Water extract from Pinus densiflora (WPD) was investigated for its antioxidant activity and its ability to provide protection from DNA damage. A series of antioxidant assays, including a 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay, a reducing power assay, a metal-chelating assay, a superoxide radical scavenging assay, and a nitrite scavenging ability, as well as a DNA damage protection assay were performed. Total phenolic content was found to be 211.32 mg Tan/g WPD. The extract scavenged 50% DPPH free radical at a concentration of 21.35 ${\mu}g/mL$. At that same concentration, the reducing power ability of WPD was higher than that of ${\alpha}$-tocopherol. The extract chelated 68.9% ferrous ion at the concentration of 4 mg/mL. WPD showed better nitrite scavenging effect at the lower pH. Meanwhile, WPD exhibited a strong capability for DNA damage protection at 1 mg/mL concentration. Taken together, these data suggest water extract from Pinus densiflora could be used as a suitable natural antioxidant.
The aim of this study were to evaluate tannin resorcinol formaldehyde (TRF) for the preparation of cross-laminated timbers (CLTs) made from fast-growing tree species and to analyze the physical and mechanical properties of CLTs. TRF copolymer resin was prepared by using the bark extracts of Swietenia mahagoni (L.) Jacq. It was observed that the TRF adhesive possessed less solid content (23.59%), high viscosity (11.35 poise), and high pH values (10.0) compared to the standard phenol resorcinol formaldehyde. The TRF adhesive was applied to produce CLTs with the addition of 15% tapioca and flour as an extender. The six-layered CLTs were produced from sengon (Falcataria moluccana Miq.), jabon [Anthocephalus cadamba (Roxb) Miq.], coconut (Cocos nucifera L.), and the combination of coconut-jabon and coconut-sengon wood. The analysis of variance revealed that the layer composition of CLT significantly affected the physical and mechanical properties of the beam. While the modulus of rupture met the standard, the moisture content and modulus of elasticity values did not fulfill JAS 1152-2007. All of the CLTs produced in this study demonstrated low formaldehyde emission, ranging from 0.001 mg/L to 0.003 mg/L, thereby satisfying the JAS 1152 for structural glue laminated timber.
The cultural characteristics and some factors such as nutrient sources and supplements effecting on mycelial growth and density were investigated to study the possibility of an artificial cultivation of P. cocos. The optimum pH for P. cocos was 4.0-4.5. The optimal growth temperature ranged from $25^{\circ}C$ to $29^{\circ}C$. Myceial growth of P. cocos was better in SPD than PD media. Adding the nurient sources such as dextrose, yeast and potato infusion to pine extract media practically stimulated the mycelial growth and density of P. cocos comparing to pine extract media alone. When P. cocos was cultured on sawdust media added 3 different supplements composed of corn meal, rice bran and wheat bran, corn meal was the best and its percentage was 30 (w/w) for mycelial growth. On culturing in sawdust media added by varying the mixture ratio of them, the media mixed corn meal and wheat bran (3:1, w/w) supported more vigours for mycelial growth. In inoculation test to pine stem, the fungal growth was good in under or inside pine bark and xylem, but the sclerotium was not observed in the stem. Mycelial growth was also observed in central part of pine stem by cross section.
Ulmus pumila L. bark underwent distilled water extraction under three temperature condition ($4^{\circ}C$, room temperature, or $80^{\circ}C$) and two extraction times (1, or 5 min) in order to develop a functional beverage products. Changes in yield, pH, color, total phenolic (TP) content, tannin content and antioxidant activity of the aqueous extracts were evaluated for each extraction temperature and duration. Extraction conditions did not affect yield or pH value of the extracts; however CIE $b^*$ values were high in extracts prepared under high extraction temperature ($80^{\circ}C$) and long extraction duration (5 min) conditions. Both extraction temperature and duration affected the TP and tannin contents of the extracts; however, all extraction conditions resulted in ${\geq}450\;mg\;GAE/g$ TP content and ${\geq}80\;mg\;CE/g$ tannin content. All extracts exhibited ABTS and DPPH radical scavenging ability similar to that of vitamin C. Nitric oxide inhibition activity was lower in the 5 min duration sample than in the 1 min sample. The $4^{\circ}C$ extraction temperature produced an extract with the highest reducing power and hydrogen peroxide values. Extraction temperature also affected sensory evaluation results with the $80^{\circ}C$ extraction temperature producing significantly higher flavor, bitterness, and color score, than those obtained under $4^{\circ}C$ and room temperature extraction conditions.

