Bacteriological Characteristics of Unidentified Vibrio sp., Hemolysin Producer Isolated from Brackish Water -2. Bacteriological Characteristics of Vibrio sp. E10 Similar to Vibrio mimicus- (기수에서 분리된 용혈독소를 생산하는 미분류 Vibrio sp.의 세균학적 특징 -2. Vibrio mimicus와 유사한 Vibrio sp. E10의 세균학적 특성-)

  • A hemolysin producing bacterial strain which belong to Vibrio species was isolated from the Kum River estuary. In the process of identification, the strain did not show characteristics of known Vibrio species; thus, the strain was designated as Vibrio sp, E10 (V. kunsan) tentatively and further identification study was carried out by comparing its bacteriological characteristics. Morphologically Vibrio sp, E10 was comma shaped rod with a polar flagellium. Clear hemolysis zones were observed with the strain against human and sheep blood agar. Hemollytic toxicity was confirmed by strong vascular Permeability and fatal toxicity against mouse was also observed. Therefore the strain was a pathogenic vibrio. Growth conditions for Vibrio sp. E10 were ranged salinity of 0$\~$$4.5\%$, pH of 6.2$\~$9.2, temperature of 14$\~$42$^{\circ}C$, respectively, 16S rDNA partial sequence of Vibrio sp, E10 showed $99\%$ homology with dozens of V. cholerae species including V, cholerae El Tor N16961 and V, snmisnfus ATCC 33653T. This strain belonged to Proteobacteria; gamma subdivision; Vibrionacea: Vibrio. But, among knorn Vibrio species no identical styains were found when using automatic bacteria identification system ($MicroLog^{TM}$system, release 4.0, Biolog Inc., USA) which evaluated the ability of metabolizing 95 kinds of carbon and nitrogen sources. Vibrio sp, E10 showed 18 and 11 different responses as compared to V. mimicus and V, cholerae, respectively.

In Vitro Antagonistic Effects of Bacilli Isolates against Four Soilborne Plant Pathogenic Fungi

  • Twenty isolates of Bacillus spp. obtained from livestock manure composts and cotton-waste composts were tested for in vitro antagonistic effects against soilborne plant pathogenic fungi, Fusarium oxysporum, Phytophthora capsici, Rhizoctonia solani AG-4, and Sclerotinia sclerotiorum. Seven isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of F. oxysporum tested. The bacterial isolate RM43 was the most effective to inhibit the mycelial growth of the fungal isolates. Twelve isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of P. capsici tested. The bacterial isolates M34 and M47 were very effective to inhibit the mycelial growth of the fungal isolates. Thirteen isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of R. solani AG-4 tested. The bacterial isolates M27 and M75 were very effective to inhibit the mycelial growth of the fungal isolates. Fourteen isolates of Bacillus sp. had antagonistic effects on mycelial growth of all the isolates of S. sclerotiorum tested. The bacterial isolates M49 and M75 were very effective to inhibit the mycelial growth of the fungal isolates. The antagonistic effects of most Bacillus spp. isolates against the isolates of the four fungi differed depending on the fungal species and the isolates of each fungus. The bacterial isolates M27 and M75 were the most effective to inhibit the mycelial growth of all four fungi.

Molecular Phylogenetic Diversity and Spatial Distribution of Bacterial Communities in Cooling Stage during Swine Manure Composting

  • Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and subsequent sub-cloning and sequencing were used in this study to analyze the molecular phylogenetic diversity and spatial distribution of bacterial communities in different spatial locations during the cooling stage of composted swine manure. Total microbial DNA was extracted, and bacterial near full-length 16S rRNA genes were subsequently amplified, cloned, RFLP-screened, and sequenced. A total of 420 positive clones were classified by RFLP and near-full-length 16S rDNA sequences. Approximately 48 operational taxonomic units (OTUs) were found among 139 positive clones from the superstratum sample; 26 among 149 were from the middle-level sample and 35 among 132 were from the substrate sample. Thermobifida fusca was common in the superstratum layer of the pile. Some Bacillus spp. were remarkable in the middle-level layer, and Clostridium sp. was dominant in the substrate layer. Among 109 OTUs, 99 displayed homology with those in the GenBank database. Ten OTUs were not closely related to any known species. The superstratum sample had the highest microbial diversity, and different and distinct bacterial communities were detected in the three different layers. This study demonstrated the spatial characteristics of the microbial community distribution in the cooling stage of swine manure compost.

