및 인 공급원으로 사용된 $(NH_4)_2SO_4$$K_2HPO_4$가 제외되고 $(NH_4)_2HPO_4$가 대체된 M16 배지를 사용하였을 때 Fe(II) 산화 속도의 감소가 없음은 물론 jarosite 생성이 거의 없음을 확인할 수 있었다. 또한 M16 배지의 초기 pH 변화에 따른 철 산화 거동 및 jarosite 생성 측면을 조사한 결과 초기 pH 1.8이 최적임을 확인할 수 있었다. 다음으로 celite beads에 세포를 고정화 한 후 jarosite 생성 최소화 배지(M16 배지)에서의 고정화 세포에 의한 철 산화 거동을 고찰하였다. 반복 회분식 배양 결과, 회분식 배양의 결과 최대 Fe(II) 산화 속도에 이르러서는 거의 일정해지는 결과를 보였다. 반분회분식 배양의 결과 최대 Fe(II) 산화 속도가 2.33 g/L . h이었다. 연속 조업을 수행한 결과 현탁 세포의 최대 비성장 속도보다 높은 희석속도에서 최대 Fe(II) 산화 속도가 결정되었다. 최대 Fe(II) 산화속도는 2.14 g/L . h이었으며, 이??의 희석속도는 0.25 $h^{-1}$이었다. 반복 회분식 및 연속 조업기간 동안 Fe(total)의 농도는 초기 Fe(II) 농도와 거의 비슷하게 유지되었고 이러한 사실로부터 jarosite가 거의 생성되지 않았음을 확인하였다.

주정오니를 활용한 수중의 카드뮴(Cd) 흡착 특성 (Adsorption Characteristics of Cadmium ions from Aqueous Solution using by-product of Brewing)

  • 본 연구는 맥주제조공장에서 발생하는 주정오니의 영양염류 용출특성 및 폐수중 존재하는 카드뮴(Cd)이온에 대한 흡착특성을 알아보고 이를 통하여 생물흡착제의 적용가능성을 알아보기 위하여 수행하였다. 흡착실험에 사용된 흡착제는 회화를 하지 않은 원시료(M-Raw)와 $300^{\circ}C$, $400^{\circ}C$, $500^{\circ}C$로 회화된 시료를 준비하여 증류수를 이용하여 용출실험을 하였으며 그 결과 원시료(M-Raw)의 경우 수중에 용출된 인 질소의 양은 $400^{\circ}C$로 회화시킨 시료(M-400)에 비해 각각 7배, 11배 이상의 용출량을 나타내었으며 이는 회화로 인하여 시료를 구성하고 있는 관능기가 대기중으로 방출되어 수중에 용출되는 양이 감소한 결과로 생각된다. TGA를 이용한 회화 실험결과 $260^{\circ}C\sim380^{\circ}C$범위에서는 급격한 무게감량이 일어 났으며, $380^{\circ}C$ 이후에는 완만한 무게감량이 일어나는 것을 볼 수 있는데 이는 생물흡착제를 구성하고 있는 성분들(cellulose, hemicellulose, lignin)의 서로 다른 분해온도로 인한 결과 로 생각된다. 또한, 회화 전 후의 표면변화를 관찰한 결과 회화를 거치지 않은 시료는 표면이 거칠고 불순물이 존재하는 반면, 회화를 끝낸 시료의 경우 표면의 불순물이 제거되고, 동시에 표면의 갈라짐을 관찰할 수 있었다. 회화 전 후의 생물흡착제 표면변화 및 특성을 알아보기 위하여 미세영상장비와 FT-IR을 사용한 결과, 회화온도가 올라갈수록 흡착제표면의 갈라짐이 관찰되었으며 관능기의 개수는 점차로 줄어드는 결과를 나타내었다. 회화 전 후 시료의 FT-IR 분석결과 회화전 시료의 경우 3300 $cm^{-1}$ 부근에서 hydroxyl group 이 나타났으며 1080~1730 $cm^{-1}$ 범위에서는 여려종류의 관능기를 관찰할 수 있었다. 회화 전 1600~1080 $cm^{-1}$ 부근에서 관찰된 primary amine, secondary amine, aliphatic nitro compounds, aromatic phosphate 등의 작용기는 회화 온도가 증가할수록 spectrum이 점차로 줄어드는 결과를 나타내었다. 중금속 흡착 실험결과 본 실험에서 사용된 생물흡착제의 카드뮴이온 제거율은 농도위 20 mg/L 이하에서 60~91%로 나타났으며, 흡착 평형을 이루는 시간은 3시간이 소요되었다. 실험결과를 Freundlich 및 Langmuir 모델에 적용시킨 결과 Langmuir 모델에 더 잘 부합되는 결과를 나타내었으며, 최대흡착량($Q_{max}$)은 회화를 시키지 않은 시료(M-Raw)의 경우 28.17 mg/g으로 매우 높은 수준을 나타내었다.

