Unrecorded bacterial species belonging to the phylum Actinobacteria originated from Republic of Korea

  • As a subset study for the collection of Korean indigenous prokaryotic species, 62 bacterial strains belonging to the phylum Actinobacteria were isolated from various sources. Each strain showed higher 16S rRNA gene sequence similarity (>98.75%) and formed a robust phylogenetic clade with closest species of the phylum Actinobacteria which were defined with valid names, already. There is no official description on these 62 actinobacterial species in Korea. Consequently, unrecorded 62 species of 25 genera in the 14 families belonging to the order Actinomycetales of the phylum Actinobacteria were found in Korea. Morphological properties, basic biochemical characteristics, isolation source and strain IDs are described in the species descriptions.

Isolation and Characterization of Bacteriophages Infecting Ralstonia solanacearum from Potato Fields

  • Bacterial wilt caused by Ralstonia solanacearum is one of the most devastating diseases in major Solanaceae crops. The pathogen is easily disseminated and survives for many years in plant farming system. Although chemicals are applied to control the disease, they are of limited efficacy and cause several problems. Therefore, the use of phage therapy has been suggested to control the disease as a biological agent. In this study, we discovered bacteriophages lysing diverse Ralstonia isolates from plant and soil samples obtained from the potato cultivated field in Jeju. Three times repeated pickings of plaques resulted in obtaining 173 single phages showing diverse spectrum of host-specificity. With the results, 12 core phages were selected and dendrogram was generated. Genetic diversity of the selected phages was also confirmed by AFLP (Amplified Fragment of Length Polymorphism) fingerprinting. The stability of the phages was investigated in various temperatures and various conditions of pH in vitro. The phages were stable at $16^{\circ}C-44^{\circ}C$ and pH 6-10. Morphological characterization of the phages revealed they were all classified into the Podoviridae, but had diverse head sizes. The results of this research will contribute to control the disease and further researches regarding genetic and molecular aspects will facilitate understanding phage and bacteria interaction.

  • 한국의 토양에서 분리한 균 중 bacterial $\alpha$-amylase에 저해효과가 있는 균주를 분리하여 DMC-47 균주라 명명하였고, 이 균주는 Streptomyces 속의 균임을 확인하였다. 이 균주를 옥수수 전분 배지에서 진탕 배양한 결과 4일후에 최대 저해 효과를 나타내었다. 이 균주가 생성한 저해물질은 bacterial $\alpha$-amylase, pancreatic $\alpha$-amylase, salivary $\alpha$-amylase, glucoamylase에 저해효과를 보였고, $\beta$-amylase 에는 저해효과가 없었다.

A report of 22 unrecorded bacterial species in Korea in the phyla Bacteroidetes and Rhodothermaeota

  • A total of 22 bacterial strains belonging to the phylum Bacteroidetes were isolated primarily from aquatic environments such as seawater, freshwater, lagoon and tidal flat. One of these 22 strains was isolated from ginseng soil. Phylogenetic analyses based on 16S rRNA gene sequences revealed that 21 strains showed the high sequence similarities(${\geq}98.7%$) to the closest type strains and formed robust phylogenetic clades with closely related species in the phylum Bacteroidetes. One strain, which had been previously classified as Balneola vulgaris in the phylum Bacteroidetes, was identified as a member of the newly described phylum Rhodothermaeota. These strains had not been previously reported in Korea. Here, we report 21 species of 13 genera in the phylum Bacteroidetes and one species in the phylum Rhodothermaeota which were not reported in Korea. Morphological, biochemical, and physiological characteristics, isolation sources, and NIBR deposit numbers are described in the species descriptions.

Isolation and Structural Characterization of an Oligosaccharide Produced by Bacillus subtilis in a Maltose-Containing Medium

  • Among 116 bacterial strains isolated from Korean fermented foods, one strain (SS-76) was selected for producing new oligosaccharides in a basal medium containing maltose as the sole source of carbon. Upon morphological characterization using scanning electron microscopy, the cells of strain SS-76 appeared rod-shaped; subsequent 16S rRNA gene sequence analysis revealed that strain SS-76 was phylogenetically close to Bacillus subtilis. The main oligosaccharide fraction B extracted from the culture supernatant of B. subtilis SS-76 was purified by high performance liquid chromatography. Subsequent structural analysis revealed that this oligosaccharide consisted only of glucose, and methylation analysis indicated similar proportions of glucopyranosides in the 6-linkage, 4-linkage, and non-reducing terminal positions. Matrix-assisted laser-induced/ionization time-of-flight/mass spectrometry and electrospray ionization-based liquid chromatography-mass spectrometry/mass spectrometry analyses suggested that this oligosaccharide consisted of a trisaccharide unit with 1,6- and 1,4-glycosidic linkages. The anomeric signals in the $^1H$-nuclear magnetic resonance spectrum corresponded to ${\alpha}$-anomeric configurations, and the trisaccharide was finally identified as panose (${\alpha}$-D-glucopyranosyl-1,6-${\alpha}$-D-glucopyranosyl-1,4-D-glucose). These results suggest that B. subtilis SS-76 converts maltose into panose; strain SS-76 may thus find industrial application in the production of panose.

