Comparative Responses of Sugar Beet Cyst Nematode, Heterodera schachtii and Clover Cyst Nematode, H. trifolii to Different Nematicides (클로버씨스트선충과 사탕무씨스트선충 알과 유충에 대한 약제 반응 비교)

  • This study aimed to investigate the differences in the effects of nematicidal materials (Burkholderia rinojensis, fluazaindolizine, fluopyram, fosthiazate, hydrogen peroxide, and imicyafos) on eggs and larvae of the sugar beet cyst nematode, Heterodera schachtii and clover cyst nematode, H. trifolii, which have recently become problematic in highland Chinese cabbage cultivation areas in Korea. Comparison of the hatching rates according to the test materials showed differences between treatments. In the treatment with fluopyram, fosthiazate, and imicyafos, a very high hatching inhibitory effect of 99% was observed in both nematodes, and there was no difference in the efficacy of treatments between nematode species except for that with hydrogen peroxide. Regarding the nematocidal activity of treatments against hatched larvae, only the fluazaindolizine treatment, despite showing a high hatching inhibitory effect, showed a low nematicidal effect of less than 30% against both nematodes, whereas a mortality rate of more than 90% was observed in the other treatments. There was no difference between nematode species in the reactions of the 2nd instar juveniles to all nematicidal test materials after 24 and 48 h of treatment. Although there are limitations to the laboratory experiment, the reaction to the nematicidal materials of the two cyst nematodes that damage the same host was similar, and simultaneous control is suggested in the areas where the two nematodes occur together.

Histological and Biochemical Studies on the Rooting of Hard-wood Cuttings in Mulberry (Morus species) (뽕나무 古條揷木의 發根에 關한 組織 및 生化學的 硏究)

  • Rootability of the hardwood cuttings of mulberry was related not only histological characteristics but dependent on biochemical properties. In this connection, the characteristics of the hardwood cuttings were histologically observed and the growth substances produced by the cuttings were also identified by means of mung bean bioassay. Amino acid, carbohydrate, nucleic acid contents, and the C/N ratio were also analysed. The results are summarized as follows. 1. There were differences in rootability of cuttings between mulberry species and varieties Among the three mulberry species tested, Morus Lhou Koidz. showed the highest rootability while M. bombycis showed the lowest one. In varietal differences in rootability, it was shown that the varieties could be grouped according to rootability: high varieties(above 80%), medium(41~79%), and low(below 40%). The higher varieties were Kemmochi, Nakamaki, Kosen, and Wusuba roso. 2. The histological characteristic of the hardwood cuttings most closely related to rootability was cell layer arrangement in the sclerenchyma tissue. The lower rootability varieties developed two or three overlapping cell layers in the bark tissue and in the higher rootability varieties they were scattered over the primary cortex. 3. In the higher rootability varieties, there was a positive correlation between the development of root primodia and rootability of the hardwood cuttings. It was also shown that there was a close relationship between the size of primodia and the surface area of the lenticel with rootability of the cuttings. 4. Effect of growth substances extracted from the hardwood cuttings were determined by mung bean bioassay. The higher rootability varieties usually showed higher activities of the growth substances, in contrast the lower rootability varieties showed higher activities of the inhibitory substances. 5. It was evident that the substance separated by paper chromatography was identified as indole acetic acid with $R_f$ value ranging from 0.3 to 0.5. The other substances detected at a $R_f$ value ranging from 0.8 to 1.0 and origin to 0.1 were also responsible for rooting. 6. There exists a quantitatively different distribution of growth substances in a synergistic system in the tissues of cuttings, and the balance between growth and inhibitory substances gives rise to the development of rooting. Particularly, no descent of the substances from winter buds resulted in no rooting of cuttings but these substances were produced a week after planting in a warm environment. 7. It was shown that there were positive correlations between carbohydrate ($r=0.72^*$) and total sugar ($r=0.67^*$) and rootability, respectively, but there were negative correlations between reducing sugars ($r=-0.75^*$) and rootability. 8. High C/N ratio gave rise to high rootability($r=0.67^*$). The latter therefore depended on high amount of carbohydrate rather than nitrogen in the cuttings. 9. The content of RNA and DNA in the cuttings was not changed for upto two weeks after the cuttings were planted. Then an increase in RNA content took place in only the high rootability varieties. 10. There were quantitative and qualitative differences in the compositions of the amino acids between the high rootability varieties and the low rootability varieties. More aspartic acid and cystine were found in the higher rootability varieties than in the low rootability varieties.

