Physiological Characteristics and Anti-obesity Effect of Lactobacillus plantarum K10

  • This study aimed to investigate the physiological characteristics and anti-obesity effects of Lactobacillus plantarum K10. The ${\alpha}-amylase$ inhibitory activity, ${\alpha}-glucosidase$ inhibitory activity, and lipase inhibitory activity of L. plantarum K10 was $94.66{\pm}4.34%$, $99.78{\pm}0.12%$, and $87.40{\pm}1.41%$, respectively. Moreover, the strain inhibited the adipocyte differentiation of 3T3-L1 cells ($32.61{\pm}8.32%$) at a concentration of $100{\mu}g/mL$. In order to determine its potential for use as a probiotic, we investigated the physiological characteristics of L. plantarum K10. L. plantarum K10 was resistant to gentamycin, kanamycin, streptomycin, ampicillin, ciprofloxacin, tetracycline, vancomycin, and chloramphenicol. It also showed higher Leucine arylamidase, Valine arylamidase, and ${\beta}-galactosidase$ activities. Moreover, it was comparatively tolerant to bile juice and acid, exhibiting resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus with rates of 90.71%, 11.86%, 14.19%, and 23.08%, respectively. The strain did not produce biogenic amines and showed higher adhesion to HT-29 cells compared to L. rhamnosus GG. As a result of the animal study, L. plantarum K10 showed significantly lower body weight compared to the high-fat diet group. The administration of L. plantarum K10 resulted in a reduction of subcutaneous fat mass and mesenteric fat mass compared to the high-fat diet (HFD) group. L. plantarum K10 also showed improvement in gut permeability compared to the HFD positive control group. These results demonstrate that L. plantarum K10 has potential as a probiotic with anti-obesity effects.

Optimizing the fermentation condition of low salted squid jeotgal by lactic acid bacteria with enhanced antioxidant activity

  • Lactic acid bacteria (LAB) are widely used as starter culture in food fermentation due to their harmless entity and health beneficial properties along with the ability to change texture, aroma, flavor and acidity of food products. In this study, five different LAB (FB003, FB058, FB077, FB081, and FB111) isolated from different Korean traditional fermented foods, assigned to Lactobacillus plantarum, Pediococcus pentosaceus, Weissella viridescens, Lactobacillus sakei, and Leuconostoc mesenteroides, respectively, on the basis of their physiological properties and 16S rRNA sequence analysis, to use as fermentation starter and check their ability to fasten the ripening time as well as the overall optimization in the fermentation condition. To check their suitability as starters, their safety, acid and bile tolerance, NaCl and temperature resistance, susceptibility to common antibiotics, and antimicrobial activities were determined. Squid jeotgal samples were prepared by adding $10^8CFU/g$ of each strain in different samples, which were then kept for fermentation at $4^{\circ}C$ and checked for their antioxidant activities at 0, 7, 15, and 21-day intervals. The samples fermented with FB003 and FB077 displayed the highest antioxidant activity. This study revealed two effective starter cultures (FB003, FB077) for squid jeotgal fermentation, which presented increased functionalities. The results of this study will lead to the development of novel industrial-scale production avenues for jeotgal preparation, and offer new insights into the prevention and control of chronic diseases.

Stress Tolerance of Bifidobacterium infantis ATCC 27920 to Mild-heat Adaptation

  • Two-dimensional gel electrophoresis (2-DE) was employed to assess the thermo-tolerance characteristics of Bifrdobacterium infantis ATCC 27920 to mild heat adaptation. When exposed to various heat levels, pH, and hydrogen peroxide ($H_2O_2$) stress conditions, B. infantis ATCC 27920 exhibited high level of stress resistance. Under mild-heat treatment ($46^{\circ}C$), no significant change in viability level was observed after 2 hr. Interestingly, improved viability was observed in mild-heat adapted ($46^{\circ}C$ for 1 hr) cultures exposed to $55^{\circ}C$, in comparison to control experiments. Viability was not affected by pH, bile, and $H_2O_2$ stress conditions. 2-DE analysis revealed those mild-heat adaptation up-regulated 4 proteins and down-regulated 3 proteins. Among these protein spots, isopropyhnalate dehydratase (leuD), glycosyltransferase (glgA), and ribosomal protein L5 (rp1E) were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALD1-TOF/MS).

