Genetic Characteristics of 207 Microsatellite Markers in the Korean Population and in other Asian Populations

  • Microsatellites, short tandem repeats, are useful markers for genetic analysis because of their high frequency of occurrence over the genome, high information content due to variable repeat lengths, and ease of typing. To establish a panel of microsatellite markers useful for genetic studies of the Korean population, the allele frequencies and heterozygosities of 207 microsatellite markers in 119 unrelated Korean, Indian and Pakistani individuals were compared. The average heterozygosity of the Korean population was 0.71, similar to that of the Indian and Pakistani populations. More than 80% of the markers showed heterozygosity of over 0.6 and were valuable as genetic markers for genome-wide screening for disease susceptibility loci in these populations. To identify the allelic distributions of the multilocus genetic data from these microsatellite markers, the population structures were assessed by clustering. These markers supported, with the most probability, three clustering groups corresponding to the three geographical populations. When we assumed only two hypothetical clusters (K), the Korean population was separate from the others, suggesting a relatively deep divergence of the Korean population. The present 207 microsatellite markers appear to reflect the historical and geographical origins of the different populations as well as displaying a similar degree of variation to that seen in previously published genetic data. Thus, these markers will be useful as a reference for human genetic studies on Asians.

Development of microsatellite markers to assess the genetic diversity of the red-tongue viper, Gloydius ussuriensis (Reptilia: Viperidae) on the Korean Peninsula

  • The red-tongue viper(Gloydius ussuriensis) is one of only three species of the genus Gloydius found in South Korea. Gloydius ussuriensis has a narrow activity radius and is distributed nationwide, and this species was reported to have the largest population among the Korean species in genus Gloydius. Preliminary results of a phylogenetic analysis using part of the mitochondrial DNA indicated that domestic G. ussuriensis is not comprised of monophyletic groups, and morphological analysis showed differences between domestic populations. In this study, we developed 17 microsatellites for the analysis of G. ussuriensis genetic diversity based on these characteristics. These microsatellites were developed using six multiplex panels, which could be employed to validate 80 G. ussuriensis specimens from different geographical regions in South Korea. The average number of alleles per locus was 12.2 and ranged from 4 to 25 alleles; the observed heterozygosity ranged from 0.238 to 0.950 and the expected heterozygosity ranged from 0.213 to 0.933. As a result of assessing four inland populations, a high level of genetic diversity was confirmed. These newly developed markers will be useful for further studies on the population structure and evolutionary history of the G. ussuriensis.

Identification of Quantitative Trait Loci Associated with Traits of Soybean for Sprout

  • The identification of quantitative trait loci (QTL) has the potential to enhance the efficiency of im- proving food processing traits of soybean. In this study, 92 restriction fragment length polymorphism (RFLP) loci and two morphological markers (W$_1$ and T) were used to identify QTL associated with food processing traits of soybean for sprout in 83 F$_2$-derived lines from a cross of 'Pureun' x 'Jinpum 2'. The genetic map consisted of 76 loci which covered about 760 cM and converged into 20 linkage groups. Eighteen markers remained unlinked. Phenotypic data were collected for hypocotyl length, abnormal seedling rate, and sprout yield seven days after seed germination at 2$0^{\circ}C$. Based on the single-factor analysis of variance, eight independent markers were associated with hypocotyl length. Four of seven markers associated with abnormal seedling rate were identified as independent. Seven loci were associated with sprout yield. For three different traits, much of genetic variation was explained by the identified QTL in this population. Several RFLP markers in linkage group (LG) Bl were detected as being associated with three traits, providing a genetic explanation for the biological correlation of sprout yield with hypocotyl length (r=OA07***) and with abnormal seedling rate (r=-406***).

Development and characterization of 21 microsatellite markers in Daphne kiusiana, an evergreen broad-leaved shrub endemic to Korea and Japan

  • Microsatellite markers were isolated for Daphne kiusiana var. kiusiana (Thymelaeaceae), an evergreen broad-leaved shrub endemic to Korea and Japan. Because its populations in Jeju Island are morphologically controversial, and consistently threatened by anthropogenic pressures, taxonomic delimitation and conservation effort are required at the genetic level. We developed 21 polymorphic microsatellite loci from Next Generation Sequencing data. The primer set included di-, tri-, and tetra-nucleotide repeats. Variability in the markers was tested for 80 individuals of D. kiusiana from three natural populations in Jeju Island and Japan. Among the 21 loci, three were unavailable for population JKJU of Japan. The Neighbor-Joining tree based on microsatellite markers described here classified the three populations into two groups according to geographical or morphological traits. These will be a powerful genetics tool for determining the taxonomic boundary and establishing suitable conservation strategies for D. kiusiana in Jeju Island.

