  • The isoelelectric points of Cd-BP(l) and Cd-BP(II), cadmium-binding proteins, were 6.01 and 5.35, respectively. Both of them contained zinc. As for the amino acid composition, Cd-BP(I) contained a lot of glycine and lysine but none of such aromatic amino acids as tyrosine and phenylalanine.. On the other hand, Cd-BP(II) contained leucine, histidine, asparti cacid and alanine but no aromatic amino acids.

Study of Novel Markers for Early Diagnosis of Cardiovascular Diseases

  • In our country, cardiovascular disease (CVD) and Coronary heart diseases (CHD) are the leading causes of death. It is well known that CHD is multifactorial, involving environmental factors such as diet, level of exercise and cigarette smoking, and inherited factors. According to the statistical data in 2003, the cause of death with the highest mortality was including hypertension, ischemic heart disease and atherosclerosis, which accounted for $24.7\%$ of total mortality. In spite of, there have been few study reports on the change of biochemical markers and mechanisms concerned. The development of biochemical markers is required for an early diagnosis and treatment of cardiovascular diseases that are related with dietary habits of Korean people enjoying mixtures of traditional dietary style and westernized food-styles. Therefore, the most efficient cost-saving biochemical marker was established in this study, through analysis of biochemical markers related with dietary habits which are susceptibly being changed in association to cardiovascular diseases from the pre-disease phase, and through reanalysis and assessment of early diagnosis of and preventive effects of diagnosis of cardiovascular diseases by demographical character including sex, age, and socioeconomic level with use of biochemical markers that are identified and selected among the parameters in consideration of the effectiveness and appropriateness of early diagnosis of diseases. The appropriateness of biochemical markers was reviewed by professionals (medical, pharmaceutical area and food/ nutrition area) and CRP(C-Reactive Protein) and was identified to be possible in Korea. It is thought that these biochemical markers may be used as the basic data for early diagnosis and prevention of cardiovascular diseases (CVD) which may be used for Korean people.

낙동강 하구 간석지에서 분리된 세균의 층별 종조성 및 특성에 관하여 (Vertical composition and character analysis of saprophytic bacteria isolated from the mudflat of Nakdong river estuary)

  • Bacterial identification was performed with morphological, physiological and biochemical tests to the isolates from the mudflat of 30cm depth sampled in Nakdong river estuary in March and June, 1985. Flavobacterium and Cnterobacteriaceae were regarded as dominants. Pseudomonas, Bacillus, Micrococcus, Vibrio, Aerococcus, Aerononas, Acinetobacter and Staphylococcus were founded in various depth. Vertical composition of bacterial genera in March was more diversiform than that of June. Character analysis was carried out with the calculation of similarity index (S). At a level of 85% similarity, the isolates were clustered into 5 groups and ungrouped 2 strains. Classifying groups of bacterial strains with determination schemes and groups from similarity index were in good agreement.

Combining Ability for Morphological and Biochemical Characters in Mulberry (Morns spp.) under Salinity Stress

  • A line x tester analysis was carried out in mulberry (Morns spp.) under different salinity levels to determine the changes in the genetic interaction of various morpho-biochemical characters. Five mulberry genotypes, 3 females and 2 males, differing in salt tolerance were selected for the study. Clones of these parents along with clones of the F1 hybrids were planted in earthen pots and subjected to different levels of salinity (0.0%, 0.25%, 0.50%, 0.75% and 1.00% NaCl). Data on morphological and biochemical characters were subjected to line x tester analysis. The result revealed significant variation among the parents studied. The prominence of non-additive gene effect under control condition suggests the need for well chalked out breeding program to exploit the non-fixable variance of components for improvement of plant height, leaf size and leaf yield, chlorophyll and photosynthesis in mulberry. However, under salinity stress a shift from non-additive gene effect to additive gene effect for the above said character further suggests the need for a change in breeding strategy. The general combining ability (GCA) analysis has identified English black as the best combiner among the parents and the specific combining ability analysis (SCA) found crosses of English black X C776 and Rotndiloba x Mandalaya were good for Plant height and leaf size and English black X C776 and Rotundiloba x C776 were good for biochemical proline and chlorophyll. From the performance of parents and their crosses under different salinity levels and also under normal cultural conditions it is concluded that in mulberry different approaches are required to develop varieties for the irrigated and saline conditions.