${\beta}$-lapachone(${\beta}$-Lap), a natural o-naphthoquinone, presents in the bark of the Lapacho tree. ${\beta}$-Lap is cytotoxic against a variety of human cancer cells and it potentiates the anti-tumor effect of Taxol. In addition, ${\beta}$-Lap has been reported to radiosensitize cancer cells by inhibiting the repair of radiation-induced DNA damage.In the present study, we investigated the cytotoxicity of ${\beta}$-Lap against RKO human colorectal cancer cells as well as the combined effect of ${\beta}$-LaP and ionizing radiation. An incubation of RKO cells with 5 ${\mu}$M of ${\beta}$-Lap for 4 h killed almost 90% of the clonogenic cells. An incubation of RKO cells with 5 ${\mu}$M of ${\beta}$-Lap for 4 h or longer also caused massive apoptosis. Unlike other cytotoxic agents, ${\beta}$-Lap did not increase the expression of p53 and p21 and it suppressed the NFkB expression. The expression of Caspase 9 and 3 was minimally altered by ${\beta}$-Lap. Radiation and ${\beta}$-Lap acted synergistically in inducing clonogenic cell death and apoptosis in RKO cells when ${\beta}$-Lap treatment was applied after but not before the radiation exposure of the cells. Interestingly, a 4 h treatment with 5 ${\mu}$M of ${\beta}$-Lap starting 5 h after irradiation was as effective as that starting immediately after irradiation. The mechanisms of ${\beta}$-Lap-induced cell killing is controversial but a recent hypothesis is that ${\beta}$-Lap is activated by NAD(P)H: quinone-onidoreductase (NQO1) in the cells followed by an elevation of cytosolic Ca$\^$2+/ level and activation of proteases leading to apoptosis. It has been reported that NQO1 level in cells is markedly up-regulated for longer than 10 h after irradiation. Indeed, using immunological staining of NQO1, we observed a significant elevation of NQO1 expression in RKO cells 5h after 2-4 Gy irradiation. Such a prolonged elevation of NQO1 level after irradiation may be the reasons why the ${\beta}$-Lap treatment applied S h after irradiation was as effective as that applied immediately after irradiation in killing the cells. In view of the fact that the repair of radiation-induced damage is usually completed within 1-2 h after irradiation, it is highly likely that the ${\beta}$-Lap treahment applied 5 h after irradiation could not inhibit the repair of radiation-induced damage. For in vivo study, RKO cells were injected S.C. into the hind-leg of Nu/Nu mice, and allowed to grow to 130 mm3 tumor. The mice were i.p. injected with ${\beta}$-lapachone or saline 2 h after irradiation of tumors with 10 Gy of X-rays. The radiation induced growth delay was increased by 2.4 $\mu\textrm{g}$/g of ${\beta}$-lapachone. Taken together, we may conclude that the synergistic interaction of radiation and ${\beta}$-Lap in killing cancer cells is not due to radiosensitization by ${\beta}$-Lap but to an enhancement of ${\beta}$-Lap cytotoxicity by radiation through an upregulation of NQO1. The fact that NQO1 is elevated in tumors and that radiation causes prolonged increase of the NQO1 expression may be exploited to preferentially kill tumor cells using ${\beta}$-Lap in combination with radiotherapy.
This experiment was carried out to examine the MBC adsorption on clay minerals by the slurry method. The adsorption of MBC on Kaolinite nearly reached equilibrium after shaking lot 2hrs. but on Bentonite and Zeolite, 10hrs., respectively. The adsorption amount was in the order of Zeolite>Bentonite>Kaolinite, and Na-saturated clay mineral adsorbed more MBC than Mg-saturated clay mineral did. The more the amount of clay minerals, the more MBC was adsorbed by clay minerals, whereas the amount of MBC adsorbed per unit gram of clay minerals reduced and Kd value cf MBC was greatest on the Na-Zeolite. The lower the pH of the water-clay system at each concentration, the more MBC was adsorbed by clay minerals. The pH dependence of the adsorption of MBC may be due to protonation of MBC in acid solution.
