MicroTom is a miniature tomato plants with various properties that make it as a model system for experiments in plant molecular biology. To extend its utility as a model plant to study a plant - bacterial wilt system, we investigated the potential of the MicroTom as a host plant of bacterial wilt caused by Ralstonia solanacearum. We compared the disease progress on standard tomato and MicroTom by two inoculation methods, root dipping and soil drenching, using a race 1 strain GMI1000. Both methods caused the severe wilting on MicroTom comparable to commercial tomato plant, although initial disease development was faster in root dipping. From the diseased MicroTom plants, the same bacteria were successfully reisolated using semiselective media to fulfill Koch's postulates. Race specific and isolate specific virulence were investigated by root dipping with 10 isolates of R. solanacearum isolated from tomato and potato plants. All of the tested isolates caused the typical wilt symptom on MicroTom. Disease severities by isolates of race 3 was below 50 % until 15 days after inoculation, while those by isolates of race 1 reached over 50% to death until 15 days. This result suggested that MicroTom can be a model host plant to study R. solanacearum - plant interaction.
The global warming by increased $CO_2$ will effect of plant pathogenic microorganisms and resistance of host plants, and it is expected to affect host-pathogen interactions. This study used Capsicum annuum L. and Xanthomonas euvesicatoria, a pathogenic bacteria of pepper, to investigate interactions between hosts and pathogens in a complex environment with increasedcultivation temperature and drought stress. As a result, the bacterial spot disease of C. annuum L. caused by X. euvesicatoria was $35^{\circ}C$ higher than $25^{\circ}C$. In addition, the effect on water potential on bacterial spot disease was much greater water potential -150 kPa than -30 kPa. The disease progress and severity higher than water potential -30 kPa. This result will useful for understanding interaction with red pepper and X. euvesicatoria under the complex environment with increased cultivation temperature and in water potential -150 kPa drought stress in the future.
Purpose: To explore differences in the subgingival microbiome according to the presence of periodontitis and/or type 2 diabetes mellitus (T2D), a metagenomic sequencing analysis of the subgingival microbiome was performed. Methods: Twelve participants were divided into 4 groups based on their health conditions (periodontitis, T2D, T2D complicated with periodontitis, and generally healthy). Subgingival plaque was collected for metagenomic sequencing, and gingival crevicular fluids were collected to analyze the concentrations of short-chain fatty acids. Results: The shifts in the subgingival flora from the healthy to periodontitis states were less prominent in T2D subjects than in subjects without T2D. The pentose and glucuronate interconversion, fructose and mannose metabolism, and galactose metabolism pathways were enriched in the periodontitis state, while the phosphotransferase system, lipopolysaccharide (LPS) and peptidoglycan biosynthesis, bacterial secretion system, sulfur metabolism, and glycolysis pathways were enriched in the T2D state. Multiple genes whose expression was upregulated from the red and orange complex bacterial genomes were associated with bacterial biofilm formation and pathogenicity. The concentrations of propionic acid and butyric acid were significantly higher in subjects with periodontitis, with or without T2D, than in healthy subjects. Conclusions: T2D patients are more susceptible to the presence of periodontal pathogens and have a higher risk of developing periodontitis. The pentose and glucuronate interconversion, fructose and mannose metabolism, galactose metabolism, and glycolysis pathways may represent the potential microbial functional association between periodontitis and T2D, and butyric acid may play an important role in the interaction between these 2 diseases. The enrichment of the LPS and peptidoglycan biosynthesis, bacterial secretion system, and sulfur metabolism pathways may cause T2D patients to be more susceptible to periodontitis.
