  • 수량(收量) 등급별(等級別) 영양기준설정(榮養基準設定)을 위하여 전국적(全國的)으로 실시된 수도삼요소시험중(水稻三要素試驗中) 3 개년간(個年間)$(1967{\sim}1969)$의 고(藁) 또는 수분석결과(穗分析結果)에 의(依)하여 수량등급별(收量等級別)로 생육시기별(生育時期別) N,P,K 및 Si의 함량(含量)을 조사(調査)하였다. 수량증가(收量增加)에 따라 양분농도(養分濃度)가 증가(增加)하는 경향은 N>P>K>Si의 순(順)으로 감소(減少)하였다. 그 경향(傾向)은 양분(養分)에 따라서 해에 따라 생육시기별(生育時期別) 보다는 정조수량등급(正租收量等級)에 양분(養分)의 농도변화(濃度變化)가 더 밀접(密接)한 관계(關係)를 보였다.

Unrecorded prokaryotic species belonging to the class Actinobacteria in Korea

  • For the collection of indigenous prokaryotic species in Korea, 35 strains within the class Actinobacteria were isolated from various environmental samples (animals and clinical specimens) in 2017. Each strain showed high 16S rRNA gene sequence similarity (>98.8%) and formed a robust clade with recognized actinobacterial species. The isolates were assigned to 35 species, 22 genera, 15 families, and 8 orders of the class Actinobacteria. There are no official descriptions of these 35 bacterial species in Korea. Morphological properties, basic biochemical characteristics, isolation source, and strain IDs are included in the species descriptions.

  • To study the biodiversity of bacterial species, here we report indigenous prokaryotic species of Korea. A total of 23 bacterial strains affiliated to the class Alphaproteobacteria were isolated from various environmental sources including seaweeds, seawater, fresh water, wetland/marsh, tidal sediment, plant roots, sewage and soil. Considering higher than 98.8% 16S rRNA gene sequence similarities and formation of a well-defined phylogenetic clade with named species, it was confirmed that each strain belonged to the predefined bacterial species of the class Alphaproteobacteria. There is no official report of these 23 species in Korea; 20 species of 16 genera (Mameliella, Yangia, Paracoccus, Ruegeria, Loktanella, Phaeobacter, Dinoroseobacter, Tropicimonas, Lutimaribacter, Litoreibacter, Sulfitobacter, Roseivivax, Labrenzia, Hyphomonas, Maricaulis, Thalassospira) in the order Rhodobacterales and 3 species of a single genus (Brevundimonas) in the order Caulobacterales. Gram-staining, cell morphology, basic biochemical characteristics, isolation sources, optimum temperature, growth media, and strain IDs are detailed in the species description as well as Table 1.

수지상세포의 항원제시 능력 및 항암활성에 미치는 Lipofectin의 영향 (Effect of Lipofectin on Antigen-presenting Function and Anti-tumor Activity of Dendritic Cells)

  • Background: Dendritic cells (DC) are professional antigen-presenting cells in the immune system and can induce T cell response against virus infections, microbial pathogens, and tumors. Therefore, immunization using DC loaded with tumor-associated antigens (TAAs) is a powerful method of inducing anti-tumor immunity. For induction of effective anti-tumor immunity, antigens should be efficiently introduced into DC and presented on MHC class I molecules at high levels to activate antigen-specific $CD8^+$ T cells. We have been exploring methods for loading exogenous antigens into APC with high efficiency of Ag presentation. In this study, we tested the effect of the cationic liposome (Lipofectin) for transferring and loading exogenous model antigen (OVA protein) into BM-DC. Methods: Bone marrow-derived DC (EM-DC) were incubated with OVA-Lipofectin complexes and then co-cultured with B3Z cells. B3Z activation, which is expressed as the amount of ${\beta}$-galactosidase induced by TCR stimulation, was determined by an enzymatic assay using ${\beta}$-gal assay system. C57BL/6 mice were immunized with OVA-pulsed DC to monitor the in vivo vaccination effect. After vaccination, mice were inoculated with EG7-OVA tumor cells. Results: BM-DC pulsed with OVA-Lipofectin complexes showed more efficient presentation of OVA-peptide on MHC class I molecules than soluble OVA-pulsed DC. OVA-Lipofectin complexes-pulsed DC pretreated with an inhibitor of MHC class I-mediated antigen presentation, brefeldin A, showed reduced ability in presenting OVA peptide on their surface MHC class I molecules. Finally, immunization of OVA-Lipofectin complexes-pulsed DC protected mice against subsequent tumor challenge. Conclusion: Our data provide evidence that antigen-loading into DC using Lipofectin can promote MHC class I- restricted antigen presentation. Therefore, antigen-loading into DC using Lipofectin can be one of several useful tools for achieving efficient induction of antigen-specific immunity in DC-based immunotherapy.

