Transformation Characteristics of Chlorinated Aliphatic Hydrocarbon (CAH) Mixtures in a Two-Stage Column: 1st Chemical Column Packed with Zinc Natural Ore and 2nd Biological Column Stimulated with Propane-Oxidizing Microorganisms (아연 광석과 프로판산화 미생물을 이용한 이단 고정상 반응기에서의 염소계 지방족 탄화수소 혼합물 분해 특성)

  • This study was conducted to develop a combined method for remediating a Chlorinated Aliphatic Hydrocarbons (CAHs) mixtures-contaminated aquifer. The process is consist of two processes. A chemical process (1st) using natural zinc ores for reducing higher concentrations of CAH mixtures to the level at which biological process is feasible; and A biological process (2nd) using aerobic cometabolism for treating lower concentration of CAH mixtures (less than 1 mg/L). Natural zinc ore showed relatively high transformation capacity, average dehalogenation percentage, and the best economic efficiency in previously our study. To evaluate the feasibility of the process, we operated two columns in series (that is, chemical and biological columns). In the first column filled with natural zinc ore and sand, CAH mixtures were effectively transformed with more than 95% efficiency, the efficiency depends on the Empty Bed Contact Time (EBCT) and the mass of zinc ore packed. Scanning Electron Microscope (SEM), X-Ray Diffraction (XRD) analysis were performed to make sure whether natural zinc ore played an key role in the dechlorination of the CAH mixtures. The characteristics of zinc metal surface changed after exposure to CAHs due to oxidation of $Zn^0$ to $Zn^{2+}$. In the biological column injecting propane, DO and effluent of the chemical column, only 1,1,1-TCA was cometabolically transformed. Consequently, the combined process would be effective to remediate an aquifer contaminated with high concentrations of CAH mixtures.

Characteristics of Pesticide Residues in Leafy Vegetables Collected from Wholesale and Traditional Markets in Cheongju (청주지역 도매시장과 재래시장 유통 엽채류 중 농약의 잔류 특성)

  • This study was carried out to survey the residual characteristics of pesticides and assess their safety. Nineteen agricultural commodities, collected from wholesale and traditional markets in Cheongju. Nineteen agricultural commodities including perilla leaves were collected from the markets on October 29th, 2010. Total 240 pesticides which can be analyzed by multiresidue analysis method by GLC and HPLC were monitored and the pesticides detected were confirmed by GC-MSD and LC-MS. Five pesticides, alachlor, bifenthrin, endosulfan, procymidone and triflumizole, were detected from five samples, such as welsh onion, leek and celery in case of wholesale market and perilla leaves and welsh onion in case of traditional market. Detection rate of 13.2% was obtained as a result of pesticide analysis but 2.6% of the pesticides detected exceeded their maximum residue limits. The estimated daily intakes (EDIs) and maximum permissible intakes (MPIs) of the pesticides detected were less than 26% and 0.05% of their acceptable daily intakes (ADIs) respectively, representing that residue levels of the pesticides detected would be safe.

Eighteen unreported radiation-resistant bacterial species isolated from Korea in 2018

  • In 2018, as a subset study to discover indigenous prokaryotic species in Korea, a total of 18 unreported bacterial strains were discovered. From the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade, it was determined that each strain belonged an independent and predefined bacterial species. There were no official report that these 18 species were previously described in Korea; therefore, one strain of Williamsia, one strain of Rhodococcus, three strains of Microbacterium, three strains of Agromyces, one strain of Arthrobacter, one strain of Paeniglutamicibacter, one strain of Pseudarthrobacter, one strain of Nocardioides, one strain of Fibrella, one strain of Hymenobacter, one strain of Deinococcus, two strains of Fictibacillus, and one strain of Paenibacillus are described as unreported bacterial species in Korea. Gram reaction, basic biochemical characteristics, and colony and cell morphologies are described in the species description section.

  • The objective of this study was to develop kinetic model for the MBR and investigate kinetic characteristics of the gravitational flow transverse direction MBR system. Kinetic model was derived by mass balance of substratc and biomass combined with empirical membranc filtration rerm for the MBR. To find kinctic values, permeale flux and COD removal were analyzed through the laboratory, MBR operation as different solids retention times. Permeate flux was ranged 2.5-5.0 LMH (L/m$^2$/hr) as sludge characteristics in each run. Although the soluble COD in the bioreactor was changed, the effluent COD was stable as average 99% removal rate during the experimental periods. Y$_g$ of this MBR system was higher than those of cross-flow MBR processes. The kinetics of this MBR showed that smaller k, larger b, and larger K$_s$ values than the conventional activated sludge process. These results indicated that substrate was used for cell maintenance rather than growth in this MBR system.

