Experimental animals have been used to biological and medical purposes and the animals must be, for these purposes, healthy and clean to microbial infection. However, the animals can be easily exposed to pathogenic microorganism via several routes. Of the routes, environmental conditions are the most important factors to keep the animals healthy and clean, especially air condition. Monitoring of air-condition has been required to keep the animal healthy and clean. However, any guideline is not available for experimental conditions with pigs. Therefore, the sampling times and points were compared in different conditions to establish an optimal protocol for monitoring of air borne bacteria. Tryptic soy agar(TSA), blood agar containing 5% defibrinated sheep blood and Sabraud dextrose agar(SDA) were used as media to capture total bacteria, pathogenic bacteria and fungi, respectively. Two methods, compulsive capture using an air-sampler and capturing fall-down bacteria were used to capture the microorganisms in the air. The points and time of capturing were different at each experiment. Air borne microorganisms were captured at three and five points in the open and closed equipments, respectively. Air was collected using an air-sampler for 1 min and 5 min and the agar plates as open status were left from 30 min to 2hr. At first, we monitored an experimental laboratory which dealt with several pathogenic bacteria and then, a protocol obtained from the investigation was applied to open or close experimental conditions with pigs. Number of bacteria was high from 10:00 to 15:00, especially on 13:30-15:30 but sharply decreased after 17:00. The tendency of the number of bacteria was similar between two methods even though the absolute number was higher with air sampler. Critical difference in the number of cells was observed at 5 min with air sampler and 2 hr with fall-down capturing method. However, 1 min with air sampler and 1 hr with fall-down capturing were the best condition to identify bacterial species collected from the air. Number of bacteria were different depending on the sampling points in closed condition but not in opened condition. Based on our results, a guide-line was suggested for screening air-borne microorganism in the experimental conditions with pigs.
This study was performed to know the hypocholesterolemic effect of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$. The changes of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and phospholipid levels in serum, as well as the atherogenic index were elucidated in rats for 35 days. In order to evaluate single-dose toxicity of HAG-50 in rats via oral routes, induction any mortalities and abnormal signs in clinical finding, body weight, gross findings and histopathological finding, and estimation of $LD_{50}$ were elucidated. Total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and phospholipid levels in the serum significantly decreased and resulted in the decrease of atherogenic index in rats fed diets containing $5\%\;and\;10\%$ HAG-50 (p<0.01). HAG-50 did not induce any mortalities and abnormal signs in clinical findings, body weights, gross findings and histopathological findings in single-dose toxicity test via oral routes of rats, after which it was impossible to estimate $LD_{50}$ values.
Background: Korean Red Ginseng (KRG) has been used in Asia for its various biological effects, but no studies have investigated the safety of its long-term intake. Therefore, the present study evaluated the safety of KRG intake for 24 weeks. Methods: We randomized 1,000 participants in a 1:1 ratio into two groups, which were treated daily with 2 g of KRG or a placebo for 24 weeks. The primary endpoint was all adverse events and adverse drug reactions (ADRs) that occurred after KRG or placebo administration, which were reported at week 4, 12, and 24 after the baseline visit. Results: In total, 192 and 211 participants experienced adverse events in the KRG and placebo groups (39.2% and 42.0%, respectively; p = 0.361), and 59 and 57 KRG- and placebo-treated individuals reported ADRs (12.0% and 11.4%, respectively; p = 0.737). The frequently occurring ADRs were pruritus (2.0%), headache (1.6%), diarrhea (1.4%), and dizziness (1.2%) in the KRG group and pruritus (2.0%), headache (1.8%), dizziness (1.6%), rash (1.4%), and diarrhea (1.2%) in the placebo group. Discontinuation of drug administration due to ADRs was reported in 13 participants, six (1.2%) and seven (1.4%) in the KRG and placebo groups, respectively (p = 0.814). No significant abnormal changes were revealed by anthropometric, laboratory, and vital sign measurements in the KRG group compared with those in the placebo group. Conclusion: The present study confirms the safety and tolerability of daily intake of 2 g of KRG for 24 weeks by healthy adults.
