Structure and Biological Activity of K(H2O)L (L = 5,7-Dihydroxy-6,4'-dimethoxyisoflavone-3'-sulfonate)

  • Potassium(I) with 5,7-dihydroxy-6,4'-dimethoxyisoflavone-3'-sulfonate (L) assembles to K($H_2O$)L (L = 5,7-dihydroxy-6,4'-dimethoxyisoflavone-3'-sulfonate). It was characterized by single-crystal X-ray diffraction, element analysis, IR and $^1H$ NMR spectroscopy. It crystallizes in the monoclinic space group $P2_1$/n and reveals a seven-coordinate complex. Polyhedra potassium chains, C-H${\cdot}{\cdot}{\cdot}\pi$ and C-H${\cdot}{\cdot}{\cdot}$O and O-H${\cdot}{\cdot}{\cdot}$O hydrogen bonds lead K($H_2O$)L to a three-dimensional network structure. The biological activity of resistance to hypoxia was tested, and the results showed that the biological activity of resistance to hypoxia of K($H_2O$)L is as good as that of its precursor, irisolidone.

Characterization and Antifungal Activity against Candida albicans of Vaginal Lactobacillus spp. Isolated from Korean Women (질 내 유산균의 Candida albicans에 대한 항진균 효과 연구)

  • Vulvovaginal candidiasis (VVC) is one of the urogenital infections occurring in women worldwide. Candida albicans is generally observed among various types of microorganisms causing VVC. Antibiotic therapy is typical, and the use of Lactobacilli probiotics is to be recognized as a promising alternative. The aim of this study was to select vaginal lactobacilli with probiotic properties against C. albicans. In a previous study, we isolated 38 lactobacilli from vagina of Korean women and 20 isolates were shown to inhibit C. albicans. We further selected 10 isolates which were able to inhibit C. albicans less than $10^5CFU/mL$. Among these selected strains, Lactobacillus salivarius MG242 (identified by 16s rRNA sequencing) was finally selected based on its strong anti-candidal activity, acid/bile salt resistance and adhesion property. Indirect adhesion activity of MG242 measured by auto-aggregation assay showed more than 60% auto-aggregation after 5 h standing. Taken these results together, the selected strain MG242 may have potential for application in vagina health related products.

Quick Detection of Firefly Luciferase Gene Expression in Live Developing Bovine Embryos by Photoncounting

  • The present study was designed, fIrst to develop the new methodology to measure the bioluminescence activity easily in live developing bovine embryos by photoncounting, and secondly to compare the expression efficiency of four luciferase reporter genes in bovine embryos at four- to 16-cell stages. In experiment 1, equimolar pSVlacZ and pSVEluc were microinjected into the pronucleus of fertilized bovine oocytes. At 2 days after micro injection, bioluminescence activity of these embryos was measured by photoncounting with a luminometer for 1 min, and lacZ gene expression in the same embryos was assayed by X-gal staining. All the luciferase-positive oocytes showed some bacterial ${\beta}$-galactosidase activity irrespective of the intensity. In experiment 2, four firefly luciferase genes (pTKEluc, pTK6WEluc, pSVEluc and pMiwluc) were introduced by micro injection, and the injected embryos were cultured for the following 2 days. Detection of the luciferase gene expression was done by photoncounting at 5 to 55 min. Over the measurement period, the luciferase activity was almost constant irrespective of the transgenes microinjected. The luciferase activity and expression efficiency at 2 days after microinjection were not significantly affected by the difference in the microinjected transgenes. The present results demonstrated that the bioluminescence activity in live developing bovine embryos could be measured quickly by photoncounting.

Determination of Phytochemical Contents and Biological Activities from the Fruits of Elaeagnus multiflora

  • The purpose of this study was to investigate the phytochemical contents and biological activities, including $\alpha$-glucosidase inhibitory activity and antioxidant activity, from the fruits of Elaeagnus multiflora using different solvent systems. The 50% ethanol extract of this species showed the most potent $\alpha$-glucosidase inibitory activity (88.5%) at a concentration of 1 mg/mL. Moreover, this extract also displayed the strongest antioxidant activity in each assay, showing 96.3% in DPPH scavenging activity, 98.2% in ABTS scavenging activity, and 2.5% in reducing power. The highest total phytochemical contents, including flavan-3-ols, phenolic acids, and flavonoids, were observed in the 50% ethanol extract. Interestingly, flavan-3-ols (3.150 mg/g) were detected at a significantly higher total content than those of phenolic acids (0.380 mg/g) and flavonoids (not detected). Additionally, the contents of individual phytochemicals showed remarkable differences, especially the epicatechin gallate (2.008 mg/g) and gallic acid (0.099 mg/g), which were the predominant constituents of each phytochemical type. Therefore, our results suggest that the 50% ethanol extract of E. multiflora fruits has strong biological activities, which are correlated with high phytochemical contents.