하.폐수 고도처리시 F/M비 변화에 따른 유기산 잔류 농도가 슬러지 침강성에 미치는 영향 (Influence of Organic Acids Residual Concentration by the Change of F/M Ratio on Sludge Settleability in Advanced Sewage Treatment Processes)

  • 생물학적 고도처리는 혐기, 무산소, 호기의 조건을 반복하거나 재배열함으로서 이루어지는데 이때 혐기조와 무산소조에서는 유기산이 발생한다. 여기서 발생한 유기산은 질소와 인의 제거와 슬러지침강성에 중요한 인자로 작용한다. 따라서 본 연구에서는 하 폐수 고도처리 시 F/M비 변화에 따른 유기산의 발생정도와 잔류농도 변화와 기에 따른 유기물과 질소의 제거 특성과 처리효율 및 슬러지 침강성과의 관계를 모색하기 위해 특성을 검토 하였다. 고도처리를 위해 $A^2/O$ 공정을 사용하였고 MLSS의 농도의 변화로 F/M비를 조절하였다. F/M비를 $0.16{\sim}0.08$ kg-BOD/kg-MLSS/day로 변화 시켰을 때 F/M비에 감소에 따라 회분식 반응기에 의한 유기산의 생성량의 증가하였고 잔류 유기산 농도는 감소하였다가 증가하였다. F/M비가 $0.16kg/kg{\cdot}d$ 실험 조건에서 SVI와 SS는 높게 나타났으며 F/M비가 $0.11{\sim}0.13kg/kg{\cdot}d$로 높아짐에 따라 감소하여 양호한 상태를 보이다 F/M비가 증가함에 SVI와 SS도 지속적으로 증가하여 $0.08kg/kg{\cdot}d$에서 높은 SVI와 SS 농도를 나타내었다. 무산소조의 유기산 잔류 농도 그리고 탈질률을 비교한 결과 무산소조의 유기산 잔류량이 적을수록 탈질률은 증가하였다. 슬러지 침강성과 질소 제거효율을 고려한 최적의 유기산 잔류 농도는 $1.4{\sim}2.2$ mg/L이며 이때의 F/M비는 $0.11{\sim}0.13$ kg-BOD/kg-MLSS/day범위로 나타났다.

혈소판유래성장인자-BB가 성견 치근이개부병변의 조직재생에 미치는 효과 (THE EFFECTS OF THE PLATELET-DERIVED GROWTH FACTOR-BB ON THE PERIODONTAL TISSUE REGENERATION OF THE FURCATION INVOLVEMENT OF DOGS)

  • 치주질환 진행의 최종적인 결과는 하반 지지조직의 소실로 치아상실을 초래하는 것이다. 이러한 치주질환의 이상적인 치유형태는 부착상실 예방을 비롯 상실된 조직의 재상 즉 신생 치조골과 백악질이 형성되고 두 조직사이에 치주인대가 재형성되는 것이다. 최근까지의 치주조직재생을 위한 처치법으로 이환된 병소부의 단순제거, 치관변위판막술, 약제의 치근면처리법, 조직유도재생술, 골전도물질 삽입, 골유도 혹 골형성물질 사용등 다양한 방법이 제시되었으나 아직 이상적인 치료법은 밝혀지지 않고 있다. 치아지지의 가장 중요한 역할을 하는 치조골의 재생에 대해 많은 연구들과 아울러 조직화학적 연구에서 Polypeptide Growth Factor(PGF) 가 다양한 종류의 세포증식과 이주 및 기질합성에 촉진효과가 있다고 하여 조직재생에 사용 가능성이 보고된 바 있었다. 이 PGF중 혈소판유래성장인자가 섬유아세포와 골아세포의 유사분열 및 단백질합성에 촉진작용이 있으며 조직재생에 영향을 미친다는 보고도 있었다. 본 연구의 목적은 총 8 마리의 실험동물을 이용하여 치근이개부병변을 형성한 후 혈소판유래성장인자를 처리하고 기존 조직결손부에 사용하였던 Tricalcium Phosphate(TCP) 와 콜라젠을 성장인자 함유매개체로 하여 이개부병변에 병용삽입한 경우 치유과정에 미치는 효과를 규명하여 임상적용의 가능성을 규명하고 실제 임상적용방법을 개발할 목적으로 실시하였다. 실험동물 Pentobarbital Sodium으로 전신마취를 시킨후, 초음파치석제거기등을 이용하여 구강위생을 청결하게 한 다음, 치은열구절개를 이용하여 전층판막을 형성하였다. 피질골과 치조골을 삭제하고 형성된 이개부 결손부에 치근면활택술을 시행하였으며, 구연산으로 치면처리 후, 계획된 재료를 삽입하고, 치관변위판막술과 유사한 형태의 봉합을 시행하였다. 실험동물을 2, 4, 8, 12 주에 관류고정과 아울러 회생시켜 악골을 채취하고 통법에 의해 후고정, 탈회, 탈수과정을 거쳐 파라핀으로 포매한 후 $7{\mu}m$두께로 절편하고 H & E 염색후 광학현미경으로 관찰하여 다음과 같은 결과를 얻었다. PDGF-BB 만 처리한 군에서는 2주 소견에서부터 결손부 전체에 걸쳐 활성도가 높은 조골세포들이 균일하게 분포하면서 이들로부터 생성된 골양조직이 기초적인 골소주의 형태를 이루고 있음이 관찰되었으며 그후 매우 빠른 골형성이 계속되어 8주 소견에서는 결손부 정상에 이르기까지 성숙된 치밀골이 채워져 있었다. 신생골형성의 전반적 형태는 치근면의 외형에 따라 치근이개부상단부로 형성되는 양상이었다. PDGF-BB 군에서 신생백악질의 형성은 2주소견 에서는 미약하였으나 4주이후 치근면에 수직으로 배열된 교원섬유들과 함께 균일한 두께로 형성되기 시작하여, 8주에 이르러서는 비 손상부위에서보다 더욱 두터운 신생백악질이 형성되었고 전형적인 샤피스씨 섬유가 완성되어 있음이 관찰되었다. PDGF-BB와 TCP를 병용한 경우 및 PDGF-BB와 콜라젠을 병용한 경우에서는 PDGF-BB군에 비해 신생골 및 신생백악질의 형성, 치주인대강의 발달이 상대적으로 미약하였으며 이러한 현상은 수술후 초기단계에서 더욱 두드러져 함유매개체로서의 기능을 기대하였던 이들 이식재들이 도리어 급속하게 분화 증식되는 세포들의 이동에 장애물로서 작용하는 것으로 나타났다. 결론적으로 PDGF-BB의 치근면 처리가 치근이 개부병변 치료에 있어 전반적으로 조직재생의 속도가 빠르고 그 치유양상도 시간경과에 따라 치주조직 고유형태로 진행됨이 관찰되어 동일 병소치료에 응용가능성을 확인하였다. 또한 차후 결손부에 주입방법과 성장인자의 관리법 및 적용량 그리고 적응중의 규명을 위해 연이은 연구가 있어야 할것으로 사료된다.

