Re-evaluation of the Genus Antrodia (Polyporales, Basidiomycota) in Korea

  • The wood decay fungi Antrodia P. Karst. play important ecological roles and have significant industrial and economic impacts as both wood degraders and sources of pharmaceutical and biotechnological products. Although each Antrodia species has distinct morphological characteristics, the misidentification rate is especially high due to their simple morphological characters. A combination of morphological and internal transcribed spacer region sequence analyses revealed that 27 of 89 specimens previously identified by morphology alone were correct, whereas 35 of these specimens were misidentified as other Antrodia species. We report here that seven Antrodia species exist in Korea (A. albida, A. heteromorpha, A. malicola, A. serialis, A. sinuosa, A. sitchensis, and A. xantha) and based on these specimens, we provide taxonomic descriptions of these species, except for A. serialis, which was only confirmed by isolate.

Distribution and Antifungal Activity of Endophytic Fungi in Different Growth Stages of Chili Pepper (Capsicum annuum L.) in Korea

  • This study investigated the distribution of endophytic fungi obtained from the surface sterilized tissues of leaves, stems and roots of chili pepper ($Capsicum$ $annuum$ L.) plants in seedling, flowering and fruiting stages in Korea and their antifungal activity. A total of 481 isolates were recovered and were identified using molecular techniques. Based on rDNA ITS gene sequence and phylogenetic analysis, 21 fungal genera were characterized, belonging to 16 Ascomycota and 5 Basidiomycota. $Penicillium$ in seedling stage, $Fusarium$ in flowering stage, $Colletotrichum$ followed by $Fusarium$, $Alternaria$ and $Xylaria$ in fruiting stage was predominant and $Alternaria$, $Cladosporium$ and $Fusarium$ were common in all growth stages. Among 481 endophytes 90 phenotypes were evaluated for the antimicrobial activity against three major pathogens ($Phytophthora$ $capsici$, $Colletotrichum$ $acutatum$ and $Fusarium$ $oxysporum$) of chili pepper. Among them 16 isolates inhibited the growth of at least one test microorganisms. Three strains showed a broad spectrum antifungal activity and displayed strong inhibition against chili pepper pathogenic fungi.

  • 1995년 4월부터 1997년 10월까지 주로 한라산 일대를 중심으로 버섯을 채집한 결과 총 127속 292종이 동정되었다. 이들 중 담자균문의 41속 103종과 자낭균문의 13속 88종이 새롭게 제주도에 기록되게 되었으며 Typhula phacorriza(제주노랑부들국수버섯), Coryceps crinalis(깊은주름동충하초), Cordyceps cochlidiicola(쐐기나방번데기동충하초), Coryceps longissima(제주긴뿌리동충하초), Cordyceps prolifica(나무가지동충하초) 등 5종이 새롭게 한국기록종으로 등록되었다.

The Optimal Culture Conditions for the Mycelial Growth of Oudemansiella radicata

  • Oudemansiella radicata, one of edible mushrooms belonging to Tricholomataceae of Basidiomycota, has been known to exhibit outstanding therapeutic effects on the hypertension caused by high blood pressure and inhibitory effects on the sarcoma 180 and Erhrlich carcinoma of mice. As one of preliminary experiments for producing fruiting-body of O. radicata, this study was carried out to obtain the basic information for culture conditions of mycelial growth of the fungus. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and pH 6, respectively. The medium for favorable mycelial growth of O. radicata was shown in the Lilly medium, whereas compact mycelial density was found in Hamada medium. The carbon and nitrogen sources promoting for mycelial growth of O. radicata were xylose and alanine, respectively. The optimum C/N ratio was about 20 : 1 in case that 3% glucose was supplimented to the basal medium as a carbon source.

Macrofungal Survey of the Tian Shan Mountains, Kyrgyzstan

  • The Tian Shan mountain system is one of the large mountain ranges located in Central Asia. This region is globally recognized as mountain ranges, offering inestimable wealth in fauna and flora with significant biodiversity values. We surveyed macrofungal diversity of Tian Shan in Kyrgyzstan from 2016 to 2018. A collection of macrofungi was made, and these were subjected to sequence comparisons and phylogenetic analysis to ensure the identity of the collected macrofungi. Of those collected, 95 out of 100 specimens were successfully sequenced and compared with those of other related species retrieved from GenBank. The sequenced specimens were classified into 2 phyla, 8 orders, 24 families, 47 genera, and 57 species, based on current taxonomic concepts (combining morphology and phylogeny). To the best of our knowledge, this study provides the first well-documented checklist and phylogenetic analysis of macrofungi recovered from the Tian Shan mountains in Kyrgyzstan.

Identification of Two Fungal Endophytes Associated with the Endangered Orchid Orchis militaris L.

