당근 지상부 추출물 유래 항염 및 항균 활성 성분 (Anti-inflammatory and Anti-bacterial Constituents from the Extracts of Daucus carota var. sativa Aerial Parts)

  • 본 연구에서는 당근 지상부 추출물 및 용매 분획물의 항염, 항균 활성을 확인하고 유효성분을 분리하여 화학구조를 동정하였다. 당근 지상부 에탄올 추출물 및 용매 분획물의 항염 활성을 측정하기 위해 LPS로 자극된 RAW 264.7 세포를 이용하여 NO 생성 억제 활성을 확인하였다. 그 중 에틸아세테이트 분획물이 NO의 생성을 농도 의존적으로 감소시키는 효과가 나타났고, 추가적인 항염 기전 연구를 위해 에틸아세테이트 분획물에 대해 $PGE_2$, 전염증성 사이토카인의 생성량 및 iNOS, COX-2 단백질의 발현량을 측정하였다. 그 결과, 에틸아세테이트 분획물이 $PGE_2$, IL-$1{\beta}$, IL-6의 생성을 감소시키고, iNOS, COX-2 단백질의 발현도 억제 시키는 효과가 있음을 확인하였다. 또한 표피포도상구균과 여드름균을 이용한 활성 실험 결과, 헥산 및 에틸아세테이트 분획물에서 항균 활성이 나타났다. 이와 같은 결과를 바탕으로 에틸아세테이트 분획물에 대해 컬럼 크로마토그래피를 수행하여 3개의 화합물을 분리하였으며, $^1H$$^{13}C$ NMR 데이터 분석과 문헌을 통하여 화학구조를 동정하였다; diosmetin (1), disomin (2), cynaroside (3). 분리된 화합물 1-3에 대해 항염 및 항균 활성을 측정하였으며, 그 결과 화합물 1-3 모두 NO의 생성을 저해시키는 효과가 있음을 확인하였다. 또한 화합물 3은 여드름균에 대해 항균 활성이 우수하였다. 이상의 연구 결과를 바탕으로 당근 지상부 추출물을 이용한 항염 및 항균 효과를 갖는 천연 화장품 소재로의 개발이 가능할 것이라 사료된다.

Isolation and Characterization of Microbial Strains with Hydrolytic Enzyme Profile from Clay Minerals

  • A total of 262 bacterial strains were isolated from clay minerals, bentonite and zeolite, in Gyeongsangbukdo, Republic of Korea, and their hydrolytic enzyme activities were analyzed. Most of the isolated strains belonged to Micrococcales and Bacillales order. Of strains, 96 strains produced α-amylase activity, 42 strains showed cellulase activity, 111 strains had pectinase activity, and 70 strains showed protease activity. Among them, 177 isolates exhibited one or more of the hydrolytic enzyme activities and in particular Bacillus cereus MBLB1321, B. albus MBLB1326 and KIGAM017, B. mobilis MBLB1328, MBLB1329 and MBLB1330 showed all of the enzyme activities. These results demonstrate the diversity of functional Bacillus species in clay minerals as vital sources for the discovery of industrially valuable hydrolytic enzymes, which have a great commercial prospect in various bio-industrial applications.

환경시료에 존재하는 미생물 배양을 위한 filter plate micro trap의 개발 (Filter Plate Micro Trap as a Device for in situ Cultivation for Environmental Microorganisms)

  • 난배양성 미생물의 배양과 보다 다양한 미생물의 분리를 위하여 Filter plate microbial trap (FPMT)을 개발하였고, 이 장치를 사용하여 Greenland의 토양에서 세균을 분리하였다. FPMT는 배지와 하단부의 membrane filter(0.45 ${\mu}m$ pore size)로 구성되어 있다. 시료에 포함된 세균과 화학물질이 FPMT 내로 이동하고, 세균이 증식하는 원리이다. 그 결과, FPMT를 이용한 새로운 in situ 배양방법을 통해 통상적인 Petri dish 배양방법보다 더 많은 종과 기존의 방법으로는 배양할 수 없는 세균을 분리하였다. 이 장치는 앞으로 다양한 환경에서 난배양성 미생물을 분리하는데 유용함을 확인하였다.