Serotonin (5-HT; 5-hydroxytryptamine) exerts multiple effects on central nervous system as well as behaviors such as mood and appetite. The signaling of serotonin is mediated by 7 families of serotonin receptors, designated 5-HT$_1$ to 5-HT$_{7}$. Six families of this receptor are G-protein coupled 7TM receptors, and the third intracellular loop of these receptors is proposed to interact with specific types of G-proteins. To investigate the specific interaction between the third intracellular loop of 5-HT$_{6}$ with G$\square$s, we have constructed a chimera protein that represent the third intracellular loop of 5-HT$_{6}$ within a leucine zipper motifs, In addition an alpha subunit of human G-protein that interact with 5-HT$_{6}$ was cloned into a bacterial expression vector. The two proteins were expressed in E. coli and purified in homogeneity. The interaction of the prepared proteins was examined by ELISA assay. The affinity between the two proteins and effect of insertion mutations were discussed.ussed.d.
Dermcidin is a human antibiotic peptide that is secreted by the sweat glands and has no homology to other known antimicrobial peptides. As an initial step toward understanding dermcidin's mode of action at bacterial membranes, we used homonuclear and heteronuclear NMR to determine the conformation of the peptide in 50% trifluoroethanol solution. We found that dermcidin adopts a flexible amphipathic $\alpha$-helical structure with a helix-hinge-helix motif, which is a common molecular fold among antimicrobial peptides. Spin-down assays of dermcidin and several related peptides revealed that the affinity with which dermcidin binds to bacterial-mimetic membranes is primarily dependent on its amphipathic $\alpha$-helical structure and its length (>30 residues); its negative net charge and acidic pI have little effect on binding. These findings suggest that the mode of action of dermcidin is similar to that of other membrane-targeting antimicrobial peptides, though the details of its antimicrobial action remain to be determined.
Indole-3-acetic acid (IAA) production is a typical mechanism of plant growth promotion by some rhizobacteria. However, a functional genomic study is necessary to unravel the function and mechanism of IAA signaling during rhizobacteria-plant interactions. In this study, the expression of SlIAA1 and SlIAA9 among the auxin response genes in tomato was examined during the interaction between IAA-producing Acinetobacter guillouiae SW5 and tomato plants. When 3-day grown tomato seedlings were treated for 30 min with 10~100 µM of IAA produced by bacteria from tryptophan, the relative mRNA levels of SlIAA1 and SlIAA9 increased significantly compared with those of the control, demonstrating that IAA produced by this bacterium can induce the expressions of both genes. Inoculation of live A. guillouiae SW5 to tomato seedlings also increased the expressions of SlIAA1 and SlIAA9, with more mRNA produced at higher bacterial density. In contrast, treatment of tomato seedlings with dead A. guillouiae SW5 did not significantly affect the expression of SlIAA1and SlIAA9. When 3-day bacterial culture in tomato root exudates was administered to tomato seedlings, the relative mRNA level of SlIAA1 increased. This result indicated that the plant may take up IAA produced by bacteria in plant root exudates, which may increase the expression of the auxin response genes, with resulting promotion of plant growth.
The hydroxylation of methane (CH4) is crucial to the field of environmental microbiology, owing to the heat capacity of methane, which is much higher than that of carbon dioxide (CO2). Soluble methane monooxygenase (sMMO), a member of the bacterial multicomponent monooxygenase (BMM) superfamily, is essential for the hydroxylation of specific substrates, including hydroxylase (MMOH), regulatory component (MMOB), and reductase (MMOR). The diiron active site positioned in the MMOH α-subunit is reduced through the interaction of MMOR in the catalytic cycle. The electron transfer pathway, however, is not yet fully understood due to the absence of complex structures with reductases. A type II methanotroph, Methylosinus sporium 5, successfully expressed sMMO and hydroxylase, which were purified for the study of the mechanisms. Studies on the MMOH-MMOB interaction have demonstrated that Tyr76 and Trp78 induce hydrophobic interactions through π-π stacking. Structural analysis and sequencing of the ferredoxin domain in MMOR (MMOR-Fd) suggested that Tyr93 and Tyr95 could be key residues for electron transfer. Mutational studies of these residues have shown that the concentrations of flavin adenine dinucleotide (FAD) and iron ions are changed. The measurements of dissociation constants (Kds) between hydroxylase and mutated reductases confirmed that the binding affinities were not significantly changed, although the specific enzyme activities were significantly reduced by MMOR-Y93A. This result shows that Tyr93 could be a crucial residue for the electron transfer route at the interface between hydroxylase and reductase.