넙치, Paralichthys olivaceus의 비특이적 방어기작에 대한 Edwardsiella tarda 균의 반응 (Responses of Edwardsiella tarda to innate immunity of olive flounder, Paralichthys olivaceus)

  • This paper aims to compare difference with the in an ability of their resistance and survival against in a non-specific defence mechanism of the olive flounder, between the virulent and the avirulent E. tarda strains. The tested E. tarda strains, we divided into the virulent and the avirulent strain groups on the basis of a value of 50% lethal dose (LD50) for the olive flounder weighed 10.3 g in average. The strains of LD50 101.6~104.2 cfu/fish were grouped as virulent strains, such as KE-1, KE-3, KE-5 and FSW910410. The group of avirulent strains as LD50 exceeded 108.7 cfu/fish were included the strains, SU100 and AL92448. A test was conducted to understand the survival ability of each strain in the mucus of the skin and the intestine of olive flounders. The results showed KE-1, KE-3, KE-5 and FSW910410 were highly to survive between 6 hours and 24 hours in intestine. The survival ability in the bile of olive flounder the number of avirulent strains declined during incubation but the virulent strain showed the number of alive bacteria having sustained or increased. In the test for the survival of bacteria in fresh sera of olive flounder, the virulent strains also had tendency to multiply. Concerning the tested bacteria internalization into the head kidney macrophages and the intracellurar replication in the macrophages of olive flounder. The virulent strains exhibited strong internalization, followed high rate replication. According to the results, virulent strains of E. tarda revealed more ability to resist and survive in the face of humoral and cellular defence factors than avirulent strains.

백김치 유래 유산균을 이용한 요구르트의 Anti-Helicobacter pylori 활성 (Anti-Helicobacter pylori Activity of Yogurt Fermented with Lactic Acid Bacteria from Baikkimchi)

  • 백김치로부터 분리된 유산균으로 제조된 요구르트를 냉장 보관하는 동안 미생물학적 및 물리화학적 특성 및 Helicobacter pylori ATCC 43504에 대한 항균 활성을 조사하였다. 요구르트의 유산균수, 적정산도, 점도 및 총 고형물 함량은 사용된 균주에 따라 유의적인 차이가 있었으나, 발효 직후부터 7일간 저장하는 동안 유의할만한 차이 없이 일정하게 유지되었다. Lactobacillus brevis BK11과 Lactobacillus paracasei BK57 균주로 발효시킨 요구르트는 인공 위액과 담즙액에 대해 다른 균주들 보다 강한 저항성을 보였다. 한편, 이들 유산균으로 제조한 요구르트 내에 존재하는 유산 생성량은 상대적으로 높았으므로 H. pylori와 혼합 배양한 결과 대조구에 비해 유의적인 항균 효과를 나타낸 것으로 추정되었다. 특히, L. brevis BK11에 의해 발효시킨 요구르트에 의해선 AGS 세포에 대한 H. pylori의 부착을 억제할 수 있었고, 이들이 생산하는 urease의 활성을 낮추는데도 효과적이라는 것을 확인하였다.

사료용 생균제 개발을 위한 마늘 내성 유산균의 분리 (Isolation of Garlic Resistant Lactic Acid Bacteria for Feed Additives)

  • 마늘의 항균력에 저항성을 갖는 유산균을 분리하기 위해서 마늘 추출물이 첨가된 배지에서 생육하는 112 균주를 김치, 젓갈, 장아찌류로부터 분리하였다. 이 중에서 14균주에 대해서 내산성 및 내담즙성 그리고, 가축 병원성 세균인 Salmonella choleraesuis, Escherichia coli, Staphylococcus aureus, Shigella flesneri에 대해 항균력을 조사한 결과, 파김치 유래의 P'GW50-2 균주가 가장 우수한 특성을 지니고 있어 생균제로 이용 가능한 균주로 선발하였다. 선발된 균주는 16S rRNA 유전자의 염기서열 분석결과 Lactobacillus plantarum으로 동정되었고 L. plantarum TJ-LP-002으로 명명하였다. L. plantarum TJ-LP-002는 Bacillus cereus, S. aureus, Clostridium perfringens와 같은 Gram 양성균과 Aeromonas hydrophila, E. coli, Pseudomonas, Salmonella, Shigella와 같은 Gram 음성균에 대해서 비교적 넓은 항균활성을 나타내었다.

