In this study, the marine ecological impacts of dephosphorized slag and steel slag on the initial life cycle of the rotifer Brachionus plicatilis and benthic copepod Tigriopus japonicus (in marine trophic structure as a first consumer) exposure to slag extracts have been considered using a marine ecotoxicological assessment. In the results of a screen test on slag extracts, the pH of an undiluted solution was measured to have high alkalinity (pH 8.89-12.16), but a toxic reaction to this undiluted solution before and after aging was divided according to test species. For non-aged slag, the toxic effect ($LC_{50}$) of neonate on B. plicatilis was seen to be severe with dephosphorized slag (20.8 %) than steel slag (63.8 %) with under pH-uncontrolled conditions. The toxic values of dephosphorized and steel slag were estimated to be 35.3 % and 36.0%, respectively, for nauplius with T. japonicus. However, the toxicity of slag extracts before and after aging were different for T. japonicus than for B. plicatilis based on the characteristics of the test materials, with pH-controlled conditions. In conclusion, the results of this study suggest that slag can be relatively stable after aging and may not be likely to influence marine environments, even given repetitive extracting under pH-uncontrolled conditions. This study confirms that a marine ecotoxicological assessment method applied to mechanically activated samples can give an idea of the resistance a marine environment has against the introduction of hazardous materials due to precipitation and weathering.
Several bioassays have been performed for assessment of the impact of polluted sediments. The direct exposure method using sediments is limited by difficulty controlling feeding and its effects on organisms. Furthermore, only macro-organisms and benthic organisms are used. To evaluate the potential application of sediment elutriate as a complementary strategy for impact assessment, copepods, small organisms with a short life cycle, were exposed to sediment elutriates, and several end-points were measured. As a result, sediment elutriates prepared from polluted sites caused growth retardation in marine copepods. In terms of molecular biomarkers, antioxidant-related and chaperone protein gene expression levels were increased in a dose- and time-dependent manner. Thus, we suggest that sediment elutriate tests can provide an effective alternative for toxicity assessment using whole sediment samples. Further studies are required to obtain sufficient data for future applications.
The purpose of this study was to assess cross-generational effects of bisphenol A exposure in benthic copepods, Tigriopus west. Nauplii (<24 hours old) were exposed to graded concentrations of bisphenol A, and toxicity end-points such as survival, development, sex ratio, and fecundity were measured. $F_1$ generations were grown under innoxious conditions, and similarly assessed. Significant differences were observed in development of nauplii and copepodites, between exposed and non-exposed copepods; however, there were no differences in survival of nauplii or copepodites, sex ratio, or brooding rate in parental generation. In contrast, in the $F_1$ generation, there were significant differences between the control group and exposed group in survival and development of nauplii. Length, width, and biomass of parental and $F_1$ generations were reduced in the exposed group compared to the control group. In addition, some deformities, such as swelling of the prosome, abnormally shaped egg sac, and dwarfism were observed after exposure to bisphenol A. So, our study demonstrates that a cross-generation toxicity test and monitoring of morphological deformities in harpacticoid copepods, can be useful for development of potential bioindicators for environmental monitoring, and assessment of chemical impact.

Harpacticoid copepod Tigriopus japonicus is widely distributed in coastal waters of Korea and plays important role in marine trophic structure as a first consumer. In this study, a series of experiment were conducted to test the potential of the species as a standard test species for marine bioassay. Tolerance on salinity and pH, sensitivity on the reference materials(copper sulfate and cadmium chloride) and response on the ocean dumping materials(waste sludge) we re tested to identify if the species satisfy the basic criteria as standard species for marine bioassay. The nauplius of the species($100{\sim}200{\mu}m$) showed wide tolerance on salinity with >90.0% survival rates exposed to $5.0{\sim}35.0psu$ for 48 h. Wide adaptability on pH's were also observed from 6.3 to 8.2 with >90.0% survival rates during the test. $LC_{50}$ values for copper sulfate and cadmium chloride were $3.6{\pm}0.7ppm,\;1.7{\pm}0.8ppm$, respectively. The variations in mortality between replicates were less than 10.0%. Comparison of $LC_{50}$ values indicated that T. japonicus nauplius was lower sensitive to copper sulfate than the most marine crustaceans included copepods, however, the sensitivity of test animal to cadmium chloride higher than the adults of copepod T. japonicus, Paracalanus parvus, and marine rotifer Brachinonus plicatilis. There were significant concentration-response relationship in the mortality of T. japonicus nauplius using the elutriates of three ocean dumping materials(industrial waste sludge). 48 h $LC_{50}$ values we re $31.1{\pm}1.1%$ for the elutriate of sludge from leather processing company and $54.4{\pm}15.1%$ for that of dye production company. Based on the above experimental results, bioassay using benthic harpacticoid T. japonicus nauplius must be a good estimation tool for marine ecotoxicological assessment of waste or chemicals. Wide tolerance on the salinity and pH, and significant linear relationship between concentration and response(mortality) supported the high potential of the species as a standard test species.