Toxic Effects of Phenol on Survival and Oxygen Consumption of the Abalone Juvenile, Haliotis discus hannai (참전복, Haliotis discus hannai 치패의 생존과 산소소비에 미치는 phenol의 독성 영향)

  • This study was carried out to estimate toxic effects of phenol on survival and metabolism of the abalone juvenile, Haliotis discus hannai. The experiment was conducted by renewal bioassay procedure with different salinities at $20^{\circ}C$. The $LC_{50}$ of the juvenile exposed to phenol in the range of 0.5 and $100mg/\ell\;was\;34.3\~6.5mg/\ell\;at\;2.4\%_{\circ}\;and\;52.2\~9.3m/\ell\;at\;32\%_{\circ}$ salinity with exposure time from 24 hours to 96 hours. $LT_{50}$ was remarkablely reduced with increase of phenol conentration and decrease of salinity. Lethal toxicity or phenol was higher at low salinity than at high salinity. Therefore, salinity is likely to be one of factor to increase phenol toxicity. The oxygen consumption of the juvenile was reduced with increase of phenol concentration and with decrease of salinity. In spite of phenol toxicity, the oxygen consumption of the juvenile exposed to phenol of low concentration was high and similar as compared with that of control group. Survival rates of the abalone kept in phenol-free sea water after exposure to phenol concentration of 5, 10 and $20mg/\ell$ for 96 hours were reduced with decrease of salinity. Durations required to recover the normal metabolic rate of the juvenile, which was exposed to phenol concentration of 5, 10 and $20mg/\ell$ for 96 hours, were made longer with increasing phenol concentration. In the case of the juvenile exposed to sublethal concentration of phenol for 15 days, it were elongated as compared with that of the abalone exposed to phenol concentration caused acute toxicity. The result of this experiment indicated that relatively low concentration of phenol can impact on the abalone juvenile in marine ecosystem.

Acute and Chronic Toxicity of Phenol to Mysid, Archaeomysis kokuboi (곤쟁이, Archaeomysis kokuboi에 미치는 phenol의 급성 및 만성독성)

  • Acute and chronic toxicity of phenol on the mortality, long-term survival and respiration rates of the mysid, Archaeomysis kokuboi adult and juvenile were examined. This experiment was condurted by static bioassay procedure with the different salinity at $25^{\circ}C$ In lethal test, the test animals were exposed to 6 different phenol concentrations to determine $LC_{50}$ and I$LT_{50}$ (median lethal concentration and time) values. The $LC_{50}$ values with the exposure time for the mysid adult ranged from 31.31ppm to 1.49ppm phenol and for the mysid juvenile ranged from 6.90ppm to 0.26ppm in all experimental groups. Mortality was increased with the decrease of salinity, The $96hr-LC_{50}$ values at 16, 24 and $32\%o$ salinity for the mysid adult were 1.49, 2.71 and 4.53ppm phenol, white the values for the mysid juvenile were 0.26, 0.56 and 0.71ppm, respectively. The ratios of $96hr-LC_{50}$ values for the mysid adult to those for the mysid juvenile at 16, 24 and $32\%p$ salinity were 5.73, 4.84 and 6.38, respectively. The mysid juveniles were more sensitive to phenol than the mysid adults. Compared $LT_{50}$ values for the mysid adult with those for the mysid juvenile, the $LT_{50}$ values for the mysid adult ranged from 384.7 to 29.0 hours at 1.7-127ppm phenol concentrations and for the mysid juvenile ranged from 132.2 to 18.7 hours at 0.5~6.Oppm phenol concentrations. The lowest $LT_{50}$ values for the mysid adult and juvenile were showed at the combination of the highest experimental concentration of phenol and the lowest experimental salinity. The mysid juveniles showed lower $LT_{50}$ values than those of adults. The chronic effects of phenol on the mysid at the sublethal effective concentration of phenol were lower in the $32\%o$ salinitr group than 16 or $24\%o$ salinity groups. Oxygen consumption rates of the mysid adult were decreased with the increase of phenol concentration and exposure time, and decreased significantly in lower salinity at the same concentration or phenol.

