Studies on the Change of Minerals during Yakju Brewing(I) (Aspergillus kawachii를 이용한 약주 양조법에 따른 무기물의 변화 I)

  • Calcium and magnesium in polished rice, polished barley, what flour, and corn which were used for Yakju fermentation were determined by atomic absorption spectrophotometry and their quantitative changes in the filtrates of mashes were checked at 24 hours intervals dueing the whole brewing period. The results obtained were as follows. 1) Mineral contens of raw materials(mg% of each anhydrous materials). Ca : polished rice 8.21, polished barley 26.11, wheat flour 66.10, corn 86.63 2) Mineral contents of kojies made from raw materials (mg %of each anhydrous materials). Ca : rice koji 26.36, barley koji 97.61, what flour koji 87.69, corn koji 16.13, seeding koji 28.76 Mg: rice koji 29.29, barley koji 39.84, what flour koji 244.50, corn koji 102.64, seeding koji 143.79 3) Quantitative changes of minerals in the fitrates of mashes. Calcium contents in the filtrates of mashes were increased gradually after mashing in the first stage but unchanged till the mashes were ripened after mashing in the second stage. On the other hand, magnesium cotents were decreased gradually after increase in the the other hand, magnesium contens were decreased gradually after increase in the first stage but showed a tendency to increase gradually in the second stage. 4)Mineral contents of Yakjues produced, marketing Yakju, and natural water for brewing (g/ml). Ca : rice Yakju 72.38, barley Yakju 84.08, what flour Yakju 105.32, corn Yakju 71.26, marketing Yakju 71.50, natural water for brewing 51.25 Mg :rice Yakju 93.67, barley Yakju 62.39, wheat flour Yakju 273.34, corn Yakju 321,60, marketing Yakju 90.00, natrual water for brewing 20.00 5) Mineral contents of Yakju residues (mg% of each anhydrous matrials).   • The manufacture of riboflavine fortified Dwen-Jang has been tried with an exceedingly riboflavine productive Aspergillus oryzae #612 mutant which has been developed by the authors. Both the rice and barley koji of this mutant and Aspergillus sojae have been prepared. Their riboflavine production, saccharifying and protease activities have begin compared The riboflavine fortified Dwen-Jang has been manufactured using the barley koji of riboflavine productive mutant. Their riboflavine content and qualities have been studied comparing with an ordinary Dwen-Jang which has been prepared with the barley kojo of A. sojae strain. The following results have been obtained. (1) The baley koji was superior in riboflavine production and protease activity, inferior in saccharifying ability than rice koji both with A. oryzae #612 and A. sojae. (2) In barley koji, the mutant, A. oryzae #612, produces 1.5 times riboflavine than A. sojae and shows stronger saccharifying and protease activities than the latter. (3) The riboflavine fortified Dwen-Jaug manufactured contained $5.2{\gamma}/g$ of riboflavine, about 1.5 times that of A. sojae. The higher contents of free sugar and free amjno nitrogen have been observed than the ordinary Dwen-Jang manufactured with A. sojae.

  • Studies on the brewing of Takju with polished barley was carried out. The results obtained and as follows. 1. Polished barley koji was not available because of low acidit production. 2. No significant differences of Takju quality observed among treatments of polished barley, but barleyflour Takju was remarkably inferior to other materials originated Takju in quality and color. 3. Yield per compressed barley Takju was higher than that of other type of materials. 4. The most desirable enzyme sources ratio in the brewing with polished barley were koji 20-30%, Kokja 2.5%, bunkuk 2% respectively. 5. This results indicates that Takju berwing with polished barley is possible.

  • The enzymic activities of Kojis which were prepared by the various combination of raw-materials, soybean and barley, were measured and the ripening process of soybean paste made from the kojis was pursued. 1). The koji made by the admixture of soybean, barley and wheat at the ratio of 40-60 : 60-40:20 was superior to the Kojis made from soybean or barley alone, in the aspect of dextrinizing, saccharifying and proteolytic activities. 2). The soybean paste mashed with the admixed koji with higher enzymic activities caused the more and rapid formation of reducing sugars and proteolysis than the soybean pate from barley-koji. 3). The effect of salt on the activities of ${\alpha}-amylase$, saccharification and proteases was examined and it was noticed that the three was inhibited to the different extent respectively.

  • This study was carried out to investigate the effect of Monascus barley koji powder addition on the preparation of chocolate(milk, white). The viscosity, moisture contents, color, rancidity and sensory evaluation were observed. When the ratio(0, 2, 4, 6%) of Monascus barley koji powder were varied on the preparation of chocolate, the milk chocolate was not changed respect to viscosity, moisture contents, color and rancidity. Under the same condition, the white chocolate was observed as follows, viscosity was increased a little bit, moisture contents were decreased. In color, L(lightness) and a(redness) value were increased obviously but the b value(yellowness) was decreased. And there are no changed in rancidity. In sensory evaluation, the milk chocolate with Monascus barley koji powder(0∼6%) addition did not make a significant difference in color, taste and flavor but texture and acceptability. The white chocolate with Monascus barley koji powder(0∼2%) addition was observed no significant difference in taste, smell, texture and acceptability except for color.

