Genogroup position of aquabirnavirus GC-1 isolated from rockfish Sebastes schiegeli in Korea

  • The cDNA of the aquabirnavirus, GC-1 isolated from rockfish Sebastes schlegeli in Korea, was synthesized using the reverse transcriptase-polymerase chain reaction. The nucleotide and deduced amino acid sequences were determined from cDNA of the VP2-NS-VP3 coding region of genome segment A. The nucleotide sequences of the segment A were 3,086 base pairs (bp) in length and contained large open reading frame (ORF) and terminal sequences. The large ORF was comprised of 2,916 bp nucleotides and composed of 972 deduced amino acid sequences. Pairwise comparisons were made with other aquabirnavirus sequences published previously. The study of genetic relationships between GC-1 and aquabirnaviruses in the large ORF and VP2 coding regions demonstrated that the GC-1 has the nearest genetic relationship with the marine birnaviruses (MABV strains), and the GC-1 and MABV strains can be clustered as the same genogroup. GC-1 can be included in MABV, which is the 7th genogroup of family Aquabirnaviridae.

Synthesis of N-Alkylated 4-Fluoro-5-phenylpyrrole-2-carboxylate via Isolable Pyrroline Ionic Intermediates

  • Organic fluorine chemistry produces many useful products. This paper elucidates the reaction of ethyl-4,4- difluoro-2-iodo-5-oxo-5-phenylpentanoate (2) with primary amines in a one-pot scheme. The reaction produced a series of β-fluoropyrrole derivatives at ambient temperatures. In this reaction, the less bulky the primary amine the higher was the resultant yield. When (2) and aqueous methylamine (40%) were allowed to react below 0 oC, 5-(ethoxycarboxyl)-1-methyl-3,3-difluoro-2-hydroxy-2-phenylpyrrolidine, an intermediate molecule for 2-ethyl-4-flouro-1-methyl-5-phenylpyrrole-2-carboxylate (5), was isolated first. Then, (5) reacted with hydroperchloric acid and acetic anhydride to form 5-(ethoxycarboxyl)-1-methyl-3,3-difluoro-2- phenylpyrrolinium perchlorate (6), which was converted to 2-ethyl-4-flouro-1-methyl-5-phenylpyrrole-2- carboxylate gradually in the presence of a base. Our experiments demonstrate that the formation of 2-ethyl-4- flouro-1-methyl-5-phenylpyrrole-2-carboxylate occurs via both one-pot schemes and stepwise pathways, depending on the reaction conditions. The isolation and characterization of the isolated intermediate (6) suggest an anionic pathway for this reaction.

  • DNA topoisomerase I(TOP1) helps the control of DNA replication, transcription and recombination by assist­ing breaking and rejoining of DNA double strand. Camptothecin (CPT) and its derivative, topotecan, are known to inhibit TOP1 by intercalating into TOP1-DNA complex. Recently various non-CPT intercalators are synthesized for a new class of TOP1 inhibitors. In this study, six compounds isolated from Poria cocos were investigated for their interaction with TOP1­DNA complex using the flexible docking program, FlexiDock. The binding modes were analyzed and compared with the TOP1 inhibition activities. The compounds that showed potent activity were intercalated between the + 1/-1 base pairs of DNA, located near the active site phosphotyrosine723 and formed hydrogen bonds with active site residues. On the other hand, compounds with no activity were not docked at all. The binding modes were well correlated with the inhibition activity, suggesting the possibility that potent inhibitors can be designed from the information presented by the docking study.

Complete Nucleotide Sequence of Cytochrome P450 2El Expressed in the Rat Brain

  • From the RT-PCR amplifications using mRNA templates isolated from Sprague-Dawley rat brains, we isolated a cDNA fragment of 1,524 bp which covered the full coding information of the rat brain CYP2El. Its nucleotide sequence was identical to the previously reported rat liver CYP2El mRNA except for the difference of one base (A to C at the nucleotide position 73). This difference also altered the amino acid Lys to GIn. However, no insertion or deletion of nucleotide(s) which could alter the reading frame was found within the structure of this rat brain CYP2El. This study should provide the molecular basis regarding the pathophysiological function of CYP2El in the brain.

Rearing managements of pig farms and survey on pneumonia of slaughtered pigs (양돈장 사양관리와 도축돈 폐 병변조사)

  • This studies were carried out to investigate the rearing managements of pig farms and survey on pneumonia of the slaughtered pigs from 5 selected herds located in Jangsu, Jeonbuk. Isolated aerobic microorganisms from pneumonic lung were examined antibiotic susceptibility and tested serological antibody titers of the herd base. Prevalence rate of pneumonia were examined according to rearing and health management conditions of pig farms. Prevalence rate of pneumonia were detected in 78.8% and enzotic pneumonia. In 47.7%, pleuropneurnonia in 31.1%. In serological antibody titers showed the positive reaction with 54.4% in Actinobacillus pleuropneumonia serotype 5, 44.8% in Pasteurella multocida, 36.8% in A pleuropneurnonia serotype 2, 13.6% in Mycoplasmal pneumonia. Isolated aerobic microorganisms were examined antibiotic susceptibility and showed the high activity in gentamicin (58.3%), enrofloxacin (53.3%), norfloxacin (51.6%), cephalothin (41.7%) and low activity in amoxycillin (98.3%), oxytetracycline (98.3%), penicillin G (90.0 %), tetracycline (88.4%), ampicillin (88.3%). Farm managements were deficient effect of humidity in swine house but ammonia gas all appeared the 10 ppm that were recommendation density, below.