Polyethylene glycol(PEG) 처리에 의한 Agrobacterium tumefaciens spheroplast와 연초 엽육 protoplast의 상호작용을 연구하기 위하여, 효소적 방법으로 분리한 연초엽육 protoplast와 carbenicillin 및 lysozyme의 처리에 의해 제조된 Agrobacterium tumefaciens ATCC 15955 spheroplast를 섞어서 polyethylene glycol (PEG) 및 high pH-high $Ca^{2+}$ buffer를 처리한 후 시료를 취하여 전자현미경으로 관찰한 결과, spheroplast는 초기 단계에서 protoplast membrane에 부착하고, 시간이 경과함에 따라 endocytosis에 의해 protoplast의 세포질 내부로 도입된 다음, 점차로 그 형체(cell integrity)가 파괴되어지는 것을 관찰할 수 있었다. 이러한 관찰 결과로부터 spheroplast는 polyethylene glycol(PEG)에 의해 protoplast내부로 endocytosis되어짐을 알 수 있었다.
순천만 일대 칠면초 군락의 근권 토양에 존재하는 근권 세균의 다양성 분석을 위해 몇 개 지점을 선정한 후 샘플링을 실시하였다. 채취한 토양시료는 marine broth, tryptic soy broth 한천배지를 이용하면서 세균 집락 간 형태학적인 구분을 통해 순수분리 되었다. 분리된 세균의 genomic DNA를 획득한 후, 각각의 16S rDNA 염기서열을 증폭 분석하여 총 29 strain이 부분동정 되었다. 이들의 유연관계 확인을 위해 계통수를 작성한 결과, 이들은 각각 firmicutes문 (44.8%), gamma-proteobacteria group (27.6%), alpha-proteobacteria group (10.3%), bacteriodetes 문(10.3%), actinobacteria 문(6.8%)에 속하였다. 최우점하는 firmicutes 문에서는 Bacillus 속이, 차 우점하는 gamma-proteobacteria group에서는 각각 Marinobacterium, Halomonas, Vibrio 속이 집중 분포하는 것으로 나타났다. 또한 채취 지점별로 몇 가지 척도를 사용하여 다양성 지수를 도출하였을 때, 채취 지점 간 지수의 차이를 보여 전체 순천만 갯벌 중 위치에 따라 상이한 미생물상을 가지는 것으로 추측된다. 분리된 균들 중 일부는 갯벌 특유의 극한환경을 극복하면서 칠면초 근권에서 생존하며 물질순환, 식물생장 촉진 및 병원체 방어작용 유도 등 식물생장에 긍정적 역할을 수행할 것으로 생각된다.

Chitosan-based film-forming gel is regarded as a promising vehicle for topical delivery of antimicrobial agents to skin wounds, since it protects from microbial infection and the cationic polymer itself possesses antibacterial activity. In this study, possible synergistic interaction against common skin pathogens between the cationic polymer and tyrothricin (TRC), a cyclic polypeptide antibiotic, was investigated, by determining the concentration to inhibit 90% of bacterial isolates (MIC). The addition of the polysaccharide to TRC dramatically reduced the MIC values of TRC by 1/33 and 1/4 against both methicillin-resistant and methicillin-susceptible Staphylococcus aureus, respectively. The synergism of TRC and chitosan combination against both strains was demonstrated by the checkerboard method, with a fractional inhibitory concentration index below 0.5. Moreover, co-treatment of TRC and chitosan exhibited antibacterial activity against Pseudomonas aeruginosa, due to the antibacterial activity of chitosan, whereas TRC itself did not inhibit the gram-negative bacterial growth. These findings suggested that the use of chitosan-based film for topical delivery of TRC could be an alternative to improve TRC antimicrobial activity against strains that are abundant in skin wounds.
