Isolation and characterization of Bacillus amyloliquefaciens TK3 inhibiting causative bacteria of atrophic rhinitis and fowl typhoid (돼지위축성비염과 가금티푸스 병원균을 저해하는 Bacillus amyloliquefaciens TK3의 분리 및 특성 조사)

  • For prevention of atrophic rhinitis of swine by Bordetella bronchiseptica and fowl typhoid by Salmonella gallinarum, bacterial strains showing antimicrobial activity against those pathogenic bacteria were isolated from various samples collected at animal farms. Among 372 bacterial isolates strain TK3 showed the highest antibacterial activity against both pathogens, and was identified as Bacillus amyloliquefaciens by 16S rRNA gene sequence analysis. B. amyloliquefaciens TK3 could inhibit growth of both pathogens by secretion of antibacterial compounds such as siderophore, rhamnolipid and antimicrobial peptide. Production radius of siderophore on Chrome azurol S agar plate by strain TK3 was 0.53 cm after 14 days of incubation, and concentration of siderophore in King's B medium was 1.06 mmol/ml. It also secreted 82.4 mg/L of rhamnolipid, and antimicrobial peptide that completely inhibited growth of both pathogens at concentration of $30{\mu}l/ml$ in LB medium.

  • The functional properties of novel sugar derivative sweeteners, fructo-oligosaccharide, maltitol, sorbitol and high maltose syrup(HMS) were examined for their humactant effect, lactic acid bacterial growth, Streptococcus mutants growth and relative sweetness compared to sucrose. Sorbitol exhibited remarkably high water activity reducing capacity, whereas fructo-oligosaccharide and maltitol showed the same level as sucrose. Maltitol showed distinct anti-bacterial(bacteriocidic) effect against Stc. mutants and most of lactic acid bacteria tested except for L. plantarum. The molar basis relative sweetness of sugar derivatives in comparison with 1%(w/w) level of sucrose were 0.69 for Neosugar(fructo-oligosaccharide), 0.21 for sorbitol, 0.50 for maltitol and 0.27 for HMS.

The Skin-Related Biological Activities of Aerially Extract of Oenothera lamarckiana (큰달맞이꽃 지상부 추출물의 피부 관련 생리활성 효과)

  • Skin plays important roles in protecting the internal organs from the chemical-biological risk factors and ultraviolet light. Exposure to the chemical and biological stimuli has a detrimental effect on skin's structure and physiological regulation. Therefore, much attention has been paid to natural products that show biological activities such as anti-oxidation, anti-aging and anti-bacterial activities. In this study, we investigated the skin-related biological activities of Oenothera lamarckiana aerial part extract. The extract contained 229.35 mg TAE (tannic acid equivalents)/g total polyphenolic compounds and the extract showed relative high antioxidant activity ($SC_{50}$ value: $8.52{\mu}g/mL$). The $IC_{50}$ value against tyrosinase and elastase were 307.94 and $181.51{\mu}g/mL$, respectively. This suggested that O. lamarckiana can be applied to whiten skin and slow the aging of skin. O. lamarckiana extract showed a growth inhibitory effect on Staphylococcu epidermidis (minimum inhibitory concentration: $250{\mu}g/mL$). Interestingly, O. lamarckiana extract showed no inhibitory effect on Pseudomonas aeruginosa on the paper disc assay. Yet the extract inhibited the growth of Pseudomonas aeruginosa on the broth dilution assay in a dose-dependent manners. Taken together, O. lamarckiana could have good potential for development as an additive in the cosmetic industry.

Biotransformation of Intestinal Bacterial Metabolites of Ginseng Saponin to Biologically Active Fatty-acid Conjugates

