Study on analytical method of residual benzimidazole anthelmintics in meat by LC/MS (LC/MS를 이용한 식육중 잔류 벤지미다졸계 구충제 분석법 연구)

  • Recently, mass spectrometry coupled with liquid chromatography (LC/MS) has been a preferred technique for determination of organic compounds in complex matrixes. LC/MS provides a high degree sensitivity and specificity of the compounds of interest. The purpose of this study was to confirm analytical method of residual 6 benzimidazoles (thiabendazole, oxfendazole, mebendazole, albendazole, flubendazole and fenbendazole) in meat by LC/MS. Benzimidazoles were analyzed by LC/MS on XTerra $C_{18}$ column with 0.01% trifluoroacetic acid-acetonitrile (TFA) in a gradient mode as mobile phase, and that were identified by electrospray ionization with selected ion recording mode at 150-350 amu mass range. Residual benzimidazoles were extracted from tissue with ethylacetate, and elute benzimidazoles with $50\%$ acetonitrile. In the LC/MS analysis of benzimidazoles, signal to noise ratio was showed relatively high in the positive mode and special ion in the quality analysis was determined via $[M+H]^+$ and Fragment ions. A spectrum of benzimidazoles was showed from all 6 benzimidazoles

  • This stud was attempted to detect six benzimidazoles[thiabendazole(TBZ), oxfendazole(OFZ), mebendazole(MBZ), flubendazole(FLBZ), albendazole(ABZ), and febendazole(FBZ)] in eggs using high performance liquid chromatography(HPLC) with photodiode array detector(DAD) simultaneously. The eluates were determined by HPLC on a waters X-Terra$^{TM}\;C_{18}$ reverse-phase column($4.6{\times}250nm,\;5{\mu}m$) with DAD at 295nm. The mobile phase was 0.04M ammonium phosphate(pH 7.5)/ACN(62.28 v/v) run isocratically. The calibration curves were linear(r>0.999) for six benzimidazoles. Average recovery rate from eggs were 94.54% for TBZ, 97.71% for OFZ, 88.82% for MBZ, 93.71% for FLBZ, 78.61% for ABZ, and 75.09% for FBZ, respectively. The limits of detection and quantification were 2.27ppb, 3.88 for TBZ, 9.34ppb, 15.61ppb for OFZ, 28.53ppb, 45.15ppb for MBZ, 27.39ppb, 40.95ppb for FBZ, 8.61ppb, 13.95ppb for ABZ and 12.79ppb, 22.34ppb for FBZ, respectively.

  • Several studies of terbenzimidazoles and bibenzimidazoles suggested that benzimidazoles, especially 5-nitro-2-(para-methoxyphenyl)benzimidazole. possess topoisomerase I inhibition activities. In order to find out the structure activity relationship of 5-nitro-2-phenyl-benzimidazoles. eight derivatives that are substituted at the para position of 2-phenyl moiety were selected, synthesized & evaluated considering their electronic or lipophilic parameters.   • TBACl (10 mol %) was found to be a useful catalyst for the synthesis of substituted benzimidazoles. The method was proved to be simple, convenient and the product was isolated with good yield.

  • Three new peptide analogs 5a-c were obtained through coupling of 5-Amino benzimidazoles 2a-c with L-phenylalanine. For the purpose ${\alpha}$-amino group was blocked with phthalic anhydride and activation of ${\alpha}$-carboxy group of phenylalanine was carried out by preparing phthaloyl-L-phenylalanyl chloride 4. After developing a successful peptide linkage, the phthaloyl group was removed by treating 5a-c with hydrazine hydrate to get free peptides 6a-c, purified through a column of Amberlite (IR-4B). All of these compounds 2a-c and 5,6a-c have been characterized on the basis of their IR, 1H NMR and EIMS analyses. Antibacterial activity of these compounds is also been reported.

  • A series of substituted benzimidazoles and benzothiazoles were prepared through the one-pot reaction of ophenylenediamine and o-aminothiophenol with various aldehydes in the presence of ferric hydrogensulfate both in EtOH and water as solvent. The reactions proceed smoothly in excellent yield, high chemoselectivity and with an easy work-up.

  • Solid silica supported ferric chloride ($SiO_2-FeCl_3$) catalyzed one-step synthesis of various benzimidazoles from o-phenylenediamine and aldehydes using $H_2O_2$ as the oxidant. The salient features of this method are simple and convenient procedure, easy purification and shorter reaction times.
