A Proposal on the New Genotyping of Sacbrood viruses for the Definition of korean Sacbrood Virus (kSBV) (한국형 낭충봉아부패병 바이러스(kSBV)의 정의를 위한 새로운 SBV 유전자형의 제안)

  • korean Sacbrood Virus (kSBV) was firstly found since 2008 in Korea, and it might be a main reason why 99% of populations belonged to Apis cerana in Korea were disappeared now. In this study, full length, reported DNA sequences of 32 Sacbrood Viruses (SBVs) were analysed based on alignments using nucleotides and/or deduced amino acid sequeces. In this analysis, variable deletions were found that are located around 2,100 bases in each CDS of SBVs. The genotyping depended on these deletions might be related with infection-patterns by these pathogens. In addition, it is not escaped our notice that the genotyping we have proposed immediately suggests a possible origin of kSBV for the quarantine and protection against further invasion.

  • This paper reviews a current view regarding the association between periodontitis and atherosclerotic cardiovascular diseases (ACVD). Many evidences have suggested that there exist biological mechanisms by which periodontitis can lead to ACVD. Periodontal infection can lead to direct bacterial invasion into endothelial tissues through the blood stream, then the bacteria can activate the host inflammatory response followed by atheroma formation, maturation and exacerbation. Also, chronic periodontal infections may indirectly induce endothelial activation or dysfunction through a state of systemic inflammation as evidenced by elevated plasma acute proteins, IL-6 and fibrinogen as well. There is moderate evidence that periodontal treatment can reduce systemic inflammation and improvement of both clinical surrogate markers. But there is no periodontal intervention study available on primary ACVD prevention. There is consistent and strong epidemiologic evidence, including in vitro, animal and clinical studies, that periodontitis imparts increased risk for future ACVD. However, evidences from intervention trials to date are not sufficient to confirm the multi directional causality of periodontitis in ACVD etiology. Well-designed intervention trials on the impact of periodontal treatment on the prevention of ACVD outcomes are needed.

  • Recently, onset of bacterial disease, especially infiltration of chronic respiratory diseases have been increasing at high rate. The main cause of these diseases are originated from inappropriate sanitary management and slow progress in introducing system and herd free system, which are the base of productivity improvement in bigger pig management, Methods for the prevention and removal of these diseases, are divided into four categories. The first category includes prevention and removal of infectious diseases by organizing strict animal quarantine, enforcing vaccination, and legal regulation and disposal at the time of outbreak. The second category includes improvement of production systems. This purpose can be achieved by discontinuing the open herd system, which brings on the invasion of foreign pathogens and replacing the system by the closed type of farm. To Continue eradication process step-wisely by performing the all-in all-out system at every pig pen or herd level is also effective for this purpose. The third category includes introduction of herd free system can be achieved by repopulating conventional pigs with SPF pigs. If these means are unrealizable from economic or technical viewpoint, medicated early weaning system is perfomed. This system consists of disinfection of pigsty at pre-postpartum stage, application of drugs to pregnant sows and newborn piglets in accordance with the medication program, and early weaning and rearing of newborn piglets in isolation. The fourth category is sanital leading by veterinarians, aimed to eliminating incentives for diseases through diagnoses. Their main activities include periodical monitoring and improvement leading on each responsible subject.

Visualization of periodontopathic bacteria within crevicular epithelial cells with fluorescence in situ hybridization (형광제자리부합법을 이용한 치은열구세포 내의 치주염 유발 세균의 관찰)

  • Purpose: Periodontal pathogens can invade the host tissue. Morphologic studies have revealed bacteria within the pocket epithelium, gingival connective tissues, alveolar bone, and oral epithelium. The objective of this study was to visualize and evaluate presence of Porphyromonas gingivalis and Tannerella forsythia in crevicular epithelial cells of periodontally healthy subjects and chronic periodontitis patients. Materials and Methods: A total of 666 crevicular epithelial cells in the samples obtained from 27 chronic periodontitis patients and 9 healthy volunteers were examined. Specific probes for P. gingivalis and T. forsythia and a universal probe for detection of all eubacteria targeting 168 rRNA for fluorescence in situ hybridization was used in conjunction with confocal laser scanning microscopy. Results: 98.99% of sulcular epithelial cells from healthy volunteers and 84.40% of pocket epithelial cells from periodontitis patients were found to harbor bacteria. P. gingivalis and T. forsythia were discovered more often in crevicular epithelial cells from periodontitis patients. Conclusion: P. gingivalis and T. forsythia can invade crevicular epithelial cells and intracellular bacteria may act as a source of bacteria for persistent infection.

Host Cell Nuclear Localization of Shigella flexneri Effector OspF Is Facilitated by SUMOylation

  • When Shigella infect host cells, various effecter molecules are delivered into the cytoplasm of the host cell through the type III secretion system (TTSS) to facilitate their invasion process and control the host immune responses. Among these effectors, the S. flexneri effector OspF dephosphorylates mitogen-activated protein kinases and translocates itself to the nucleus, thus preventing histone H3 modification to regulate expression of proinflammatory cytokines. Despite the critical role of OspF, the mechanism by which it localizes in the nucleus has remained to be elucidated. In the present study, we identified a potential small ubiquitin-related modifier (SUMO) modification site within OspF and we demonstrated that Shigella TTSS effector OspF is conjugated with SUMO in the host cell and this modification mediates the nuclear translocation of OspF. Our results show a bacterial virulence factor can exploit host post-translational machinery to execute its intracellular trafficking.