Characterization of Biological Treatment by an Isolated Phenol-Degrading Bacterium (페놀분해세균의 분리 및 생물학적 처리 특성)

  • 20 bacterial strains capable of growing on phenol minimal medium were isolated from soil and wastewater by the enrichment culture technique, and among them, one isolate which was the best in the cell growth was selected and identified as Bacillus sp. SH3 by its characteristics. Strain SH3 could grow with phenol as the sole carbon source up to 15 mM, but did not grow in minimal medium containing above 20 mM of phenol. The optimal conditions of temperature and initial pH for growth and phenol degradation were 30$^{\circ}$C and 7.5, respectively. This strain could grow on various aromatic compounds such as catechol, protocatechuic acid, gentisic acid, o-, m-, p-cresol, benzoic acid, p-hydroxybenzoic acid, anthranilic acid, phenyl acetate and pentachlorophenol, and the growth-limiting log P value of strain SH3 on organic solvents was 3.1. In batch culture, strain SH3 degraded 97% of 10 mM phenol in 48 hours. In continuous culture under the conditions of 20 mM of influent phenol concentration and 0.050 hr$^{-1}$ of dilution rate, the treatment rate of phenol was 94%.

  • Salmonella enterica serovars Gallinarum(SG, causative agent of fowl typhoid) and S Pullorum(SP, causative agent of pullorum disease) are very important bacterial pathogens in poultry industry. They share some common antigenic properties though the characteristics of outbreaks are quite different. To discriminate between SG and SP, we developed two rapid diagnostic methods, allele-specific real-time PCR and 3'-tailed PCR over 2 single nucleotide polymorphisms ($237^{th}\;and\;598^{th}$). In both methods, $237^{th}$ allele was found to be a good target for differential diagnosis, while $598^{th}$ allele produced some non-specific reactions.

  • Applications of probiotics to shrimp ponds were carried out to determine their effects on water quality. Fermented solutions consisting of Bacillus spp. and Nitrosomonas spp. were applied to a 4 ha shrimp (Fenneropenaeus chinensis) pond from July to September, 2000. In the pond treated with probiotics, daily variations of DO and pH, and concentrations of DIN and DIP were lower than those in the ponds without probiotic treatment. Concentration of phytoplankton was less variable and the number of species was more variable in the probiotic-treated pond than those in the control pond. Variation of bacterial numbers and the number of Vibrio spp. were lower in the treated pond than those in the control pond. It is confirmed that the probiotics can be used to improve water quality of the shrimp ponds.

  • A bacterial strain No.71, which produced alkaline protease, was isolated from soil and identified to the genus Bacillus. With the successive mutation, a mutant strain No. M-71, having high alkaline protease productivity, was obtanined from the parental strain No 71. Alkaline protease productivity of mutant strain No. M-71 was about 50 times as much as that of the parental strain No.71. The enzyme preparations showed strong activities toward casein, the optimum pH being 11.0 and the optimum temperature about $55^{\circ}C$.

Biocontrol of Fusarium Crown and Root Rot and Promotion of Growth of Tomato by Paenibacillus Strains Isolated from Soil

  • In this study, bacterial strains were isolated from soils from 30 locations of Samcheok, Gangwon province. Of the isolated strains, seven showed potential plant growth promoting and antagonistic activities. Based on cultural and morphological characterization, and 16S rRNA gene sequencing, these strains were identified as Paenibacillus species. All seven strains produced ammonia, cellulase, hydrocyanic acid, indole-3-acetic acid, protease, phosphatase, and siderophores. They also inhibited the mycelial growth of Fusarium oxysporum f. sp. radicis-lycopersici in vitro. The seven Paenibacillus strains enhanced a range of growth parameters in tomato plants under greenhouse conditions, in comparison with non-inoculated control plants. Notably, treatment of tomato plants with one identified strain, P. polymyxa SC09-21, resulted in 80.0% suppression of fusarium crown and root rot under greenhouse conditions. The plant growth promoting and antifungal activity of P. polymyxa SC09-21 identified in this study highlight its potential suitability as a bioinoculant.

  • A bacterial glutathione S-transferase from Pseudomonas sp. DJ77 has been crystallized. The crystals diffract to at least $2.3\;\AA$ resolution, and belong to the orthorhombic space group $P2_{1}2_{1}2_{1}$, with cell parameters $a=97.4\;\AA,\;b=100.3\;\AA$, and $c=46.0\;\AA$. There is one dimer molecule of pGST per crystallographic asymmetric unit. with the crystal volume per protein mass of $2.34\;\AA^3/dalton$ and a solvent content of about 47% (v/v).

  • The udders of 28 culled dairy cows were removed at slaughter and bacteria isolated. Isolations of major known mammary pathogens comparised Staphylococcus sp. (40.7%), non-identified Gram's negative rods (33.2%) and E. coli (7.4%). In the mixed infection, Staphylococcus sp. was most frequently involved. Drug sensitivity tests were performed on several bacterial species by disc method. These tests indicated that the microorganisms isolated from clinical mastitis of dairy cows were more resistant to antimicrobial agents than the organisrms isolated from milk samples.