This study aimed to isolate and identify L-(+)-lactic acid-producing bacteria from tree barks collected in Thailand and evaluate the potential strain as probiotics. Twelve strains were isolated and characterized phenotypically and genotypically. The strains exhibited a rod-shaped morphology, high-temperature tolerance, and the ability to ferment different sugars into lactic acid. Based on 16S rRNA gene analysis, all strains were identified as belonging to Weizmannia coagulans. Among the isolated strains, BKMTCR2-2 demonstrated exceptional lactic acid production, with 96.41% optical purity, 2.33 g/l of lactic acid production, 1.44 g/g of lactic acid yield (per gram of glucose consumption), and 0.0049 g/l/h of lactic acid productivity. This strain also displayed a wide range of pH tolerance, suggesting suitability for the human gastrointestinal tract and potential probiotic applications. The whole-genome sequence of BKMTCR2-2 was assembled using a hybridization approach that combined long and short reads. The genomic analysis confirmed its identification as W. coagulans and safety assessments revealed its non-pathogenic attribute compared to type strains and commercial probiotic strains. Furthermore, this strain exhibited resilience to acidic and bile conditions, along with the presence of potential probiotic-related genes and metabolic capabilities. These findings suggest that BKMTCR2-2 holds promise as a safe and effective probiotic strain with significant lactic acid production capabilities.
Objective of this research was to determine the effect of application rate of a slow release fertilizer (SRF) in three root media, peatmoss+vermiculite (1:1, v/v; PV), peatmoss+composted rice hall (1:1, v/v; PR), and peatmoss+composted pine bark (1:1, v/v; PB), on growth and nutrient contents of potted chrysanthemum 'Lima Honey'. All media contained polyacrylic acid sodium salt at a rate of $4.5g L^{-1}$. The fresh and dry weights at 43 days after transplanting did not show statistical differences among treatments in each root media. Elevated application rate of SRF increased fresh and dry weights at 80 days after transplanting in PV and PB media, but not in PR medium. Elevated application rates of SRF resulted in the increase of tissue phosphorus content and decrease of tissue Ca, Na, and Zn contents at both 43 and 80 days after transplanting. Elevated application rates of SRF resulted in the decrease of pH and increase of EC and concentrations of ${NO_3}^-$ and ${P_2O_5}^{3-}$, K, Ca, and Mg in the soil solution of PV and PR media. The trends of those in PR media were also similar except ${NO_3}^-$. The differences among treatments in EC at 80 days after transplanting were less significant as compared to those at 43 days after transplanting in three media.

The heartwood of Rhus verniciflua has been known to be effective for lingering intoxication and diabetes mellitus and rheumatoidal arthritis in traditional folk medicine in Korea. We have previously reported that antimutagenic effect of flavonoids derived from the heartwood extract of R. verniciflua, and sulfuretin was the active component. Recently, we have demonstrated that sulfuretin could be an anti-inflammatory principle of the R. verniciflua heartwood partially dependent on cyclooxigenase-inhibitory activity. The present study was undertaken to demonstrate the anti-rheumatoidal arthritis effect of the R. verniciflua heartwood extract, its EtOAc fraction and the main flavonoids, sulfuretin and fustin. All the test samples showed variably significant inhibitory effects on hind paw edema induced by Freund's complete adjuvant reagent (FCA reagent). Sulfuretin treatment with 10 mg/kg (i.p.) for seven days showed the inhibitory effect of 54.2${\pm}$3.0%, Similar trends in RA- and CRP tests, vascular permeability test and trypsin inhibitor test were also found. In addition, no dead mouse was found even when the dose was increased up to 5,000 mg/kg (i.p.). Treatment with 250-1,500 mg/kg on normal rats did not show any marked toxicological significances in the tests of body weight gain, wet weight of organs and hepatic functions. These results suggested that the heartwood of R. verniciflua could be an adequate crude drug for rheumatoidal arthritis with an active component of sulfuretin. The toxicological safety of the heartwood of R, verniciflua is contrasted to known severe allergenic action of the stem bark or its exudate.
This study was conducted to investigate the inhibitory effect of Ecklonia cava (EC) and Eisenia bicyclis (EB) ethanol extract on histamine production in mackerel. Changes in viable cell counts, histamine contents, pH and VBN of mackerel fillet treated with ethanol extracts during 25 days at 4 were measured. Treatments of EC and EB ethanol extract had reduced growth of viable cells by 2 log cycles during storage. Production of histamine was decreased by EC and EB extracts (115 and 96 ppm) when compared to the control at 5 days (384 ppm). The pH of mackerels treated with EC and EB extracts were no different, while the pH of the control increased during storage. Furthermore, the VBN of mackerels treated with EC and EB extracts were significantly decreased when compared to the control. In conclusion, EC and EB extract may reduce scombroid fish poisoning by decreasing histamine production in mackerel during refrigerated storage.
