In Machine Learning Methods (ML) utilizes phonetic highlights with apply notable ML algorithm. In this paper, in classification and identification be complete base under in optimizations technique called sequential minimal optimization with Random Forest algorithm (SMORF) for expanding the exhibition and proficiency of sentiment classification framework. The three existing classification algorithms are compared with proposed SMORF algorithm. Imitation result within experiential structure is Precisions (P), recalls (R), F-measures (F) and accuracy metric. The proposed sequential minimal optimization with Random Forest (SMORF) provides the great accuracy.

Complete genome and two plasmids sequences of Lactiplantibacillus plantarum L55 for probiotic potentials

  • In this study, we report the complete genome sequence of Lactiplantibacillus plantarum L55, a probiotic strain of lactic acid bacteria isolated from kimchi. The genome consists of one circular chromosome (2,077,416 base pair [bp]) with a guanine cytosine (GC) content of 44.5%, and two circular plasmid sequences (54,267 and 19,592 bp, respectively). We also conducted a comprehensive analysis of the genome, which identified the presence of functional genes, genomic islands, and antibiotic-resistance genes. The genome sequence data presented in this study provide insights into the genetic basis of L. plantarum L55, which could be beneficial for the future development of probiotic applications.

  • In this study, we performed whole-genome sequencing of Levilactobacillus brevis KL251 (KL251) isolated from kimchi. The KL251 genome, characterized by a circular chromosome spanning 2,345,062 base pairs with a GC content of 45.78%, was analyzed. KL251 contains 2,275 coding DNA sequences (CDSs), 56 transfer RNAs (tRNAs), and 4 ribosomal RNAs (rRNAs). Genes associated with gamma-aminobutyric acid (GABA) production and CRISPR-Cas systems were identified and could potentially be used for GABA synthesis and defense against foreign DNA. Additionally, the presence of functional genes involved in isoprenoid biosynthesis, glutathione generation, and redox sensing showed that cellular metabolism and stress responses were important characteristics of this genome. These genomic findings suggest that the KL251 strain could potentially have several applications, including food fermentation, probiotics, dairy product starters, and the development of health-enhancing products.

  • Pullorum disease and Fowl typhoid are kind of poultry specific disease for poultry. The peculiar character of these poultry specific diseases is that it can be infected by transmitting vertically and horizontally, also it is hard to be discovered by clinical sign, and pathology or immunology. So, to develop the PCR method which distinguishes these two genetically similar diseases of separated the specific DNA fragment from each strain and use it for differential diagnosis by subtraction PCR method. Standard strain of S gallinarum and S pullorum, and field isolation strain were verified by biochemistry, It confirmed existence of plasmid by using the PFGE. Then, Isolated DNA from it and used it as materials for the experiment. After cutting genomic DNA of two strains by using Sau 3Al, It ligated primer to tester DNA for PCR amplification and separated specific DNA fragment bacteria with method of subtraction PCR. And, It confirmed that it is a piece of unique DNA in every bacteria using base sequence of separated DNA fragment. 1. The six specific DNA fragment were separated from the DNA of S gallinarum and S pullorum by the subtraction PCR method. 2. In the result of comparison after setting base sequence of each fragment, each separated base sequence of DNA fragment they did not correspond to each other 3. As the result of each DNA fragment is derived from the each strain of DNA, and there was no homology of genomic DNA level in mutual. 4. The fragment originated in plasmid and includes S pullorum did not separate. 5. In the result of searching base sequence in Genebank, it partially shows homology in Salmonella enterica, S typhimurium, S dublin, Escherichia coli, Shigella flexneri, Yersinia pestis, Klebsiella pneumoniae. 6. Primer design by S gallinarum DNA 2, 3 fragment used PCR, They are positive reaction in only S gallinarum at 276, 367 bp position.