Marine Environmental Characteristics on the Dinoflagellate Cysts Distribution in Surface Sediments in the Southwest Sea, Korea (한국남서해역 표층퇴적물 중의 와편모조류 시스트 분포에 영향을 미치는 해양환경요인)

  • Marine environmental characteristics on the dinoflagellate cysts distribution in surface sediment of the southwest sea of Korea were investigated from 21 stations in September 2003, and 36 stations in June, 2004. The water mass characteristics indicated that the southwest sea of Korea is characterized by various oceanographic conditions due to coastal waters of Korea and China. The Tsushima warm currents and the cold bottom water of the Yellow Sea. Mud contents and chlorophyll a concentrations were higher in central region such as, Namhaedo, Yeosu and Goheung coast than in western region such as, Wando, Haenam and Jindo coast in the South Sea of Korea. A total of 35 taxa of dinoflagellate cysts were identified representing 21 genera, 33 species, 2 unidentified species. Cyst abundance ranged from 13 to 527 cysts $g-dry^{-1}$, showing higher abundance in the coastal areas than in western region of the South Sea of Korea. From the result of the PCA analysis, the distribution of dinoflagellate cysts was not only related to the different water masses which appeared in the southwestern sea but also to physical and biological parameters such as water temperature, light, surface sediment faces and phytoplankton biomass.

  • Functional genomics aims at uncovering useful information carried on genome sequences and at using it to understand the mechanisms of biological function. Elucidating the unknown biological functions of new genes based upon the genomics rationales will greatly speed up the extensive understanding of biocatalysis and biodegradation in biological world including microorganisms. DNA microarrays generate a system for the simultaneous measurement of the expression level of thousands of genes in a single hybridization assay. Their data mining (transcriptome) strategy has two categories: differential gene expression and coordinated gene expression. Furthermore, measurement of proteins (proteome) generates information on how the transcribed sequences end up as functional characteristics within the cell, and quantitation of metabolites yields information on how the functional proteins act to produce energy and process substrates (metabolome). Various composite functional genomics databases containing genetic, enzymatic and metabolic information have been developed and will contribute to the understanding of the life blue print and the new discoveries and practices in biocatalysis and biodegradation that could enrich their industrial and environmental applications.

Automatic Addition Control of the External Carbon Source by the Measurement of ORP in Biological Nitrogen Removal Process (생물학적 질소 제거공정에서 ORP 측정을 통한 외부탄소원의 자동 주입 제어)

  • For the cost-effective biological nitrogen removal (BNR) process whose characteristics of influent have low COD/N ratios, the automatic control system for the addition of external carbon based on oxidation-reduction potential (ORP) data in an anoxic reactor has been developed. In this study, it was carried out with a pilot-scale Bardenpho process which was consisted of anoxic 1, aerobic 1, aerobic 2, anoxic 2, aerobic 3 tank and clarifier. Firstly, the correlation coefficient ($R^2$) of the dosage of external carbon source and ORP value was about 0.97. Consequently, the automatic control system using ORP showed that the dosage of external carbon source was decreased by about 20% compared with a stable dosage of 75 mg/L based on the COD/N ratio of the anoxic influent.

  • This study aimed to obtain fundamental data for demonstrating biodiversity of vegetation of East Asian alder (Alnus japonica) Forest Wetland of Heonilleung Royal Tombs. A total of 166 vascular plants (159 species, three subspecies, three varieties, and one cultivar) belonging to 132 genera and 59 families were found, accounting for 8.3% of 1,996 vascular plant species found in Seoul. Therophyte was the most common life-form of plants in Heonilleung Wetland. As for rare plant species, one Least Concern (LC) species was found. There were 15 floristic regional indicator species in the research area. Three of them belonged to floristic grades III and IV. This indicates that their habitats are discontinuous and isolated to some degree. Nineteen invasive alien plant species were found, most of which were introduced from North America after the year 1964 with a spread rate of V (widespread, WS).

  • An investigation of species of the genus Aspergillus present in arthropod, freshwater, and soil led to the discovery of seven undescribed species in Korea. Based on their morphological characteristics and molecular phylogeny analyses using a combined data set of β-tubulin (BenA) and calmodulin (CaM) sequences, the isolated strains CNUFC IGS2-5, CNUFC YJ1-19, CNUFC WD27, CNUFC U8-70, CNUFC AS2-24, CNUFC S32-1, and CNUFC U7-48, were identified as Aspergillus brunneoviolaceus, A. capensis, A. floccosus, A. inflatus, A. parvulus, A. polyporicola, and A. spelaeus, respectively. In the present study, the detailed morphological descriptions and phylogenetic relationships of these species are provided.

Effect of Temperature Change on the Respiration Characteristics of Vegetables

  • The effect of fluctuating temperature on the respiration of vegetables has been investigated. Spinach was selected as the experimental material because of its high respiratory activity and kept under the condition that temperature changed alternately at low and high levels every 4 hours. The low-high level temperature combination was set in $1-10^{\circ}C,{\;}1-20^{\circ}C{\;}and{\;}1-30^{\circ}C$. Respiration was evaluated in terms of quantity of $CO_2$ evolved from spinach. The evolution rate of $CO_2$ was determined by a change in its concentration. The evaluation rate of $CO_2$ followed closely the temperature change. In the temperature combinations at $1-10^{\circ}C{\;}and{\;}1-20^{\circ}C$, the relationship between $CO_2$ evolution rate and temperature was found to be able to express by Arrhenius law, while at $1-30^{\circ}C$, it did not obey the law.