전만해안지역의 비브리오 패혈증균(Vibrio vulnificus) 분포 (Distribution of Vibrio vulnificus in Chonnam Coastal Area)

  • Vibrio vulnificus, a normal bacterial inhabitant of estuaries, is of concern because it can be a potent human pathogen, a causing septicemia, wound infections and gastrointestinal disease in susceptible host. In this survey, total 431 samples were obtained from different sites of the Chonman coastal area during the periods from Mar. 1997 to Feb. 1998. Vibrio vulnificus was isolated from the middle of May to the begining of November of 1997 in Chonman coastal area, as the seawater temperature was at 20$^{\circ}C$ and 15$^{\circ}C$, respectively and was rapidly increased to above 40% from July to September. The isolation rates of V. vulnificus from sediment, seawater, raw seafoods and aquarium water were 52.1%, 49.1%, 32.5%, and 27.3%, respectively and isolation rate was highest in oyster among various collected samples. V.vulnificus was also isolated from 73.1%(38/52) of sampling sites of Chonman coastal area.

  • 경기도 일대의 가금류 공장부근 토양로부터 단백질 분해활성이 높은 균주를 분리한 후 이것들 가운데 Keratinolytic protease 생산성이 우수한 균주는 KP-364를 선별하여 효소생산을 위한 최적 배양조건을 검토하였다. 선별된 KP-364 는 형태학적 생화학적 특성을 종합한 결과 Pseudomonas sp 로 동정되었으며 편의상 Pseudomonas sp. KP-364로 명명하였다. 효소생산을 위한 최적 배지조성은 Keratin 0.1% glucose 2.0% soybeam meal 0.5% NaNO3 0.5% KCI 0.2%이었고 초기 pH 6.5 배양온도 $37^{\circ}C$, 48시간진탕 배양할때 효소생산이 가장 우수하였으며 이때의 효소의 역가는 1,270 U/ml/hr이었다.

Isolation and Characterization of an Extremely Thermophilic Sulfur-metabolizing Bacterium from a Deep-sea Hydrothermal Vent System

  • A water sample was taken from a black smoker chimney of a deep-sea hydrothermal vent by using an unmanned submersible "Dolphin 3K". The temperature of the hydrothermal fluid from the black smoker was $276^{\circ}C$. After isolation by repeated serial dilutions, An extremely thermophilic bacterial strain was selected. The strain designated as DT1331, was an anaerobic, non-motile, coccoid shaped bacterium with about 0.5 to $1.0\;\mu\textrm{m}$ in diameter. The strain DT1331 could grow up to $93^{\circ}C$, but the optimum temperature of this strain was $80^{\circ}C$. The growth occurred in the pH range of 4.5 to 8.5 and the optimum pH was 6.0. The strain DT1331 required 1% to 5% NaCl for growth and cell lysis was observed below 1% NaCl concentration. The bacterium could grow on polypeptides such as tryptone, peptone, soytone and on proteins such as casein or gelatin. However, no growth was observed on single amino acids, sugar and organic acids. Hydrogen gas was detected slightly during growth. This bacterium obligately required elemental sulfur and hydrogen sulfide gas was produced during growth.

내열성 Xylanase를 생산하는 Paenibacillus sp. DG-22 균주의 분리 및 효소 특성 (Isolation and Characterization of Thermostable Xylanase-producing Paenibacillus sp. DG-22.)