Studies on the Bioavailable Amino Acid of Feather Meals Processed by Different Methods - Available Amino Acid on the Meal Feeding of Semipurified and Purified Diet with Chick - (가공방법을 달리한 우모분의 아미노산 이용율에 관한 연구(I) -순수사료와 준순수사료의 Meal Feeding 하에서의 아미노산 이용율 -)

  • This study was conducted to bioassay of amino acid availability of feather meal processed by a different methods, commercial feather meal and raw feather meal, The feather meals were processed by labolatory pressure cooker(autoclave) at 2kg/$\textrm{cm}^2$ for 30 minutes ; 3kg/$\textrm{cm}^2$ for 90 minutes : 4kg/$\textrm{cm}^2$ for 120 minutes. Chick employed in the present experiment were Abor Acre strain, male or meat type (body weight, 100-140g), fed with semipurified diet and protein free diet was given during the determination of the metabolic and endogeneous amino acid. The contents of amino acid of samples were investigated by ion-exchange chromatography. The results were as follows; 1. The amino acid availability of raw, 2kg/$\textrm{cm}^2$ for 30 minutes, commercial, 4kg/$\textrm{cm}^2$ for 120 minutes and 3kg/$\textrm{cm}^2$ for 90 minutes of feather meal were -3.09, 63.28, 67.47, 71.22 and 73.75% respectively. 2. The essential amino acid availability of raw, 2kg/$\textrm{cm}^2$ for 30 minutes, commercial, 4kg/$\textrm{cm}^2$ for 120 minutes and 3kg/$\textrm{cm}^2$ for 90 minutes of feather meal were 2.55, 66.78, 66.89, 72,56 and 73.62%, respectively. 3. In individual amino acid of the different processing loather meal and commercial feather meal, biovailabilities were increased methionine, phenylalanine, leucine, arginine, threonine, isoleucine, however, histidine, lysine and aspartic acid were remarkely decreased. In conclusion, the bioavailability or amino acid for the feather meal processed at 3kg/$\textrm{cm}^2$ for 90 minutes was superior to those of other treatment or raw feather meal.

The Effect of Hypoxia on the Release of Endothelium-derived Relaxing Factor in Rabbit Thoracic Aorta (토끼 대동맥 혈관내피세포에서 저산소증이 내피세포성 이완인자의 분비에 미치는 영향)

  • Background: To clarify the effect of hypoxia on vascular contractility, we tried to show whether hypoxia induced the release of endothelium-derived relaxing factor (EDRF) and the nature of the underlying mechanism for this release. Material and Method: Isometric contractions were observed in rabbit aorta, and the released EDRF from the rabbit aorta was bioassayed by using rabbit denuded carotid artery. The intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) in the cultured rabbit aortic endothelial cells was recorded by a microfluorimeter with using Fura-2/AM. Hypoxia was evoked to the blood vessels or endothelial cells by eliminating the $O_2$ in the aerating gases in the external solution. Chemical hypoxia was evoked by applying deoxyglucose or $CN^-$. Result: Hypoxia relaxed the precontracted rabbit thoracic aorta that had its endothelium, and the magnitude of the relaxation was gradually increased by repetitive bouts of hypoxia. In contrast, hypoxia-induced relaxation was not evoked in the aorta that was denuded of endothelium. In a bioassay experiment, hypoxia released endothelium-derived relaxing factor (EDRF) and the release was inhibited by L-NAME or the $K^+$ channel blocker tetraethylammonium (TEA). In the cultured endothelial cells, hypoxia augmented the ATP-induced increase of the intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) and this increase was inhibited by TEA. Furthermore, chemical hypoxia also increased the $Ca^{2+}$ influx. Conclusion: From these results, it can be concluded that hypoxia might induce the release of NO from rabbit aortic endothelial cells by increasing $[[Ca^{2+}]_i$.

