Antimicrobial Persistence of Silver Diamine Fluoride and Silver Fluoride against Streptococcus mutans

  • Purpose: To evaluate the antimicrobial persistence of silver diamine fluoride (SDF) and silver fluoride (AgF) on Streptococcus mutans. Materials and Methods: An in vitro experiment was conducted to observe changes in the diameter of the inhibition zone of various materials, including AgF (Riva Star AquaTM step 1; SDI), potassium iodine (Riva star aquaTM step 2; SDI), Fluor protector® (FP, Ivoclar Vivadent), SDF (Riva starTM step 1; SDI), Ampicillin (Sigma-Aldrich), Amphotericin B (Nexstar) and negative control on S. mutans. Result: SDF, AgF and FP exhibited significant antimicrobial persistence over the 4 weeks period (P<0.05). At day 28, the diameter of inhibition zone was larger in SDF than in AgF. Conclusion: SDF and AgF have significant antibacterial durability against bacteria commonly associated with dental caries, with the antimicrobial effect lasting for at least 4 weeks. Further clinical studies are needed to validate these findings in vivo.

  • Bacillus subtilis의 생육 억제에 미치는 보존료의 병용효과 및 식염과의 상승효과를 조사하기 위하여 tryptone glucose-yeast extract 액체배지에 potassium sorbate와 sodiun bnzoate를 각각 최고허용농도의 1/2인 0.1%, 0.03%로 처리하고 이에 미생물의 내염성 기준도인 2% 식염을 첨가하여 진탕배양(4시간) 또는 정치배양 (4일)하면서 생육 상태를 조사한 결과, 두 보존료의 상승효과가 정치 및 진탕배양에서 모두 나타나지 않았으나 potassium sorbate와 식염의 병용처리구에서는 상승효과가 현저하게 나타났다.

Electrochemical, Antifungal, Antibacterial and DNA Cleavage Studies of Some Co(II), Ni(II), Cu(II) and Zn(II)-Copolymer Complexes

  • Cyclic voltammetric measurements were performed for Co(II), Ni(II), Cu(II) and Zn(II) complexes of 1 : 1 alternating copolymer, poly(3-nitrobenzylidene-1-naphthylamine-co-succinic anhydride) (L) and Ni(II) and Cu(II) complexes of 1 : 1 alternating copolymer, poly(3-nitrobenzylidene-1-naphthylamine-co-methacrylic acid) ($L^1$). The in vitro biological screening effects of the investigated compounds were tested against the fungal species including Aspergillus niger, Rhizopus stolonifer, Aspergillus flavus, Rhizoctonia bataicola and Candida albicans and bacterial species including Staphylococcus aureus, Escherichia coli, Klebsiella pneumaniae, Proteus vulgaris and Pseudomonas aeruginosa by well diffusion method. A comparative study of inhibition values of the copolymers and their complexes indicates that the complexes exhibit higher antimicrobial activity. Copper ions are proven to be essential for the growth-inhibitor effect. The extent of inhibition appeared to be strongly dependent on the initial cell density and on the growth medium. The nuclease activity of the above metal complexes were assessed by gel electrophoresis assay and the results show that the copper complexes can cleave pUC18 DNA effectively in presence of hydrogen peroxide compared to other metal complexes. The degradation experiments using Rhodamine B dye indicate that the hydroxyl radical species are involved in the DNA cleavage reactions.

Antimicrobial Activity of Methyl Gallate isolated from the Leaves of Glochidion superbum Against Hospital Isolates of Methicillin Resistant Staphylococcus aureus

  • An antimicrobial compound has been isolated from the leaves of Glochidion superbum. The compound was determined as methyl 3, 4, 5-trihydroxybenzoate (methyl gallate), based on ultraviolet (UV), infrared (IR), nuclear magnetic resonance (NMR) and mass spectroscopy (MS) analysis. The isolated compound exhibited potent antimicrobial activity against three clinical isolates of methicillin resistant Staphylococcus aureus (MRSA) by qualitative agar disc diffusion method and quantitative broth dilution method. Agar disc diffusion was done in a dose-dependent manner for each bacterial isolate at disc potencies of 25, 50, 100, and $150{\mu}g/disc$. The zones of inhibition were on average equal to 12.27, 14.20, 15.43, and 24.17 mm respectively. The inhibition zones were compared with that of vancomycin disc at $30{\mu}g$ as a reference standard. The MIC and MBC values were $50{\mu}g/ml$ and $100{\mu}g/ml$ respectively. The results of anti MRSA activity were analyzed using one-way ANOVA with Turkey's HSD and Duncan test. In conclusion, methyl gallate which was isolated from G. superbum showed the inhibition activity against methicillin resistant S. aureus.

