Anti-inflammatory Effect of Bear's Gall in Rat Microglia

  • We hypothesize that bear's gall may have a certain role in anti-inflammation through a preventive effect of pro-inflammatory potentials. Secondly, we tried to connect the experimental results to Alzheimer's disease (AD), which chronic inflammation is a main cause of the disease. For this theme, we designed to elucidate the efficacy of bear's gall in suppressing the pro-inflammatory mediators, such as nitric oxide (NO) and $interleukin-1{\beta}\;(IL-1{\beta})$ in rat microglia. From the study, we concluded that bear's gall plays a positive role in suppressing such pro-inflammatory repertoire from rat microglia comparing to normal and positive control, such as culture media and cyclosporine. Interestingly, bear's gall showed a prolonged effect of anti-inflammation comparing with cyclosporine when time goes by up to 48h with a significant suppression at $1.2\;mg/m{\ell}$. Therefore, we can consider that bear's gall in part can be applied to AD therapy in that it suppresses the expression of pro-inflammatory mediators as well as its continued effect.

Effect of bear's gall on mammalian cell growth (웅담이 mammalian세포의 생육에 미치는 영향)

  • In the present investigation we have studied the effect of bear's gall on mammalian cells and demonstrated that COS-7 cells, which were derived Monkey kidney cells, had shown almost same extent of growth with 78 hrs in 10% FCS Dulbecco's Modified Eagle's medium with bear's gall and without bear's gall. But the hybridoma cells which were fused murine myeloma cells and the rat spleen cells for monoclonal antibody production died almost within 48 hrs. To investigate the effect of biosynthetic mechanism, cDNA were transfected to COS-7 cells, and it was shown that cDNA-transfected COS-7 cell had produced 30-40% less the amount of recombinant protein than the medium without bear's gall.

Determination of Trace Metals in Fel Ursi (시판 웅담의 미량금속 정량에 관한 연구)

  • Nine trace elements in Bear Galls(Fel Ursi) studied in this paper are Na, Mg, K, Ca, Cu, Zn, Fe, Pb and Ge. The contents of Na, Mg, K, Ca, Cu, Zn, Fe, Pb are determined by atomic absorption spectrometry and Ge is determined by graphite atomic absorption spectrometry. In both methods, the sample is digested with nitric-perchloric acid and then ashed. The results of analysis are shown in table and the result obtained from this study are as follows. a. The highest content of trace element of Bear Gall is Na and other elements are getting lower in order of K, Mg, Ca, Ge, Zn, Fe, Cu, and Pb. b. Particularly the content of Ge in Bear Gall was determined.

Anti-oxidative and Cytoprotective Effect of Ursodeoxycholic Acid, an Active Compound from the Bear's Gall, in Mouse Microglia (생쥐 뇌소교세포주에서 웅담추출활성성분(우르소데옥시콜린산)의 항산화 및 세포보호효과)

  • The in vitro cytoprotective and anti-oxidative effects of ursodeoxycholic acid, a major active compound from bear's gall were investigated in mouse brain microglia. In the present study, we wished to scrutinize the potential role of UDCA as an anti-neurodegenerative agent in neurodegenerative disease such as Alzheimer's disease. This concept was supported by the multiple preliminary studies in which UDCA has an anti-inflammatory effect in microglial cells. In the study, we found that $7.5\;{\mu}g/mL$ UDCA was effective in the protection of cells from $H_2O_2$ damage, a reactive oxygen, and the resuIt was coincided with the anti-apoptotic effect in DAPI staining. Moreover, the metal-catalyzed oxidation study showed that UDCA has antioxidant effect as much as ascorbic acid at $50{\sim}100\;{\mu}g/mL$. In conclusion, these study results suggested that neuro-degenerative diseases such as Alzheimer's disease probably caused by over-expressed beta amyloid peptide in elderly people can be controled by UDCA through an anti-inflammatory, anti-oxidative and anti-apoptotic effect. The evidences showed in the study may be references for more in-depth in vivo and clinical studies for a candidate of anti-neurodegenerative therapy in the near future.

Case Series Study on the Use of BU Pharmacopuncture Treatment in Patients with Acute Lumbar Sprain

  • Objective: The purpose of this study was to investigate the clinical effects of BU pharmacopuncture therapy consisting of bear's gall(fel ursi) and ox bezoar(bovis calculus) on acute lumbar sprain. Methods: 12 patients diagnosed as acute lumbar sprain in 6 designated local Korean medicine clinics from October 2017 to February 2018 were treated by BU pharmacopuncture. Several acupoints in abdomen and lumbar region were selected by clinicians at their own discretion. The effectiveness of the therapy was evaluated using VAS and ODI. After that we reviewed the medical records of all these patients to evaluate the effectiveness and safety of the therapy. Results: The VAS and ODI scales were significantly decreased after BU pharmacopuncture therapy. And no major complications and adverse effects were reported. Conclusion: BU pharmacopuncture showed rapid pain relief in patients with acute lumbar sprain. It is possible to shorten the treatment period of acute lumbar sprain and prevent progressing to chronic back pain in advance. To establish the effects of BU pharmacopuncture therapy, more succeeding clinical and laboratory studies are needed.

Three Cases of Menopausal Hot Flush and Sweating Treated by Ascending Kidney Water and Descending Heart Fire (AKDH) Pharmacopuncture Treatment (약침을 이용한 수화조절법으로 호전된 갱년기 상열감 및 발한과다 환자 치험 3례)

  • Objectives : The purpose of this study is to evaluate the clinical effects of ascending kidney water and descending heart fire pharmacopuncture treatment for hot flush and sweating. Methods : Ascending kidney water and descending heart fire pharmacopuncture is achieved by injecting pharmacopuncture on a specific acupoint to change from the state of water-fire disharmony to harmonious state. Hwangryunhaedok-tang (黃連解毒湯) pharmacopuncture is injected on both side Gyeonjeong (GB 21 ), Pungji (GB 20 ). Total 0.4 cc was injected by 0.1 cc each point. BUM (Bear’s gall bladder, ox bezoar and musk) pharmacopuncture is injected on both side Jeonjung (CV 17 ), Jungwan (CV 12 ), Gihae (CV 6 ). Total 0.15 cc was injected by 0.05 cc each point. Treatment was done daily. The method of evaluation are Hot Flush Score (HFS), VAS and sweating areas. Results : In case 1, after treatment hot flush score was reduced from 24 to 4 points. VAS scale was reduced from 7 to 2. It took about two weeks for the symptoms to decrease by half. Associated symptoms almost did not stay at discharge. In case 2, after treatment hot flush score was reduced from 28 to 2 points. VAS scale was reduced from 10 to 3. And emotional symptoms were reduced about 70%. In case 3, after treatment hot flush score was reduced from 8 to 1 points. VAS scale was reduced from 6 to 1. And headache and chest discomfort symptoms have disappeared. Conclusions : Ascending kidney water and descending heart fire pharmacopuncture treatment is effective for improve ascending kidney water and descending heart fire energy. Therefore, it will be used to alleviate hot flush and sweating.