Distribution of Quinolones (Ciprofloxacin, Norfloxacin and Oxolinic acid) after Oral Administration in Carp (Cyprinus carpio) (잉어에 있어서 Quinolones (Ciprofloxacin, Norfloxacin 및 Oxolinic acid)의 경구투여에 따른 장기내 분포상)

  • The concentrations of quinolones (oxolinic acid; OXA, norlloxacin: NRF & ciprofloxacin: CPF) after oral administration of single doses (20 mg/kg B.W.) were investigated in carp (Cyprinus carpio) kept in freshwater at 20-23$^{\circ}C$. The distribution of the drug was studied after treatment. At points timed, from 1 h to 96 hrs after administration, blood (B), liver (L), kidney (K) and muscle (M) from 5 individuals in each group were collected for analyse with microbiological bioassay method. The peak concentrations were measured at 8 h (L), 12 h (B and K) and 24 h (M) after administration regardless of treated drugs. Considerably high concentrations of CPF (13.8-19.6${\mu}g/m{\ell}$) NRF (11.8-16.9${\mu}g/m{\ell}$) and OXA (10.8-13.9 ${\mu}g/m{\ell}$) were revealed during the 24 h. At the last time point of the experiment (96 h), concentrations of all three quinolones were: OXA, 2.3-6.3 ${\mu}g/m{\ell}$ ; NRF, 3.1-4.5 ${\mu}g/m{\ell}$ ; CPF, 3.0-5.5${\mu}g/m{\ell}$ in samples. The concentrations decreased subsequently, indicating a first rapid redistribution, followed by a slow phase of elimination. The steady state was observed in blood (12-36 h), liver (12-96 h) and muscle (36-96 h) after the initiation of treatment with OXA. Concerning the compartmental concentrations, (L, K. and M/B concentration ratio), the fluctuation of the ratio was founded at different time points, among drugs. For CPF, highest tissue ratios were prolonged in the order of L>K>M (0.65-1.2/0.82-0.93/1.0-1.7) during the experiments. On the other hand, NRF presented L>K>M (0.65-1.3/0.86-1.0) till 24 h, but L>M>K (0.89-1.26) at 36-96 h. OXA showed L>K>M (0.95-2.1) at 1-8 h, M>K>L (0.51-1.0) at 12-36 hand M>L>K (1.0-2.3) at 48-96 h, respectively.

Field Bioassays On Shellfish To Assess Environmental Pollution Levels Of The Masan Bay (마산만(馬山灣)의 환경오염(環境汚染) 평가(評價)를 위한 야외(野外) 생물(生物) 오염(汚染) 시험(試驗) 연구(硏究))