  • 경주지역에 있는 목재 저장소의 토양으로부터 내별성 xylanase를 생산하는 호열성 세균인 DG-22 균주가 분리되었다. 형태적, 생화학적, 계통학적 연구에 근거하여 이 분리균은 Paenibacillus 속하는 종으로 판명되었다. 이 균주에서 xylanase의 생산은 성장배지에 xylan을 첨가함으로서 유도되었고 glucose또는 xylose에 의해서 억제되었다. Cellulase 활성은 탐지되지 많았다. 효소 활성을 위한 최적온도와 pH는 각각 $80^{\circ}C$와 5.0-5.5이었다. 이 조효소는 $60^{\circ}C$매서 안정하였고 $70^{\circ}C$에서는 2시간 추에도 60%의 활성을 유지하였다. 배양상등액의 zymogram 분석을 통해 22 kDa과 30 kDa 크기의 xylanase활성 band를 확인하였다

Naphthalene 분해균주 Alcaligenes sp. A111의 분리 및 특성 (Isolation and Characterization of a Naphthalene-Degrading Strain,Alcaligenes sp,A111)

  • A bacterial strain which formed a distinct colony on agar plate containing naphthalene as a vapor phase and grew well ina liquid minimal medium was isolated and identified as Alcaligenes sp. A111. Optimum temperature and pH for the cultivation of Alcaligenes sp. A111 were 30$\cir$C and 7.0, respectively. Cell growth increased dramatically from 12 hours after inoculation and revealed a stationary phase at about 48 hours. Relative growth rate ($\mu$')increased hyperbolically depending on the conceration of naphthalene up to 500 ppm and reached to the maximum value pf 2.8/day, but $\mu$' didn't change within a range of 500~4000 ppm naphthalene. NH$_{4}$Cl or NH$_{4}$NO$_{3}$ was preferrd as a nitrogen source and a P : N ratio by weight og 6 : 1 was favorable to cell growth. Alcaligenes sp. A111 utilized the intermediates of degradation of naphthalene and showed tolerance to benzene, toluene, and octane. therefore, it is suggested that Alcaligenes sp. A111 could be effectively used for the biological treatment of wastewater containing naphthalene in the presence of some aromatic compounds.

Therapeutic Effects of Bacteriophages Against Salmonella gallinarum Infection in Chickens

  • In this study the isolation and characterization of three bacteriophages (ST4, L13, and SG3) infecting Salmonella gallinarum were carried out. They were further tested for their in vivo efficacy in phage therapy. All three phages belong to the Siphoviridae family with isometric heads and non-contractile tails. They have a broad host range among serovars of Salmonella enterica. The burst sizes were observed to be 1670, 80, and 28 for ST4, L13, and SG3, respectively. The in vivo efficacy of the phages was tested in chickens. Layer chickens were challenged with S. gallinarum, whereas contact chickens were cohabited without direct challenge. Each bacteriophage was orally inoculated in the form of feed additives. Mortality was observed and S. gallinarum was periodically re-isolated from the livers, spleens, and cecums of the chickens. Bacterial re-isolation from the organs and mortality decreased significantly in both challenged and contact chickens treated with the bacteriophages compared with untreated chickens serving as the control. The three bacteriophages may be effective alternatives to antibiotics for the control of fowl typhoid disease in chickens.

The Current Status and Prospect of Sericultural Byproduct Industry in China

  • Sericulture is a traditional agro-industry, which involves mulberry cultivation and silkworm rearing, has made great contributes to the human civilization. With the development of national economy and modem technology, mulberry and silkworm are being used to develop products with functionality besides the traditional cocoon production in China. In this paper, we brief the current developing situation of sericultural byproducts with functionality in the following aspects. (1) Functional products from silkworm larvae: silkworm powder, white muscardine silkworm, isolation and purification of anti-bacterial proteins from the larvae and production of medically valuable substances by Bombyx mori nuclear polyhedrosis virus (BmNPV) vector. (2) Utilization of silkworm feces: for pillow and for isolation of chlorophyll etc. (3) Production of valuable Chinese traditional medicine like Cordyceps sinensis with pupae, functional utilization of pupa protein and chitin. (4) Silk as additives to cosmetics, silk food and medical materials. (5) Functional utilization of mulberry: cultivation of edible fungus on mulberry shoots as medium, mulberry fruit drinks, mulberry tea, etc. The prospect of sericultural byproduct industry in China is also discussed.
