The Effect of Nutrient Amendments on Biodegradability of Kerosene and Growth of Kerosene-degrading Microorganisms (영양원 변화가 Kerosene 분해율 및 분해균주 성장에 미치는 영향)

  • Bioremediation is the technology to harness nature's biodegradative capabilities to remove or detoxify pollutions that threaten public health as environmental contaminants. Composting may become one of major bioremediation technologies for treating soils contaminated with petroleum if the fate of contaminants during composting is better understood Most composting research of petroleum was primarily focused on removing contaminant by optimizing composting conditions. Accordingly, laboratory feasibility studies may be useful to establish a realistic basis in co-composting complex substrate such as petroleum hydrocarbons. The purpose of this study was to assess the optimal conditions of kerosene biodegradation following supplementation with nutrient amendments under simulated composting conditions. Although it increased the growth of bacterial consortium, addition of co-substrates 0.5%(w/v) such as acetic acid, citric acid, glucose, and malic acid was not beneficial. Combination of nitrogen and phosphorous source enhanced kerosene biodegradation and reduced VOC evolution. These results showed that kerosene was able to utilize in bioremediation technology.

  • Persicaria hydropiper juice was applied to develop a novel meidum for the mass cultivation of an antifungal bacterium, Bacillus lentimorbus G-74. In this research, a optimized culture condition of B. lentimorbus G-74 were detenninecl. Persicaria hyclropiper juice was contained 1.76% nitrogen, 0.15% phosphorus, 18,230 ppm potassium, 13,753 ppm calcium and 4,844 ppm magnesium. The optimal growth was obtained when the juice was a 30%(v/v) level. The optimum temperature and pH for the bacterial growth were $35^{\circ}C$ and 5.0, respectively.

  • Treated sewage could enable growth by providing key nutrients or seeding the sediments with enterococci strains that can grow in the environment. This study is to test the hypothesis that the flux of bacteria into the water column is rate-limited by the transfer of bacteria across the sediment/water interface. Two conceptual models are derived for the transfer of bacteria to the water column from the sediment/water interface: convective diffusion of isolated bacteria and resuspension of particle-associated bacteria. The model predictions are directly tested together with field measurements of bacteria and sediment in an effluent-dominated stream where high concentrations of enterococci in this stream originate primarily from growth of the bacteria in stream sediments. The results reveal that high concentrations of enterococci in this stream are transported primarily by resuspension of particle-associated bacteria accumulated at the sediment/water interface, either in the form of bacterial aggregates or in the form of inorganic particles.

  • The hairy root cultures of Centella asiatica were established by infection leaf explants with Agrobacterium rhizogenes A4, 15834 in 1/2 Murashing and skoog liquid medium supplemented with 50 $\mu$M acetosyringone. The induced hairy roots were subjected to paper electrophoresis for the detection of opine and opine-positive clones which were considered to have been transformed. Five hairy root clones were selected according to the different bacterial strains used, growth rate and pattern. Among media tested, MS basal medium substituted phosphate concentration by 2.5mM K2HPO4 showed the highest growth rate in the dark condition.

  • Cadimium ion-resistant microorganism was isolated from the sludge of wastewater. The physiological, morphological and other cultural data showed that this strain belonged to Citrobacter freudii. A clearcut distinction of growth among nutrient broth, typtic soy broth and synthetic medium was demonstrated. The resistant cells showed only slight mutagenic action. During the growth of bacterial population in resting state, the organisms reduced the initial level of resistance to cadmium ions when they were not kept in contact with cadmium ions in bacteral multiplication. And cadmium ion-resistant and cadmium ion-sensitive strain were found to show equal, lower or higher sensitivity to other heave metals.

Rhizobacterial Exopolysaccharides Elicit Induced Resistance on Cucumber

  • The role of exopolysaccharides (EPSs) from a plant growth-promoting rhizobacterium, Burkholderia gladioli IN26, on elicitation of induced systemic resistance was investigated. A purified EPS induced expression of PR-1a::GUS on tobacco and elicited induced resistance against Colletotrichum orbiculare on cucumber. The maximum level of disease protection was noted when seeds were soaked in 200 ppm of the EPS. Our results indicate that EPS from specific rhizobacteria can elicit induced resistance and suggest that bacterial EPS might be a useful elicitor of resistance under field conditions.

  • A strain of Enterobacter cloacae isolated from soil showed the evidence of growth in medium containing 1,500 ppm of Cd$\^$++/ though its growth was inhibited at high cadmium concentrations. This strain accumulated 59% of cadmium into the cells during incubation in medium containing 0.5 ppm of cadmium and its uptake was nearly proportional to the dry weight of cells, the average being 7.8 mg cadmium per g dry cells. It was also found that 60-70% of cadmium accumulated in cells were distributed in the cell wall fraction.

  • Henri-Michaelis-Menten model for predicting the behavior of culture of Brevibacterium flavum under carbon limiting condition has been evaluated on a chemostat growing in the presence of the phenol derivatives: guaiacol, o-vanillin and vanillin. It is found that Henri-Michaelis-Menten model could be applicable to the evaluation of the growth rate of cells in the phenol derivatives. The marked enhancing abilities of the cells treated by the phenol derivatives during fermentation were ascribed to the formation of charge-transfer complex between the phenol-derivatives and oxygen which supplies oxygen effectively to the fermentation system.

  • Takju is a traditional alcoholic beverage that has been prepared by fermenting the cooked rice and Nuruk (Korean-style bran koji). During fermentation. bacterial contamination is a problem which inhibits the growth of yeast and thus lowers the ethanol production from starch of rice, and causes souring. Major contaminants were known to be gram-positive acid producers at the early stage of fermentation. This problem would be solved if the contaminated bacteria could be controlled. Nisin, a GRAS-grade preservative, was added at the level of 500 iu/g as it retards the growth of the gram-positive bacteria. It was possible to control acid and ethanol production during fermentation. This process increased the ethanol production by 2 % comparing with control.

  • The use of aortic valve homograft has been developed since 1962 when Ross and Barratt - Boyes independently replaced a diseased aortic valve with an orthotopically inserted homograft valve. And also surgical treatment of complex congenital cardiac malformations utilizing homograft extracardiac conduit has been tried with better result than any other prosthetic material. The present study was undertaken to clarify the safety tissue viability, sterility, after following our protocol of procurement of heart, dissection of aortic and pulmonic homograft, sterilization, cryopreservation, thawing and dilution, and transplantation on experimental animal, sheep. Tissue viability of valve and great artery was assessed by tissue culture. Sterility was evaluated by bacterial and fungal culture. The method used was proven no deleterious effect on the integrity of the valve. Tissue culture of valve tissue before, and after cryopreservation process resulted that active fibroblast growth was observed from homograft sterilized with antibiotics. And culture of the transplanted homograft from sacrificed animal showed active fibroblast growth. Pathologic examination of implanted valve tissue from sacrificed sheep showed mild calcification and minor change, but there were moderate and severe calcification of wall of great arteries.