Isolation and Identification of Antimicrobial Compound from Amarantus lividus (참비름 추출물에서 항균성 물질의 분리 및 동정)

  • Isolation and identification of pathogens from slaughter and meat processing plant were investigated. Antimicrobial activity of Amaranthus lividus against isolated pathogens such as Aeromonas sobria, Escherichia coli, Escherichia coli O157, Listeria monocytogenes, Salmonella spp., and Staphylococcus aureus was investigated. Among the chloroform, ethyl acetate and buthanol fraction of amaranthus lividus showed inhibitory effect against Aeromonas sobria CLFM1 and Escherichia coli CLFM2. Antimicrobial substance in chloroform fraction was isolated by silica gel adsorption column chromatography, sephadex LH-20 column chromatography and silica gel partition column chromatography. The antimicrobial compound of amaranthus lividus was identified as diethyl phtalate by HPLC, GC-MS, H-NMR and C-NMR.

Molecular genetic analysis of phytochelatin synthase genes in Arabidopsis

  • This study has investigated the biosynthesis and function of the heavy metal binding peptides, the phytochelatins, in plants. PCs are synthesised enzymatically from glutathione by the enzyme PC synthase in the presence of heavy metal ions. Using Arabidopsis thaliana as a model organism cadmium-sensitive, phytochelatin-deficient mutants have been isolated and characterised in previous studies. The cadl mutants have wildtype levels of glutathione, are PC deficient and lack PC synthase activity. Thus, the CADl gene has been proposed to encode PC synthase. The CADl gene was isolated by a positional cloning strategy The gene was mapped and a candidate identified. Each of four cadl mutants had a single base pair change in the candidate gene and the cadmium-sensitive, cadl phenotype was complemented by the candidate gene. This demonstrated the CADl gene had been cloned. A homologous gene in the fission yeast, Schizosaccharomyces pombe was identified through database searches. A targeted-deletion mutation of this gene was constructed and the mutant, like cadl mutants of Arabidopsis, was cadmium-sensitive and PC-deficient. A comparison of the redicted amino acid sequences reveals a highly conserved N-terminal region Presumed to be the catalytic domain and a variable C-terminal region containing multiple Cys residues proposed to be involved in activation of the enzyme by metal ions. Similar genes were also identified in animal species. The Arabidopsis CADl/AtPCSl and S. pombe SpbPCS genes were expressed in E. coli and were shown to be sufficient for glutathione-dependent, heavy metal activate PC synthesis in vitro, thus demonstrating these genes encode PC synthase enzymes. Using RT-PCR, AtPCSl expression appeared to be independent of Cd exposure. However, at higher levels of Cd exposure a AtPCSl-CUS reporter gene construct appeared to be more highly expressed. Using the reporter gene construct, AtPCSl was expressed most tissues. Expression appeared to be greater in younger tissues and same higher levels of expression was observed in some regions, including carpels and the base of siliques. AtPCS2 was a functional gene encoding an active PC synthase. However, its Pattern of expression and the phenotype of a mutant (or antisense line) have not been determined. Assuming the gene is functional then it has clearly been maintained through evolution and must provide some selective advantage. This implies that, at least in some cells or tissue, it is likely to be the dominant PC synthase expressed. This remains to be determined

  • The boundary reaction method(BRM) is a substructure time domain method, it removes global iterations between frequency and time domain analyses commonly required in the hybrid approaches, so that it operates as a two-step uncoupled method. The BRM offers a two-step method as follows: (1) the calculation of boundary reaction forces in the frequency domain on an interface of linear and nonlinear regions, (2) solving the wave radiation problem subjected to the boundary reaction forces in the time domain. In the time domain analysis, the near-field soil is modeled to simulate the wave radiation problem. This paper evaluates the performance of the BRM according to modeling extent of near-field soil for the nonlinear SSI analysis of base-isolated NPP structure. For this purpose, parametric studies are performed using equivalent linear SSI problems. The accuracy of the BRM solution is evaluated by comparing the BRM solution with that of conventional SSI seismic technique. The numerical results show that the soil condition affects the modeling range of near-field soil for the BRM analysis as well as the size of the basemat. Finally, the BRM is applied for the nonlinear SSI analysis of a base-isolated NPP structure to demonstrate the accuracy and effectiveness of the method.

