Molecular Docking Studies of Wolbachia Endosymbiont of Brugia Malayi's Carbonic Anhydrase Using Coumarin-chromene Derivatives Towards Designing Anti-filarial Agents

  • Filariasis causing nematode Brugia malayi is shown to harbor wolbachia bacteria as symbionts. The sequenced genome of the wolbachia endosymbiont from B.malayi (wBm) offers an unprecedented opportunity to identify new wolbachia drug targets. Hence the enzyme carbonic anhydrase from wolbachia endosymbiont of Brugia malayi (wBm) which is responsible for the reversible interconversion of carbon dioxide and water to bicarbonate and protons (or vice versa) is chosen as the drug target for filariasis. This enzyme is thought to play critical functions in bacteria by involving in various steps of their life cycle which are important for survival, The 3D structure of wBm carbonic anhydrase is predicted by selecting a suitable template using the similarity search tool, BLAST. The BLAST results shows a hexapeptide transferase family protein from Anaplasma phagocytophilum (PDB ID: 3IXC) having 77% similarity and 54% identity with wBm carbonic anhydrase. Hence the above enzyme is chosen as the template and the 3D structure of carbonic anhydrase is predicted by the tool Modeller9v7. Since the three dimensional structure of carbonic anhydrase from wolbachia endosymbiont of Brugia malayi has not yet solved, attempts were made to predict this protein. The predicted structure is validated and also molecular docking studies are carried out with the suitable inhibitors that have been solved experimentally.

  • Porous SiC samples were prepared with three types of wood (poplar, pine, big cone pine) by simply embedding the wood charcoal in a powder mixture of Si and $SiO_2$ at 1600 and $1700^{\circ}C$. The basic engineering properties such as density, porosity, pore size and distribution, and strength were characterized. The samples showed full conversion to mostly $\beta$-SiC with good retention of the cellular structure of the original wood. More rigid SiC struts were developed for $1700^{\circ}C$. They showed similar bulk density ($0.5{\sim}0.6\;g/cm^3$) and porosity (81~84%) irrespective of the type of wood. The poplar sample showed three pore sizes (1, 8, $60\;{\mu}m$) with a main size of $60\;{\mu}m$. The pine sample showed a single pore size ($20\;{\mu}m$). The big cone pine sample showed two pore sizes (10, $80\;{\mu}m$) with a main size of $10\;{\mu}m$. The bend strength was 2.5 MPa for poplar, 5.7 MPa for pine, 2.8 MPa for big cone pine, indicating higher strength with pine.

  • Chemokine receptor antagonists have potential applications in field of drug discovery. Although the chemokine receptors are G-protein-coupled receptors, their cognate ligands are small proteins (8 to 12 kDa), and so inhibiting the ligand/receptor interaction has been challenging. The application of structure-based in-silico methods to drug discovery is still considered a major challenge, especially when the x-ray structure of the target protein is unknown. Such is the case with human CCR2 and CCR5, the most important members of the chemokine receptor family and also a potential drug target. Herein, we review the success stories of combined receptor modeling/mutagenesis approach to probe the allosteric nature of chemokine receptor binding by small molecule antagonists for CCR2 and CCR5 using Rhodopsin as template. We also urged the importance of recently available ${\beta}2$-andrenergic receptor as an alternate template to guide mutagenesis. The results demonstrate the usefulness and robustness of in-silico 3D models. These models could also be useful for the design of novel and potent CCR2 and CCR5 antagonists using structure based drug design.

P123-Templated Co3O4/Al2O3 Mesoporous Mixed Oxides for Epoxidation of Styrene

  • $Co_3O_4$, $Al_2O_3$ and $Co_3O_4$/$Al_2O_3$ mesoporous powders were prepared by a sol-gel method with starting matierals of aluminum isopropoxide and cobalt (II) nitrate. A P123 template is employed as an active organic additive for improving the specific surface area of the mixed oxide by forming surfactant micelles. A transition metal cobalt oxide supported on alumina with and without P123 was tested to find the most active and selective conditions as a heterogeneous catalyst in the reaction of styrene epoxidation. A bBlock copolymer-P123 template was added to the staring materials to control physical and chemical properties. The properties of $Co_3O_4$/$Al_2O_3$ powder with and without P123 were characterized using an X-ray diffractometer (XRD), a Field-Emission Scanning Electron Microscope (FE-SEM), a Bruner-Emmertt-Teller (BET) surface analyzer, and $^{27}Al$ MAS NMR spectroscopy. Powders with and without P123 were compared in catalytic tests. The catalytic activity and selectivity were monitored by GC/MS, $^1H$, and $^{13}C$-NMR spectroscopy. The performance for the reaction of epoxidation of styrene was observed to be in the following order: [$Co_3O_4$/$Al_2O_3$ with P123-1173 K > $Co_3O_4$/$Al_2O_3$ with P123-973 K > $Co_3O_4$-973 K>$Co_3O_4$/$Al_2O_3$-973 K > $Co_3O_4$/$Al_2O_3$ with P123-1473 K > $Al_2O_3$-973 K]. The existence of ${\gamma}$-alumina and the nature of the surface morphology are related to catalytic activity.