Comparative Analysis of the Three Classes of Archaeal and Bacterial Ribonucleotide Reductase from Evolutionary Perspective

  • The Ribonucleotide reductases (RNR) are essential enzymes that catalyze the conversion of nucleotides to deoxynucleotides in DNA replication and repair in all living organisms. The RNRs operate by a free radical mechanism but differ in the composition of subunit, cofactor required and regulation by allostery. Based on these differences the RNRs are classified into three classesclass I, class II and class III which depend on oxygen, adenosylcobalamin and S-adenosylmethionine with an iron sulfur cluster respectively for radical generation. In this article thirty seven sequences belonging to each of the three classes of RNR were analyzed by using various tools of bioinformatics. Phylogenetic analysis, dot-plot comparisons and motif analysis was done to identify a number of differences in the three classes of RNRs. In this research article, we have attempted to decipher evolutionary relationship between the three classes of RNR by using bioinformatics approach.

수업에 의해 변화되는 9학년 유전 개념의 생태 지위 분석 (Analysis of Ecological Niche in 9th Graders' Genetic Concepts after Instruction)

  • 개념 생태에서 개념은 독립적으로 존재하는 것이 아니라 생태적 환경 하에 놓여 생태 지위를 차지한다. 생물학의 많은 개념들 중에서 유전 개념은 유전 단원 외에도 여러 단원과 관련되어 있으며, 유전 단원 내에서도 영역 사이의 개념 간 의존도가 높은 단원이므로 생태 지위적 접근을 통해 개념의 다양성과 개념 근접성을 분석하였다. 이를 위하여 9학년 학생 995명을 대상으로 하였다. 설문지의 설문 영역은 유전자, 염색체, 체세포분열, 감수분열로 4가지 유전 관련 개념을 대상으로 하였다. 설문지는 각 영역과 관련된 생물학적 개념이나 용어를 제시하게 하고, 제시한 생물학적 개념이나 용어를 각 영역과의 관련된 정도를 1에서 30의 점수로 표시하게 하였다. 9학년 학생들을 대상으로 수업을 통한 유전개념의 변화를 생태 지위적 접근 방법으로 분석하였다. 수업을 통해 전체 개념 수는 감수분열, 체세포분열, 염색체, 유전자 영역 순으로 모든 영역에서 증가하였고, 전체 개념 수의 증가로 인해 상대밀도는 감소하였다. 개념 다양성 지수는 감수분열, 체세포분열, 염색체, 유전자 순으로 모두 증가하였다. 그리고 학습 후 개념들의 상대밀도의 차이가 감소하였으며, 관련성 점수의 차이도 감소하여 개념 간 유사성이 증가하였다. 이 결과를 바탕으로 다음의 결론을 얻을 수 있었다. 첫째, 학습을 통해 유전개념의 개념 다양성이 증가하였다. 둘째, 학습을 통해 유전개념의 개념 근접성이 증가하였다.

Westerdykella reniformis: A New Record from Field Soils in Korea

  • During a survey of fungal diversity in different provinces of South Korea in 2017, a new fungal isolate was discovered. This fungal isolate was identified as Westerdykella reniformis, based on its morphological characteristics and phylogenetic analysis, using internal transcribed spacer (ITS) and 28S ribosomal DNA (28S rDNA) sequence data. To our knowledge, W. reniformis has not previously been reported in South Korea. Thus, in this study, we report a new record of a species from the Dothideomycetes class in Korea, and provide a detailed description with morphological illustrations.

  • In 2014, as a subset study to discover indigenous prokaryotic species in Korea, a total of 33 bacterial strains assigned to the class Gammaproteobacteria were isolated from diverse environmental samples collected from soil, tidal flat, freshwater, seawater, oil-contaminated soil, and guts of animal. From the high 16S rRNA gene sequence similarity (>98.5%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 33 species have been described in Korea; therefore, 1 strain of the Aeromonadales, 6 strains of the Alteromonadales, 3 strains of the Chromatiales, 5 strains of the Enterobacteriales, 4 strains of the Oceanospirillales, 11 strains of the Pseudomonadales, and 3 strains of the Xanthomonadales within the Gammaproteobacteria are described for unreported bacterial species in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, and isolation sources are also described in the species description section.

A report of 21 unreported bacterial species in Korea, belonging to the Betaproteobacteria

  • As a subset investigation to discover indigenous prokaryotic species in Korea, a total of 21 bacterial strains assigned to the class Betaproteobacteria were isolated from a wide range of environmental samples which collected from fresh water, roots of plants, mineral water and soil from ginseng farm. Phylogenetic analysis based on 16S rRNA gene sequences indicated that 21 isolated strains were most closely related to the class Betaproteobacteria, with high 16S rRNA gene sequence similarity (>99.1%) and constructed a robust phylogenetic clade with the closest species in the class Betaproteobacteria. These isolated species have no previous report or publication in Korea; therefore 17 species in 14 genera of 6 families in the order Burkholderiales, 1 species in the order Methylophilales, 2 species in 2 genera of 1 family in the order Neisseriales are reported for betaproteobacterial species found in Korea.