  • Carbonic anhydrase isozymes are a widespread, zinc-containing metalloenzyme family. The enzyme catalyzes the reversible inter-conversion of $CO_2$ and $HCO_3$. This reaction is the main role played by CA enzymes in physiological conditions. This enzyme has been found in virtually all organisms, and at least 16 isozymes have been isolated in mammals. Unlike mammals, there is little information available regarding CA isozymes in the tissues of non-mammalian groups, such as fish. Carbonic anhydrase is very important in the osmotic and acid-base regulation in fish. It is well-known that the gills of fish play the most important role in acid-base relevant ion transfer, the transfer of $H^+$ and/or $HCO_3^-$, for the maintenance of systemic pH. Rainbow trout, Oncorhynchus mykiss, is the most important freshwater fish species in the aquaculture industry of Korea, with annual production increasing each year. In addition, environmental toxicology research has shown that rainbow trout is known to be the species that is most susceptible to environmental toxins. Consequently, carbonic anhydrase was detected in rainbow trout, Oncorhynchus mykiss. The isolated protein showed the specific band with a molecular weight of 30 kDa and pI of 7.0, and it was identified as being carbonic anhydrase. The immunohistochemical result demonstrated that the carbonic anhydrase was located in the epithelial cells of the gills.

A Study on the Nonlinear Modeling of Lead Rubber Bearings by a Neural Network Theory (신경망 이론을 적용한 납삽입 적층 고무베어링의 비선형 모델링 기법에 관한 연구)

  • In this paper, a nonlinear modeling of lead rubber bearings(LRBs) was presented by a neural network theory. An shaking table test for a scaled frame model, of which base was isolated by the LRBs, was performed to verify numerical accuracies of the neural network model. White noise and three types of seismic records were adoped as base loads of the shaking table in order to train and generalize the neural network in case of seismic loads, numerical results of the neural network model were evaluated according to different magnitudes of PGA. As results, it is concluded that the presented neural network model has given a good agreement with the experimental data in details and can be useful to a nonlinear modeling of LRBs within prescribed domains.

Comparison of seismic behavior of long period SDOF systems mounted on friction isolators under near-field earthquakes

  • Friction isolators are one of the most important types of bearings used to mitigate damages of earthquakes. The adaptive behavior of these isolators allows them to achieve multiple levels of performances and predictable seismic behavior during different earthquake hazard levels. There are three main types of friction isolators. The first generation with one sliding surface is known as Friction Pendulum System (FPS) isolators. The double concave friction pendulum (DCFP) with two sliding surfaces is an advanced form of FPS, and the third one, with fully adaptive behavior, is named as triple concave friction pendulum (TCFP). The current study has been conducted to investigate and compare seismic responses of these three types of isolators. The structure is idealized as a two-dimensional single degree of freedom (SDOF) resting on isolators. The coupled differential equations of motion are derived and solved using state space formulation. Seismic responses of isolated structures using each one of these isolators are investigated under seven near fault earthquake motions. The peak values of bearing displacement and base shear are studied employing the variation of essential parameters such as superstructure period, effective isolation period and effective damping of isolator. The results demonstrate a more efficient seismic behavior of TCFP isolator comparing to the other types of isolators. This efficiency depends on the selected effective isolation period as well as effective isolation damping. The investigation shows that increasing the effective isolation period or decreasing the effective isolation damping improves the seismic behavior of TCFP compared to the other isolators. The maximum difference in seismic responses, the base shear and the bearing displacement, for the TCFP isolator are calculated 26.8 and 13.4 percent less than the DCFP and FPS in effective isolation damping equal to10%, respectively.