To observe the seasonal fluctuation of the meiobenthic fauna community around the coastal area of Jeju island, 20 stations were selected and seasonal surveys were conducted. Three-replicate meiobenthic samples were collected from each station in April, July, and November 2017; February, May, August, and November 2018; and February in 2019, in a total of eight months from 2017 to 2019. The total density of meiobenthos at each station ranged from 733 to 2,505 ind. 10 cm-2. The month in which the highest habitat density appeared in most stations was April 2017 and the month in which the lowest habitat density was seen was February 2019. Nematodes were the most dominant faunal group among the representative 13 meiofaunal groups at all stations. The subdominant groups were benthic harpacticoid copepods and nauplius. The nematode/copepod (N/C) ratio, reflecting the health indices within the ecosystem, varied from 0.02 to 87.40 over the entire season and the average station values ranged from 2.00 to 16.80. The lowest N/C ratio value was found in April 2017 (0.02) and the highest N/C ratio was found at Station 11 in February 2019, which was 87.40, indicating the highest level of pollution. Seasonal and regional meiobenthos community structure similarity was divided into three groups through group analysis and multi-dimensional scaling (MDS) analysis, which showed that the meiobenthos community structure was affected by seasonal variations, rather than by regional differences.

Six standard methods for marine ecotoxicological tests were established(or applicated) using marine decomposer, primary producers and consumers. Development processes referred to the standard methods established by USEPA(United States Environmental Protection Agency), international organizations and European methods. However, the standard test species were selected among the domestic species generally found in the Korean waters and sediments. The test methods provide the culture/maintenance of test species, test methods, reproducibility and quality acceptance criteria etc. A total of nine test species were designated including bioluminescent bacteria(Vibrio fischeri), diatom(Skeletonema costatum), seaweed(Ulva pertusa), rotifer(Brachionus plicatilis), benthic copepod(Tigriopus japonicus), benthic amphipods(Mandibulophoxus mai, Monocorophium acherusicum), and fishes(Oryzias latipes, Paralichthys olivaceus). These test species represent the decomposer, primary producer and consumers in marine trophic system in Korean coastal ecosystems, and we recommend the "battery test" including at least one species from the each trophic level for marine ecotoxicological test.
The mixotrophic marine ciliate Mesodinium rubrum possesses a highly modified algal endosymbiont as a nutrition source for the species. Accordingly, we assumed that the species can reflect the ecotoxicity on marine producer (as phytoplankton) and consumer (as zooplankton) both. A series of experiments were conducted to identify the potential of the species as a standard test species for marine ecotoxicological study. The comparison of species sensitivity on reference toxic materials was made using potassium dichromate for phytoplankton and copper chloride for zooplankton. The ciliate revealed the highest sensitivity on both reference materials among the seven test species including phytoplankton, benthic copepod and rotifer species. The toxicity end point of the species was 72hr-$EC_{50}$=1.52 mg/L (as potassium dichromate) estimated by population growth inhibition (PGI), which is more sensitive than the most sensitive phytoplankton Skeletonema costatum (72hr-$EC_{50}$=3.05 mg/L). As comparison to rotifer, it also revealed higher sensitivity on copper chloride; 72hr-$EC_{50}$=0.38 mg/L for ciliate and 48hr-$EC_{50}$=0.48 mg/L for rotifer. Also, the elutriate toxicity test of various ocean disposal wastes were conducted to identify the potential of ciliate toxicity test application using industrial waste sludges. The toxicity of leather processing waste sludge was highest on the ciliate, followed by dyeing waste sludge and dye production waste sludge as an increasing order of toxicity. 72h-$EC_{50}$ of ciliate PGI test was 1.83% and that of S. costatum 3.84% for leather waste sludge which showed highest toxicity. The toxicity test results also revealed that the highest sensitivity was observed on ciliate species on ocean disposed sludge wastes. Also, ciliate toxicity test well discriminated the degree of toxicity between sludge sources; 72h-$EC_{50}$ values were 1.83% for leather processing waste sludge, 16.75% for dye production waste sludge and 27.75% for textile production waste sludge. Even the laboratory culture methods of the species were not generally established yet, the species has high potential as the standard test species for marine toxicity test in terms of the dual reflection of phyto- and zooplankton toxicity from single test, sensitivity and test replicability.
The community structure of meiofauna was surveyed from six stations in sand mining area near around Sungapdo, Taean five times from April-December, 2007. Ten meiofaunal taxa in total, comprising 5-9 taxa per station were recovered. Nematodes were dominant from four stations in April and from five stations in May. In June, copepods were dominant from all stations except station two, from which sarcomastigophorans were predominant. Copepods were also dominant from three stations in October and four stations in December. The mean density of meiofauna per $10\;cm^{-2}$ was the highest in October (130 individuals) and lowest (68 individuals) in April. Meiofauna mean biomass per $100\;cm^{-2}$ was $3.80-75.26{\mu}gC$ from each station, with the monthly mean biomass being lowest in April ($11.97{\mu}gC$) and highest in October ($39.50{\mu}gC$). The highest biomass was recorded at station five in October ($75.26{\mu}gC$). Eleven families of harpacticoid copepods occurred Ameiridae and Ectinosomatidae were the most frequent. The mean diversity index was 1.21 from all the stations, 1.22 from the sand mining stations, and 1.19 from the reference stations. The nematode:copepod (N/C) ratio was 1.57 from the sand mining stations and 1.37 from the reference stations. Non-metric multidimensional scaling analysis divided the community into group A (station one, non-sand mining and reference area) and groups B and C in the sand mining area. The present study reports similar numbers of taxa but lower density and biomass of meiofauna compared to previous studies in Korean waters. The variability of the diversity index and N/C ratio may indicate large fluctuation of meiofauna depending on temporal and spatial conditions of the environment in the study area.