  • A study on field bioassay test using four species of commercially important shellfish was carried out to assess the effect of pollutants and determine the extent of marine environmental pollution of the Masan Bay from 9 to 15 August 1978. Water quality analysis and planktological examination of sea water were made during the experiment and the examination gave the following results. The water temperature was so high at 31.7$^{\circ}C$ in maximum and rather subject to change on weather condition of the land than on the effect of the water mass from outer bay. The range of DO,COD and SS at the stations were 0.3-7.08cc/l, 0.07-3.31ppm and 5.5-117ppm, respectively with the high values of COD and SS at the stations 7 and 1. The concentrations of the dissolved inorganic nitrogen in sea water, NH$\sub$4/-N, NO$\sub$2/-N, NO$\sub$3/-N, and PO$\sub$4/-P were 18.90-99.80, 2.48- 19.60, 13.00-39.00 and 1.04-14.0$\mu\textrm{g}$ at/l, respectively with decrease of their values in the outer part of the Bay. The high values mentioned above were caused by the sewage and industrial activities. The effects of organic waste are increased oxygen demand, nutrient concentration, turbidity and a higher input of pathogens, leading to structural changes in the marine ecosystems and to a considerable hazard to public health. The percentage composition of phytoplankton standing crop between diatom and dinoflagellate was characterized by making a difference between the two groups in respect of location: a decrease of diatom and a increase of dinoflagellate in numerical abundance toward inner part from outer part of the bay. Namely phytoplankton organisms were composed of 80% of diatom and 20% of dinoflagellate in outer bay, on the contrary, only 4% of diatom and 96% of dinoflagellate occupied by 94% of prorocentrum micans known as tolerant species to polluted reaas in the inner bay. On the occurrence and composition of zooplankton, there are two significant communities in the bay:one is characterized by the predominance of Oithona nana and the other by Favella sp They were composed of a range from 84% to 90% of the total organisms and monotonously constituted of themselves only at most inner station 3 even small numbers. From the results mentioned above, Oithona nana, Favella and prorocentrum micans recommed themselves as valuable indicators for judging the extent of the marine pollution.During the period of the biossays Mytilus edulis showed the highest mortality and Tapes japonica the lowest one between the four test species. The highest death rate by stations was found at most inner stations 3'and 4near Masan Free Export Zone with the most sensitive response and the lowest one occured at outer station 13 where no death specimen of oyster and arkshell was found during the whole test period.As for mussel,85 percent death rate appeared after 72 hours and 100percent rate after 120hours at station 4. It was found that the significant high mortality of the test shellfish mentioned above was caused by severe pollution with mainly organic pollutants from domestic sewage and industrial wastes from the results of too much higher concentrations of dissolved inorganic nitrogen especially ammonia-N,COD,SS and lack of dissolved oxygen,and furthermore occurrence and abundance composition of Prorcentrum,Favella and Oithona nana by stations, valuable indicator species of coastal pollution by orgnic and boilogical pollutants.

Analysis of Algal Response Effects Using Nutrient Enrichment Bioassays in Shingu Reservoir (신구저수지 식물플랑크톤성장에 대한 제한 영양염 분석)

  • Nutrient Enrichment Bioassays (NEBs) were conducted in the laboratory during June $22{\sim}28$, 2006 in order to determine primary limiting factor on the phytoplankton growth. For the NEBs, the water was sampled using a 10L polyethylene-lined container and dispensed into 2.5L container in the laboratory. The algal growths response in the control (C) and three treatments of phosphorus (P), 2-fold phosphorus (2P), and nitrate nitrogen $(NO_3-N)$ were monitored during 7 days. In the cubitainers which were spiked as P (T1) and 2P (T2) Chl-${\alpha}$ concentrations were decreased during the test period and the final concentrations was low than initial values. However, Chl-${\alpha}$ in the cubitainers which were spiked as $NO_3$(T3) and $P+NO_3$(T4) showed significant increases compared to the initial values, indicating that in the short-term experiments, nitrogen seemed to be a primary limiting nutrient during the periods of NEBs experiment. Long-term ambient nutrient data of TP and TN, and TN:TP mass ratios, however, showed a potential phosphorus limitation on phytoplankton growth and previous other researchers showed a variations of limiting nutrients by nitrogen or phosphorus depending on the seasons sampled and locations. In this study nitrogen as primary limiting nutrient in the NEBs seem to be an seasonal effect rather than the consistent nitrogen limitation.