  • A survey of the endangered orchid Orchis militaris populations was carried out in north-eastern Italy. The occurrence of fungal root endophytes was investigated by light and electron microscopies and molecular techniques. Two main sites of presence were individuated in the Euganean Hills, differing as to the percentage of flowering individuals and of capsules completing maturity. Fluorescence microscopy revealed an intracellular cortical colonization by hyphal pelotons. Two ITS PCR products co-amplified. Sequencing revealed for the former an identity and a high similarity (99%) with a Tulasnellaceae (Basidiomycota) fungus found within tissues of the same host in independent studies in Hungary and Estonia, suggesting an interesting case of tight specificity throughout the Eurosiberian home range. The second amplicon had 99% similarity with Tetracladium species (Ascomycota) recently demonstrated as potential endophytes. TEM revealed two different hyphal structures. Double fungal colonization appears to occur in Orchis militaris and the possible requirement of a specific fungal partner throws light on the causes of this plant's rarity and threatened status.

한국산 개암버섯속의 미기록종 보고 (Notes on Some Unrecorded Species in Korean Hypholoma)

  • 본 연구는 한국산 주름버섯목의 종 다양성과 분포상을 조사하는 과정에서 경기도 용문산에서 한국미기록 긴대개암버섯 "Hypholoma elongatum(Pers.) Ricken"을 채집하여 형태적 특성에 준하여 분류동정 하였다. 자실체의 형태적 특징에 대한 상세한 기술과 미세구조를 도해하고 한국명을 신칭하여 보고하고자 한다. 본 연구에 사용된 표본은 국립농업과 학원 식물균류표본보존센타(HCCN)에 보존되어 있다.

Diversity of Endophytic Fungi Associated with Taraxacum coreanum and Their Antifungal Activity

  • Endophytic fungi were isolated from healthy leaf and root samples of Taraxacum coreanum. Of the 72 isolates recovered, 39 were from leaves and 33 from roots with an isolation frequency of 54% and 46%, respectively. Based on ITS sequence analysis, 72 isolates were classified into 19 genera of which 17 were under the phylum Ascomycota and 2 were under Basidiomycota. Diverse genera were found and Alternaria, Cladosporium, Fusarium and Phoma were dominant. Out of 19 genera, Apodus, Ceriporia, Dothideales, Leptodontidium, Nemania, Neoplaconema, Phaeosphaeria, Plectosphaerella and Terfezia were new to Korea. Seventy two isolates were screened for antifungal activity, of which 10 isolates (14%) were found active at least against one of the tested fungi. Isolate 050603 had the widest antifungal spectra of activity, and isolates 050592 and 050611 were active against three plant pathogenic fungi.

Isolation of wild yeasts from soils collected in Pochoen-si, Korea and characterization of unrecorded yeasts

  • In 2019, as a subset study to discover indigenous yeast species in Korea, a total of 20 yeast species were isolated from soil samples collected in Pochoen-si. Among them, eight strains were unreported species. From the high 26S rRNA gene sequence similarity and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to independent and predefined yeast species. The 20 strains were assigned to the genera Aureobasidium (1 strain) and Meyerozyma (1 strain) of the phylum Ascomycota and Cystofilobasidium (2 strains), Filobasidium (1 strain), Naganishia (2 strains), Bullera (3 strains), Leucosporidium (9 strains) and Sampaiozyma (1 strain) of the phylum Basidiomycota. There is no official report of the following species in Korea: Leucosporidium creatinivorum (4 strains), Leucosporidium escuderoi(2 strains), Leucosporidium golubevii(1 strain) and Leucosporidium intermedium (2 strains). Basic biochemical characteristics, colony and cell morphology are also described in the species description section.

Ethanol Production from Various Sugars and Cellulosic Biomass by White Rot Fungus Lenzites betulinus

  • Lenzites betulinus, known as gilled polypore belongs to Basidiomycota was isolated from fruiting body on broadleaf dead trees. It was found that the mycelia of white rot fungus Lenzites betulinus IUM 5468 produced ethanol from various sugars, including glucose, mannose, galactose, and cellobiose with a yield of 0.38, 0.26, 0.07, and 0.26 g of ethanol per gram of sugar consumed, respectively. This fungus relatively exhibited a good ethanol production from xylose at 0.26 g of ethanol per gram of sugar consumed. However, the ethanol conversion rate of arabinose was relatively low (at 0.07 g of ethanol per gram sugar). L. betulinus was capable of producing ethanol directly from rice straw and corn stalks at 0.22 g and 0.16 g of ethanol per gram of substrates, respectively, when this fungus was cultured in a basal medium containing 20 g/L rice straw or corn stalks. These results indicate that L. betulinus can produce ethanol efficiently from glucose, mannose, and cellobiose and produce ethanol very poorly from galactose and arabinose. Therefore, it is suggested that this fungus can ferment ethanol from various sugars and hydrolyze cellulosic materials to sugars and convert them to ethanol simultaneously.