β-Sitosterol Contributes in the Resistance to Invasion and Survival of Brucella abortus 544 within RAW264.7 Cells, and Cytokine Production with Reduced Susceptibility to Infection in BALB/c Mice

  • We previously identified β-sitosterol (BS) as one of the most abundant compounds found in Korean red ginseng oil. BS is a widely prevalent vegetable-derived phytosterol with many known health benefits. Here, we investigated the efficacy of BS against Brucella (B.) abortus infection. BS showed no effect on bacterial growth but attenuated internalization, intracellular survival and MAPKs-linked intracellular signaling in RAW264.7 cells. BS treatment in cells is also associated with increased nitrite concentration during infection at 24 h. Slightly enhanced resistance to B. abortus infection was observed in mice orally given BS, which could be mediated by induced production of proinflammatory cytokines. Taken together, our study demonstrates the contribution of BS treatment against B. abortus infection although further investigation is encouraged to maximize its beneficial effects against intracellular infection.

Oridonin Suppresses Proliferation of Human Ovarian Cancer Cells via Blockage of mTOR Signaling

  • Oridonin, an ent-kaurane diterpenoid compound isolated from the traditional Chinese herb Rabdosia rubescens, has shown various pharmacological and physiological effects such as anti-tumor, anti-bacterial, and anti-inflammatory properties. However, the effect of oridonin on human ovarian cancer cell lines has not been determined. In this study, we demonstrated that oridonin inhibited ovarian cancer cell proliferation, migration and invasion in a dose-dependent manner. Furthermore, we showed oridonin inhibited tumor growth of ovarian cancer cells (SKOV3) in vivo. We then assessed mechanisms and found that oridonin specifically abrogated the phosphorylation/activation of mTOR signaling. In summary, our results indicate that oridonin is a potential inhibitor of ovarian cancer by blocking the mTOR signaling pathway.

  • Pyogenic liver abscess with metastatic septic complications is a rare and serious infectious disease if not treated properly. Pyogenic liver abscesses are caused by bacterial, fungal, or parasitic organisms. Escherichia coli used to be the predominant causative agent, but Klebsiella pneumoniae emerged as a major cause in the 1990s. Liver abscesses are caused by hepatic invasion via many routes, such as, the biliary tree, portal vein, hepatic artery, direct extension, or penetrating trauma. Furthermore, diabetes mellitus and malignant conditions are established important risk factors of K. pneumoniae liver abscesses and of septic metastasis, and several recent studies have asserted that K. pneumoniae liver abscess might be a presentation of occult or silent colon cancer. We report a case of K. pneumoniae liver abscess, metastatic septic pulmonary embolism, and endophthalmitis associated with diabetes and advanced gastric cancer.

Isolation of Anaerobic Bacteria from Clinical Specimens (임상검사물에서의 혐기성세균 분리)

  • Anaerobic bacteria are the major residents of human skin and mucous membrane. The importance, as opportunistic pathogens, of anaerobic bacteria are well recognized because of more population. with decreased defense against bacterial invasion due to chemotherapy, rediation therapy, extensive surgical operation etc. are dealt at hospitals. An analysis of the anaerobe isolation results at Yonsei University Medical Center during the January 1974-May 1975 period was made and the following results were obtained. 1) From 118 patients 146 strains of anaerobes were isolated. Among these 81.3% were nonsporforming anaerobes. Most frequently isolated anaerobes were Pc. asaccharolyticus, Ps. anaerobius, Ps. intermedius, B. fragilis and Cl. perfringens. 2) Anaerobes were frequently isolated from wound, female genital, intraabdominal, and pleuropulmonary specimens. Fewer anaerobes were isolated from blood, spinal fluid and liver specimens. 3) The ratio of anaerobe isolation to total bacteria isolation were; liver 66.7%, intraabdominal 33.3%, pleuropulmonary 28.9%, spinal fluid 5.0% and blood 4.2%. 4) Among the 118 anaerobe isolated patients, 48.3% yielded anaerobes only and rest of them yielded anaerobes together with aerobes. 5) Most of the gram-positive anaerobes were susceptible to the antibiotics tested. Exception was to tetracycline to which appreciable number showed resistance. It was noteworthy that only 48% of B. fragilis was susceptible to tetracycline.

Overexpression of rice NAC transcription factor OsNAC58 on increased resistance to bacterial leaf blight (전사인자 OsNAC58 과발현을 통한 벼 흰잎마름병 저항성 증진 벼)

  • Bacterial blight in rice caused by Xanthomonas oryzae pv. oryzae (Xoo) greatly reduces the growth and productivity of this important food crop. Therefore, we sought to increase the resistance of rice to bacterial blight by using a NAC (NAM, ATAF, and CUC) transcription factor, one of the plant-specific transcription factors that is known to be involved in biotic/abiotic stress resistance. By isolating the OsNAC58 gene from rice and analyzing its biological functions related to Xoo resistance, phylogenetic analysis showed that OsNAC58 belongs to group III. To investigate the biological relationship between bacterial leaf blight (BLB) and OsNAC58 in rice, we constructed a vector for overexpression in rice and generated transgenic rice. The expression analysis resulting from use of RT-PCR showed that OsNAC58-overexpressed transgenic rice exhibited higher levels of OsNAC58 expression than wild types. Further, subcellular localization analysis using rice protoplasts showed that the 35S/OsNAC58-SmGFP fusion protein was localized in the nuclei. Thirteen OsNAC58-overexpressed transgenic rice lines, with high expression levels of OsNAC58, showed more resistant to Xoo than did the wild types. Together, these results suggest that the OsNAC58 gene of rice regulates the rice disease resistance mechanism in the nucleus upon invasion of the rice bacterial blight pathogen Xoo.