Egg development and early life history of the Korean spine loach, Iksookimia pumila was investigated to provide basic information regarding biological characteristics and restoration. Adult fish were sampled using spoon net in the Baek Stream, Sangseo-myeon, Buan-gun, Jeollabuk-do, Korea, 7 July 2010. Eggs and sperms were obtained from the females and male with Ovaprim injecting (0.5 mL/kg) and then fertilized using the dry method in the laboratory. Number of spawned eggs were 1,107 (352~1,440). Spawned eggs were slightly adhesive, light yellowish coloring and measured $1.3{\pm}0.04$ mm (mean${\pm}$SD) in diameter. Spawned eggs hatched out 52 (47~55) hours after fertilization at water temperature of $23^{\circ}C$, and newly hatched larvae an average were $4.7{\pm}0.14$ mm in total length. At 5 days after hatching, larvae averaged $7.1{\pm}0.20$ mm in total length and their yolk sacs had been completely absorbed. Beginning at 17 days after hatching, fish entered the juvenile stage and reached $11.0{\pm}0.50$ mm in total length. At 100 days after hatching, the band patterns and external form of juvenile fish were similar to those of adults, and they averaged $31.3{\pm}3.98$ mm in total length.
Background: MicroRNAs (miRNAs) have demonstrated their potential as biomarkers for lung cancer diagnosis. In recent years, miRNAs have been found in body fluids such as serum, plasma, urine and saliva. Circulating miRNAs are highly stable and resistant to RNase activity along with, extreme pH and temperatures in serum and plasma. In this study, we investigated serum miRNA profiles that can be used as a diagnostic biomarker of non-small cell lung cancer (NSCLC). Methods: We compared the expression profile of miRNAs in the plasma of patients diagnosed with lung cancer using an miRNA microarray. The data from this assay were validated by quantitative real-time PCR (qRT-PCR). Results: Six miRNAs were overexpressed and three miRNAs were underexpressed in both tissue and serum from squamous cell carcinoma (SCC) patients. Sixteen miRNAs were overexpressed and twenty two miRNAs were underexpressed in both tissue and serum from adenocarcinoma (AC) patients. Of the four miRNAs chosen for qRT-PCR analysis, the expression of miR-23a was consistent with microarray results from AC patients. Receiver operating characteristic (ROC) curve analyses were done and revealed that the level of serum miR-23a was a potential marker for discriminating AC patients from chronic obstructive pulmonary disease (COPD) patients. Conclusion: Although a small number of patients were examined, the results from our study suggest that serum miR-23a can be used in the diagnosis of AC.
Laboratory trials were conducted for control of onion maggot and tobacco cutworm with ento¬mogenous nematodes. The onion maggot, Delia antiqua, was exposed to Steinernema feltiae at concentration of 0,30,60, 120 or 240 nematodes per larva, and to Heterorhabditis heliot-hidis at concentration of 0, 10, 20, 40 or 80 nematodes per larva. Mortalities of the maggot ranged from 80 to 100% in S. feltiae and from 63.3 to 100% in H. heliothidis. The tobacco cutworm, SpodoPtera litura was exposed to S. feltiae at concentration of 0,50,100,200 or 400 nematodes per larva and to H. heliothidis at concentration of 0,20,40,80 or 160 nemat¬odes per larva with or without kale in petri dish. The 3rd instar larvae of the tobacco cutworm was more susceptible to both nematode species than the 4th or 5th instar at low concentration. Mortalities of the 3rd instar were 100% in S. feltiae and 67.7-100% in H. heliothidis while those of 4th and 5th instar ranged 76.7-100% and 43.3-100% in S. feltiae, and 36.7-90% and 3.3-90% in H. heliothidis, respectively. Mortalities of the tobacco cutworm larvae decreased when the nematodes were sprayed on the kale leaves in petri dish except 3rd instar.

