Fermentative characteristics of yogurt using lactic acid bacteria isolated from Korean traditional fermented food (전통 발효 식품에서 분리한 유산균을 이용한 yogurt 발효특성)

  • The objective of this study was to select yogurt starter from Korean traditional fermented foods. The 2 strains (KM24, KM32) among 50 strains of isolated lactic acid bacteria selected as starter based on milk clotting ability, antimicrobial activity against various pathogens, tolerance in artificial gastric and bile juice and growth in 10 % skimmed milk. The strains were identified as Lacobacillus plantarum (KM32) and Pediococcus pentosacesus (KM24) by 16S rRNA gene sequencing. Viable cell number of yogurt fermented with mixed strains (KM24 and KM32) was 9.66 log CFU/mL after fermentation for 48 h and maintained $10^9CFU/mL$ during fermentation for 72 h at $37^{\circ}C$. The pH and titratable acidity of mixed cultured yogurt were 4.25% and 0.83% after fermentation for 48 h at $37^{\circ}C$, respectively. The physico-chemical characteristics of mixed cultured yogurt after fermentation for 48 h were $38.45{\mu}g/mL$ (polyphenol content), 48.57% (DPPH radical scavenging activity) and 465.40 cp (viscosity), respectively. The mixed cultured yogurt maintained $10^9CFU/mL$ of lactic acid bacteria during storage 10 days at $4^{\circ}C$. The viable cell number of yogurt prepared with mixed culture(KM32+KM24) maintained higher and than that of control (L. casei) during storage. These results indicated the potential use of selected strains (KM32+KM24) isolated from kimchi as a yogurt starter with strong acid tolerance and probiotics properties.

Yogurt Production Using Exo-polysaccharide-producing Leuconostoc and Weissella Isolates from Kimchi (김치유래 exo-polysaccharide 생성능 Leuconostoc 및 Weissella균을 이용한 발효유 제조)

  • The purpose of this study was to investigate the effect of exopolysaccharide (EPS)-producing Leuconostoc and Weissella isolates from kimchi as a probiotic starter and replacement for thickening agents such as pectin and gums in yogurt. Potential probiotic isolates were first screened for their acid and bile tolerance, and then evaluated for antimicrobial activity against Escherichia coli and Salmonella Typhimurium. When the selected Leuconostoc or Weissella isolates were co-inoculated in yogurt without a thickening agent, the yogurt with 4% sucrose produced lower syneresis values than the control and had higher EPS yields. The isolates were able to survive at a level of $10^6CFU/mL$ when incubated at $4^{\circ}C$ for 12 days. This study shows that EPS-producing Leuconostoc and Weissella strains have the potential to produce a synbiotic yogurt.

Probiotic Properties of Enterococcus durans LP44 Isolated from Pigs Feces (돼지의 분변에서 분리한 Enterococcus durans LP44의 생균제로서의 특성)

  • We have screened the microorganisms from pigs feces for the development of probiotics which have acid-and bile-tolerance. Among them, a strain which was identified as Enterococcu durans LP44 was selected. Sixty Duroc$\times$Yorkshire$\times$Landrace pigs (6.30$\pm$0.04 kg average initial body weight) were used in a 14 days growth assay to determine the effects of Enterococco duran Lf44 culture on growth performance of weaned pigs. Dietary treatments included 1) DF (dry feeding),2) W (wet feeding),3) WF+5ED (wet feeding+5% Enterocucu duran LP44 culture),4) WF+ 10ED (wet feeding +10% Enterococcus durans LP44 culture). For overall periods, average daily gain in pigs fed % diet tended to increase compared to pigs fed DF diet without significant difference. Also, there was no significant difference in gain/feed. At 5 days after the onset of the experiment, pigs fed WF+5ED and WF+10ED diets were significantly increased in dry matter and nitrogen digestibilities compared to pigs fed DF and W diets (p<0.05). At 14 days after the onset of the experiment, pigs fed WF+ 10ED diet were significantly increased in dry matter digestibility compared to pigs fed DF diet (p<0.05). Also, pigs fed WF + 10ED diet were significantly increased in nitrogen digestibility compared to other treatments (p<0.05). In conclusion, supplemental Enterococcus durans LP44 was an effective means of improving ADG and nutrient digestibility.

Probiotic Effects of Lactobacillus plantarum Strains Isolated from Kimchi (김치에서 분리한 Lactobacillus plantarum 균주들의 프로바이오틱 효과)

  • Probiotic effects of Lactobacillus plantarum pF1 NITE-P1462 (Lp-pF1), L. plantarum KCCM 11352P (Lp-PNU), L. plantarum CBT LP3 KCTC 10782BP (Lp-CB), and L. plantarum KCTC 3099 (Lp-3099) isolated from kimchi and Lactococcus lactis KFCC 11510P (L-lactis) isolated from Doenjang were studied. Resistance to gastric and bile acid, adhesion to intestines in colon cells, thermal stability, and antioxidative and in vitro anticancer effects in HT-29 cancer cells were evaluated. L. plantarum strains showed improved tolerance of gastric and bile acids than L-lactis. Lp-pF1 had better adhesion ability in the intestine than Lp-PNU, Lp-3099, and L-lactis. Lp-pF1 also showed better heat resistance at $50^{\circ}C$, $70^{\circ}C$, and $80^{\circ}C$ than Lp-CB, Lp-3099, and L-lactis. In addition, Lp-pF1 exhibited greater antioxidant activity by scavenging DPPH radicals or hydroxyl radicals and anticancer effects in MTT assay than others. Taken together, these results suggest that L. plantarum isolated from kimchi showed higher probiotic activities with antioxidant and anticancer properties than Lac. lactis isolated from Doenjang. Lp-pF1 revealed the best probiotic activities among L. plantarum and could be used as a promising potential probiotics.

Potential probiotics activity of Bacillus spp. from traditional soybean pastes and fermentation characteristics of Cheonggukjang (전통장류유래 Bacillus spp.의 프로바이오틱스 활성과 청국장 발효 특성)

  • In this study, we tried to screen the Bacillus strain having safety probability by isolation of strains from traditional fermented food, measurement of probiotic properties, and the fermentative characteristics of Cheonggukjang. We isolated 400 Bacillus-like isolates from traditional fermented foods. Selected strains examined on the prevalent characteristic such as extracellular enzyme and antibacterial activities, and their safety probability was confirmed by biogenic amine productivity, hemolytic, and harmful substances and enzyme productivity. We selected the 5 strains by analysis of biogenic amine, antibacterial and B. cereus toxic associated gene. Five selected strains were examined on cell surface hydrophobicity, and bile and acid tolerance, and we selected the SRCM100730 as the final strain. SRCM100730 was confirmed B. amyloliquefaciens by 16S rRNA sequencing, and named the B. amyloloquefaciens SRCM100730 (KCCM11966P). Finally, we manufactured Cheonggukjang using SRCM100730 for confirmation of fermentation properties. Manufactured Cheonggukjang did not contain B. cereus, and showed that ${\gamma}$-PGA and extracellular enzyme activities were superior to commercial Chunggukjang. Amino nitrogen content was 544.02 mg% and 26 free amino acid were detected, and the bitterness-related amino acid content was lower than commercial Cheonggukjang. Especially, the amount of GABA was 3 fold higher than commercial Cheonggukjang. These results suggest that SRCM100730 have high availability in commercial probiotics market and fermented food industry.