  • For the automation of Koji incubation process, microcomputer based Koji incubation system was built and applied to acquisition of the process variables, and to control of the Koji incubation process. The incubation variables included the relative humidity and Koji weight. And data measured were sent to the microcomputer by the interface device built with MC 6821 PIA. Incubation environment conditions -temperature and humidity- were controlled by the actuation of heater and mist sprayer with on/off signal generated by ASIC program. Aspergillus oryzae as a starter of the Koji and steamed barley as media were used and Koji was successfully manufactured both at $25^{\circ}C,\;70%$ RH and at $27^{\circ}C,\;80%$ RH. During the Koji preperation, the temperature was linearly increased and substrate was consumed stepwise showing 3 steps in the weight loss curve.

Effects of Barley Koji and Legumes on the Quality and Fibrinolytic Activity of Korean Traditional Rice Wine (보리 입국과 두류 첨가가 전통주의 품질과 혈전용해활성에 미치는 영향)

  • Effects of cereal kojis and legumes on alcohol fermentation and fibrinolytic activities of Korean traditional wines were investigated. The Korean traditional wine which was brewed by addition of 10% barley koji into the mash showed the greatest fibrinolytic activity of 20.0 U and good ethanol productivity (16.8%). The fibrinolytic activity was increased up to 26.0 U by addition of 50% of mungbean and its acceptability were improved by addition of jujube (3%) into the mash.

  • The chemical changes and the quality of the soysauce which was Bade by new convenient method using vinyl teat instead of koji room, replacing wheat with milled barley koji were investigated. Protease activity of the koji was higher as tile ratio of soybeans was increased but amylase activity was higher as the ratio of milled barley was increased. The amounts of total nitrogen showed substantial increases until 80 days and afterwards, their rates of increases were lower. The more its content of soybeans, the higher its amount of total nitrogen, The amount of amino nitrogen increased slowly in all samples and the sample which had a large amount of soybeans was higher than those with small amounts of soybeans. The amount of ammonia nitrogen showed increases until 120 days tut decreased after then. The amount of reducing sugar showed increases until 80 days and decreased after then and the greater its source of carton, the higher its amount of reducing sugar. The total acid contort had large increases until 100 days in all samples and afterward they did not show any particular changes. The changes of pH decreased for 100 days and afterwards, they increased a little. In the original soysauce, combined ratio of soybean 3 to milled barley 2 and the sample of those ratio 4 : 1 are better than others and in diluted soysauce combined ratio of soybean 4 to milled barley 1 is better than the others. In original soysauce the use of $20{\sim}40%$ barley koji, which is added to soybeans, is superior and diluted soysauce the use of 20% barley koji is superior.

  • Kojis were prepared from the different starch raw materials such as glutinous rice, barley rice, wheat flour and sweet potato, and were tested in an effort to reduce production cost and to improve quality of Kochuzang(red pepper paste). During the starting period of Koji preparation, there were less significant changes in pH; however, pH decreased somehow in the case of sweet potato, whereas it increased for the other Kojis. In general, the highest acidity was obtained after 72 hours of Koji preparation. Total nitrogen, soluble nitrogen and amino-nitrogen content increased in order of wheat flour, barley rice, and glutinous rice; Kojis they were markedly produced between 72 and 96 hours of Koji preparation. The maximum amount of reducing sugar was observed between 48 and 72 hours of Koji preparation during this period the reducing sugar content varied widely depending on starch source. Sweet potato Koji produced the highest level of ethyl alcohol content after 72 hours of Koji preparation; for the other Koji the same trends were observed after 24 hours. The starch liquefying activities have reached the highest level after 96 hours and for glutinous rice and barley rice; however, they kept on increasing until 120 hours for wheat flour and sweet potato. Koji Starch liquefying and saccharogenic amylase activities tended to increase in order of sweet potato, glutinous rice, barley rice and wheat flour. Kojis Various protease activities were measu-red during the Koji preparation, and they increased in order of alkali, neutral and acidic protease.

Utilization of Soymilk Residue for Barley Doenjang (두유박을 이용한 보리 된장 제조)

  • For the effective use of soymilk residue (SR), by-product of soymilk production, barley Doenjang was manufactured. As the results, the contents of reducing sugar increase3 for 50 days in all samples and decreased thereafter. The contents of amino nitrogen increased prominently for 50 days, but increasing ratio was slightly slowdowned thereafter. The higher mixing ratio of SR, the less were the contents of reducing sugar and amino nitrogen. The contents of total acid also increased prominently for 30 days, but were slowdowned thereafter and the differences among all treatments were negligible. As the mixing ratio of SR was increased, the lightness and the yellowness increased and the redness decreased in 80days aged samples. In sensory evaluation of 90 days aged samples, the control was the best in soy sprout Doenjang soup. As the mixing ratio of SR increased, the taste was dropped slightly but no significant differences were obtained among sample A(barley koji 100, soybean 75, SR 25), B(barleykoji 100, soybean 50, SR 50) and C(barley koji 100, soybean 25, SR 75). In the original taste of Doenjang, the control was the best and as the mixing ratio of SR was increased, the point was dropped but these were no significant differences among control, A and B, and among A, B and C, and among B, C and D(barley koji 100, soybean 0, SR 100).