Isolation, Identification, and Characterization of Bacillus strains from the Traditional Korean Soybean-fermented Food, Chungkookjang

  • A total of 45 bacterial strains were isolated from the traditional Korean soybean-fermented food, Chungkookjang. Among these strains, seven strains were selected and identified based on morphological, physiological, and biochemical characteristics, as well as phylogenetic analysis using 16S rDNA sequences. All strains were Gram-positive, aerobic, motile, oxidase-positive, rod-shaped, and endospore-forming bacteria, and produced extracellular enzymes such as amylase, cellulase, lipase, protease, and xylanase. The isolates were grown in the presence of 0-11% (w/v) NaCl. Growth was optimal at pH 6-9 and at temperatures of $30-45^{\circ}C$. According to VITEK automicrobic system tests and supplementary tests, the isolates were similar to several species of the genus Bacillus. The phylogenetic analysis of seven bacterial strains based on comparisons of 16S rDNA sequences, revealed that the strains were closely related to Bacillus species. The identification of strains that produced surfactin was also carried out, based on PCR screening of the sfp gene. Among the seven isolated strains, six yielded a surfactin-positive result with PCR.

A report of 12 unrecorded prokaryotic species isolated from gastrointestinal tracts and feces of various endangered animals in Korea

  • In 2016 and 2017, as part of a comprehensive investigation to identify the prokaryotic species in Korea, a total of 12 bacterial strains were isolated from the gastrointestinal tract and/or fecal samples of four endangered species, including reptile, bird, and marine and terrestrial mammals. Phylogenetic analysis with the 16S rRNA gene sequence was used to assign these strains to the phyla, Firmicutes, Actinobacteria or Proteobacteria. Furthermore, most of the strains Firmicutes belonged to the order Lactobacillales. Interestingly, 12 of the isolated strains have not been previously reported from the Korean Peninsula. Also, based on their high 16S rRNA gene sequence similarities(>98.7%) and formation of strong monophyletic clades with the closest type species, each isolated strain of isolates was assigned to an independent, predefined bacterial species. Gram-stain reaction, colony and cell morphology, biochemical characteristics, isolation source, and NIBR IDs are described in the species description section.

전분폐수의 생물학적 처리에 관한 연구 1. 소맥 전분포수 처리균의 분리와 처리효과 (Biological Treatment of Starch Waste Part 1. Isolation of Wheat Starch Waste Decomposing Organisms and Their Efficiency on Waste Treatment)

  • Activated sluge를 폐수처리와 사료로 이용하기 위하여 고구마와 소맥 전분폐수에서 채집한 32종의 균원을 선별한 결과 2종의 우수한 균원을 확인하고 N-9균원으로부터 7종의 세균J-3으로부터 6종의 세균을 분리하여 최종 선별 결과 3종의 세균 J-3-1, J-3-2, J-9-2를 선별하였다. 1. N-9의 혼합 배양의 경우 J-3보다는 floc형성능이 좋았으며 N-9로부터 Eugelena속의 원생동물 1종이 J-3에서는 Bodo 속의 원생동물 1종이 동정되었다. 2. 3종의 선별된 균에 대한 형태학적 생리학적 특성을 검사했다. 3. Sample N-9의 혼합균 배양에 의해 99%의 폐수의 B.O.D가 제거되었으며 세균 단독 배양일 경우 92% 이상의 B.O.D.가 제거되었다. 4. 원료소맥 전분폐수의 B.O.D가 8,000 ppm이하에서도 B.B.D. 제거율과 sludge에서 생산된 protein 량은 폐수의 B.O.D. 와 처리하는 균에 따라 상이하였다.

Isolation and Characterization of an Antifungal and Plant Growth-Promoting Microbe

  • Fungal diseases including anthracnose, stem rot, blight, wilting, and root rot of crops are caused by phytopathogens such as Colletotrichum species, Sclerotinia sclerotiorum, Phytophthora species, and Fusarium oxysporum and F. solani which threaten the production of chili pepper. In this study, to identify biological control agents (BCAs) of phytopathogenic fungi, potentially useful Bacillus species were isolated from the field soils. We screened out five Bacillus strains with antagonistic capacity that are efficiently inhibiting the growth of phytopathogenic fungi. Bacillus species were characterized by the production of extracellular enzymes, siderophores, and indole-3-acetic acid (IAA). Furthermore, the influence of bacterial strains on the plant growth promoting activity and seedling vigor index were assessed using Brassica juncea as a model plant. Inoculation with Bacillus subtilis SRCM 121379 significantly increased the length of B. juncea shoots and roots by 45.6% and 52.0%, respectively. Among the bacterial isolates, Bacillus subtilis SRCM 121379 showed the superior enzyme activities, antagonistic capacity and plant growth promoting effects. Based on the experimental results, Bacillus subtilis SRCM 121379 (GenBank accession no. NR027552) was finally selected as a BCA candidate.

Enteric Pathogens in Pediatric Patients with Acute Gastroenteritis in Gyeongju, Korea: A Retrospective Study for 7 Years in a Regional Hospital

  • Acute diarrhea is a global health problem that causes high morbidity and mortality in children. Notably, enteric pathogen co-infections have been suggested to play an important role in gastroenteritis. In this study, we analyzed 1150 stool specimens of patients who visited the pediatric ward of Dongguk University Hospital in Gyeongju province from January 2011 to December 2017. The average isolation rate of potential stool pathogens over 7 years was 37.3% (429/1150), and coinfections were observed in 51 patients (51/429; 11.9%). In the 51 co-infection cases, the most frequent type of co-infection was found to be that of virus-bacteria (33/51). The most frequently detected bacterial pathogen among the co-infected cases was Clostridium spp. (22/51), out of which Clostridium perfringens was found to be the main pathogen (16/22; 72.7%). Escherichia coli spp. were the second most common bacterial pathogens found in 12 cases (12/51; 23.5%), with 10 cases of E. coli EPEC. Furthermore, the most frequently implicated viral pathogen among the co-infected cases was norovirus (16/51), followed by rotavirus (12/51).