  • Purpose: The purpose of this study was to investigate the effects of a 12-week Tai Chi on the bone mineral density and bone metabolic markers in postmenopausal women Methods: Data were collected from March to July, 2009. Fifty postmenopausal women were recruited for the study. Twenty two women were allocated to experimental group, and 28 to control group. The experimental group underwent Tai Chi exercise twice a week for twelve weeks. The control group was only notified with results of bone mineral density and bone metabolic markers. Bone mineral density was measured by using of DTX-200 (Osteometer MediTech, Hawthorne, CA, USA) at distal radius site and bone metabolic markers were measured by radioimmunoassay method. Collected data were analyzed by t-test, $X^2$-test, and Mann-Whitney test. Results: After 12 weeks of treatment, the Tai Chi group showed a significant difference in bone mineral density compared to control group but no significant effect on osteocalcin and deoxypyridinoline level. Conclusion: Our results suggest that 12 weeks of Tai chi may delay bone loss in postmenopausal women.

  • Objectives Memory impairment is a very important mental health issue for elderly and adults. Mild cognitive impairment (MCI) is a prodromal stage of Alzheimer's disease (AD). Early detection of the prodromal stage of patients with AD is an important topic of interest for both mental health clinicians and policy makers. Methods Electroencephalograpgy (EEG) has been used as a possible biological marker for patients with MCI, and AD. In this review, we will summarize the clinical implications of EEG and ERP as a biological marker for AD and MCI. Results EEG power density, functional coupling, spectral coherence, synchronization, and connectivity were analyzed and proved their clinical efficacy in patients with the prodromal stage of AD. Serial studies on late event-related potentials (ERPs) were also conducted in MCI patients as well as healthy elders. Even though these EEG and ERP studies have some limitations for their design and method, their clinical implications are increasing rapidly. Conclusion EEG and ERP can be used as biological markers of AD and MCI. Also they can be used as useful tools for early detection of AD and MCI patients. They are useful and sensitive research tools for AD and MCI patients. However, some problems remain to be solved until they can be practical measures in clinical setting.

Development and Characterization, and Application of Ten Polymorphic Microsatellite Markers in the Crested Ibis Nipponia nippon from South Korea

  • The Asian crested ibis Nipponia nippon is one of the world's most endangered species. Except for the Sanxii population from China, it is known that all of the crested ibis populations from East Asia have been extinguished. In these days, most of them are being inbred as captive populations in China, South Korea, and Japan, which caused their low expected genetic diversity. Microsatellite markers are well known as a suitable DNA marker for exploring genetic diversity among captive populations of a variety of endangered species. In the present study, ten microsatellite markers were developed for the captive populations of the South Korean crested ibis, which were employed to examine the level of genetic diversity with the two founders from Sanxii, China and the 70 descendants of them. As a result, the mean number of gene diversity, observed heterozygosity, and expected heterozygosity of the captive population were 0.70, 0.84, and 0.70 respectively. It revealed that the captive population of South Korea is as genetically more stable than we expected. In addition, the principal coordinates analysis and genetic structure analyses showed that the captive population of N. nippon can be divided into the two different genetic groups. The developed microsatellite markers here could be helpful for crested ibis conservation in East Asian countries such as China and Japan as well as South Korea.

  • Purpose: This study was undertaken to explore sex differences in the risk markers of cardiovascular disease (CVD) and psychosocial factors and the relationships among these variables for the community-residing adults. Methods: A cross-sectional study was conducted with a total of 114 participants consisting of 65 men aged 42.85 years old and 49 women aged 46.86 years old. Participants were selected by a convenient sampling of at community cultural centers and churches. They completed a structured questionnaire including demographic characteristics, physiologic risk markers of CVD; lifestyle factors; and depression, social support, and stress. Results: The male participants showed significantly higher systolic and diastolic blood pressures than their counterparts. Significantly higher percent of women were currently non-smokers, and practiced moderate alcohol intake, physical activity, and low fat diet. Among women, the higher the scores of depression and stress were and the lower that of the social support was, the higher blood pressure, waist circumference and BMI were. Among men, those who were less depressed and with higher scores of social support had more physical activity. Conclusion: The findings suggest further studies to explore sex differences of CVD risk markers and psychosocial factors, and their relationships in a larger sample, and to find the mechanism underlying these differences with a prospective design.

Relationship between Personality and Biological Reactivity to Stress: A Review

  • Objective Personality traits can be the basis for individual differences in the biological response of stress. To date, many psychobiological studies have been conducted to clarify the relationship between personality and biological reactivity to stress. This review summarizes the most important findings in this area of research. Results Key findings related to the relationship between personality factors and stress-sensitive biological systems in four research models have been summarized; model of psychosocial characteristics, model based on Rumination and Emotional Inhibition, Eysenck's biopsychological model, and Five-Factor Approach of Personality. Conclusion According to the results of this review, it can be concluded that personality typology of individuals influenced their biological reactivity to stressful events. Understanding the biological basis of personality can help to better understand vulnerability to stress. Future research can be continuing based on framework of the four models.