  • Ginsenosides are metabolized (deglycosylated) by intestinal bacteria to active forms after oral administration. 20(S)-Protopanaxadiol $20-O-{\beta}-D-glucopyranoside$ (M1) and 20(S)-protopanaxatriol (M4) are the main intestinal bacterial metabolites (IBMs) of protopanaxadiol- and protopanaxatriol-type glycosides. M1 was selectively accumulated into the liver soon after its intravenous (i.v.) administration to mice, and mostly excreted as bile; however, some M1 was transformed to fatty acid ester (EMl) in the liver. EM1 was isolated from rats in a recovery dose of approximately $24mol\%.$ Structural analysis indicated that EM1 comprised a family of fatty acid mono-esters of M1. Because EM1 was not excreted as bile as Ml was, it was accumulated in the liver longer than M1. The in vitro cytotoxicity of M1 was attenuated by fatty acid esterification, implying that esterification is a detoxification reaction. However, esterified M1 (EM1) inhibited the growth of B16 melanoma more than Ml in vivo. The in vivo antitumor activity paralleled with the pharmacokinetic behavior. In the case of M4, orally administered M4 was absorbed from the small intestine into the mesenteric lymphatics followed by the rapid esterification of M4 with fatty acids and its spreading to other organs in the body and excretion as bile. The administration of M4 prior to tumor injection abrogated the enhanced lung metastasis in the mice pretreated with 2-chloroadenosine more effectively than in those pretreated with anti-asialo GMl. Both EM1 and EM4 did not directly affect tumor growth in vitro, whereas EM1 promoted tumor cell lysis by lymphocytes, particularly non-adherent splenocytes, and EM4 stimulated splenic NK cells to become cytotoxic to tumor cells. Thus, the esterification of IBM with fatty acids potentiated the antitumor activity of parental IBM through delay of the clearance and through immunostimulation. These results suggest that the fatty acid conjugates of IBMs may be the real active principles of ginsenosides in the body.

Complete genome sequencing of Pseudomonas parafulva PpaJBCS1880, a biocontrol and plant growth promoting agent (식물 병 방제 및 생육촉진 효과를 나타내는 Pseudomonas parafulva PpaJBCS1880균주의 유전체 염기서열)

  • Pseudomonas parafulva PpaJBCS1880 (PpaJBCS1880) isolated from rice seeds showed strong antagonistic activity against bacterial plant pathogens by producing lipopeptide. Furthermore, the strain controlled the incidence of bacterial pustule in soybean plants and promoted the growth of rice plants. Here we present complete genome sequence of PpaJBCS1880. The genome comprises of 5,208,480 bp with GC content of 63.4%, which includes 4,487 predicted protein-coding genes, 19 rRNAs, and 74 tRNAs. Genome analysis revealed genes encoding antimicrobial secondary metabolites such as lipopeptide, pyoverdine, phenazine, and hydrogen cyanide, which are known to play essential roles in biocontrol of plant diseases.

Antibacterial Effect of Chitosan-Modified Fe3O4 Nanozymes on Acinetobacter baumannii

  • The aim of this study was to determine whether the antibacterial activity of chitosan-modified Fe3O4 (CS@Fe3O4) nanomaterials against Acinetobacter baumannii (A. baumannii) is mediated through changes in biofilm formation and reactive oxygen species (ROS) production. For this purpose, the broth dilution method was used to examine the effect of CS@Fe3O4 nanoparticles on bacterial growth. The effects of CS@Fe3O4 nanoparticles on biofilm formation were measured using a semi-quantitative crystal violet staining assay. In addition, a bacterial ROS detection kit was used to detect the production of ROS in bacteria. The results showed that CS@Fe3O4 nanoparticles had a significant inhibitory effect on the colony growth and biofilm formation of drug-resistant A. baumannii (p < 0.05). The ROS stress assay revealed significantly higher ROS levels in A. baumannii subjected to CS@Fe3O4 nanoparticle treatment than the control group (p < 0.05). Thus, we demonstrated for the first time that CS@Fe3O4 nanoparticles had an inhibitory effect on A. baumannii in vitro, and that the antibacterial effect of CS@Fe3O4 nanoparticles on drug-resistant A. baumannii was more significant than on drug-sensitive bacteria. Our findings suggest that the antibacterial mechanism of CS@Fe3O4 nanoparticles is mediated through inhibition of biofilm formation in drug-resistant bacteria, as well as stimulation of A. baumannii to produce ROS. In summary, our data indicate that CS@Fe3O4 nanoparticles could be used to treat infections caused by drug-resistant A. baumannii.