苦蔘抽出物이 毛髮成長 促進 및 面疱 抑制에 미치는 영향 (Studies on the effect of Sophora flavescens extract on the hair growth stimulation and acne inhibition)

  • In the course of screening natural extracts for hair growth, we found that the extract of dried root of Sophora flavescens has the prominent hair growth promoting effect. After topical application of Sophora flavescens extract to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. In addition, the Sophora flavescens extract revealed to possess potent inhibitory effect on $5{\alpha}$-reductase Ⅰ and Ⅱ activity. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by Sophora flavescens extract treatment. RT-PCR analysis showed that Sophora flavescens extract induced mRNA levels of growth factors such as insulin-like growth factor-Ⅰ and keratinocyte growth factor in dermal papilla cells, suggesting hair growth promoting effect of Sophora flavescens extract is mediated through inhibition of $5{\alpha}$-reductase type Ⅱ activity and the regulation of growth factors in dermal papilla cells. Furthermore, Sophora flavescens extract also showed anti-bacterial effect on Propionibacterium acnes. These results suggest that Sophora flavescens can be used as a potent treatment agent for helping hair growth stimulation and acne inhibition.

Antimicrobial Effect of Polyphenon 60 against Streptococcus mutans and Streptococcus sobrinus

  • Polyphenon 60 refers to the mixture of catechins present in green tea. The aim of this study was to investigate the antimicrobial activities of polyphenon 60 against 4 strains of Streptococcus mutans and 2 strains of Streptococcus sorbrinus, which are the major causative bacteria of dental caries. The minimum bactericidal concentration (MBC) values of polyphenon 60 for S. mutans and S. sobrinus were determined and the effect of biofilm formation inhibition of that was evaluated. The MBC value of polyphenon 60 against the bacterial strains was 2.5 mg/ml except for one particular strain, S. mutans KCOM 1128 for which the value was 1.25 mg/ml. The results of biofilm formation inhibition assay revealed that polyphenon 60 inhibited biofilm formation more than 90% at a concentration of 2.5 mg/ml. It was apparent that polyphenon exhibited biofilm formation inhibition activity along with bactericidal effect against S. mutans and S. sobrinus. Therefore, it is proposed that polyphenon 60 as one of the components of bactericidal agents could be useful in developing oral hygiene products, toothpaste or gargling solution.

Antifungal Activities of Copper(II) with Biosensitive Macrocyclic Schiff Base Ligands Derived from 4-Aminoantipyrine Derivatives

  • Novel copper(II) complexes have been synthesized from the macrocyclic Schiff bases derived from Knoevenagel condensed ${\beta}$-ketoanilides (obtained by the condensation of acetoacetanilide and substituted benzaldehydes), 4-aminoantipyrine and ophenylene diamine. The structural features have been determined from their analytical and spectral data. All the Cu(II) complexes exhibit square planar geometry. Their high molar conductance values support their 1 : 2 electrolytic nature. The magnetic moment data provide evidence for the monomeric nature of the complexes. The X-band ESR spectra of the |$CuL^1$|$(OAc)_2$ in DMSO solution at 300 and 77 K were recorded and their salient features are reported. Structural Studies of Porcine Myeloid Antibacterial Peptide, PMAP-23 in DPC micelles by NMR Spectroscopy

  • Leukocytes are important elements in the host defense against microbial infections. A variety of antimicrobial peptides named as the cathelicidin family have been identified from leukocytes. PMAP-23 derived from porcine myeloid cells is an antimicrobial peptide belong to the cathelicidin family. PMAP-23 was reported to have potent growth inhibition activity against bacterial and tumor cells with no hemolytic activity.(omitted)

  • By coculturing E. coli HGU-3 with Bifidobacterium KH-2 or Streptococcus faecalis HGO-7 with Bifidobacterium KH-2, the productivity of $\beta$-glucuronidase and $\beta$-glucosidase was inhibited. When lactulose, growth factor of lactic acid bacteria, was added into this medium, the productivity of these enzymes and pH of the medium were dramatically decreased. When intestinal microflora of human and rat were inoculated in the medium containing lactulose, the enyzme productivity and pH of the medium were dramatically decreased. By s.c. injecting DMH into mice, $\beta$-glucuronidase of intestinal bacteria was induced, but the production of the enzymes was inhibited by adminstering lactulose.