Effect on Viability of Microencapsulated Lactobacillus rhamnosus with the Whey Protein-pullulan Gels in Simulated Gastrointestinal Conditions and Properties of Gels

  • Lactobacillus rhamnosus GG (LGG) has low resistance to low pH and bile salt in the gastrointestinal juice. In this study, the gel made from whey protein concentrate (WPC) and pullulan (PUL) was used as the wall material to prepare the microencapsulation for LGG protection. The gelation process was optimized and the properties of gel were also determined. The results showed the optimal gel was made from 10% WPC and 8.0% PUL at pH 7.5, which could get the best protective effect; the viable counts of LGG were 6.61 Log CFU/g after exposure to simulated gastric juice (SGJ) and 9.40 Log CFU/g to simulated intestinal juice (SIJ) for 4 h. Sodium dodecyl sulphite polyacrylamide gel electrophoresis (SDS-PAGE) confirmed that the WPC-PUL gel had low solubility in SGJ, but dissolved well in SIJ, which suggested that the gel can protect LGG under SGJ condition and release probiotics in the SIJ. Moreover, when the gel has highest hardness and water-holding capacity, the viable counts of LGG were not the best, suggesting the relationship between the protection and the properties of the gel was non-linear.

김치에서 분리한 Lactobacillus plantarum 균주들의 프로바이오틱 효과 (Probiotic Effects of Lactobacillus plantarum Strains Isolated from Kimchi)

  • 본 연구에서는 김치에서 분리한 L. plantarum pF1 NITE-P1462(Lp-pF1), L. plantarum KCCM 11352P(Lp-PNU), L. plantarum CBT LP3 KCTC 10782BP(Lp-CB)와 L. plantarum KCTC 3099(Lp-3099)의 프로바이오틱 효과(내산 및 내담즙성 측정, 장 부착능과 열 안정성, 항산화 및 항암 기능성의 측정)를 된장에서 유래한 Lac. lactis KFCC 11510P(L-lactis)와 비교하여 측정하였다. 그 결과 4종의 김치 L. plantarum이 Lac. lactis보다 전반적으로 높은 프로바이오틱 효과를 나타내었다. 4종의 김치 L. plantarum 중 내산 및 내담즙성 측정에서 Lp-pF1이 유의적으로 가장 높은 안정성을 나타냈고, 장 부착능 측정에서도 Lp-pF1이 Lp-CB와 함께 가장 뛰어난 부착능을 보였으며, 열 안정성 측면에서는 Lp-pF1과 Lp-PNU가 가장 높은 안정성을 가진 것으로 나타났다. 또한, DPPH 라디칼 및 수산화 라디칼소거 활성 측정을 통해 Lp-pF1이 모든 유산균 중 유의적으로 가장 높은 항산화 활성을 나타내었고, HT-29 인체 대장암세포에서도 가장 높은 암세포 성장 억제 효과를 나타냈다. 따라서 Lp-pF1의 프로바이오틱으로서의 이용가능성을 확인하였으며, Lp-pF1은 특히 높은 항산화 및 항암 활성을 가지고 있어 향후 기능성 프로바이오틱으로 활용할 가능성이 높다고 할 수 있다. 또한, Lp-pF1을 앞서 연구했던 나노화된 Lp-pF1과 함께 섭취하였을 때 단독으로 Lp-pF1만 섭취하는 것보다 더 높은 건강 기능적 효과가 기대되며, 이에 대한 연구도 추가적으로 필요할 것으로 생각된다.