Immobilization of Trypsin onto Silk Fibroin Fiber via Spacer Arms

  • Trypsin can be immobilized on silk fibroin fiber (SFF) by introducing several spacer arms, such as ethylene diamine (ED), bovine serum albumin (BSA) and silk sericin (SS). Direct immobilization on silk fiber (SFFGA) has low activity because of the steric hindrance between the trypsin and substrate. The introduction of spacer arms onto SFF-GA can enhance the activity of trypsin by reducing the steric hindrance. When ED is used as a spacer arm, the activity of trypsin has increased but its stability decreased due to the increased hydrophobicity of SFF. BSA and SS, as a spacer arm, have better results in both activity and stability. SFF-BSA shows some decrease in the specific activity due to improper immobilizatin. SFF-SS maintained 90% of its initial activity even after 12 hrs incubation at $50^{\circ}C$. In the case of repeated hydrolysis of silk sericin with immobilized trypsin, SFF-GA and SFF-ED lost 50% of their initial activity right after first run, whereas SFF-BSA and SFF-SS maintained 80% of their initial activities even after 5 runs. Higher operational stability is due to increased hydrophilicity of SFF by introducing hydrophilic spacer arms such as BSA and SS. The high content of serine in SS increases the hydrophilicity of SFF resulting the best results among other spacer arms.

  • Mycelial growth inhibition activity of forty-one N-substituted phenyl 5-chloro-1,3-dimethylpyrazole-4-carboxamides against Rhizoctonia solani was analysed quantitatively by multiple regression analysis using physicochemical parameters of substituents as independent variables and $pEC_{50}$ as dependent variable. As a result, a quantitative structure-activity relationship was formulated using eight physicochemical parameters, which explains 83% of variance of the fungicidal activity. The most important parameter for the biological activity was log k', as related to the penetration and transport processes in the biological system. The activity also correlated with other hydrophobic parameters$({\pi}_2,\;{\pi}_3)$, an electronic parameter$({\Sigma}{\sigma})$, and steric parameters$(STERIMOL\;parameters\;L_3,\;L_4)$.

  • The purpose of this study was to investigate the ultra sonic extraction method of phenolic compounds and antioxidant activity from Rumex crispus. Rumex crispus was fractionated with hexane, ethyl acetate, butanol and water. The amount of phenol compounds and antioxidant activity was presented higher content in ethyl acetate fraction then others.

  • It has been known that the novel diterphenoids, hericenone and erinacine isolated from the fruiting body and cultured mycelia of Hericium erinaceus showed potent stimulating activity of nerve growth factor (NGF)-synthesis. To investigate the biological activities of extracts from fruiting body, cultured mycelium and cell-free broth of H. erinaceus, the activity experiments of antioxidation and AChE inhibition were carried out. Sephadex G-10 gel filtration followed by HPLC on a ${\mu}Bondapak$ $C_{18}$ column of EtOAc extract from cultured mycelium showed a biological activity.

  • The germination of Lactuca sativa seeds was significantly inhibited by the water extract of the fresh leaves of Pulsatilla koreana N. including abundant ranunculin. Germination inhibitory activity increased in a dose-dependantly. Protoanemonin, produced from ranunculin by enzymatic action during maceration process of leaves, was proved to be the active principle with inhibitory activity was above 90% at concentration of 1 mg/ml.

  • The effect of nonlamellar-prone lipids, diacylglycerol (DG) and phosphatidylethanolamine (PE), on the ATPase activity of SecA was examined. When Escherichia coli (E. coli) PE of the standard vesicles composed of 60 mol% of this lipid and 40 mol% of dioleoylphosphatidylglycerol (DOPG) is gradually replaced with either dioleoylgylcerol (DOG) or dioeloyl PE(DOPE), the ATPase activity of SecA present together increased appreciably.(omitted)