炭川의 大型水生植物群集의 分布와 環境 (Environmental Factor and the Distribution of Aquatic Macrophytes Community in Tanchon)

  • The distribution of aquatic macrophytes in TanChon basin, a stream of the Han River, were investigated in terms of environmental gradient from June 1989 to March 1990. In the basin, 12 species of aquatic macrophytes were listed and four communities of Potamogeton crispus community. $P. octandrus$ community, $Hydrilla verticillata$ community and $Vallisneria$ asiatica community were recongized by character species. $P. crispus, P. octandrus$ and $V. asiatica$ were found in rapids while $H. verticillata , Ceratophyllum demersum$ and $Trapa japonica$ were done in pools. The depth of sediment $TanCh\u{o}n$ was showed as a exponential function of water velocity, Bd=exp (-K Wv). The values of Biochemical Oxygen Demand(BOD), Chemical Oxygen Demand(COD) and Suspendid Solid(SS) were recorded as range of 3.2~121.0mg/1, 4.2~54.5mg/1 and 4.1~114.0mg/1, respectively. And the linear positive correlation between BOD(X) and COD(Y) were expressed as Y=3.904+0.4308 X with $R^2$=0.9808 and also the correlation between BOD value(X) and SS value(Y) were done as Y=5.333+0.9606X with $R^2$=0.9700. In two dimensional analysis of BOD and water velocity, their clusters were showed similar types matching to communities classified by character species. However, no aquatic macrophyes were found at the site with BOD$\geq$50mg/l or DO$\leq$0.2mg/l.

꼬시래기 유래 아가로즈와 블루베리 열매 추출물을 이용한 화장용 에멀젼 개발 (Development of Cosmetic Emulsion Using Blueberry Fruit Extract and Agarose from Gracilaria verrucosa)

  • The need for natural cosmetic ingredients has been increasing over the world nowadays. Agarose, a natural polymer from red seaweeds, has high hydrophilic character and a function of scaffolder. As skin moisturizer, agarose is adequate for percutaneous absorption. While, blueberry fruits extract possesses rich procyanidins and anthocyanins which show health benefits, anti-oxidant effect, anti-aging and anti-melanogenesis. Stability, sensory preference, skin trouble of the emulsion formula are important for cosmetic product development. In this study, we manufactured an emulsion formula for skin moisturizers using the two ingredients and tested emulsion stability and skin trouble. Total phenolic contents of the blueberry fruits extract were evaluated as well as tyrosinase inhibitory and collagenase inhibitory activities. $IC_{50}$ values of blueberry fruits extract for anti-tyrosinase and anti-collagenase activities were 168 and $112{\mu}g/mL$, respectively using gallic acid as a control ($64.8{\mu}g/mL$). The stability (pH and viscosity) of the formula containing 2% blueberry fruits extracts and 0.1% agarose was measured at five different temperatures (room temp., $25^{\circ}C$, $55^{\circ}C$, $45^{\circ}C$, $55^{\circ}C$) under the sun light at 2 day intervals for 12 days. There has been little pH change at the different temperatures. According to the sensory evaluation, there was no significant flavor, discoloration and physical changes of the formula at $25-65^{\circ}C$. These results suggest that emulsion formula containing blueberry extract and agarose could be used as a candidate for lotion and essence products.

금강 하구 퇴적토에서의 종속영양 세균의 분포에 미치는 환경요인과 계절적 우점속에 관하여 (Environmental factors affecting seasonal destribution of heterotrophic bacteria in Kum river estuarine sediments)

  • Dominant genera of heterotrophic bacteria were investigated seasonally in Kum River estuarine sediments. Numerical taxonomy on the isolated heterotrophic bacteria were carried out according to their morphological, physiological, nutritional and biochemical characters. Heterotrophic bacteria in the depths of 0 to 10 cm of sampled sediments were almost facultative anaerobes, and 77.9% to 86.9% of them were Gram negative rods. Kominant genera among the isolates were Pseudomonas, Flavobacterium, Micrococcus, Cnterobacteria and Vibrio during sampling periods. According to their character analyses by similarity index(S), the isolates were clustered in fourteen groups at clay type sediments (Site 1) and four groups at sand type sediments (Site 2).

Biochemical and Genetic Variation of Hordein Subunits in Korean Barley

  • One-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (1D SDS-PAGE) was used to determine whether it would provide improved resolving power of hordein proteins concomitant with improved identification of Korean barley cultivars and germplams. This system gave rapid and reproducible separations of hordein polypeptides. Total fourteen of clear and easily scorable subunits were identified in Korean barley cultivars and germplasms and their polymorphic constitutions could provide biochemical genetic information in progeny analysis and endosperm quality improvement in barley breeding programs. Each hordein polypeptides residing in B, C, and D hordein pattern designations were scored to prepare a cultivar catalogue of protein patterns. On the basis of this character, 7 hordein polypeptide patterns were constructed from 108 barley cultivars and experimental lines. The molecular weight of hordein subunits in Korean barley cultivars and experimental lines varied in the range of 98 to 48 kDa. In contrast, less polymorphic hordein polypeptides were found in the low protein barley lines including malting barleys than those found in Korean barley cultivars and experimental lines.

