Management of Myzus persicae using Beauveria bassiana and Environment-friendly agricultural materials (EFAM) on pepper

  • This study was carried out to development the environment-friendly control method of Myzus persicae using Beauveria bassiana and Environment-friendly agricultural materials (EFAM). Germination rate the dried conidia of B. bassiana was 90 percent at $24{\sim}27^{\circ}C$ temperature condition. Mortality of M. persicae was 56.7% at $1{\times}10^7$ cfu/ml conidia concentration. Four EFAM such as Pachungtan, Wangjungwang, Newbiodakill, Muginchon were low toxicity to B. bassiana growth. Among them, Muginchon (derris extract) and Wangjungwang (neem extract) could have synergistic effect to aphid mortality when mixing treat with B. bassiana, which protection value was 99% in 3 days after treat.

  • Some biological activities of Beauveria bassiana were studied to elucidate pharmacological function of B. bassiana-infected larva of the silkworm. The mycelium consisted mainly of carbohydrate (65.8%), followed by protein (15.9%) and fat (8.3%). Glucose (68.8%), mannose (7.1%), and galactose (6.1%) were major components in carbohydrates. Ten amino acids including glutamine, threonine, valine, aspartic acid, alanine, leucine, serine, glycine, arginine, and isoleucine were found in protein as major amino acids. Various extracts were prepared from the freeze-dried mycelium of B. bassiana by systemic extraction and their biological activities were investigated. Among tested fractions, the hot-water extract (HW) contributed significantly to the anti-coagulant activity, anti-complementary activity, and stimulation of intestinal immune system. The methanol extract (ME) increased acetylcholinesterase (AChE) inhibition activity and reactive oxygen species (ROS) scavenging activity.

  • Beauveria bassiana, one of the most common entomopathogenic fungi, has been isolated, pre defined and characterized in-house from soil of tea cultivation area. Experiments have been performed to verify the presence of chitinase as intracellular metabolite and its release as extracellular product rendering the spores with biopesticide activity. Although there are many responsible enzymes for the pest killer action of B. bassiana, binding property of chitinase depending on presence as well as absence of serine supplemented in the media has been studied with respect to the production and kinetics. A programmed investigation conclusively indicates that the isolated spore (hyphae) of B. bassiana has been metabolically enriched with the enzyme chitinase in presence of an externally added amino acid serine with its inhibitory kinetics.

Evaluation of Strains of Metarhizium anisopliae and Beauveria bassiana against Spodoptera litura on the Basis of Their Virulence, Germination Rate, Conidia Production, Radial Growth and Enzyme Activity

  • Ten strains of the entomopathogenic fungi Metarhizium anisopliae and Beauveria bassiana were evaluated to find the most effective strain for optimization studies. The first criterion tested for strain selection was the mortality (> 50%) of Spodoptera litura larvae after inoculation of the fungus for 4 days. Results on several bioassays revealed that B. bassiana BNBCRC showed the most virulence on mortality S. litura larvae (80% mortality). B. bassiana BNBCRC also showed the highest germination rate (72.22%). However, its conidia yield ($7.2{\times}10^8$ conidia/mL) was lower than those of B. bassiana B 14841 ($8.3{\times}10^8$ conidia/mL) and M. anisopliae M6 ($8.2{\times}10^8$ conidia/mL). The highest accumulative radial growth was obtained from the strain B14841 (37.10 mm/day) while the strain BNBCRC showed moderate radial growth (24.40 mm/day). M. anisopliae M6 possessed the highest protease activity (145.00 mU/mL) while M. anisopliae M8 possessed the highest chitinase activity (20.00 mU/mL) during 96~144 hr cultivation. Amongst these criteria, selection based on virulence and germination rate lead to the selection of B. bassiana BNBCRC. B. bassiana B14841 would be selected if based on growth rate while M. anisopliae M6 and M8 possessed the highest enzyme activities.

  • This study was conducted to observe the effects of pesticides on the pathogenicity of entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae isolated from Korea. Spomlation of entomopathogenic fungi in SMAY medium that mixed different concentrations of pesticides was similar to control in metalaxyl and tolclofos- methyl treatment but in half recommanded treatment of fenitrothion and mepronil very poor spomlation was showed. Pathogenicity of entomopathogenic fungi dipped in all tested pesticide solution for 1 to 5hr was 100% pathogenicity of M. aniropliae in the pesticide treated chinese cabage fields were similar to control, Bt, teflubenzuron, metalaxyl, and carbofuran treatment but alachlol was below 80%. Pathogenicity and persistence of entomopathogenic fungi in turfgrass when pesticides were posttreatmented were continued to 4 months in iprodione+thiram and tolclofos-methyl treated plots but those of B. bassiana and M. anisopliae was losted in fenitrothion pathogenicity and persistence of in mepronil were nil M. anisopliae from 3 months after treatment.

