Synergistic Inhibition by Bacteriocin and Bacteriophage against Staphylococcus aureus

  • Staphylococcus aureus is a representative pathogenic bacterium carefully controlled in the dairy industry because it causes bovine mastitis and thus, can enter the dairy chain. Furthermore, the emergence of multi-drug resistant S. aureus is a big problem. We previously isolated a Lactococcus lactis strain producing a bacteriocin that exhibited strong antimicrobial activity against S. aureus. In this study, we investigated the synergistic inhibition of S. aureus by the bacteriocin and a bacteriophage (SAP84) which is specific to the organism. The bacteriocin (12.5-100 AU/mL) inhibited the growth of S. aureus KCTC 3881 in a dose-dependent manner, as did the bacteriophage SAP84 (0.001-1 MOI; multiplicity of infection). Co-treatment with the bacteriocin (100 AU/mL) and the bacteriophage (0.1 MOI) significantly inhibited the growth of S. aureus compared to each treatment alone (bacteriocin or bacteriophage), indicating the two components showed synergistic inhibition of S. aureus. Therefore, the bacteriocin and bacteriophage combination can be used as a good strategy for controlling pathogenic bacteria.

  • Lactococcus sp. 1112-1 균주가 생산하는 bacteriocin을 정제하고 특성을 조사하였다. CM-sephadex ion exchange column chromatography, Sephadex G-50 gel filtration, SDS-PAGE를 행함으로서 수율 16.2 정제도 123배의 단일물질을 얻었다. 이 물질은 열에 비교적 안정하여 $60^{\circ}C$에서 60분간 열처리 할 때 38의 잔존활성을 유지하였으며 알카리쪽에서는 비교적 불안정하여 pH 8.0에서 48시간 처리시 77의 활성을 소실하였다. 이 물질은 26개의 아미노산 잔기를 이루고 있었다.

  • 동치미로부터 분리된 Latobacillus sp. FF-3의 Bacteriocin 생산에 대한 최적 배양조건과 배양 특성을 조사하였다. Bacteriocin 생산을 위한 최적 생산조건은 MRS(proteose peptone 1%, beef extract 1%, yeast extract 0.5%, glucose 2%, polysorbate 80 0.1%, ammonium citrate 0.2%, sodium acetate 0.5%, magnesium sulfate 0.01%, manganese sulfate 0.005%, dipotassium phosphate 0.2%) 배지를 기본으로 하여 21시간 배양한 결과, 배양온도는 30∼37$^{\circ}C$, 초기 pH는 7.0 으로 확인되었다. 최적 생산배지조성으로 탄소원은 glucose 3%, 질소원은 tryptone 4%, 무기염류는 manganese sulfate 0.005%가 bacteriocin 생성에 최적 조성으로 나타났으며, 최적 배지상에서 항균활성은 최대 484 BUf/mL로 나타났다.

  • Lactrococcus sp. HY449균줄르 M17-glucose broth에 배양하여 배양 상등액으로부터 propanol-actone 침전 ion-exchange chromatography gel-filtration chromatography 및 reverse-phase chroamtography 등을 통하여 비활성 25,600,000 BU/mg 인 순수한 bacteriocin 을 정제하였다. 정제 과정 주에서 ion-exchange chromatography 단계에 서는 35.3%의회수율이 7.3%로 감소하였다. Reverse-Phase chromatography에선 3.3%의 회수율을 보였고 활성도는 413.5배로 증가하였다. Tricine-SDS 전기영도 결과 bacteriocin 은 단일 밴드로 나타났으며, N-말단 아미노산 서열 분석을 수행한 결과 $NH_2$-IIe-Leu-Pro-GIn로 확인되었다. 아미노산조정 분석결과를 바탕으로 분자량을 예측한 결과 본 bacteriocin은 32개의 아미노산으로 이루어져 있으며 분자량은 3.6kDa인 것으로 추정되었다.

Evaluation of Optimum Conditions for Bacteriocin Production from Lactobacillus sp. JB-42 Isolated from Kimchi

  • Bacteriocin-producing microorganism was isolated from Kimchi. The microorganism was identified as a Lactobacillus sp. and named Lactobacillus sp. JB-42. The optimum conditions for the bacteriocin production from the isolated microorganism were evaluated. For the maximum yield of bacteriocin from Lactobacillus sp. JB-42, the cell should be harvested at the early stationary phase and the temperature, pH and NaCl concentration should be $30^{\circ}C$, pH 7 and without the addition of NaCl, respectively. Sucrose, glucose or fructose should be used as a carbon source and organic nitrogen such as tryptone should be used as a nitrogen source for the best yield. The production of bacteriocin was related to the cell growth of Lactobacillus sp. JB-42 indicating the role of Lactobacillus in the Kimchi fermentation process.

  • A bacteriocin was isolated from the supernatant fluid of M17G broth culture of Lactococcus sp. HY 449 strain, which showed strong inhibitory activity against the growth of selective indicator strain, Lactobacillus fermentum IFO3023. When the bacteriocin wasa added to the growing indicator cells or cell suspensions, viable cells and optical density were density were decreased, indicating bacteriolytic mode of action. Electron microscopic observation of indicator cells treated with bacteriocin revealed apparent damages on the cell surface and eventual lysis of cell walls.