제주연안해역의 Vibrio parahaemolyticus 분포에 관한 연구 (Study on the Distribution of Vibrio parahaemolyticus along Cheju Coast)

  • 제주도내 주요 해안지역인 화북, 삼양, 대포, 중문, 표선, 안덕, 애월, 곽지에서 생산되는 해수, 갯벌, 해산물 등에서 2002년 1월부터 12월까지 V. parahaemolyticus 분포상황을 조사하였다 가검물은 총 2,880건으로 각각 해수 960건, 갯벌 960건, 해산물 960건이었으며, 분리된 V. parahaemolyticus에 대하여 생화학적, 혈청학적 실험을 실시하였다. 총가검물 2,880건에 대한 분리 동정에서 417건(14.5%)이 분리되었다. 0.85% NaCl용액을 희석 액으로 사용하여 API 20E kit를 이용한 V. parahaemolyticus 100균주의 중요한 생화학적 특성은 전 균주가 lysine, ornithine, indole, glucose, mannitol은 양성반응을 보였고, ONPG, arginine, Sodium citrate, $H_2S$, urea, tryptophane, VP, inositol, sorbitol, rhamnose, sucrose, melibiose 반응에서 음성반응을 보였고, gelatin, amygdalin, arabinose 반응에서 음성 혹은 양성반응을 보였다. 검체별 분리율은 해수가 960건 중 161건(16.8%), 갯벌이 960건 중 137건(14.3%), 해산물이 960건 중 119건(12.4%)이었다. 지역별 분리율은 화북 14.4%(52/360), 삼양 15.3%(55/360), 대포 13.6%(49/360), 중문 18.3%(66/360), 표선 13.1%(47/360), 안덕 16.4% (59/360), 애월 12.5% (45/360), 곽지 12.2% (44/360)이었다. 분리된 V. parahaemolyticus 417균주를 대상으로 지역별, 검체별 분포를 조사한 결과 중문 지역이 18.3%(66/360)로 가장 높았고, 검체별로는 해수가 16.8% (161/960)로 가장 높았다.

수영만에서 분리된 Bacillus cereus Group LS-1 의 세균학적 특성에 관한 연구 (Studies on Bacterial Characteristics of Bacillus cereus Group LS-1 Isolated from Suyeong Bay)

  • 수영만에서 분리된 Bacillus cereus group LS-1 의 형태학적, 생화학적 성상 및 지방산 조성 분석을 통하여 세균학적인 특성을 밝혔다. B. cereus group LS-1 은 면양혈구배지에서 비용혈성의 점조성 집략과 convex 하고 가장자리가 규칙적인 둥근형태로 운동성이 없고 glucose, maltose, sucrose 와 gelatin 을 이용하고 trehalose 와 salicin 을 분해하지 않으며 6.5% NaCl 에서 자라지 않는 Gram 양성의 중심성 아포형성간균으로 표준균주 B. cereus group 과 다소 차이를 나타내었다. 지방산 조성 분석에서 chain 의 길이가 $C_{12}$ 에서 $C_{17}$로 iso $C_{15}$와 iso $C_{13}$의 branched chain 이 우점하는 B. cereus group 의 전형적인 특징을 나타내었으며 $nC^{15}$가 검출되지 않는 B.mycoides GC subgroup B 로 0.312 의 similarity index(SI) 를 지칭하여 다른 연구 결과와도 일치하였다. 한편 API system (API 50 CHB & API 20E) 의 ATB computer profile 에서 "Doubful Profile" 99.7% 의 B. firmus 로 나타내어 큰차이를 나타내었다. 67 개의 biochemical character 로 B. mycoids S-12 는 각각 42%, 42% 59% 와 52% 의 similarity matrix 를 나타내었다. B. cereus group 간의 아주 낮은 similarity 를 나타내어 상당한 차이가 있음을 인식하였다. 따라서 key test 와 지방산 조성등을 종합하여 볼때 B. ceresu group 중의 B. mycoides 로 새로운 biotype 인 것으로 사료되며 지방산 조성 분석으로 동정함이 훨씬 용이하였다.