  • Following natural disasters, there is a need to develop temporary housing through the rapid installation of mass-produced prefabricated housing. This study has developed a model construction manual for prefabricated temporary housing installable in underground spaces. First, the temporary housing prototype was drawn based on opinion poll results from earlier studies. $4.32m^2$ was calculated as the basic unit of the temporary housing living space for one person. Units included panel type, box type and mixed-type, for a variety of field conditions and scales. In order to enable space expansion and movement desirable to the user, the prototype was combination of panel and box types. The drawn prototype was utilized as the basic design template for producing prototypes and for the manual. Second, the possibility of utilizing subway stations as temporary housing was verified through the production of the prototype, and the construction process was summarized in the manual. The manual was developed in the form of a checklist to provide a guideline for users to build with prefab materials.

  • We present results from an experimental study of conductivity change of poly (3,4-ethylenedioxythiophene) poly(styrenesulfonate) (PEDOT:PSS) film according to the surface structuring. We demonstrate that the patterned structure was enhanced with approximately five times conductivity in comparison with non structure of PEDOT:PSS film. In order to patterning, we have fabricated polystyrene (PS) colloidal monolayer as a template with sphere diameter of 780nm and 1.8um. Structure has honeycomb shape and it provide shorter path way to flowing of electron. Pattern size was controlled by PS diameter and varied by Transformer Coupled Plasma (TCP) etching system. Conductivity was converted from sheet resistance which measured by 4-point prove. Film thickness was derived using Field Emission Scanning Electron Microscopy (FE-SEM) images.

  • 오늘날 유기고분자기반 태양전지는 다른 태양전지와 비교될 정도로 낮은 광변환효율로 인해 효율향 상을 위한 많은 연구들이 진행되어 왔다. 그중 패터닝을 통한 광포집률과 charge carrier 수집효율이 증가되었다는 많은 보고들이 있었다. 따라서 우리는 200~1,400 nm polystyrene bead를 합성하여 air-liquid interfacial 방법을 이용해 2차원 육방조밀구조를 갖는 template를 형성하고 Nanosphere lithography (NSL)를 이용하여 대면적으로 균일한 poly(3,4-ethylenedioxythiophene) poly(styrenesulfonate) (PEDOT:PSS)를 패턴화하였다. 균일한 패턴형성을 측정하기위해 Field Emission Scanning Electron Microscopy (FE-SEM), image를 얻었으며, Atomic Force Microscopy (AFM)를 통해 형성된 패턴의 낙차 높이를 얻었고, Near IR-UV-Vis을 통해 bead size 변화에따라 얻어진 PEDOT:PSS 패턴의 반사율을 측 정하였다.

  • In general, the durability performance of a large-sized military truck has been checked through a field durability test which required many man-hours and costs. To reduce these expenses, the durability analysis using a VTL(Virtual Test Lab) at an initial design stage was introduced recently. In this paper, the VTL with a multi-post testrig template for a large-sized truck was developed to compute the load histories transferred to cabin and chassis frame. The VTL consisted of trimmed FE models of cabin, chassis frame, and deck, dynamic models of front and rear suspensions, and a 8-post testrig template. The basic characteristics of the VTL were correlated with experimental results which had been extracted from actual driving test, modal test, and static weight test. The fatigue analysis using MSM(Modal Superposition Method) was applied to evaluate the durability performance of a large-sized military truck. From a series of analytic methods, it is shown that the fatigue analysis process using the VTL could be a useful tool to estimate the fatigue lives and weak areas of a large-sized military truck.

The Effects of Kaolin Addition on the Properties of Reticulated Porous Diatomite-kaolin Composites (고령토의 첨가가 3차원 망상 구조를 가지는 다공성 규조토-고령토 복합재의 기본 특성에 미치는 영향)

  • In this study, the effects of kaolin addition on the properties of reticulated porous diatomite-kaolin composites are investigated. A reticulated porous diatomite-kaolin composite is prepared using the replica template method. The microstructure and pore characteristics of the reticulated porous diatomite-kaolin composites are analyzed by controlling the PPI value (45, 60, and 80 PPI) of the polyurethane foam (which are used as the polymer template), the ball-milling time (8 and 24 h), and the amount of kaolin (0-50 wt. %). The average pore size decreases as the amount of kaolin increases in the reticulated porous diatomite-kaolin composite. As the amount of kaolin increases, it can be determined that the amount of inter-connected pore channels is reduced because the plate-shaped kaolin particles connect the gaps between irregular diatomite particles. Consequently, a higher kaolin percentage affects the overall mechanical properties by improving the pore channel connectivity. The effect of kaolin addition on the basic properties of the reticulated porous diatomite-kaolin composite is further discussed with characterization data such as pore size distribution, scanning electron microscopy images, and compressive strength.