Effects of the growth inhibition against plant pathogenic bacteria using Beauveria bassiana (백강균(Beauveria bassiana)을 이용한 식물병원성세균 생장억제효능 검증)

  • This study was performed to investigate the antibacterial activities and the minimal inhibitory concentration (MIC) of 6 strains of Beauveria bassiana against mulberry pathogenic bacteria. The antibacterial activities and the MIC were measured using paper disc method and broth dilution method, respectively. The antibacterial activities were found out just B. bassiana J200, and shown at 13 mm from Erwinia rhapontici KACC 10407 and at 17 mm from Pseudomonas syringae KACC 10390 and Xanthomonas campestris KACC 12134. The MIC were all observed at 4.0% from E. rhapontici KACC 10407, P. syringae KACC 10390 and X. campestris KACC 12134. The results suggest that B. bassiana could play a good role for biological control against mulberry pathogenic bacteria.

  • This study was conducted to observe the influence of chemical pesticides and environmentally friendly agricultural materials (EFAMs) used in tomato cultivation on the pathogenicity of the entomopathogenic fungus, Beauveria bassiana. B. bassiana mycelium didn't grow on PDA media containing 13 fungicides including chlorothalonil and colonies were not formed on PDA media containing 12 fungicides. B. bassiana mycelium grew and colonies were formed on all PDA media containing insecticides and EFAMs, but mycelial growth and colony formation on most PDA media were significantly inhibited compared to the control. The insecticidal activity of B. bassiana against Trialeurodes vaporariorum was decreased when fungicides (polyoxin B, mandipropamid) and EFAMs containing sulfur were added, but insecticides (pyridaben, dinotefuran) and EFAMs originated from plant extracts did not have any influence on the insecticidal activity of B. bassiana. The pathogenicity of a mixture of B. bassiana and polyoxin B against T. vaporariorum was lower than that of B. bassiana alone under greenhouse conditions.

Development of Liquid Culture Media for the Mass Production of Beauveria bassiana Blastospore (흰굳음병균(Beauveria basiana) 단균사의 대량배양생산을 위한 액체배지 개발)

  • To develope a liquid culture media for the mass culture of Beauveria bassiana blastospore, wheat bran and soybean cake were selected as a carbon and nitrogen source, respectively. The composition and final concentration of wheat bran and soybean cake in liquid culture media were determined by the growth of B. bassiana blastospore. The results showed that the growth of B. bassiana blastospore was effective in the rate of 3 wheat bran: 2 soybean cake and in 5% of final concentration of the liquid culture media, suggesting that the liquid culture media using wheat bran and soybean cake are available.

  • This study was conducted to investigate agents inducing conidial germination of an entomopathogenic fungus, Beauveria bassiana KK5. Different chemicals including carbohydrates were mixed with conidia of B. bassiana and incubated on water agar for 12 hours. Fructose, mannose and skim milk were useful for spore germination compared to other chemicals. Bioassays against green peach aphids were conducted with the fungal conidia suspended in 1% fructose, mannose and skim milk. Of them, a mixture of skim milk plus conidia of B. bassiana KK5 showed the highest mortalities against $3^{rd}$ instar of green peach aphid.

  • The greenhouse whitefly, Bemisia tabaci, is considered one of the most destructive pests of crops. In this study, we aimed to determine the optimal liquid culture conditions in shake flasks for maximal sporulation of Beauveria bassiana M130 using rice bran. The optimal initial pH for the spore production of B. bassiana using extracted rice bran medium was 5.2 and $28^{\circ}C$. The screening in shake flasks of carbon and nitrogen sources resulted in the identification of an optimal medium based on 0.5% $(NH_4)_2SO_4$, with extracted rice bran 8:1. Using this medium, a production level of $2.15{\times}10^9$ spores per ml was obtained after six days from culture inoculation at $28^{\circ}C$ in a rotary shaking incubator at 130 rpm. In addition, the specific activities of extracellular enzymes of chitinase and protease were $4,296{\mu}mol$ and $375{\mu}mol$, respectively. These results suggest that Beauveria bassiana M130 could be a bio-controller for the greenhouse whitefly.