Entomopathogenic nematode, Steinernema carpocapsae GSN1 strain was evaluated for the environmentally sound control of Tebenna issikii (Lepidoptera: Choreutidae) in the laboratory. The corrected mortality of Tebenna issikii larvae was 100% at the 40 infective juveniles (Ijs)/larva 3 days after treatment with S. carpocapsae GSN1 strain in Petri dish. $LC_{50}$ value of S. carpocapsae GSN1 strain against Tebenna issikii was 5.7 Ijs. The mean penetration numbers of Ijs of S. carpocapsae GSN1 strain at the 5, 10, 20, 40 and 80 Ijs/larva in a Tebenna issikii larva were 1.4, 1.4, 3.2, 5.6 and 11.9 Ijs/larva, respectively. However penetration rate of Ijs of S. carpocapsae GSN1 strain at 5 Ijs/larva was the highest among other nematode concentrations. Progeny of S. carpocapsae GSN1 strain in a Tebenna issikii larva was higher with increasing nematode concentration.

Ostrinia larvae feed the pods and stem of red bean and seriously damage the bean production from farmers. In this study we investigated biological and developmental characteristics including field collection, host feeding preference, artificial rearing diet, morphological and molecular taxonomical identification, and pheromone analysis for an Ostrinia sp. in Korea. The male adults have massive tibia in the middle legs and 3-lobed uncus in the genitalia. The partial nucleotide sequences of mitochondrial cytochrome oxidase I (COI) and II (COII) were not corresponded to those DNA sequences from other Ostrinia species reported previously in Japan and China. Host plants for this species are also different from the previous species reported. In the gas chromatography (GC) analyses, (Z)-9-tetradecenyl acetate was not detected from the pheromone gland of our species while the component as a sex pheromone was found in O. zaguliaevi and O. zealis, With taken results, we conclude this Ostrinia species in Korea is Ostrinia scapulalis or closely related species. When larvae collected in a fall were incubated in the outdoor condition, they emerged to adult between June and July in the next year. The result indicates that the winter diapause could be started in late larval stage. In addition, we developed a semi-synthetic artificial diet adopted for mass rearing of the O. scapulalis in laboratory.
Eggs development and early life history of the endangered Korean dwarf loach, Kichulchoia brevifasciata (Teleostei: Cobitidae) was investigated to provide basic information regarding biological characteristics and restoration. Adult fish specimens were sampled using a spoon net at Geurnsan-myeon, Goheung-gun, Jeollanam-do, Korea from June to July 2011. Since, spawning characteristics were analyzed, and females were induced to spawn by injecting Ovaprim (0.5 mL/kg) and their eggs were artificially fertilized with sperms by the dry method in the laboratory. Total length of mature female were 46~76 mm with GSI $9.6{\pm}3.77%$, and total length of mature male was 42~52 mm with GSI $3.5{\pm}1.04%$. Sex ratio (♂/♀) was 0.10, and there were no secondary sexual characteristics. The number of mature eggs was averaged $60{\pm}28.7$ per female. The lemon yellow eggs were slightly adhesive $1.46{\pm}0.07mm$ in diameter. The embryo hatched approximately 66 h after fertilization at $25^{\circ}C$, and the hatched larvae were averaged $5.5{\pm}0.07mm$ in total length (TL). At 6 days after hatching, the larvae averaged $9.0{\pm}0.29mm$(TL) and their yolk sac was completely absorbed. At 17 days after hatching, they entered the juvenile stage and reached $12.6{\pm}0.24mm$ (TL). At 80 days after hatching, the band patterns and external form of the juveniles were similar to those of adults, and they averaged $33.0{\pm}2.19mm$(TL).

Eggs were obtained from the females with Ovaprim (0.5 mL/kg) injecting and then fertilized using the dry method in the laboratory. Number of spawned eggs were $1,015{\pm}199$. Spawned eggs were strongly adhesive, opaque and grayish and measured $0.74{\pm}0.08mm$ (mean${\pm}$SD) in diameter. Fertilized egg size continuously increase until $2.94{\pm}0.10mm$ for 9 h 30 min at $23^{\circ}C$. Fertilized eggs hatched 26 h after fertilization, and newly hatched larvae an average were $3.4{\pm}0.07mm$(TL). The yolk sacs of the larvae were of most absorbed of 3 days after hatching and $4.4{\pm}0.1mm$ (TL). About at 13 days after hatching, they became to juvenile stage and reached $16.8{\pm}0.50mm$ (TL). At 80 days after hatching, the band patterns and external form of juveniles were similar to those of adults, and they averaged $27.7{\pm}2.32mm$(TL).
