Study on Extraction Condition and Analysis Methods of Benzopyrene in Black Ginseng (흑삼에서 벤조피렌의 추출 조건 및 분석 방법에 관한 연구)

  • To develop fundamental data of herbal materials with heat treatment, we studied analytical and extraction methods of benzopyrene in black ginseng and validated by HPLC analysis. Benzopyrene was successfully separated in mobile phase of acetonitrile:water (80 : 20) and detection of excitation 294 nm, emission 404 nm. The calibration curve of benzopyrene was linear over the concentration range of 1.17~37.50 ng/ml with correlation coefficient of above 0.999. The limit of detection (LOD) and quantitation (LOQ) of benzopyrene was 0.25 and 0.75 ng/ml, respectively. Hexane extraction method was used as a new extraction method for benzopyren and the efficient of extraction was over 95%. In conclusion, analytical method and extraction method were suitable for the determination of benzopyrene in the black ginseng and could be applied to fundamental study and guideline of herbal materials with heat treatment.

  • Benzopyrene is known to be one of the most powerful carcinogens which can build intercalated motif between base pairs in damaged DNA. The dimension of benzopyrene itself is much bigger than any of the DNA bases and thus the question whether the lesion of some base pair by insertion of benzopyrene can happen with or without a dramatic distortion of the helical structure is a highly interesting theme. In this work we used a molecular mechanics simulation using AMBER program package to go into the conformational characteristics. The condition of the insertion process of the benzopyrene motif from minor groove of the starting structure between the base pairs in the internal area of double helix was investigated using the molecular dynamics simulation at elevated temperature.

HPLC Analysis of Organic Acids, Phenol, and Benzopyrene in Wood Vinegar (HPLC에 의한 목초액 중의 유기산, 페놀 및 벤조피렌의 분석법 개발)

  • The organic acids (formic acid, acetic acid and propionic acid), phenol and benzopyrene in wood vinegar were determine by HPLC. An $Atlantis^{TM}dC_{18}$ column with a acetonitrile: 0.1% phosphoric acid (5: 95, organic acids), acetonitrile: water (10 : 90, phenol), 100% acetonitrile (benzopyrene) as a mobile phase was used. Retention time of acetic acid, formic acid, propionic acid, phenol and benzopyrene was 4.77, 3.73, 9.08, 30.97 and 6.10 min, respectively. The calibration curves of organic acids, phenol, benzopyrene were linear over the concentration range of $30{\sim}500,\;60{\sim}1000$, and $3{\sim}50\;{\mu}g/ml$ with correlation coefficient of above 0.999. The limit of detection (LOD) and limit of quantitation (LOQ) of acetic acid, formic acid, propionic acid, phenol, and benzopyrene was 1.71 and 5.19, 1.11 and 3.35, 4.87 and 14.74, 6.45 and 19.55, 0.08 and $0.24\;{\mu}g/ml$, respectively. The coefficients of variation for intra- and inter-day assay were $0.21{\sim}4.14$ and $0.07{\sim}1.19%$, respectively and the precision was $95{\sim}115%$. From this study, we suggest that HPLC is suitable method for the determination of organics in wood vinegar and could be applied to quality control of wood vinegar.

Conformational and Molecular Dynamical Properties of Damaged DNA (손상된 핵산의 구조와 분자동력학적 특성)

  • Some of the benzopyrene (BP)-DNA adduct are known to build intercalated motif between flanking base pairs in damaged DNA depending on the structural condition. The size of benzopyrene itself is definitely not comparable with any of the DNA bases and thus the question whether the lesion of some base pair by insertion of benzopyrene can happen with or without a dramatic distortion of the helical structure is a highly interesting theme. In this work we used a molecular dynamics simulation based on the theory of molecular mechanics. The specific consequences about the structural properties of the intercalated structures and benzopyrene motif in minor groove of the double helix are deduced after 5 ns simulation time.

Antimutagenic Effects of Bifidobacteria (Bifidobacteria에 의한 항돌연변이 효과)

  • The antimutagenic properties of twenty-one strains of Bifidobacterium were examined using Salmonella typhimurium TA98 in an in vitro assay system. The mutagens utilized for testing included Trp-P-1 (3-amino-1, 4-dimethyl-$^{5}H-pyrido$ (4, 3-b) indole), benzopyrene, IQ (2-amino -3-methylimidazo [4,5-f] quinoline), and NQO. The lyophilized cells of strain showed inhibitory effect of 64, 38, 29 and 20% in average against Trp-P-1, benzopyrene, NQO and IQ, respectively. There was no marked variation between each strain or growth stage in the degree of antimutagenicity against Trp-P-1, benzopyrene and IQ. Twelve hour grown cells showed higher antimutagenicity against NQO than 5-day grown cells. The results indicate that Bifidobacterium cells had a antimutagenic effect against several well-known mutagens to some degree.

  • Background: Cigarette smoking is an important risk factor for chronic bronchitis and COPD. Airway epithelial cells exposed to cigarette smoke components such as nicotine, cotinine and benzopyrene can generate reactive oxygen species (ROS) and be subject to oxidative stress. This oxidative stress can induce the inflammatory response in the lung by the oxidant itself or by the release of proinflammatory cytokines. It has been reported that nicotine stimulates ROS, which are associated with NF-${\kappa}B$. Methods: Beas2B cells were treated with nicotine, cotinine and benzopyrene. RT PCR was used to measure the expression of several antioxidant factors using the total RNA from the Beas2B cells. The level of superoxide dismutase(CuZnSOD), thioredoxin, glutathione reductase expression was examined. Results: 0.5 to 4 hours after the benzopyrene, nicotine and cotinine theatments, the level of thioredoxin and glutathione reductase expression decreased. Longer exposure to these compounds for 24 to 72 hours inhibited the expression of most of these antioxidant factors. Conclusion: During exposure to smoke compounds, thioredoxin and glutathione reductase are the key antioxidant factors induced sensitively between 0.5 and 4 hours but the levels these antioxidants decrease between 24 hour and 72hours.

Determination of benzo(a)pyrene in olive oils (올리브유 중 벤조피렌 분석)

  • To determine levels of benzopyrene in olive oils, a selective analytical method of HPLC/FLD has been applied. After removing fat in food samples with hexane, it was extracted in aqueous N,N-DMF solution, cleaned-up on florisil SPE cartridge and analyzed by the instrumental analysis. The mobile phase was a mixture of acetonitrile and water in 8:2 by the isocratic elution and the excitation wavelength of fluorescence detector was 294 nm and emission wavelength of it was 404 nm. The average recovery was about 95 % and the limit of quantitation was $0.9{\mu}g/kg$. The levels of benzopyrene in the selected olive oil samples were ranged from not detected to $1.9{\mu}g/kg$, however, they were under $2.0{\mu}g/kg$, the maximum level of benzopyrene in olive oil which was established in the food code.

  • Polycyclic Aromatic Hydrocarbons(PAHs) contamination arises from several source including processing of food(smoking, direct drying, cooking) and environmental contamination of air, water or soil. A red ginseng is produced by steaming the root followed by drying. The methodology involved extraction with n-hexane and washing with water, clean-up on Sep-Pak Florisil Cartridges and determination by HPLC/FLD. The mobile phase was a mixture of acetonitrile and water in 8:2 by the isocratic elution and the excitation wavelength of fluorescence detector was 294 nm and its emission wavelength was 404 nm. The average recovery was about 105% and the relative standard deviation was 0.5. The levels of benzopyrene in the selected red ginseng beverage samples were not detected.