기성금관 수복어린이에서의 니켈내성균에 관한 연구 (A STUDY OF NI-RESISTANT BACTERIA ON THE RESTORED STAINLESS STEEL CROWN)

  • 소아의 유구치 수복에 흔히 사용하는 stainless steel crown은 주성분으로 $70\sim90%$의 니켈과 $5\sim15%$의 크롬을 함유하고 있다. 이 중 니켈 자체가 일으킬 수 있는 allergy반응이나 발암성등의 부작용이 성인에서는 활발하게 연구되고 있으며, 특히 니켈이 포함된 합금을 이용한 보철물 주변에 병원성이 있는 니켈내성 Enterococci가 존재한다는 보고가 있어 성인과는 구강환경이 다른 소아에서도 stainless steel crown 주변에 니켈내성균주가 존재하는지를 확인하고자 이 실험을 시행하였다. 유구치에 stainless steel crown(Anatom primary crown, Sankin)을 6개월 이상 장착한 환자 15명과 유구치 협면에 치아우식증이 없는 환자 15명의 치은열구액에서 채취한 시료를 BHI 한천배지, 니켈배지 및 Bile-esculin azide(BEA) 한천 배지에 도말하였다. BEA한천배지에서 자란 enterococci를 니켈내성정도를 확인하기위해 3, 5, 10, 30, 50mM 니켈배지에 접종한후 성장상태를 관찰하였다. 실험군에서는 BHI 한천배지에서 507,350개의 균주가 분리되었으며, 이 중 니켈내성세균(3mM)은 53,864 균주였다. 대조군에서는 BHI 한천배지에서 414,590 균주가, 니켈내성세균(3mM)은 37,523 균주가 분리되었다. BEA한천배지에서는 실험군의 경우 95개가 분리되었으며, 대조군에서는 20개의 균주만이 분리되었다. BHI 한천배지에서 분리된 총 균주 수를 100으로 놓았을 때 3mM 니켈배지에서 분리된 균주는 실험군에서는 10.62%, 대조군에서는 9.05%를 차지하여 두 군간에 뚜렷한 차이는 없었으며, 3mM 니켈배지에서 자란 총 균주 수와 BEA 한천배지에서 배양된 enterococci를 3mM 니켈배지로 옮겨 배양했을 때와 비교하면, 실험군에서 0.13%, 대조군에서 0.03%로 모두 1%이하의 분포를 차지하였다.

Cytotoxicity, Toxicity, and Anticancer Activity of Zingiber Officinale Roscoe Against Cholangiocarcinoma

  • Cholangiocarcinoma (CCA) is an uncommon adenocarcinoma which arises from the epithelial cells of the bile ducts. The aim of the study was to investigate the cytotoxicity, toxicity, and anticancer activity of a crude ethanolic extract of ginger (Zingiber officinale Roscoe) against CCA. Cytotoxic activity against a CCA cell line (CL-6) was assessed by calcein-AM and Hoechst 33342 assays and anti-oxidant activity was evaluated using the DPPH assay. Investigation of apoptotic activity was performed by DNA fragmentation assay and induction of genes that may be involved in the resistance of CCA to anticancer drugs (MDR1, MRP1, MRP2, and MRP3) was examined by real-time PCR. To investigate anti-CCA activity in vivo, a total of 80 OV and nitrosamine (OV/DMN)-induced CCA hamsters were fed with the ginger extract at doses of 1000, 3000, and 5000 mg/kg body weight daily or every alternate day for 30 days. Control groups consisting of 10 hamsters for each group were fed with 5-fluorouracil (positive control) or distilled water (untreated control). Median $IC_{50}$ (concentration that inhibits cell growth by 50%) values for cytotoxicity and anti-oxidant activities of the crude ethanolic extract of ginger were 10.95, 53.15, and $27.86{\mu}g/ml$, respectively. More than ten DNA fragments were visualized and up to 7-9 fold up-regulation of MDR1 and MRP3 genes was observed following exposure to the ethanolic extract of ginger. Acute and subacute toxicity tests indicated absence of any significant toxicity at the maximum dose of 5,000 mg/kg body weight given by intragastric gavage. The survival time and survival rate of the CCA-bearing hamsters were significantly prolonged compared to the control group (median of 54 vs 17 weeks). Results from these in vitro and in vivo studies thus indicate promising anticancer activity of the crude ethanolic extract of ginger against CCA with the absence of any significant toxicity. Moreover, MDR1 and MRP3 may be involved in conferring resistance of CCA to the ginger extract.