Studies on the Anti-bacterial, Anti-inflammatory and Anti-oxidant Effect of BPH (비피(鼻皮) 증류액의 항균, 소염, 항산화 작용에 대한 연구)

  • Objectives : The purpose of the study is to test the antibacterial, anti-inflammatory and antioxidant effects of BPH, which is composed of Pini Densiflorae Nodi Lignum and Querci Acutissimae Fructus, Angelicae Gigantis Radix, Cnidii Rhizoma, Angelicae Dahuricae Radix, Angelicae Tenuissimae Radix. Method : Antibacterial and anti-inflammatory effects of BPH on Propionibacterium acnes, one of anaerobic bacteria species were evaluated by measuring the levels of 2,2-diphenyl-1-picrylhydrazyl (DPPH) elimination and lipid peroxidation. Result : When BPH was applied to CCD-986sk (Human normal fibroblast) to confirm the level of cytokine(tumor necrosis factor-alpha, interleukin-8), its level increased in proportion to that of BPH's concentration, which indicated dose-dependent relationship. Using the Disk diffusion to measure the bacterial growth inhibition zone varying BPH concentration, it was found that the antibacterial effect of BPH was less than that of erythromycin, the control group, but was higher than that of saline, and it increased with higher concentrations. In a liquid culture medium containing BPH, the growth rate of Propionibacterium acnes was decreased by more than 10% at 25% BPH. After adding P. acnes to THP-1 monocyte, and treated it with BPH, and measuring the concentration of TNF-a and IL-8, it was observed that the amount of TNF-alpha and IL-8 significantly decreased depending on the level of BPH concentration. The ability to eliminate DPPH increased with higher BPH concentration. The inhibition of lipid peroxidation was increased by BHT treatment in a dose-dependent manner. Conclusion : Using Propionibacterium acnes, an anaerobic bacteria, we confirmed that BPH has antibacterial, anti-inflammatory and antioxidant effects.

Identification, Characterization, and Efficacy Evaluation of Bacillus velezensis for Shot-Hole Disease Biocontrol in Flowering Cherry

  • Though information exists regarding the pathogenesis of the shot-hole disease (SH) in flowering cherry (FC), there has been a lack of research focusing on SH management. Therefore, here, we investigated the inhibitory activities of antagonistic bacteria against SH pathogens both in vitro and in vivo as well as their biochemical characteristics and bioactive compounds. Two biosurfactant-producing bacterial antagonists, identified as Bacillus velezensis strains JCK-1618 and JCK-1696, exhibited the best effects against the growth of both bacterial and fungal SH pathogens in vitro through their cell-free culture filtrates (CFCFs). These two strains also strongly inhibited the growth of the pathogens via the action of their antimicrobial diffusible compounds and antimicrobial volatile organic compounds (VOCs). Crude enzymes, solvent extracts, and biosurfactants of the two strains exhibited antimicrobial activities. Liquid chromatography/electrospray ionization time-of-flight mass spectrometric analysis of the partially purified active fractions revealed that the two antagonists produced three cyclic lipopeptides, including iturin A, fengycin A, and surfactin, and a polyketide, oxydifficidin. In a detached leaf assay, pre-treatment and co-treatment of FC leaves with the CFCFs led to a large reduction in the severity of the leaf spots caused by Epicoccum tobaicum and Bukholderia contaminans, respectively. In addition, the two antagonists produced indole-3-acetic acid, siderophore, and a series of hydrolytic enzymes, along with the formation of a substantial biofilm. To our knowledge, this is the first report of the antimicrobial activities of the diffusible compounds and VOCs of B. velezensis against the SH pathogens and their efficiency in the biocontrol of SH.

The extracts enhanced the growth of Bifidobncterilim breve and B. longum in Media with or without carbon sources, suggesting the bifid factor (5) might be involved in the phenomenon. This effect was most pronounced with water extract of p. ginseng, the growth of 27 bifidobacteria strains belonging to B. ndolexcentium, H. longlrm, and 1. breve and B. iniuntis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and 5.fcherirhia coli had little or no ability to utilizes it (or growth. Methanol extracts of p, ginseng were found to selectively inhibit growth of various clostridia including C. perfringens and C. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake(600 mg/day for two weeks) on the fecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of fecal microflora including Bri'idobucterilim app. during the period of ginseng extract intake was significantly unaffected from the proceeding and sub sequent control periods. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The fecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobacterium spp. Other biochemical properties in faces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the pharmacological actions of P ginseng as an adaptogen.