  • A bacteriocin-producing lactic acid bacteria was isolated from contaminated milk products, which was identified by using the API50 CH kit as Lactococcus lactis subsp. lactis with reliability of 98%. Fatty and analysia of the cell membrane showed that this strain contained same fatty acids profiles as type strain, Lactococcus lactis subsp. lactis ATCC 19435. The bacteriocin of Lactococcus sp. HY 449 showed relatively wide range of inhibition spectrum against gram positive and some gram negative bacteria such as Escherichia coli and maintained the inhibitory activity between pH2.0 and pH9.0 The thermostability of this bacteriocin was higher in acidic solution than in distilled water and was stable at 60$\circ $C for 1 hour.

Optimization of Bacteriocin ST311LD Production by Enterococcus faecium ST311LD, Isolated from Spoiled Black Olives

  • Bacteriocin ST311LD is approximately 2.3 kDa in size. Low levels of bacteriocin activity were recorded in BHI and M17 broth (800 AU/ml) and in $10\%$ (w/v) soy milk (3,200 AU/ml). No bacteriocin pro-duction was recorded in $10\%$ (w/v) molasses, despite good growth. Optimal levels (12,800 AU/ml) were detected in MRS broth which had been supplemented with tryptone (20.0 g/l), saccharose (5.0 or 10.0 g/l) or vitamin C (1 ppm). Increased potassium levels did not result in higher levels of activity, and glycerol (1.0 g/l) inhibited the production of bacteriocin ST311LD.

Lactobiocin의 피부 염증 및 여드름 저해효과에 관한 연구 (Inhibitory activity of Lactobiocin on the skin inflammation and acnes)

  • The purpose of this study was to evaluate bacteriocin activity against human flora. Lactobiocin, a bacteriocin produced by Lactococcus sp. HY 449, inhibited the growth of Starphylococcus epidermidis, Starphylococcus aureus, Streptoccoccus pyogenes and Propionibacterium acnes. When crude bacteriocin was added to indicator cells during logarithmic growth, the optical density(O.D 650nm) of cells without bacteriocin increase after 5h of incubation. Whereas in the presence of bacteriocin, the O.D of cell suspensions decreased. The similar patterns were observed for absorbance readings at 280 nm and 260 nm. The release of cellular components when cell were treated with Lactobiocin suggests some degree of membrane damage or cell lysis. Scanning electron microscopy of cells following treatments with Lactobiocin in PBS buffer revealed disruptures of cell morphology. These results indicate that bacteriocin appears to cause cell lysis of tested strains. In cytotoxicity on human fibroblast, LD$\_$50/ of Lactobiocin was ca. 50 mg/ml and no change was observed cell proliferation at the same concentration. Any irritation and allergic reaction did not observed when evaluated by human patch test for Lactobiocin.

Lactobacillus sp. Oh-B3로부터 생산되는 박테리오신의 특성 (Characteristics of the Bacteriocin from Lactobacillus sp. Oh-B3.)

  • Lactobacillus sake는 냉장온도(2-4 ℃)이하에서도 성장이 가능한 통성혐기성균으로서, sausage와 진공포장육의 제조에 관여하며, 동양에서는 청주의 발효와 김치의 숙성에도 큰 비중을 차지하는 것으로 알려지고 있다. 그러나 L. sake는 이들 식품의 제조시 과도한 발효의 진행으로 식품을 변질시켜 가치를 저하시키기도 한다. 본 연구에서는 L. sake의 생육을 효과적으로 저지하는 박테리오신을 생산하는 미생물을 김치로부터 분리하여 Lactobacillus sp. Oh-B3로 명명하였다. 최대의 박테리오신을 생산하기 위한 분리균의 배양조건은 초기 pH를 8.0으로 조절한 MRS배지에 접종하여 25℃에서 24시간동안 배양했을 때 최대의 박테리오신을 생산하였다. 분리균의 배양액을 원심분리하여 균체를 제거한 상등액에 ammonium sulfate를 첨가한 뒤, 얻어진 침전물을 투석하고 여과한 뒤, -20℃에 보관하며 조박테리오신 용액으로 사용하였다. 분리균이 생산하는 박테리오신은 pH 2.0-9.0의 넓은 범위에서 비교적 안정한 활성을 나타내었으며, 그람음성균에 대해서는 항균활성을 보이지 않았고 일부 그람양성균에 대해서만 활성을 나타내었다. 조박테리오신 용액에 각각의 단백질 분해효소를 처리한 후 박테리오신의 잔존활성을 측정한 결과 분리균이 생산하는 박테리오신은 대부분의 단백질분해효소에 의해 활성이 제거되었다. 그리고 조박테리오신 용액을 열처리를 한 뒤 잔존활성을 측정한 결과 분리균이 생산하는 박테리오신은 100℃에서도 매우 안정하였으며, autoclave를 하여도 50% 이상의 활성이 유지되었다. 일반적인 박테리오신과 마찬가지로 분리균이 생산하는 박테리오신은 bacteriocidal action을 하는 것으로 관찰되었다.