Some Peroxysterols and Ceramides from "Phellinus ribis", a Korean Wild Mushroom

  • Studies on the chemical constituents from a Korean wild mushroom, Phellinus ribis, were carried out. A triterpenoid, two peroxysterols, and a chlorobenzene compound were isolated from the hexane soluble fraction of the methanol extract of dried fruiting bodies of the basidomycetes. Those compounds identifed were 3-hydroxy-20(29)-lupen-28-oic acid (betulinic acid), 5,8-epidioxyergosta-6,22-dien-3-ol(ergosterol peroxide), 5,8-epidioxyergosta-6,9(11),22-trien-3-ol (dehydroperoxyergosterol), and 1,2,4,5-tetrachloro-3,6-dimethoxybenzene. Structural studies were carried out on molecular species of a ceramide and cerebroside isolated from the chloroform soluble fraction of the methanol extract. For ceramide, the major component fatty acids were a-hydroxy fatty acid isomers of $C_{22:00}{\sim}C_{25:00};$ the predominant long-chain bases were trihydroxy sphinganine of $C_{17}{\sim}C_{18}$. The structure of a cerebroside containing mono-sugar was assumed that the long-chain base was $C_{19:2}$ sphingadienine; the major fatty acids were $C_{16}{\sim}C_{15}$ ${\alpha}$-hydroxy fatty acid isomers.

Seismic Fragility Evaluation of Isolated NPP Containment Structure Considering Soil-Structure Interaction Effect (지반-구조물 상호작용 효과를 고려한 지진격리시스템이 적용된 원전 격납건물의 지진 취약도 평가)

  • Several researches have been studied to enhance the seismic performance of nuclear power plants (NPPs) by application of seismic isolation. If a seismic base isolation system is applied to NPPs, seismic performance of nuclear power plants should be reevaluated considering the soil-structure interaction effect. The seismic fragility analysis method has been used as a quantitative seismic safety evaluation method for the NPP structures and equipment. In this study, the seismic performance of an isolated NPP is evaluated by seismic fragility curves considering the soil-structure interaction effect. The designed seismic isolation is introduced to a containment building of Shin-Kori NPP which is KSNP (Korean Standard Nuclear Power Plant), to improve its seismic performance. The seismic analysis is performed considering the soil-structure interaction effect by using the linearized model of seismic isolation with SASSI (System for Analysis of Soil-Structure Interaction) program. Finally, the seismic fragility is evaluated based on soil-isolation-structure interaction analysis results.

Antimicrobial Resistance of Clinically Important Bacteria Isolated from Burn Wound Infections in Children (화상감염 소아환자에서 분리된 주요 균종에 대한 항생제의 내성률)

  • Background & Objectives: Burn injury mortality and septic complication are frequent and well-known in burned pediatric patients. The overuse of antibiotics is the base for development of wound infection by resistant microorganisms as well as opportunist agents. Methods: We have carried out a study of the bacterial profile and antimicrobial resistance clinically important bacteria isolated from burn wound infections in children patients. The most common isolate from burn wound cultures was Pseudomonas aeruginosa (26.8%), followed by Staphylococcus aureus (25.4%), Acinetobacter baumannii (12.7%), coagulase negative staphylococcus (12.0%), Enterococcus faecium (7.7%), Escherichia coli (4.9%), Enterococcus faecalis (3.7%), Burkholderia cepacia (3.0%), Enterobacter cloacae (2.3%) and Klebsiella pneumonia (2.3%). Colistin was very significantly effective drug in gram negative organism, such as Pseudomonas aeruginosa and Acinetobacter baumannii. Results & Conclusion: The resistance rates were 65% and 98% to piperacillin, 63% and 97% to ceftazidime, 28% and 50% to levofloxacin. The most effective antibiotic in gram positive organism, such as Staphylococcus aureus, coagulase negative staphylococcus were moxifloxacin. The resistance rates were 83% and 64% to ciprofloxacin, 80% and 17% to clindamycin.

Isolation and Characterization of Major Glycosphingolipid from Rice Bran Extract (쌀겨 추출물로부터 스핑고당지질의 분리와 구조결정)

  • In order to examine the biofunctions of glycosylceramide which is representative of sphingolipid, monoglycosylceramide (cerebroside) was isolated from rice bran extract. Crude glycosylceramides were isolated in large quantities and promptly by flash system column chromatography from rice bran extract, and purified by normal-phase HPLC using an evaporative light-scattering detector. One major cerebroside was obtained by HPLC used as an eluent consisting of chloroform, methanol and water (99:11:1, v/v/v), and the constituents were analyzed by MALDI/TOF-MS, FAB-MS, GC and 600 MHz $^1$H-NMR. The component sugar was estimated to be glucose. In the ceramide, the fatty acid component consist was 2-hydroxy arachidic acid. The long-chain base component was sphinga-4,8-dienine.

Behaviour of asymmetric building with double variable frequency pendulum isolator

  • Presented in this paper is the behaviour of asymmetric building isolated by the double variable frequency pendulum isolator (DVFPI). The DVFPI is an adoption of single variable frequency pendulum isolator (VFPI). The geometry and coefficient of friction of top and bottom sliding surfaces can be unequal. The governing equations of motion of the building-isolation system are derived and solved in incremental form. The analysis duly considers the interaction of frictional forces in the two principal directions developed at each sliding surface of the DVFPI. In order to investigate the behaviour of the base isolation using the DVFPI, the coupled lateral-torsional response is obtained under different parametric variations for a set of six far-fault earthquake ground motions and criterion to optimize its performance is proposed. Further, influences of the initial time period, coefficient of friction and frequency variation factors at the two sliding surfaces are investigated. The numerical results of the extensive parametric study help in understanding the torsional behaviour of the structure isolated with the double sliding surfaces as in the DVFPI. It is found that the performance of the DVFPI can be optimized by designing the top sliding surface initially softer and smoother relative to the bottom one.