안동시 저수지에서의 대량 어류 폐사에 대한 원인과 대책에 관한 연구 (Study on Causes and Countermeasures for the Mass Death of Fish in Reservoirs in Andong-si)

  • This study focused on determining the specific causes and prevention methods of mass fish deaths occurred in five reservoirs (Gagugi, Neupgokgi, Danggokgi, Sagokji, and Hangokji) in Andong-si. For this purpose, a survey of agricultural land and livestock in the upper part of the reservoirs and analysis of water quality in the reservoir irrespective of whether it rains or not were conducted. We attempted to examine the changes in dissolved oxygen (DO) in the surface and bottom layers of reservoirs and changes in DO depending on the amount of livestock compost and time. Based on the above investigations, treatment plans were established to efficiently control the inflow of contaminated water into reservoirs. The rainfall and farmland areas in the upper part of the reservoir were investigated using Google and aviation data provided by the Ministry of Land, Infrastructure, and Transport. The current status of livestock farms distributed around the reservoirs was also examined because compost from these farms can flow into the reservoir when it rains. Various water quality parameters, such as phosphate phosphorus (PO4-P) and ammonium nitrogen (NH3-N), were analyzed and compared for each reservoir during the rainy season. Changes in the DO concentration and electrical conductivity (EC) were also observed at the inlet of the reservoir during raining using an automated instrument. In addition, DO was measured until the concentration reached 0 ppm in 10 min by adding livestock compost at various concentrations (0.05%, 0.1%, 0.3%, and 0.5% by wt.), where the concentration of the livestock compost represents the relative weight of rainwater. The DO concentration in the surface layer of reservoirs was 3.7 to 5.3 ppm, which is sufficient for fish survival. However, the fish could not survive at the bottom layer with DO concentration of 0.0-2.1 ppm. When the livestock compost was 0.3%, DO required 10-19 h to reach 0 ppm. Considering these results, it was confirmed that the DO in the bottom layer of the reservoir could easily change to an anaerobic state within 24 h when the livestock compost in the rainwater exceeds 0.3%. The results show that the direct cause of fish mortality is the inflow of excessive livestock compost into reservoirs during the first rainfall in spring. All the surveyed reservoirs had relatively good topographical features for the inflow of compost generated from livestock farms. This keeps the bottom layer of the reservoir free of oxygen. Therefore, to prevent fish death due to insufficient DO in the reservoir, measures should be undertaken to limit the amount of livestock compost flowing into the reservoir within 0.3%, which has been experimentally determined. As a basic countermeasure, minerals such as limestone, dolomite, and magnesia containing calcium and magnesium should be added to the compost of livestock farms around the reservoir. These minerals have excellent pollutant removal capabilities when sprayed onto the compost. In addition, measures should be taken to prevent fish death according to the characteristics of each reservoir.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.
